Immunomodulatory Effects of Eckol, a Pure Compound of Ecklonia Cava, on Dendritic Cells

  • Background: Eckol purified from Ecklonia cava, a brown alga has been known to have cytoprotective effects on some cell lines against oxidants and ionizing radiation. However, there is no study about the effects of eckol on immune cells. Methods: Bone marrow (BM)-derived dendritic cells (DCs) were used to demonstrate the immunomodulatory effects of eckol on DCs, such as viability, the expression of surface markers, allogeneic stimulating capacity using MTI, flow cytometric, $^3H$-thymidine incorporation assay. Results: Eckol did protect DCs against cytokine withdrawal-induced apoptosis in a concentration dependent manner based on MTT assay. And also, it increased the expression of MHC class II and CD86 (B7.2) molecules, maturation markers, on the surface of viable DCs gated in FACS analysis. Furthermore, eckol-treated DCs stimulated the proliferation of allogeneic $CD4^+$ T lymphocytes compared to imDCs in $^3H$-thymidine incorporation assay. $CD4^+$ T lymphocytes activated with eckol-treated DCs produced the larger amount of IFN-${\gamma}$ and IL-4 than those cells with imDCs. Conclusion: Taken together, we demonstrate in this study that eckol, a pure compound of Ecklonia cava, may modulate the immune responses through the phenotypic and functional changes of DCs.

NUMERICAL STUDY ON COMBINED HEAT TRANSFER IN NIR HEATING CHAMBER (근적외선 열풍기의 복합열전달에 관한 수치적 연구)

  • Numerical analysis is carried out for combined heat transfer in an indirected NIR(Near Infrared Ray) heating chamber. Reynolds number and shapes of absorbed cylinder are known as important parameters on the combined heat transfer effects. Reynolds number based on the outer diameter of the cylinder is varied from $10^3$ to $3{\times}10^5$. Four difference heat transfer regimes are observed: forced convection and radiative heat transfer on the outer surface of the cylinder, pure conduction in the cylinder body, pure natural convection and radiation between lamp surface and inner surface of the cylinder, and radiation from the lamp. Flow and temperature characteristics are presented with iso-contour lines for the absorbed circular and elliptic cylinders to compare their differences. The convective and radiative heat transfer fluxes are also compared with different Reynolds numbers. As usual, Reynolds number is an important factor to estimate increasing convective heat transfer as it increases. The shape of absorbed cylinder results overall heat transfer rates remain unchanged.

Morphometric Characterization of Honey Bee, Apis mellifera Linnaeus, Inbred Lines in Korea (국내 서양종꿀벌 순계의 형태적 특징)

  • The A, C, F colonies of Apis mellifera ligustica Spin. and D, V colony of Apis mellifera caucasia Gorb. bees were collected from 2005-2007. Consequently, inbred lines were derived from the bees of original colonies by matting in the isolated island with due regard for pure breeding. This project helps in the selection of colonies with higher production capacity, aiming to improve honey and royal jelly production and breeding programs. Twenty-three standard morphological traits of honeybee were evaluated, and samples were compared with the data of the two original subspecies. The result suggested that 8 traits partly preserved in bees of inbred lines, and the bees from A. m. ligustica preserved more traits than bees from A. m. caucasia. Among the studied inbred lines, the F line is distinguished by an increase in leg parameters, considered as a favorable phenotypic trait of inbred lines. Importantly, bred of beelines in the same area can be classified as remote and isolated areas. Therefore, we observed differences of inbred lines with the origin subspecies in description acquired with morphometric characteristics as a result of adaptation, breeding, purebred individual lines used as an important resource for breeding novel cross-breeding colonies.

Crossbreeding and parental lineage influences the diversity and community structure of rice seed endophytes

  • Seed endophytes are very remarkable groups of bacteria for their unique abilities of being vertically transmitted and conserved. As plants attain hybrid vigor and heterosis in the process of crossbreeding, this might also lead to the changes in the community structure and diversity of plant endophytes in the hybrid plants ultimately affecting the endophytes of the seeds. It would be interesting to characterize how seed endophyte composition change over time. The objective of this study is to gain insights into the influence of natural crossbreeding and parental lineage in the seed bacterial endophytic communities of two pure inbred lines exploring contributions of the two most important sources of plant endophytes - colonization from external sources and vertical transmission via seeds. Total genomic DNA was isolated from rice seeds and bacterial DNA was selectively amplified by PCR. The diversity of endophytic bacteria was studied through Terminal-Restriction Fragment Length Polymorphism (T-RFLP) analysis. Diversity between the original parents and the pure inbred line may show significant differences in terms of richness, evenness and diversity indices. Heat maps reveal astonishing contributions of both or either parents (IR29 ${\times}$ Pokkali and AT401 ${\times}$ IR31868) in the shaping of the bacterial seed endophytes of the hybrid, FL478 and IC32, respectively. Most of the T-RFs of the subsequent pure inbred line could be traced to any or both of the parents. Comparison of common and genotype-specific T-RFs of parents and their offspring reveals that majority of the T-RFs are shared suggesting higher transmission of bacterial communities common to both parents. The parents influence the bacterial community of their offspring. Unique T-RFs of the offspring also suggest external sources of colonization particularly as the seeds are cultivated in different ecogeographical locations. This study showed that host parental lines contributed greatly in the shaping of bacterial seed endophytes of their offspring. It also revealed transmission and potential conservation of core seed bacterial endophytes that generally become the dominant microbiota in the succeeding generations of plant hosts.

Perturbation of host responses by Porphyromonas gingivalis biofilm (Porphyromonas gingivalis 바이오필름에 의한 숙주 면역반응의 교란)

  • The present study was performed to evaluate how cellular and humoral immune responses were perturbed by immunization of mixed periodontal bacterial biofilms. Each group of mice was immunizared with 1) Poqhyromonas gingivalis (P. gingivaliis) grown as a planktonic culture, 2) Fusobacterium nucleatum (F. nucleatum), 3) P. gingivalis grown as a biofilm, or 4) mixed P. gingivalis plus F. nucleatum grown as a biofilm culture, respectively. Immune mouse sera were collected from each mouse. Spleens were harvested to isolate T cells and consequently stimulated with antigen presenting cells and P. gingivalis whole cell antigen to establish P. gingivalis-specific T cell lines. There were no significant differences in the mean anti- gingivalis IgG antibody titers among mouse groups. Immunization of mice with pure P. gingivalis biofilm or mixed P gingivalis plus F. nucleatum biofilm resulted in significant reduction o f antibody avidity and opsonophagocytois function. INF-$\gamma$production by P. gingivalis-specific T cell lines was also substantially recluced in mouse groups immunized with the biofilm. It was concluded that P. gingivalis biofilm perturbs the cellular and humoral immune responses in periodontal disease.

CHEMICAL ABUNDANCES OF THE SYMBIOTIC NOVA AG PEGASI

  • The high-resolution optical region spectroscopic data of the symbiotic nova AG Peg secured with the Hamilton Echelle Spectrograph at the Lick Observatory, have been analyzed along with the International Ultraviolet Explorer UV archive data. We measure about 700 line intensities in the wavelengths of 3859 to $9230{\AA}$ and identify about 300 lines. We construct pure photoionization models that represent the observed lines and the physical condition for this symbiotic nova. The spectral energy distribution of the ionizing radiation is adopted from stellar model atmospheres. Based on photoionization models, we derive the elemental abundances; C & N appear to be similar to be smaller than the Galactic planetary nebular value while O is enhanced. Our result is compared with the Contini (1997, 2003) who analyzed the UV region spectral data with the shock + ionization model. The Fe abundance appears to be enhanced than that of normal planetary nebulae, which suggests that AG Peg may have formed in the Galactic disk. The models indicate that the temperature of the central star which excite the shell gas may have fluctuated to an unexpected extent during the years 1998 - 2002.

Near-infrared studies of iron knots in Cassiopeia A supernova remnant: I. Spectral classification using principal component analysis

  • We have been carrying out near-infrared (NIR) spectroscopy as well as [Fe II] narrow band imaging observations of Cassiopeia A supernova remnant (SNR). In this presentation, we describe the spectral classification of the iron knots around the SNR. From eight long-slit spectroscopic observations for the iron-bright shell, we identified a total of 61 iron knots making use of a clump-finding algorithm, and performed principal component analysis in an attempt to spectrally classify the iron knots. Three major components have emerged from the analysis; (1) Iron-rich, (2) Helium-rich, and (3) Sulfur-rich groups. The Helium-rich knots have low radial velocities (${\mid}v_r{\mid}$ < 100 km/s) and radiate strong He I and [Fe II] lines, that match well with Quasi-Stationary Flocculi (QSFs) of circumstellar medium, while the Sulfur-rich knots show strong lines of oxygen burning materials with large radial velocity up to +2000 km/s, which imply that they are supernova ejecta (i.e. Fast-Moving Knots). The Iron-rich knots have intermediate characteristics; large velocity with QSF-like spectra. We suggest that the Iron-rich knots are missing "pure" iron materials ejected from the inner most region of the progenitor and now encountering the reverse shock.

A Study on the Combined Heat Transfer and Analysis Fire Induced Combustion Gas in a partially Open Enclosure (개구부가 있는 밀폐공간내 화재의 복합열전달 및 연소가스 분석에 관한 연구)

  • The natural convection and combined heat transfer induced by fire in a rectangular enclosure is numerically studied. The model for this numerical analysis is partially opened right wall. The solution procedure includes the standard k-$\varepsilon$ model for turbulent flow and the discrete ordinates method (DOM) is used for the calculation of radiative heat transfer equation. In numerical study, SIMPLE algorithm is applied for fluid flow analysis, and the investigations of combustion gas induced by fire is performed by FAST model of HAZARD I program. In this study, numerical simulation on the combined naturnal convection and radiation is carried out in a partial enclosure filled with absorbed-emitted gray media, but is not considered scattering problem. The streamlines, isothermal lines, average radiation intensity and kinetic energy are compared the results of pure convection with those of the combined convection-radiation, the combined heat transfer. Comparing the results of pure convection with those of the combined convection-radiation, the combined heat transfer analysis shows the stronger circulation than those of the pure convection. Three different locations of heat source are considered to observe the effect of heat source location on the heat transfer phenomena. As the results, the circulation and the heat transfer in the left region from heating block are much more influenced than those in the right region. It is also founded that the radiation effect cannot be neglected in analyzing the building in fire. And as the results of combustion gas analysis from FAST model, it is found that O2 concentration is decreased according to time. While CO and CO2 concentration are rapidly increased in the beginning(about 100sec), but slowly decreased from that time on.

Rooting Ratio of Hemp Varieties (대마 품종의 삽목 증식율)

  • In order to obtain basic information for pure line breeding of hemp which is effective on the pure line preservation and propagation, three varieties were investigative from 1995 to 1996 at Mokpo Experiment Station, Honam Agriculture Experiment Station, and cutting parts, cutting times, cutting length and storage duration after cutting of plant were observed. Higher rooting ratio were observed in cutting of shoot and later branch, in July 5, optimum harvesting time and 5 days after harvesting time, in shorter storage duration,0 and 2 days. Also there were the highest rooting ratio increased by from 12% to 16% in shorter cutting of 10cm than 15cm long cutting in shoot, lateral branch and stem cuttings. Hemp cv. Korea showed higher rooting ratio than the other varieties, Japan and Turkey. Therefore, it was concluded that hemp, a dioecious plant, could be effectively propagated for the pure lines by cutting method, especially in cv. Korea.

  • Electrothermal-Hollow Cathode Glow Discharge Spectrometry (Et-HCGDS) has been constructed in our laboratory for in-situ monitoring of traceble amounts of rare earth elements and actinides. Et-HCGDS is the portable glow discharge system that can perform the trace analysis of elements. The main structural design of Et-HCGDS was based upon the electrothermal heating and glow discharge techniques. More details on Et-HCGDS are available elsewhere. In this study, air was used as a flow gas for the glow discharge system. As a result, the emission spectra of air were collected and the assignment of air emission lines was performed with helps of pure nitrogen and oxygen emission spectra and previously published results. We found that the emission lines of air plasma were mainly due to nitrogen molecules. This paper includes the complete assignments of the air emission lines observed by using Et-HCGDS. Also, this study will be an useful reference for spectrochemical anaysis including air analysis.
