Identification of chromosomal translocation causing inactivation of the gene encoding anthocyanidin synthase in white pomegranate (Punica granatum L.) and development of a molecular marker for genotypic selection of fruit colors

  • Previous studies have not detected transcripts of the gene encoding anthocyanidin synthase (ANS) in white pomegranates (Punica granatum L.) and suggest that a large-sized insertion in the coding region of the ANS gene might be the causal mutation. To elucidate the identity of the putative insertion, 3887-bp 5' and 3392-bp 3' partial sequences of the insertion site were obtained by genome walking and a gene coding for an expansin-like protein was identified in these genome-walked sequences. An identical protein (GenBank accession OWM71963) isolated from pomegranate was identified from BLAST search. Based on information of OWM71963, a 5.8-Mb scaffold sequence with genes coding for the expansin-like protein and ANS were identified. The scaffold sequence assembled from a red pomegranate cultivar also contained all genome-walked sequences. Analysis of positions and orientations of these genes and genome-walked sequences revealed that the 27,786-bp region, including the 88-bp 5' partial sequences of the ANS gene, might be translocated into an approximately 22-kb upstream region in an inverted orientation. Borders of the translocated region were confirmed by PCR amplification and sequencing. Based on the translocation mutation, a simple PCR codominant marker was developed for efficient genotyping of the ANS gene. This molecular marker could serve as a useful tool for selecting desirable plants at young seedling stages in pomegranate breeding programs.

Direct Antimicrobial Activity and Induction of Systemic Resistance in Potato Plants Against Bacterial Wilt Disease by Plant Extracts

  • The potential of three plants extracts, to protect potato plants against bacterial wilt caused by Ralstonia solanacearum was determined under greenhouse and field conditions. All soil drenching treatments of aqueous plant extracts of Hibsicus sabdariffa, Punica granatum and Eucalyptus globulus significantly reduced the disease severity compared with inoculated control. Although the applications of all three plant extracts resulted in similar reductions of disease severity in field up 63.23 to 68.39%, treatment of E. globulus leaf extract was found greater in restricting the symptom development than other the two plant extracts in the greenhouse. More than 94% reduction in the bacterial wilt symptom was observed in potato plants. All tested plant extracts were effective in inhibiting the growth of bacterial pathogen, not only in vitro, but also in stem of potato plants as compared with the inoculated control Potato plants treated with extract of H. sabdariffa reduced bacterial growth more effectively than treatment with P. granatum and E. globulus. Activity of defence-related enzymes, including peroxidase, polyphenoloxidase and phenylalanine ammonia lyase, were significantly increased in plants treated with the plant extracts compared to the control during the experimental period. In general, the higher enzymes activities were determined in both inoculated and non-inoculated treated potato plants after 8 days from plant extracts treatment. These results suggested that these plant extracts may be play an important role in controlling the potato bacterial wilt disease, through they have antimicrobial activity and induction of systemic resistance in potato plants.

석류 과실 부위별 폴리페놀 및 안토시아닌 함량이 항산화 및 항암 활성에 미치는 영향 (Effects of polyphenol and anthocyanin contents of Punica granatum fruit parts on their antioxidant and anticancer activities)

  • 고흥산과 캘리포나이산의 석류나무(Punica granatum L.) 과실을 과육, 종자 및 외과피로 분리하여 부위별 총 페놀성 함량, 안토시아닌 함량, DPPH 라디컬 소거능에 대한 항산화 활성 및 항암활성을 비교 검토하였다. 석류과실의 총 페놀성 함량은 외과피>주스>종자 순으로 유의적인 차이로 높게 나타났다. DPPH 라디컬 소거능은 외과피>주스>종자 순으로 높았고, 특히 외과피의 항산화 활성은 vitamin C와 유사한 활성을 보였다. 폐암세포주(Calu-6)에 대한 추출물의 항암활성은 외과피>종자>과육 순으로 높게 나타났다. 고흥산 석류 과실 부위별 총 안토시아닌 함량은 외과피>주스>종자 순으로 나타났고, 함유된 주요 안토시아닌은 Cy3,5G로서 가장 높은 함량을 보였고 그 다음이 Cy3G,Dp3,5G순으로 높게 나타났다. 총 페놀성 함량과 Calu-6 폐암세포주 활성 간이 $r^2=0.9706$으로 가장 높았고, 그 다음이 총 페놀성 함량과 DPPH 라디컬 소거능 간, DPPH 라디컬 소거능과 Calu-6 폐암세포주 활성 간 순으로 상관계수($r^2$)가 각각 0.8904, 0.8309로 나타나 상호 연관이 있음을 나타냈다.

석류 껍질추출물이 식중독균 여시니아 엔테로콜리티카의 쿼럼센싱과 바이오필름 형성능 억제 (Pomegranate (Punica granatum L.) Peel Extract Inhibits Quorum Sensing and Biofilm Formation Potential in Yersinia enterocolitica)

  • 쿼럼센싱은 세포 간의 의사소통 방법이며 박테리아의 병원성과 관련된 유전자들의 조절메커니즘이다. 박테리아는 다양한 생리학적 과정들을 제어하기 위해 이 쿼럼센싱 시스템을 활용한다. 본 연구에서 석류(Punica granatum L.) 껍질 추출물이 바이오 리포터 균주인 Chromobacterium violaceum 과 C. violaceum CV026에서 쿼럼센싱 억제능을 갖는 것으로 1차 선별되어 식중독균인 Y. enterocolitica에서 편모에 의한 운동능과 바이오필름형성 억제능에 대한 석류껍질 추출물의 효과에 대한 다음 실험을 수행하였다. 추가로 N-acylhomoserine lactones (AHLs)의 합성(yenI and yenR)과 편모 레귤론(fliA, fleB and flhDC) 에 관련된 특정유전자의 발현변화를 역전사 중합효소연쇄반응법으로 평가하였다. 결과는 석류껍질 추출물이 C. violaceum CV026에서 쿼럼센싱으로 제어되는 바이오레신 생산을 78.5%까지 억제하였으며, Y. enterocolitica에서는 세포의 성장에 영향을 주지 않고 바이오필름 형성과 편모 운동성을 현저히 감소시키는 것을 확인할 수 있었다. 이러한 억제 효과는 AHLs의 합성과 운동성에 관여하는 유전자 발현을 down-regulation 하는 결과와도 일치하였다. 본 연구의 결과는 석류껍질 추출물의 임상 적용을 위하여 생체 내 특성에 대한 추가적인 연구가 필요하다는 것뿐 아니라 석류껍질 추출물이 사람의 위장관염을 방지하기 위한 잠재적인 치료제가 될 수 있다는 것을 보여준다.

꽃차용 꽃 추출물의 구강위생균에 대한 항균특성 (Antibacterial Activities of Flower Tea Extracts against Oral Bacteria)

  • In this study, we analyzed flower tea activity against oral bacteria. Lagerstroemia indica, Paeonia suffruticosa and Hemerocallis fulva showed high extract yields. Bellis perennis, Punica granatum and Cercis chinensis showed the high rates of yield by ethanol extraction. Extract yield seemed to be related to the characteristics of the specimens rather than to the solvent. Streptococcus mutans, Streptococcus obrinus, Porphyromonas gingivalis and Prevotella intermedia were used to investigate extracts activity against bacteria; the former two cause dental caries and the latter two cause halitosis. Cornus officinalis, L. indica, P. granatum and P.s uffruticosa showed high antibacterial activities against S. mutans. In specimens extracted with ethanol, P. suffruticosa, Camellia sinensis, Camellia japonica L. and Rosa hybrida showed high antibacterial activities. L. indica, P. granatum and C. officinalis showed high antibacterial activities against S. sobrinus. C. officinalis, P. granatum, L. indica and P. suffruticosa showed high activities for specimens extracted with ethanol. The results show that the warm extracts of C. officinalis, L. indica and P. granatum may be effective to prevent dental caries. In particular, the ethanol-based extracts of C. officinalis, P. suffruticosa and C. sinensis were effective to prevent dental caries and thus may be highly marketable. Chrysanthemum zawadskii, R. hybrida, P. granatum, C. japonica L. and Zinnia elegans showed high antibacterial activity against P.gingivalis. R. hybrida showed the highest ethanol extract activity, followed by P. suffruticosa, P. granatum, C. japonica L. and L. indica. R. hybrida, P. granatum, C. morifolium and C. japonica showed high activity against Pr.intermedia in the order named. C. zawadskii, P. granatum, L. indica, C. japonica and A. princeps showed high ethanol extract activity. Thus, the warm extracts of R. hybrida, P. granatum and C. japonica may be helpful to reduce halitosis. In addition, the ethanol-based extracts of P. granatum, C. japonica and L. indica are expected to be highly marketable as mouthwashes.

약용식물추출물의 아토피성피부염에 대한 항염증 및 항알레르기효과(제 1보) (Anti-inflammatory and Anti-allergic Effects of Herbal Extracts on Atopic Dermatitis(Part I))

  • Atopic dermatitis is a chronic inflammatory skin disease associated with cutaneous hyper-reactivity to environmental triggers. In order to develop effective therapeutic herbal extracts for atopic dermatitis, cytotoxicity, antioxidant, anti-inflammatory and anti-allergic activities were investigated for various herbal extracts. Among candidate extracts, we selected Aloe vera L. (AV), Viola mandshurica W. Becker (VM), Punica granatum L. (PG), Dendrobium nobile L. (DN) and mixture of the above extracts (MX) for further investigations. All of them did not show cytotoxic activities to macrophage RAW264.7 cells below the concentration of 100 ppm. All showed antioxidant, anti-inflammatory and anti-allergic effects, although to various extents. In antioxidant effects, AV showed the highest effect, followed by PG and VM, while DN did the lowest. In evaluation for anti-inflammatory activities in macrophage RAW264.7 cells, AV and DN inhibited almost completely the LPS-induced production of nitric oxide, while AV, DN and VM showed strong inhibitory activities on the LPS-induced production of TNF-${\alpha}$. In anti-allergy effect in mast cell HMC-1, DN showed the highest effect, followed by AV and PG, while VM did the lowest. In the topical allergy reaction induced by compound 48/80 in Sprague-Dawley rat, DN exhibited significant anti-allergic effect, while PG, VM and AV did slight effect. These results suggest that AV, VM, PG and DN have antioxidant, anti-inflammatory and anti-allergic activities, and thus have the potential to reduce and alleviate the symptoms of atopic dermatitis.

석류 껍질 분말을 첨가한 스펀지 케이크의 품질 특성, 노화도 분석 및 항산화 활성 (Effect of Pomegranate (Punica granatum L.) Peel Powder on the Quality Characteristics, Retrogradation and Antioxidant Activities of Sponge Cake)

  • An agricultural waste, pomegranate (Punica granatum L.) peel is known to be rich in total phenolics, which are flavonoids having strong antioxidant effects. In this study, pomegranate peel sponge cakes were prepared with varying ratios of freeze dried pomegranate peel powder (0, 1, 3, 5, 7% (w/w)) to examine their effect on quality characteristics, retrogradation and antioxidant activities. The specific gravity and moisture contents of 3, 5, and 7% pomegranate peel powder showed higher values than the control and 1% group. Addition of pomegranate peel powder increased the batter yield, while there was a significant decrease in baking loss. Increasing pomegranate peel powder content significantly decreased the lightness (L) (from 75.03 to 57.04) and pH values, whereas redness (a), yellowness (b) and ${\Delta}E$ were increased. Increasing concentration of the peel powder also increased the hardness and chewiness, while the springiness and cohesiveness decreased. Considering the Avrami equation, Avrami exponene (n) decreased from 1.8055 (control) to 0.9199 (7% pomegranate peel powder). Time constant (1/k) was lowest in control (at 17.64) and highest in the 7% group (39.84). Total polyphenol, flavonoid content, DPPH and ABTS radical scavenging activities significantly increased with increments in the content of pomegranate peel powder. A sensory evaluation by the 7-point scaling method showed that the sponge cake containing 7% pomegranate peel powder had the highest scores in color, flavor, sweetness, chewiness and overall acceptability. Hence, it is considered that sponge cake supplemented with 7% pomegranate peel powder is the most appropriate for quality characteristics, retrogradation and antioxidant activities.
