A New Method of Colorimetric Determination of Piperine Using p-Nitrophenyl Diazonium Fluoborate in Pepper (Piper Nigrum L.) (p-Nitropheneyl Diazonium Fluoborate를 이용한 후추 Piperine의 새로운 비색정량법)

  • A survey of the available methods for the determination of piperine was made, and a new method, developed based upon alkaline hydrolysis at about $140^{\circ}C$ followed by colorimetric determination of the liberated piperidine. Piperidine on treatment with p-nitrophenyl diazonium fluoborate reagent gives a red colored complex which has an adsorption maximum at 530nm. The method measures the total pungency in pepper and applicaple to piperine of black pepper and oleoresin of black pepper. The advantage of the present method is that the pungent compounds can be determined in micro samples.

  • It is previously reported that 6-paradol can induce prolonged analgesia in experimental animals. In order to investigate the mutagenicity of 6-paradol, Ames Samonella/microsome plate assay was carried out with Salmonella typhimurium strains, TA 98, TA 100, TA 1535 and TA 1538, 6-Paradol was nonmutagenic in Salmonella typhimurium with and without rat liver microsomal activation. The rec assay with Bacillus substilis strains H 17 $rec^+$ and M 45 $rec^+$ was carried out ot test 6-paradol and other compounds (1-3 mg/disc) for DNA damaging activity, 6-Paradol was also nonmutagenic in DNA damaging activity. The relative size of the inhibition zone for 6-paradol was smaller than that of capsaicin. We have also determined the pathological effects of this compound on the various tissues of rats after administrating(i.p.) with increasing doses of 4, 8, 12, 16 mg/kg at 2 hour intervals and found no significant changes in terms of histology.

Effect of [6]-Gingerol, a Pungent Ingredient of Ginger, on Osteoblast Response to Extracellular Reducing Sugar

  • Diabetes is marked by high glucose levels and is associated with decreased bone mass and increased fracture rates. To determine if [6]-gingerol could influence osteoblast dysfunction induced by 2-deoxy-D-ribose (dRib), osteoblastic MC3T3-E1 cells was treated with dRib and [6]-gingerol and markers of osteoblast function and oxidized protein were examined. [6]-Gingerol ($10^{-7}\;M$) significantly increased the growth of MC3T3-E1 cells in the presence of 30 mM dRib (p<0.05). [6]-Gingerol ($10^{-7}\;M$) caused a significant elevation of alkaline phosphatase (ALP) activity, collagen content, and osteocalcin secretion in the cells. We then examined the effect of [6]-gingerol on the production of osteoprotegerin and protein carbonyl in osteoblasts. Treatment with [6]-gingerol ($10^{-9}$ and $10^{-7}\;M$) increased osteoprotegerin secretion in osteoblastic cells. Moreover, [6]-gingerol ($10^{-9}$ and $10^{-7}\;M$) decreased protein carbonyl contents of osteoblastic MC3T3-E1 cells in the presence of 30 mM dRib. Taken together, these results demonstrate that [6]-gingerol inhibits dRib-induced damage and may be useful in the treatment of diabetes related bone diseases.

Consideration in the Interpretation of the Soyo-san Prescription (소요산의 방론에 대한 고찰)

  • Objectives : The purpose of this study was to investigate the interpretation of the Soyo-san(SYS) prescription in order to obtain the evidence for clinical applications. Methods and Result : We interpreted on the SYS-related contents based on 25 classic books, analyzed it according to followed categories ; meaning of name, alteration of herbal composition according to age, usage, origin, physiological and pathological situation of SYS, description of each herbal drug Conclusion : 1. So(逍) mean diminish(消), 遙 mean roll(搖). It reflected feature of SYS, that alleviate depression without impairing vital energy(正氣). 2. SYS was first mentioned in the prescription of peaceful benevolent dispensary(太平惠民和劑局方), its essential application was harminizing liver depression and spleen deficiency(肝鬱脾虛) 3. SYS originated from Sayuk-san. Sayuk-san was appropriate for soothe the liver and regulate the spleen(疏肝理脾), while SYS was suitable for depressed liver and blood deficiency(肝鬱血虛) 4. Most medical literature insisted that application situation of SYS is blood deficiency(血虛), depressed liver(肝鬱), spleen deficiency(脾虛). And there is strong correlation between three factors. 5. Bupleurum falcatum L.(柴胡), Mentha arvensis L.(薄荷) alleviates liver depression as "pungent dissipate-qi uprais (辛散氣升)" nature. Angelica gigas Nakai(當歸), Paeonia lactiflora Pall.(白芍藥) emoliates the liver by nourishing the blood. Atractylodes japonica Koidz. ex Kitam.(白朮), Poria cocos(茯苓), Glycyrrhiza uralensis Fisch.(甘草), Zingiber officinale Roscoe(生薑) cultivate the basis as tonifying earth(補土).

Capsaicin-Induced Apoptosis of H-Ras-Transformed Human Breast Epithelial Cells is Rac-Dependent via ROS Generation

  • Many studies have focused on the anticarcinogenic, antimutagenic or chemopreventive activi-ties of capsaicin (trans-8-methyl-N-vanillyl-6-nonenamide) which is a major pungent ingredient in red pepper. We have previously shown that capsaicin selectively induces apoptosis in H-ras-transformed MCF10A human breast epithelial cells but not in their normal cell counter-parts (Int. J. Cancer, 103, 475-482,2003). In this study, we investigated the possible roles of reactive oxygen species (ROS) and Rac1 in capsaicin-induced apoptosis of H-ras MCF10A cells. Selective induction of ROS generation by capsaicin treatment was observed only in H-ras MCF10A cells. Pretreatment of H-ras MCF10A cells with an antioxidant N-acetylcysteine(NAC) significantly reversed capsaicin-induced growth inhibition, suggesting that ROS may mediate the apoptosis of H-ras-transformed cells induced by capsaicin. Rac1 was prominently activated by H-ras in MCF10A cells. Based on the studies using a wild type Rac1 and a domi-nant negative Rac1 constructs, we propose that Rac1 activity is critical for inhibitory effect of capsaicin on growth of H-ras-transformed MCF10A cells possibly through ROS generation.

Capsaicin-Induced Apoptosis and Reduced Release of Reactive Oxygen Species in MBT-2 Murine Bladder Tumor Cells

  • Bladder cancer is a common cancer with high risk of recurrence and mortality. Intravesicle chemotherapy after trans-urethral resection is required to prevent tumor recurrence and progression. It has been known that antioxidants enhance the antitumor effect of bacillus Calmette-Guerin (BCG), the most effective intravesical bladder cancer treatment. Capsaicin, the major pungent ingredient in genus Capsicum, has recently been tried as an intravesical drug for overactive bladder and it has also been shown to induce apoptotic cell death in many cancer cells. In this study, we investigated the apoptosis-inducing effect and alterations in the cellular redox state of capsaicin in MBT-2 murine bladder tumor cells. Capsaicin induced apoptotic MBT-2 cell death in a time- and dose-dependent manner. The capsaicin-induced apoptosis was blocked by the pretreatment with Z-VAD-fmk, a broad-range caspase inhibitor, or Ac-DEVD-CHO, a caspase-3 inhibitor. In addition to the caspase-3 activation, capsaicin also induced cytochrome c release and decrease in Bcl-2 protein expression with no changes in the level of Bax. Furthermore, capsaicin at the concentration of inducing apoptosis also markedly reduced the level of reactive oxygen species and lipid peroxidation, implying that capsaicin may enhance the antitumor effect of BCG in bladder cancer treatment. These results further suggest that capsaicin may be a valuable intravesical chemotherapeutic agent for bladder cancers.

Body-fat Suppressive Effects of Capsaicin through $\beta$-adrenergic Stimulation in Rats Fed a High-fat Diet (고지방식이를 섭취시킨 흰쥐에서 $\beta$-adrenergic 활성의 증가에 의한 Capsaicin의 체지방 감소효과)

  • The effects of capsaicin, a pungent principle of hot red pepper, on body fat gain, balance serum lipid values were investigated in rats fed a high-fat(30%) diet. Administration of capsaicin by dietary administration caused a complete cessation of increased in body weight and fat gain induced by the high-fat diet. However, energy intake and body protein gain were not affected by capsaicin. Therefore, the suppression of body fat gain by capsaicn was believed due to an increased in energy expenditure. Simultaneous administration of capsaicin and a $\beta$-adrenergic blocker, propranolo, resulted in the inhibition of changes in body fat gain by capsaicin without remained unchanged, indicating an increase in the number of mitochondria in brown adipose tissue. Therefore, it appears that capsaicin possesses potent body fat suppressive effects mediated by $\beta$-adrenergic stimulation in which brown adipose tissue may be involved. On the other hand, capsaicin had no effects on serum triglyceride, total cholesterol or HDL-cholesterol levels. These results are in contrast to those reported by other investigators. Perhaps expression of the effects of capsaicin on plasma lipids is a rather complicated process, dependent on the type of diet administered, fat content of the diet, period and route of capsaicin administration, and species and strain of animals used.

Capsaicinoid Content in Fruits of Parental Cultivars and Intervarietal Hybrids of Capsicum annuum L. (고추의 친품종 및 품종간 잡종의 과실에 있어서 캡사이시노이드 함량)

  • The capsaicinoid content of F$_{1}$ hybrids between annuum L. 'California Wonder', four native Japanese cultivars, and three Korean F$_{1}$ cultivars were evaluated and the results compared with parental cultivars. The capsaicinoid content was higher in the placenta than in fruit flesh. The placenta of 'Yatsufusa', 'Taka no Tsume', 'Cheongyang', 'Sinhong', and Geoseong' were the most pungent cultivatrs, with capsaicinoid content of more than 1000mg/100g DW, while 'California Wonder', and 'Fushimi Amanaga' contained trace amounts. Capsaicinoid content in F$_{1}$ hybrids varied with reciprocal crossing. Capsaicinoid content also varied with fruit development, rapidly increasing in the fourth or fifth week after flowering, and thereafter slowly declined.

  • The onion (Allium cepa L.) is the most widely cultivated species of the genus Allium, especially it has been valued because of the pungent flavor and aroma. Allium species including onion has very large genome sizes ranging from approximately 10 to 20 Gbp, which have complicated genomic studies and precluded genome sequencing until recently. A population of 186 F2 individuals derived from a cross of 'Umjinara' ${\times}$ 'Sinsunhwang' and the two parental lines were used for this study. For the development of framework map, various types of markers including SSRs, RAPD, SNPs, and CAPS makers have been used for polymorphism test. Especially, a lot of SNP and CAPS loci were developed from the onion transcriptome sequence by RNASEQ of two parental lines. The GBS libraries have been constructed based on a modified protocol from Poland Lab using a two-enzyme system. We have been developing markers showing polymorphism between two parental lines, and genotyping for all F2 individuals were finished for a number of polymorphic markers. For the construction of GBS libraries, a set of 192 barcoded adapters were generated from complementary oligonucleotides with XhoI overhang sequence and unique barcodes of length 4-8 bp and they have been tested using two parental linesto determine the optimum conditions for GBS analysis.

  • Chili pepper (Capsicum annum) is origin of subtropical region, and has been spread all over the world. It is increasing the production and consumption in recent year. Chili peppers are readily incorporated into local South Asian cuisines perhaps because people are already familiar with pungent and spicy flavors. Chili peppers, despite their fiery "hotness", are one of very popular spices known for their medicinal and health benefiting properties. Especially in South East Asia, they grow up so many cultivars of them recently, so it is so important crop world wide. In South East Asia, there are some articles about chili pepper in Thailand and Indonesia, but in Vietnam there is not so much information about chili pepper. In this paper, we analyzed genetic diversity in Vietnamese Chili pepper through the survey of local chili pepper. As a result, we got 38 kinds of chili fruits, 26 kinds of leaves and some information from farmers all in Vietnam. And I made the phylogenetic tree by SSR with 10 DNA markers. Finally we found the genetic similarities by regions.
