Safety Investigation on Foodborne Pathogens and Mycotoxins in Honeybee Drone Pupas (수벌번데기로부터 식중독 세균 및 곰팡이독소 안전성 평가)

  • In this study, safety investigations on harmful microorganisms and mycotoxins were conducted on honeybee drone pupae as a new food material, which is rich in nutrients and capable of being mass produced in apiaries. The honeybee drone pupae produced in apiaries were collected from three different regions in Korea and frozen immediately. Subsequently, the samples were subjected to freeze-drying. According to the Korean Food Code test method, coliforms, Salmonella species, Staphylococcus aureus, and enterohemorrhagic Escherichia coli were not detected in 280 honeybee drone pupas. In addition, mycotoxins, aflatoxin $B_1$, ochratoxin A, deoxynivalenol, and zearalenone were not detected. Therefore, it is proposed that the honeybee drone pupae collected from the beehives and immediately frozen as safe from harmful microorganisms and mycotoxins and can be used as a food material.

Estimation of the Source Adult Population for Agrotis ipsilon (Lepidoptera: Noctuidae) Appearing in Early Spring in Korea: An Approach with Phenology Modeling (국내에서 이른 봄 출현하는 검거세미밤나방 성충집단의 기원 추정: 페놀로지 모형을 통한 접근)

  • The black cutworm, Agrotis ipsilon (Hufnagel), is an important crop pest worldwide that feeds more than 80 plant species including cabbage, potato, maize, wheat and bean, and this moth is a typical pest attacking underground parts of crops. It has been known in farm booklets that the larvae of A. ipsilon overwinter in the soil in Korea, but no definitive data exist yet. This study was conducted to evaluate that the specific appearance time of A. ipsilon observed actually in the field could be explained when we assumed that this pest overwinters in a form of larvae or pupae. Degree day-based phenology models were applied for tracking forward or backward to find the predicted developmental stage which developed at a specific stage found in the field. As a result of the analysis, it was confirmed that an initial population could be established in a group that does not overwinter as larvae or pupae in Korea. In other words, the appearance of adults in early March to April could not be explained by the presence of domestic overwintering populations. Populations that overwinter as larvae or pupae in Korea were able to emerge as adults in June to July at the earliest. Therefore, the group of adults appearing in early spring is highly likely to be a population that migrated from outside Korea. Taken together, it was estimated that the colony of A. ipsilon in Korea would be formed by a mixture of a migrant population through long-distance migration and a overwintering population.

  • In the silkworm fed on ginseng, the body weight and the cocoon weight were not only heavier, but also the ratio of a cocoon layer to a cocoon weight was higher than those in the control. Especially, the heavier pupae were appeared. The ginseng may be therefor available in silkworm improvement.

  • Dongchunghacho (Dong-Chong-Xia-Cho in Chinese) is one of entomogenous fungi that grow as parasites mainly to pupae or larvae. It includes many different genera such as Cordyceps, Paecilomyces, Torrubiella and Podonectria. The ethanolic extract of Cordyceps scarabaeicola, prepared from its fruiting bodies, showed significant inhibitory activity on angiogensis, which was detected by chick embryo chorioallantoic membrane (CAM) assay. The ethanolic extract of media-cultured Paecilomyces japonica also showed significant anti-angiogenic activity in CAM assay. (omitted)

Production of the Eggs with Abnormal Shape from the Domestic Silkworm, Bombyx mori, Infected with Autographa californica Nuclear Polyhedrosis Virus

  • The female pupae of the silkworms Bombyx mori, were injected with recombinant Autographa californica nuclear polyhedrosis virus (AcNPV) expressing green fluorescent protein (GFP) by percutaneous inoculation. When the 4 day-old female pupae were injected with 1x10$^{7}$ or 2${\times}$10$^{7}$ plaque forming units (pfu) of the recombinant AcNPV, oviposited number and egg weight were significantly decreased. Furthermore, the shape of the eggs was obviously divides into normal and abnormal shapes. The percentage of the eggs with an abnormal shape was 7.8% and 57.1% at 1${\times}$10$^{7}$ and 2${\times}$10$^{7}$ pfu inoculation, respectively. PCR analysis of the genomic DNA extracted from the eggs revealed that gfp and AcNPV ecdysteroid UDP-glucosyltransferase genes were amplified from both types of eggs with normal and abnormal shapes. The results demonstrate that AcNPV DNA, and gfp gene cloned into the AcNPV genome, injected in pupal stage were transmitted to eggs and remained stable through at least next generation.

Copper, Zinc-Superoxide Dismutase (Cu/Zn SOD) Gene During Embryogenesis of Bombyx mori: Molecular Cloning, Characterization and Expression

  • BmCu/Zn SOD was isolated from early embryo of Bombyx mori using microarray analysis. The BmCu/Zn SOD gene was observed during the early embryonic stage with the strongest signal found at the unfertilizaion, fertilization and blastoderm stages. The BmCu/Zn SOD gene encodes a protein of 154 amino acids with a calculated Mr of 15 kDa. The deduced amino acid sequence of BmCu/Zn SOD indicated that the residues that form on the Cu/Zn binding site are conserved and that the sequence is a 60% identity to that of M. domestica. In a phylogenetic tree, Bm SOD was also close to Drosophila SODs rather than other insect SODs. The BmCu/Zn SOD gene exists as a single copy in the genome. Transcripts of BmCu/Zn SOD cDNA were identified by northern blot analysis. The expression of the BmCu/Zn SOD gene was observed weakly in most of larvae, pre-pupae, pupae and adult tissues. Also, the BmCu/Zn SOD gene was observed in early embryonic stage. Although the roles of SODs remains to be further elucidated, the high expression of BmCu/Zn SOD gene at before 24 h post fertilization suggests that this gene is of general importance during early embryogenesis in the Bombyx mod.

Investigation of the Condition of Fruiting Body Formation by Cordyceps scarabaeicola (풍뎅이동충하초(Cordyceps scarabaeicola)의 자실체 형성 조건)

  • This experiment was carried out to study formation of fruitbody with Cordyceps scarabaeicola (EFCC C-252) isolate. This isolate was the one of best fruitbody formation on brown rice 60 g plus 30 g silkworm pupa media among EFCC C-251, EFCC C-252, EFCC C-1092 from EFCC (Entomopathogenic Fungal Culture Collection) of Kangwon National University. Fruiting body was formed only isolate EFCC C-252 among tested isolates on the medium of brown rice (60 g) and silkworm pupae (30 g). The optimal temperature and light for fruiting body formation were $25^{\circ}C$ and fluorescent light (300 lux). The maximal fruiting body formation was observed at 70 g of brown rice and 80 g of silkworm pupa medium which was treated separately. Fruiting body was formed maximally by 2 days interval of irrigation.
