Simulation Study on the Efficacy of Toxin Removal by Pulsatile Flow in Blood Purification Systems that use Semipermeable Membranes

  • Using numerical models, we investigated the efficiency of toxin removal using pulsatile flow in blood purification systems that use semipermeable membranes. The model consisted of a three-compartmental mass transfer model for the inside body and a solute kinetics model for the dialyzer. The model predicted the toxin concentration inside the body during blood purification therapy, and the toxin removal efficiencies at different flow configurations were compared quantitatively. According to the simulation results, the clearances of urea and ${\beta}_2$ microglobulin (B2M) using a pulsatile pump were improved by up to 30.9% for hemofiltration, with a 2.0% higher urea clearance and 4.6% higher B2M clearance for high flux dialysis, and a 3.9% higher urea clearance and 8.2% higher B2M clearance for hemodiafiltration. These results suggest that using a pulsatile blood pump in blood purification systems with a semipermeable membrane improves the efficacy of toxin removal, especially for large molecules and hemofiltration treatment.

  • In this paper it is concerned to develop control method using jar-test results in order to predict the optimum dosage of coaglant, PAC(PoliAluminum Chloride). Considering the relations with the reactions with the reaction of coagulation and flocculation, the five independent variables ( e, g, turbidity of raw water, water turbidity in flocculators, temperature, pH, and alkalynity) are selected out of parameters and they are put into calculation to develop a neural network model for PAC dosing process in water purification system. This model is utilized to predict optimum dosage of PAC. That is, the optimum dosage of PAC is searched in neural network model for PAC dosing process to minimize the water turbidity in flocculators. This searching is implemented by means of expert heuristics. The efficacy of the proposed contorl schemem and feasibility of acquired neural network model for PAC dosing contorl in water purification system is evaluated by means of computer simulation.

  • This study was conducted to evaluate the purification efficacy by filter and ultra violet (UV) irradiation against inlet and outlet seawater of the land based fish farm. Purification efficacies for inlet seawater (50$\ell$/min) has been examined with filter of 60$\mu$m pore size and UV irradiation at an average dose 0.5 mWS/$cm^2$ for 5 months. For outlet seawater filter of 90 $\mu$m pore size only was used. Temperature, salinity, pH, $NO_3-N,\;NO_2-N,\;NH_4-N,\;PO_4-P$, DO and COD of inlet and outlet seawater in the system were not influenced. However, the removing rate of suspended solid and turbidity of inlet seawater were appeared to be $48.7\~65.6\%$ (average $51.9\%$) and $33/3\~42/5\%$ (average $34.8\%$) after passing through screen filter respectively. Also, germicidal efficiency to the total bacteris and Vibrio species were $16.7\~20.2\%$ (average $19.2\%$) $20.0\~21.9\%$ (average $20.9\%$) respectively after passing through UV irradiation. After passing through drum filter of 90$\mu$m pore size, suspended solid and turbidity of outlet seawater were appeared to be $42.7\~52.6\%$ (average $46.9\%$), $27.7\~29.9\%$ (average $28.3\%$), respectively.

  • In the water purification plant, the raw water is promptly purified by injecting chemicals. The amount of chemicals is directly related to water quality such as turbidity, temperature, pH and alkalinity. At present, however, the process of chemical reaction to the turbidity has not been clarified as yet. Since the process of coagulant dosage has no feedback signal, the amount of chemical can not be calculated from water quality data which were sensed from the plant. Accordingly, it has to be judged and determined by Jar-Test data which were made by skilled operators. In this paper, it is concerned to model and control the coagulant dosing process using jar-test results in order to predict optimum dosage of coagulant, PAC(Polymerized Aluminium Chloride). The considering relations to the reaction of coagulation and flocculation, the five independent variables(turbidity, temperature, pH, Alkalinity of the raw water, PAC feed rate) are selected out and they are put into calculation to develope a neural network model and a fuzzy model for coagulant dosing process in water purification system. These model are utilized to predict optimum coagulant dosage which can minimize the water turbidity in flocculator. The efficacy of the proposed control schemes was examined by the field test.

  • Interest in application of ultraviolet light technology for primary disinfection of potable water in drinking water treatment plants has increased significantly in recent years. The efficacy of disinfection processes in water purification systems is governed by several key factors, including reactor hydraulics, disinfectant chemistry, and microbial inactivation kinetics. The objective of this work was to develop a computational fluid dynamics(CFD) model to predict velocity fields, mass transport, chlorine decay, and microbial inactivation in a continuous flow reactor. In this paper, It describe the how to design optimal UV disinfection device for ground water, BWT and rainwater. Spring shape instrument silver coated located in inner side of disinfection chamber. It make lead the active flowing movement target water and maximize disinfection performance. To search the optimal design method, it was performed computer simulation with 3D-CFD discrete ordinates model and manufactured prototype. Using proposed design method, performed simulation and proved satisfied performance.

  • The article discusses the critical role of chromatography in the analysis and purification of proteins in biopharmaceuticals, emphasizing the importance of comprehensive characterization for ensuring their safety and efficacy. It highlights the use of Avantor® ACE® HPLC columns for the separation and purification of proteins, focusing on the analysis of intact proteins using reversed-phase liquid chromatography (RPLC) with fully porous particles.   • Collection of large number of gonadal Primordial germ cells(gPGCs) is a prerequisite factor for improving germline transmission efficacy in the aves, In this study a magnetic-activated cell sorter(MACS) was applied for improving retrieval efficacy of quail gPGCs and the migration capacity of MACS-separated gPGCs was further examined after being transplanted to recipient embryos. We also induced germline chimeras by transfer of MACS-separated quail gPGCs at the efficiency of 17.4% on average.

Novel Anti-Mesothelin Nanobodies and Recombinant Immunotoxins with Pseudomonas Exotoxin Catalytic Domain for Cancer Therapeutics

  • Recombinant immunotoxins (RITs) are fusion proteins consisting of a targeting domain linked to a toxin, offering a highly specific therapeutic strategy for cancer treatment. In this study, we engineered and characterized RITs aimed at mesothelin, a cell surface glycoprotein overexpressed in various malignancies. Through an extensive screening of a large nanobody library, four mesothelin-specific nanobodies were selected and genetically fused to a truncated Pseudomonas exotoxin (PE24B). Various optimizations, including the incorporation of furin cleavage sites, maltose-binding protein tags, and tobacco etch virus protease cleavage sites, were implemented to improve protein expression, solubility, and purification. The RITs were successfully overexpressed in Escherichia coli, achieving high solubility and purity post-purification. In vitro cytotoxicity assays on gastric carcinoma cell lines NCI-N87 and AGS revealed that Meso(Nb2)-PE24B demonstrated the highest cytotoxic efficacy, warranting further characterization. This RIT also displayed selective binding to human and monkey mesothelins but not to mouse mesothelin. The competitive binding assays between different RIT constructs revealed significant alterations in IC50 values, emphasizing the importance of nanobody specificity. Finally, a modification in the endoplasmic reticulum retention signal at the C-terminus further augmented its cytotoxic activity. Our findings offer valuable insights into the design and optimization of RITs, showcasing the potential of Meso(Nb2)-PE24B as a promising therapeutic candidate for targeted cancer treatment.

Secretory Expression and Purification of the Recombinant Duck Interleukin-2 in Pichia pastoris

  • Interleukin-2 (IL-2) is a vital cytokine secreted by activated T lymphocytes, and plays an important role in the regulation of cellular functions and immunity of animals. In this study, the recombinant duck IL-2 (rduIL-2) was secretory expressed in Pichia pastoris (P. pastoris). The recombinant P. pastoris strain was cultured in shake flasks and then scaled up in a 5.0-l bioreactor. The result showed that the maximal fresh-cell-weight of 594.1 g/l and the maximal $OD_{600}$ of 408 were achieved in the bioreactor. The rduIL-2 was purified by two steps of purification procedures, and approximately 311 mg of rduIL-2/L fermentation supernatant was obtained. SDS-PAGE showed that the purified rduIL-2 constituted a homogeneous band of ~16 kDa or ~14 kDa corresponding to the glycosylated or non-glycosylated duIL-2 protein in size, respectively. The bioactivity of rduIL-2 was determined by lymphocyte proliferation assay. The result indicated that the rduIL-2 greatly promoted the proliferation of ConA-stimulated lymphocytes in vitro. The P. pastoris expression system described here could provide promising, inexpensive, and large-scale production of the rduIL-2, which lays the foundation for development of novel immunoadjuvants to enhance both the immunity of ducks against various infectious pathogens and vaccine efficacy.

  • Isothiazoline derivatives is widely used to food, medical drug and industrial goods, cosmetics etcs, and it makes to restrain and to sterilize a breeding of microbe as a preservative and a sterilizing agent. It differs with the raw material of paraoxybenzoic acid derivatives or imidazolydinyl urea to be in use at present, on the efficacy and effect, and has various characteristics. This synthesis makes 3,3'-dithiodipropionic chloride to add a thionyl chloride in 3,3'-dithiodipropionic acid, and 3,3'-dithiodipropionic methyl amide makes to synthesize in a reflux reaction the mono methyl amine to 3,3'-dithiodipropionic chloride. And last synthesis becomes to make chlorination-cyclization molecule doing a reflux reaction in the temperature of $90{\sim}100^{\circ}C$ to mix excessively thionyl chloride and ethylene dichloride to 3,3'-dithiodipropionic methyl amide. The last synthesis material has got in the mixture of 5-chloro-2-methyl-4-isothiazoline-3-one and 2-methyl-4-isothiazoline-3-one, and it is so-called isothiazoline derivatives. The purification of isothiazoline derivatives makes to fuse in ethyl acetate, and makes to decolorize and to deodorize in recrystallization. This experiment has been in synthesis and purification of isothiazoline derivatives, and has tried to measure on the antisepsis and sterilization function of microbe according to pH or content change.
