The following study looked at the effects of Hyangsayukgunjatang and Hyangsayukgunjatang-ga-kyenegum, moryobun on gastric ulcers in rats. In the 1st experiment, ulcers were induced by method of shay's pylorus-ligated in rats. Subjects were administered 2 kinds of medicine, Hyangsayukgunjatang and Hyangsayukgunjatang-ga-kyenegum, moryobun, which were taken orally. The quantity of gastric juice secretion, total acidity of gastric juice, HCI secretion and serumgastrin content were measured and tissues of gastric mucosa epithelium were observed. 1. The quantity of gastric juice secretion in Hyangsayukgunjatang-treated and Hyangsayukgunjatang-ga-kyenegum, moryobun-treated groups showed significant decrease incomparision with the control group. 2. In relation to total acidity of gastric juice, only Hyangsayukgunjatang -treated groupshowed significant decrease in comparison with the control group. 3. HCI secretion of Hyangsayukgunjatang-treated and Hyangsayukgunjatang-ga-kyenegum, moryobun-treated groups showed significant decrease in comparision with the control group. 4. Content of serum gastrin, which were present in Hyangsayukgunjatang -treated and Hyangsayukgunjatang-ga-kyenegum, moryobun- treated groups showed significant decrease in comparision with the control group. 5. The degree of ulcer occurrence & parietal cell numbers and chief cell numbers of Hyangsayukgunjatang-treated and Hyangsayukgunjatang-ga-kyenegum, moryobun-treatedgroups, which were observed by macrographic & histologic method showed signicant decreasein comparison with the control group. Hyangsayukguniatang-treated group prooved to be increasingly effective. From above the experiment results it can be concluded that Hyangsayukgunjatang will be more clinical effective on curing gastric ulcers induced by method of Shay's pylorus-ligated rather than that of Hyangsayukgunjatang-ga-kyenegum, moryobun.

In this study, we investigated the inhibitory activities on gastritis and gastric ulcer using liriodendrin which is a constituent isolated from Kalopanax pictus. To elucidate its abilities to prevent gastric injury, we measured the quantity of prostaglandin $E_2$ ($PGE_2$) as the protective factor, and we assessed inhibition of activities related to excessive gastric acid be notorious for aggressive factor and inhibition of Helicobacter pylori (H. pylori) colonization known as a cause of chronic gastritis, gastric ulcer, and gastric cancer. Liriodendrin exhibited higher $PGE_2$ level than rebamipide used as a positive control group at the dose of $500{\mu}M$. It was also exhibited acid-neutralizing capacity (10.3%) and $H^+/K^+$-ATPase inhibition of 42.6% ($500{\mu}M$). In pylorus-ligated rats, liriodendrin showed lower volume of gastric juice ($4.38{\pm}2.14ml$), slightly higher pH ($1.53{\pm}0.41$), and smaller total acid output ($0.47{\pm}0.3mEq/4hrs$) than the control group. Furthermore liriodendrin inhibited colonization of H. pylori effectively. In vivo test, liriodendrin significantly inhibited both of HCl/EtOH-induced gastritis (46.9 %) and indomethacin-induced gastric ulcer (46.1%). From these results, we suggest that liriodendrin could be utilized for the treatment and/or protection of gastritis and gastric ulcer.
Background: Gastric ulcer (GU) is a common gastrointestinal disease that can be induced by many factors. Finding an effective treatment method that contains fewer side effects is important. 20 (S)-ginsenoside Rg3 is a kind of protopanaxadiol and has shown superior antiinflammatory and antioxidant effects in many studies, especially cancer studies. In this study, we examined the treatment efficacy of 20 (S)-ginsenoside Rg3 on GU. Methods: Three kinds of GU models, including an alcohol GU model, a pylorus-ligated GU model, and an acetic acid GU model, were used. Mouse endothelin-1 (ET-1) and nitric oxide (NO) levels in blood and epidermal growth factor (EGF), superoxide dismutase, and NO levels in gastric mucosa were evaluated. Hematoxylin and eosin staining of gastric mucosa and immunohistochemical staining of ET-1, inducible nitric oxide synthase (NOS2), and epidermal growth factor receptors were studied. Ulcer index (UI) scores and UI ratios were also analyzed to demonstrate the GU conditions in different groups. Furthermore, Glide XP from $Schr{\ddot{o}}dinger$ was used for molecular docking to clarify the interactions between 20 (S)-ginsenoside Rg3 and EGF and NOS2. Results: 20 (S)-ginsenoside Rg3 significantly decreased the UI scores and UI ratios in all the three GU models, and it demonstrated antiulcer effects by decreasing the ET-1 and NOS2 levels and increasing the NO, superoxide dismutase, EGF, and epidermal growth factor receptor levels. In addition, high-dose 20 (S)-ginsenoside Rg3 showed satisfactory gastric mucosa protection effects. Conclusion: 20 (S)-ginsenoside Rg3 can inhibit the formation of GU and may be a potential therapeutic agent for GU.
In order to investigate the effects of Kamikwakjeongtang by using the experimental animals. the action on gastrointestinal smooth muscles. the action of gastric juice secretion, the action of antiulcer. the transport ability of intestinal contents. the action of anticatharsis and the actions on the central nervous system were studied. The results were as follow: 1. Spontaneous motility of the isolated ileum of mice was suppressed and antiacetylcho-line chloride action was recognized. 2. Anti-action on barium chloride of fundus-strip of white rat was recognized. 3. Supression effects on gastric juice secretion. free acidity was recognized. 4. Preventive effect on the ulcer induced by pylorus-ligated was recognized. 5. Supression effects of large intestinal transport ability was recognized. 6. Anti-carthartic action was shown but was not recognized. 7. Analgesic effect by the acetic-acid method and prolonged effect of the total sleep time by pentobarbital-Na were recognized. According to the above results, effects based on oriental medical reference were consistent with the actual experimental effects.
In order to investigate the efficacy of Siljangsan by using the experimental animals, the action of gastric juice secretion, antiulcer. transport rates in the intestine and anti-cathartic action were studied. The following results were obtained. 1. Preventive effects on ulcer induced by pylorus-ligated and Ethanol-HCI were recognized 2. Suppression effects on gastric juice secretinon, free of acidity and total acidity were recognized, but pepsin output was not recognized. 3. For the transport ability of intestine of mice, the ability of small intestine was activated and large intestine was suppressed in the group administrated 4. Anti-cathatic action of mice induced by castor oil was recognized. According to above results, effect based on oriental medical reference was consistent with the actual experimental effect.
Objectives : This study was performed to investigate effect of evodiae fructus on acute reflux esophigitis rat induced by pylorus and forestomach ligation operation. Methods : Twenty-four laboratory rats were divided four groups and each group had six rats ; normal intact group, acute reflux esophagitis (RE) control group, two experiment RE group treated extract of evodiae fructus 600 mg/kg (EEF600) and 300 mg/kg (EEF300). All rats was fasted for 18 hr but free water, we induced RE by pylorus and forestomach ligation operation. Intact group and RE control group rats were orally administered a distilled water and two experiment groups were orally administed with EEF 600 mg/5ml/kg and 300 mg/5ml/kg. One hour after, rats were anesthetized, intact group was cut the abdomen open and sutured with 2.0 silk thread. RE control group and EEF group were cut the abdomen open, ligated pyloric canal and forestomach with 2.0 silk thread and sutured. Six hour after the operation, rats were sacrified, collected bloods in the abdominal vein, disectted a esophagus and stomach. The stomach was washed a 1 ml PBS and the esophagus was cut longitudinally and pictured a innter mucosa area to research damages in esophagus. Results : The esophagic tissue damage percentage of reflux esophagitis rat was increased compared to that of normal intact group. But esophagic damage percentage of EEF 600 were significantly decreased compared to that of RE control group. But there was no difference on gastric juice pH between control RE, alpha-tocopherol administration rat group and EEF administration rat group. In esophagus of RE control rat, gastric damage occurred severely and injury percentage of mucosa were increased, but EEF 600 mucous inflammatory damage percentage was significantly compared to that of RE control group. Proinflammatory cytokines such as TNF-alpha, IL-1beta and IL-6 in serum on RE control group were markedly grew than those of intact rat, those of vechicle group treated with EEF 600 and EEF 300 were remarkably decreased compared to production of proinflammatory cytokine of RE control group. In microscopic observation, intact group rat had no hyperemia, mucous injury and exclusion, ulcer and edema. But it could showed mucosa damages, submucosa edema and ulcer in RE control. However, administration of EEF 600 and EEF 300 made esophagus have less inflammation and injury by gastric acid. Conclusions : The results suggest that antiinflammatory Effect of EEF could attenuate the severity of reflux esophagitis and prevent the esophageal mucosal damage, and validate its therapeutic use in esophageal reflux disease.
The novel compound KH 3218 was synthesized and evaluated for its ability to inhibit the gastric H$^{+}$+ $K^{+}$ ATPase activity in vitro as well as to lessen gastric acid secretion in vivo. KH 3218 inhibited rabbit gastric H$^{+}$+ $K^{+}$ ATPase in a concentration and time dependent manner. $IC_{50}$/ value was estimated to be about 15 $\mu$M. The inhibition of the H$^{+}$+ $K^{+}$ ATPase by KH 3218 was blocked by sulfhydryl reducing agents, dithiothreitol or $\beta$-mercaptoethanol. The inhibition of the enzyme was not reversible by 50 fold dilution of the incubation mixtures, suggesting the irreversible nature of the inactivation. In the pylorus-ligated rift, KH 3218 reduced the total acid output as compared with the control. In addition, KH 3218 was capable of inhibiting H. pylori urease activity. These data suggest that KH 3218 is a potent inhibitor for H$^{+}$+ $K^{+}$ ATPase activity as well as for gastric acid secretion, and has a potential to be developed as a novel antiulcer agent.
본 실험에서 흰쥐에 실험적으로 급성위염, 위궤양 및 십이지장궤양을 유발하고 지유 에틸아세테이트분획을 투여하여 항위염 및 항궤양에 미치는 영향을 검토하였던 바 다음과 같은 결과를 얻었다. 지유 에탄올 추출물을 n-헥산, 클로르포룸, 에틸아세테이트 및 n-부타놀과 물로 계통분획하여 농축하고 in vitro에서 항산화력을 측정한 결과 에틸아세테이트분획이 가장 우수하였다. 지유 에틸아세테이트분획 (SOL & SOH)의 HCI-EtOH급성위염에 대한 억제율은 각각 $27.7\%$, $31.7\%$로 대조약물로 사용된 cimetidine(CMT, $47.2\%$)의 억제효과에는 미치지 못했으나 CON에 비해 우수하였다. Indomethacin유발 위궤양에서, SOL & SOH의 위궤양억제율은 각각 $48.28\%$, $51.08\%$로 대조약물로 사용된 CMT($58.26\%$)의 억제효과에는 미치지 못했으나 CON에 비하며 유의 성있는 억제효과를 나타냈다. Shay 위궤양에서, 지유 에틸아세테이트분획을 십이지장에서 위내로 투여하였을 때 궤양 억제율은 SOL이 $42.86\%$로 CON에 비하여 억제되었으나 유의성 있는 효과는 아니었으며, 용량을 증량하여 투여한 SOH는 $47.71\%$로 대조약물인 omeprazol(OMP)의 저해효과($62.00\%$)에 미치지는 못했으나 CON에 비해 유의적민 효과를 나타내었다. 위액량 및 총위산분비량에서, 지유 에틸아세데이트분획 투여로 CON에 비하여 $32.11\%$와 $47.69\%$, $24.28\%$와 $34.69\%$의 감소로 유의성 있는 효과를 나타냈으며 OMP($55.69\%$, $41.62\%$)에 근접하게 감소되었다. pepsin 활성은 지유 에틸아세테이트분획 투여로 CON에 비해 각각 $19.55\%$, $\%21.45$가 저하되었으나 유의성있는 변화는 아니었다. 수침 $\cdot$ 구속 스트레스에 의해 유발된 궤양의 경우, SOL는 궤양억제율이 $9.23\%$ 로 CON와 유사하게 출혈을 동반한 깊은 궤양이 관찰되었으나 용량을 증량하여 투여한 SOH는 $48.45\%$로 CON에 비해 궤양의 깊이도 얕아졌고 출혈을 동반한 궤양도 감소되어 유의적인 억제율을 나타내었다. Cysteamine 유발 십이지장궤양에서, CMT는 유발된 십이지장 궤양을 CON에 비해 $54\%$ 정도 억제하여 우수한 항궤양효과를 나타냈고 시료투여군인 SOL 및 SOH는 CMT보다 우수하지는 않았으나 약 $35\∼42\%$ 정도의 궤양억제율을 나타냈다. 이상의 실험결과에서 지유의 항위염 및 항궤양효과는 항산화물질에 의한 결과로 판단되어지며, 앞으로 위염 및 위궤양 개선을 위한 건강 기능성 식품으로 개발이 기대된다
스트레스로 인한 위궤양형성에 중추성 교감신경의 영향여부를 추구하기 위하여 norepinephrine, epinephrine, dopamine, isoproterenol 및 clonidine을 흰쥐의 뇌실내로 투여 하고 한냉 환경 $(4^{\circ}C)$에서 4시간 구속방치하여 위 분비기능의 변동과 궤양 발생 정도를 검색하여 다음과 같은 결과를 얻었다. 1. Norepinephrine, epinephrine, dopamine및 소량의 clonidine 처치로 궤양 발생이 현저하게 감소하였다. 2. Norepinephrine또는 epinephrine 처치군에서는 위액분비, 산분비 및 펩신 분비의 감소와 궤양 발생 감소가 초래되었다. 3. Dopamine혹은 소량의 clonidine 처치군에서는 궤양 발생의 감소와 위액분비 및 산분비 감소가 초래되었으나 펩신 분비는 변동 없었다. 4. Isoproterenol처치군에서는 궤양 발생과 펩신 분비는 대조군과 차이 없고, 위액분비 및 산분비의 감소만 나타났다. 5. 대량의 clonidine 투여군에서는 궤양발생, 산분비 및 펩신분비 모두 변동없이 약간의 위액분비 감소가 나타났다 이상의 결과로 보아 중추성 교감신경자극은 궤양 형성을 억압하는 작용이 있고, 이에는 교감신경성 ${\alpha}$-수용체 및 도파민성 수용체가 관여된다고 믿어지며, 이 효과는 위액분비 감소 및 산 분비 감소작용과 아울러 또 다른 요인이 관여한다고 추측된다.
