Nanotechnology is widely considered a major technology of the twenty-first century. Nanoparticles (NPs) has been shown to pass through reproductively significant biological barriers such as the blood-testicle and placental barriers. Thus, the purpose of this study was to determine the effect of silver Nanoparticles (Ag-NPs) on sperm-egg interaction and spermatozoa quality parameters in quail spermatozoa. Semen was suspended in Ringer solution containing Ag-NPs levels at 5.5 × 106 sperm/ml (0, 0.01, 0.1, 1 and 10 ppm). The results indicated that when sperm were counted at 0.1 ppm, the number of holes formed on the inner perivitelline layer was significantly increased compared to the control. The 10 ppm group had a significant reduction in sperm viability. At 0.1 and 1 ppm, the membrane integrity was significantly decreased (P < 0.05). All treatments (except 0.01 ppm Ag-NPs) had a significant (P < 0.05) effect on the percentage of spermatozoa with an intact acrosome when compared to the control group. At 0.1, 1, and 10 ppm Ag-NPs, morphological defects in the acrosome were observed. As a result, Ag-NPs is likely capable of destroying the acrosome membrane. This research indicates that Ag-NPs may be cytotoxic to spermatozoa by impairing sperm functionality and increasing sperm mortality.
The effects of antimetabolite 6-aminonicotinamide on levels of soluble proteins, free amino acids and protease activity in various tissues of quail have been in vestigated. The levels of soluble proteins m liver, heart and pectoral muscle were markedly lowered and the specific activity of protease in kidney and pectoral muscle was markedly increased. The concentrations of aspartic acid / asparagine, alanine, valine, methionine, isoleucine, leucine and lysine in the liver were markedly increased. En the kidney the concentrations of aspartic acid I asparagine, arginine, threonine, alanine, proline and lysine were markedly increased but those of glutamic acid I glutamine were decreased. The concentrations of glutamic acid / glutamine and serine in the heart were reduced but those in glycine and methionine were increased. In the pectoral muscle the concentration of arginine was decreased but the concentration of alanine and threonine was increased. The overall results suggest that antimetabolite 6-aminonicotinamide may act to enhance concentrations of amino acids related to the generation of energy and to depress the biosynthesis of some specific amino acids.

The inhibitory effect of epigallocatechin gallate (EGCG), one of the antioxidants in green tea (Camellia sinensis), against Salmonella Enteritidis was evaluated in commercial braised quail eggs at two temperatures (4 and $25^{\circ}C$). Although S. Enteritidis was dose-dependently suppressed by EGCG in pure culture at $25^{\circ}C$, it was not inhibited in the sauce or eggs at this temperature. At low temperature ($4^{\circ}C$), S. Enteritidis was inhibited in both the sauce and eggs by $400{\mu}g/mL$ EGCG. Thus, EGCG at an appropriate concentration could be a useful food additive for inhibiting S. Enteritidis in braised quail eggs at low temperatures.
Objective: Transgenic technology is widely used for industrial applications and basic research. Systems that allow for genetic modification play a crucial role in biotechnology for a number of purposes, including the functional analysis of specific genes and the production of exogenous proteins. In this study, we examined and verified the cumate-inducible transgene expression system in chicken DF1 and quail QM7 cells, as well as loxP element-mediated transgene recombination using Cre recombinase in DF1 cells. Methods: After stable transfer of the transgene with piggyBac transposon and transposase, transgene expression was induced by an appropriate concentration of cumate. Additionally, we showed that the transgene can be replaced with additional transgenes by co-transfection with the Cre recombinase expression vector. Results: In the cumate-GFP DF1 and QM7 cells, green fluorescent protein (GFP) expression was repressed in the off state in the absence of cumate, and the GFP transgene expression was successfully induced in the presence of cumate. In the cumate-MyoD DF1 cells, MyoD transgene expression was induced by cumate, and the genes controlled by MyoD were upregulated according to the number of days in culture. Additionally, for the translocation experiments, a stable enhanced green fluorescent protein (eGFP)-expressing DF1 cell line transfected with the loxP66-eGFP-loxP71 vector was established, and DsRed-positive and eGFP-negative cells were observed after 14 days of co-transfection with the DsRed transgene and Cre recombinase indicating that the eGFP transgene was excised, and the DsRed transgene was replaced by Cre recombination. Conclusion: Transgene induction or replacement cassette systems in avian cells can be applied in functional genomics studies of specific genes and adapted further for efficient generation of transgenic poultry to modulate target gene expression.
Iron has a crucial role in growth as part of metalo-proteins like haemoglobin or myoglobin, enzymes; they are also involved in energetic reactions. Iron plays a vital role in fertility. At high doses, Iron has a harmful consequence on the reproductive system, which can be strongly reflected the final stage of spermatogenesis. Nutritional products are claiming to use nanotechnology and it is important to recognize the potential toxicity of nano-sized nutrients. Recently iron nanoparticles were proposed as a food additive for poultry. The objective of this study was to investigate the effects of L-cystein coated iron oxide nanoparticles on reproductive performance in male quails. The results of Fourier Transform Infrared Spectrometer, Alternating Gradient Force Magnetometer and Scaning Electron Microscopy showed that iron oxide nanoparticles was produced and have been coated with L-cycstein (Fe3O4-Cys NPs). A total of 100 one-week-old quail chicks were randomly placed to five groups of five replicates. Four quails (two male and two females) were raised in an individual cage for each replicate. The five experimental treatment diets consisted; negative control diet, with no Iron supplementation; positive control diet supplemented with 60 mg/kg of Fe3O4; treatment diets supplemented with 0.6, 6 and 60 mg/kg of L-cystein coated iron oxide nanoparticles. The hemoglobin, Red blood cell, mean corpuscular volume, mean corpuscular hemoglobin, mean corpuscular hemoglobin concentration, gonadal somatic index, daily sperm production, total testicular sperm and sperm viability of the male quails that were fed with diet supplemented by 0.6 mg/kg of Fe3O4-Cys NPs were improved as compare with negative control. This study showed that not only the use of the Fe3O4-Cys nanoparticles had no side effects but also it can be used as a feed additive to improve the reproductive performance in male quails.
DNA methyltransferases (DNMTs) are closely associated with the epigenetic change and the gene silencing through the regulation of methylation status in animal genome. But, the role of DNMTs in transgene silencing has remained unclear. So, we examined whether the knockdown of DNMT influences the reactivation of transgene expression in the transgenic quails. In this study, we investigated the expression of DNMT3a, and DNMT3b in blastoderm, quail embryonic fibroblasts (QEFs) and limited embryonic tissues such as gonad, kidney, heart and liver of E6 transgenic quails (TQ2) by RT-PCR. We further analyzed the expression of DNMT3a at different stages of whole embryos during early embryonic development by qRT-PCR. DNMT3a expression was detected in all test samples; however, it showed the highest expression in E6 whole embryo. Embryonic fibroblasts collected from TQ2 quails were treated with two DNMT3a-targeted siRNAs (siDNMT3a-51 and siDNMT3a-88) for RNA interference assay, and changes in expression were then analyzed by qRT-PCR. The siDNMT3a-51 and siDNMT3a-88 reduced 53.34% and 64.64% of DNMT3a expression in TQ2 QEFs, respectively. Subsequently the treatment of each siRNA reactivated enhanced green fluorescent protein (EGFP) expression in TQ2 (224% and 114%). Our results might provide a clue for understanding the DNA methylation mechanism responsible for transgenic animal production and stable transgene expression.
The weekly liveweight gain, growing and stress parameters of quails bred using two different types of lighting for 6 weeks following hatching were examined in this study. The first type of lighting was 23 L:1 D, continuous lighting (CL), widely used in the commercial system and the second was a self-photoperiod (SP) system consisting of a continuously lit chamber and a dark chamber the quails could move to as they wished. On the first 15 days, no difference was found in liveweight gain between the two breeding systems. On the $6^{th}$ week when the trial was completed, the liveweight of the male quails upon which CL lighting was used was 159.03 g while the weight of males in the SP group was 174.43 g; these values in female quails of the CL group were 179.15 g and in the SP group were 200.68 g. The CL group had lower testis volume (TVOM, $cm^{3}$) and testis weight (TW, g) than the SP group, however there was no difference between the groups in testis weight/body weight rate (BWTW %). In female quails, the ovary weight (OW, g) and the ovary weight/body weight rate (BWOW, %) values were higher in the SP group. The CL light regime was concluded to cause stress in male quails (CL, Heterophil/Lymphocyte ratio (H/L): 0.27; SP, H/L: 0.17). In conclusion; the SP system allowing the quails to regulate their light periods increased liveweight gain and enabled sexual maturity to be gained at an earlier period than in quail on the CL system and improved their welfare.
This study was carried out to obtain the menu preferences of frequently served set menu of the staff working at Samsung Medical Center. The frequently served set menus were 72 menus including 23 Korean meals, 12 bowel of rices, 18 noodles, 10 soups, 6 fried rices, 3 western meals. The subjects of this study were 624 staffs who works at Samsung Medical Center. The results are summarized as followers : The subject group for this study was composed of 23% males and 58% females, 80% of subjects are twenties and thirties. The best preferred menu was fried food 4(boiled rice with assorted mixtures, boiled quail egg). And noodles 8(cold buckwheat noodles, fried mung-beans), noodles 2(buckwheat noodles, fried vegetable), Western meals 3(hamburg steak, steamed dumpling), noodles 18(rice-cake dumping soup, fried sweet potato with sugar syrup) were preferred by the subjects. The worst preferred menu was soups 4(loach soup, fried sea food and welsh onion). In the preference each kinds of set menus, Korean meals 3(soup to relive the hangover with pollack, grilled meat, lettuce, fruit salad, Kimchi, ice tea) was the most preferred of the Koreans meals, bowel of rice 8(omelet containing fried rice, chicken salad) of the bowel of rices, noodles 8(cold buckwheat noodles, fried mung-beans) of the noodles, soups 3(spicy beef soup, chicken salad) of the soups, fried rice 5(boiled rice with assorted mixtures, boiled quail egg). There was no significant sexual difference in the menu preferences except 7 menus. There was significant age difference in the 17 menu preference, the younger age subjects were the better like noodles.

In this study, we determined major foods and food groups which contributed to the intake of folate in korean child-bearing women. Folate content of the major foods was also analyzed. A total of 91 healthy women with childbearing potential participated. They were divided into one of three groups by age; A (15∼24 yrs), B(25∼34 yrs) and C (35∼49 yrs). Food consumption data of the subjects were obtained by the 24-hr recall method. The rate of contribution of each food to the total intake of dietary folate was determined. Seventy major foods for folate were selected from the 178 foods they consumed. Two sets of the 70 foods were collected from different markets. After extracting folates from the foods by heating with a HEPES-CHES buffer, the samples were treated with α-amylase, protease and folate intakes was Korean cabbage kimchi followed by laver, soybean sprout, rice, yolmu kimchi, eggs, lettuce, perilla leaves, sea mustard and radish root. Food items that contributed to folate intake were slightly different among the age groups. Although the folate content of Korean cabbage kimchi was 51.6 ㎍/100g, but due to the high amount of intake, it was ranked the first major food prviding folate. High folate foods containing over 100㎍/100g were laver, corn flakes, spinach, mungbean, sesame, quail's eggs, small radish kimchi, kidney beans, leeks and peanuts. However, based on the serving size, spinach, corn flakes, leeks, sweet potato, pepper leaves, quail's eggs, crown daisy, small radish kimchi and perilla leaves contributed above 50㎍ of folate per serving size.
High fibre (CF, 242 g/kg) sunflower seed meal (SSM) was incorporated in standard grower and layer quail diets at 0, 50, 75 or 100 g/kg replacing part of the soybean meal (SBM) and deoiled rice bran (DORB). Each grower diet was offered to 30 quails housed in battery cages from day one to 5 weeks of age. At the end of the growing period, Twelve female quails from each of the four respective dietary groups were transferred to the individual laying cages and their laying performance from 7 to 20 weeks of age were evaluated on diets containing SSM at similar levels as that during the growing period. The results of growth bioassay revealed that the live weight gain, feed intake, nutrient retention and carcass characteristics of quails in different dietary groups did not vary significantly (p>0.05), although the quails fed SSM based diets utilized the feed less (p<0.05) efficiently than those fed the control diet. During the laying period, the egg production and egg quality traits remained unaffected due to dietary treatments but feed required per unit egg production increased (p<0.05) as the level of SSM in diet was increased to 100 g/kg. Although, the cost of feed per kg decreased due to the incorporation of SSM in diet but the same was not reflected in feed cost per unit gain, meat or egg production as such parameters were comparable on all the diets. It could thus be concluded that the SSM can be incorporated in quail diets upto 100 g/kg for part of SBM, an item usually expensive and also in short supply in developing countries including India.
