The intracellular localizations of polyamines and their related enzymes were investigated from young spinach leaves. Polyamines were present in all parts of plant cells, both in the subcellular organelles and in the soluble fraction of cytoplasm, however, polyamines were mainly located in the cytosolic fraction. Most activities of L-arginine decarboxylase(ADC) and L-ornithine decarboxylase(ODC), two important enzymes of putrescine and polyamine biosynthesis, were detected in cytosol fraction, while in subcellular organelles the activities were very low. Activities of diamine oxidase(DAO) and polyamine oxidase(PAO), the catabolic enzyme of diamine and polyamine, were not detected in spinach leaves. It was suggested that polyamines and their related synthetic enzymes were located in the soluble fraction of cytoplasm.
We examined the effect of CsCl and spermine on the induction of ornithine decarboxylase (ODC), a key enzyme in polyamine synthesis form Glycine max axes. Transcription of the ODC gene was induced by 0.1 and 1 mM of CsCl, and the amount of putrescine was increased 3.5-fold by 1 mM CsCl treatment. Spermine also induced the expression of the ODC gene in a die dependent manner. However, CsCl provoked an increase in the active phosphorylated ERK (pERK), a central element of the mitogen-activated protein kinase (MAPK) cascade. Our data demonstrates an interaction between the ODC induction and the MAPK signaling pathway, and suggests that the latter may be involved in cell signaling in salt-stressed plants.
We investigated the culture condition and effects of various growth factors on the culture of salivary gland acinar cells. Male, Sprague-Dawley rats (about 6 weeks old) were sacrificed and their submandibular, sublingual, and parotid glands were used as specimens. High oxygen level more than 90% and coating of Matrigel on culture dish were important factors to help increase the survival time of acinar cells, Proper concentration of enzymes such as collagenase and hyaluronidase during isolation steps was also important. Addition of various growth factors such as dexamethasone, insulin, transferrin, selenous acid, reduced glutathione, epidermal growth factor, isoproterenol, and putrescine in culture medium helped to increase lifetime of cultured salivary acinar cells.
The effects of polyamines on the activities of elongation factors EF-1 and EF-2, phenylalanyl-tRNA synthetase, and tRNA were investigated. The activities of EF-1 and EF-2 were mostly stimulated by spermidine among three kinds of polyamines. The activities of EF-1 and EF-2 were investigated in the presence of spermidine by 230 and 181%, respectively. The activity of phenylalanyl-tRNA synthetase was slightly increased in the presence of polyamines. The effect of spermine on the synthetase was higher than that of the other polyamines. The tRNA activity in the presence fo polyamines was increased by 206% with spermidine, by 144% with spermine, and by 114% with putrescine. According to these results, it is concluded that polyamines in higher plants stimulate the protein biosynthesis by promoting the activities of elongation factors EF-1 and EF-2, aminoacyl-tRNA synthetases, and tRNAs, but the effects of polyamines on the various components for protein biosynthesis are different in according to the kind of polyamines.
Effects of putrescine, spermidine and spermine on the activity of D-glucose-6-phosphate cyclohydrolase in the Daucus carota L. protoplast cultured for 4 days and effects of polyamines on the incorporation of D-[u-14C]-glucose treated to protoplasts in culture-medium were investigated. The activity of D-glucose-6-phosphate cyclohydrolase was increased by polyamines and among them spermine was the most effective. Polyamiens increased protein synthesis and this due to the increasing effect of the polyamines on the synthesis of glycoprotein which is one of cell wall components. The synthesis of cell polysaccharides, such sa pectic substances, hemicelluloses and cellulose was increased by polyamines, which stimulated synthesis of pectin substances, and hemicellulose more greatly than that of cellulose, and spermidine was the most effective. In the light of the above results it seems that the polyamines increase cell wall regeneration by the stimulation of enzyme activities which synthesize cell wall components.
Polyamine contents and the activities of their main biosynthetic enzymes, arginine decarboxylase (ADC) and ornithine decarboxylase(ODC), were investigated in Populus leaf segments during adventitious shoot regeneration with the addition of 1 millimolar polyamine synthesis inhibitors. From the study of polyamine synthesis inhibitors, ODC was found to be the principal route of polyamine biosynthesis in adventitious shoot regeneration of Populus leaf segments. The ADC inhibitor difluoromethyl arginine(DFMA) and ODC inhibitor difluoromethyl ornithine(DFMO) strongly reduced the putrescine content. On the contarary, DCHA, an inhibitor of spermidine synthase, increased it. Spermidine content was decreased with the treatment of each polyamine synthesis inhibitor, but the inhibitory effect of DCHA was stronger than any other polyamine inhibitor. The decreased polyamine level by polyamine synthesis inhibitors was restored with the exogenously applied polyamine. Comparing the polyamine contents with the adventitious shoot regeneration rate, were observed a close correlation between spermidine content and adventitious shoot regeneration of Populus leaf segments.
Possible roles of polyamines in the inhibition of cell elongation in Ranunculus petioles were investigated. Exogenously apoplied polyamines greatly inhibited the auxin-induced petiole growth, while treatment of the tissue with $\alpha$-difluoromethylarginine, the inhibitor of putrescine biosynthesis, further enhanced the growth in the presence of IAA. Inhibitory effect of spermine can also be apparent for fusicoccin-induced elongation, but not for growth induced by a low pH. Spermine also suppressed the ethylene-enhanced growth in the presence of auxin. Using computer-based video digitizer system, the inhibitory effects of spermine on petiole growth were kinetically analyzed. Auxin-induced growth was characterized by an initial and transient growth with a highly elevated rate followed by a steady growth with a slightly reduced rate. Spermine treatment was found to shorten the duration of the initial phase of growth, and to reduce the rates of both the initial and steady growth as well. The latent period for auxin induction was not affected by spermine.
Twelve Microorganisms capable of utilizing diaminododecane were isolated from the soil by enrichment culture technique. Seven strains of these were identified as Corynebacterium. The isolated strains were tested for the ability to utilize as carbon source, 10 different kind of alkane derivatives containing CN, $NH_2$, Cl, and SH groups. Laurylcyanide, dicyanooetane, chlorodecane, and dichlorodecane were not utilized by any of the isolated strains; putrescine dihydrochloride, cadaverine dihydrochloride, diaminododecane, and n-dodecane were utilized by all of the isolated strains; and all of the isolated strains except DAD 2-3 could utilize dodecylmercaptan. The alkane derivatives that did not serve as ,growth substrates were tested further in oxidation tests using resting cell preparation. Alkane derivatives that are being oxidized by all of the isolated strains are laurylcyanide and dichlorodecane. Dicyanooctane was also oxidized by all of the isolated strains except DAD 30L, chlorodecane was the only oxidized by the three isolated strains. The most remarkable substrate that is being oxidized is dichlorodecane containing CN groups diterminally. Evidence obtained with thin layer chromatography of ,ethyl acetate extracts of culture broth of isolated strains grown in some alkane derivatives shows that these alkane derivatives are degraded.

Gelling agent는 gelrite $0.2{\sim}0.3%$ 처리에서 발아가 촉진되었으며 $(65{\sim}70%)$ shoot 및 root의 생장도 양호하였다. 3. Cytokinin은 $0.1mg/{\ell}$의 BA와 kinetin처리에서 발아율$(65{\sim}70%)$과 신초 및 뿌리 의 길이 및 건물중이 높게 나타났다. 4. Polyamine의 효과에 대한 실험결과 putrescine $1mg/{\ell}$과 5$mg/{\ell},\;spermidine\;10mg/{\ell}$ 처리에서 체세포배의 발아(90%) 및 기관생장이 양호하였으며 분화식물의 투명화도 억제되었다.
