밀감 가공부산물에서 추출한 펙틴의 특성 (Properties of Pectin Extracted from By-product in Citrus Processing)

  • 밀감 가공부산물에서 추출한 펙틴의 특성을 조사한 결과 알콜불용성 고형물(ASS)의 함량은 albedo층이 18.1%로 가장 높고, pulp가 5.7%로 가장 낮았다. AIS 구성 다당류 중 펙틴은 pulp와 albedo층이 각각 40.5% 및 35.2%로 구성되어 있어 펙틴 추출용으로서 좋은 소재인 것으로 나타났다. 총 펙틴 함량은 부위에 따른 큰 차이를 보이지 않아 대체로 AIS에 대해 30% 내외 이었으나, 가용성 펙틴 중 염산 가용성 펙틴의 경우에는 flavedo층이 14.0%로 가장 높고, pulp가 4.4%로 가장 낮았으며, 염류 가용성 펙틴의 경우에는 pulp부위가 15%임 에 비해 과피는 7% 정도로서 부위에 따른 차이가 나타났다. 0.05N-HCI을 사용하여 $85^{\circ}C의$ 열탕에서 펙틴을 추출한 결과 60분의 가열처리로 90%이상의 펙틴이 용출되었으며, microwave의 경우 5분간 2회 조사시킴에 의해 90% 정도 추출되었으나 부위별에 따른 차이는 크지 않았고, 이들 펙틴의 에스테르화도에 있어서는 microwave 처리구의 것이 다소 높은 값을 나타내었다. 추출 펙틴의 중성 당 조성을 조사한 결과 rhamnose, arabinose, galactose, glucose 및 xylose로 구성되고 있고, 이중 galactose가 60~78% 정도를 차지한 반면 xylose는 5% 이하이었다. 펙틴의 분자량 분포를 Sepharose CL-4B를 이용, gel permeation chromatography로 조사한 결과 부위별에 따른 큰 차이는 보이지 않았으나 pulp의 경우 분리 피크가 2개로 나타나 pulp를 구성하는 펙틴은 분자량이 높은 것과 이보다 낮은 두 group으로 구성되어져 있음을 알 수 있었다.

과육이 서양삼 종자 발아에 미치는 영향 (Inhibition Effects of Pulp on Seed Germination of American Ginseng)

  • 본연구는 서양삼(西洋蔘)(Panax quinquefolium Linne) 과육(果肉)에 의한 Brassica(Brassica chinensis Linne) seed를 대상으로 발아(發芽) 억제작용을 검토하였다. 서양삼과즙(果汁)에는 발아(發芽)억제물질이 존재하고. 그 억제작용은 희석도에따라 감소하였다. 즉 희석배수가 0, 1, 2, 5, 10, 20, 50, 100일때 배근(胚根)의 길이는 각각 0, 0.32, 0.72, 3.13, 483, 16.07, 16.73, 23.50 mm이였다(CK=25.98 mm). 과육의 존재는 종배(種胚)의 성장을 현저하게 억제하였다. 수확(收穫)후 과육이 종자 표면에 부착한 시간이 0, 15, 30, 60일 때 종자 생장에 대한 억제 시간은 각각 30, 75, 135, 135일로 나타났다. 종자 썩는 비율도 높아졌다. 과육의 부착한 시간이 0, 15, 30, 60, 270일 때 종자 썩는 비율은 각각 5.47, 5.71, 19.05, 27.14, 33.33%로 나타났다. 이러한 결과는 과육에 서양삼종자휴면(西洋蔘種子休眠)을 일으키는 주요한 억제성분이 함유되어 있다는 것 을 시사하였다.

흰쥐대구치 치수노출후 치수조직내 CGRP함유 신경섬유의 분포에 관한 면역조직화학적 연구 (AN IMMUNOHISTOCHEMICAL STUDY ON THE DISTRIBUTION OF CGRP CONTAINING NERVE FIBERS AFTER PULP EXPOSURE IN RAT MOLAR)

  • The purpose of this study was to investigate the distribution of calcitonin gene-related peptide(CGRP) containing nerve fivers after pulp exposure in rats. The Spague-Dawley rats weighing about 250 - 300g were used. The animals were devided into normal control group and experimental groups. Experimental animals were sacrified on 2, 4, 7, 10 days after pulp exposure. The maxillary teeth and alveolar bone were removed and immersed in the 4% paraformaldehyde plus 0.1M phosphate buffer (pH 7.4). Serial frozen $50{\mu}m$ thick sections were cut with a cryostat. In the immunohistochemical staining procedure, the rabbit CGRP antibody was used as a primary antibody. The sections were incubated for 48 hours at $4^{\circ}C$, and placed into biotinylated anti-rabbit IgG as a secondary antibody and incubated in ABC (avidin-biotin complex), The sections were visualized by 0.05% 3.3 diaminobenzidine tetrahydrochloride. The results of this study were as follows: 1. In control group, CGRP containing nerve fibers ran parallel to the long axis of root and reached the coronal pulp. They were distributed on Raschkow plexus under the odontoblastic layer. 2. In 2 day group after pulp exposure, tissue necrosis and acute inflammation occurred and CGRP containing nerve fibers increased. In 4 day group, the necrotic tissue extended to the pulp and CGRP containing nerve fibers were distributed around the inflammation zone. 3. In 7 day group after pulp exposure, pulp necrosis occurred, and in 10 day group, the abscess under the necrotic pulp extended to the root apex area and CGRP containing nerve fibers were not observed in root canals. 4.The sprouting of CGRP nerve fibers was most remarkable at the pulp chamber under injury in 4 day group, and it was found at inflammation zone under the necrotic tissue in 7 day group and the remaining root pulp tissue in 10 day group. As mentioned above, CGRP nerve fibers had a tendency to increase around the inflammatory zone, especially around the acute inflammation tissue, when compared with control group. It is suggested that CGRP nerve fibers maybe related to the control of inflammatory response of pulp tissue.

Screening of Wood-Rotting Fungi for Efficient Decolorization of Draft Pulp Bleaching Effluents

  • In order to find fungi having high treatment activity of kraft pulp bleaching (E1) effluent without any additional nutrietns, 124 strains of white-rot fungi were isolated from decayed wood samples. The author isolated five fungi(KS-62, MZ-400 , YK-719, YK-472 and Phanerochaete sordida YK-624) having high-decolorization activity of the E1 effluent. Particularly, the fugus KS-62 show the high effect of the decolorization and the degradation of the chlorinated lignin in the E1 , effluent compared with Coriolus versicolor and Phanerochaete chrysosporium.

Effects of Surfactants on the Enzymatic Blaching of Kraft Pulp by Xylanase

  • A xylanase was purified from a commercial crude xylanase, Pulpzyme HC, and used for the bleaching of kraft pulp in the absence or in the presence of nonionic surfactants, Tween 20, Tween 80, and Igepal C930. The purified xylanase has a molecular weight of 23,500 as determined by a reducing SDS-PAGE. Tween 20 was most effective to enhance the efficiency of the enzymatic bleaching of kraft pulp by xylanase.

  • The making of high leveled smoothness for the printed electronics base paper, wood pulp and sea algal pulp were mixed. If sea algal pulp contents is increased to 9%, the smoothness was moderately increased, and more effective in Softwood mixture than Hardwood, low freeness(high wetness) than high freeness(low wetness).

Effects of Fermented Potato Pulp on Performance, Nutrient Digestibility, Carcass Traits and Plasma Parameters of Growing-finishing Pigs

  • A total of 629 Duroc${\times}$Landrace${\times}$Large White crossbred pigs were utilized in three experiments (Exp. 1, 222 pigs weighing $25.6{\pm}2.0\;kg$ BW; Exp. 2, 216 pigs weighing 5$6.2{\pm}4.3\;kg$ BW; Exp. 3, 191 pigs weighing $86.4{\pm}4.6\;kg$ BW) conducted to determine the effects of fermented potato pulp on performance, nutrient digestibility, carcass traits and plasma parameters in growingfinishing pigs. Each experiment lasted 28 d. The pigs were assigned to one of two corn-soybean meal-based diets containing 0 or 5% fermented potato pulp. The inclusion of fermented potato pulp increased weight gain (p<0.05) in experiments 1 and 2 and increased feed intake (p<0.05) in experiment 2. Feed conversion was improved (p<0.05) in experiment 2 and showed a tendency to improve (p<0.10) in experiments 1 and 3 when pigs were fed fermented potato pulp. Fermented potato pulp increased (p<0.05) dry matter digestibility in experiments 1 and 3 and energy digestibility in experiment 2. Feeding fermented potato pulp decreased plasma urea nitrogen (p<0.05) and alanine aminotransferase (p<0.05) in experiments 1 and 2, while plasma aspartate aminotransferase was decreased (p<0.05) in experiment 3. Dietary fermented potato pulp did not affect the carcass characteristics of finishing pigs. Feeding fermented potato pulp reduced (p<0.05) fecal ammonia concentration in all three experiments. In conclusion, feeding growing-finishing pigs diets containing 5% fermented potato pulp improved weight gain and feed conversion without any detrimental effects on carcass traits. The improvements in pig performance appeared to be mediated by improvements in nutrient digestibility.

The Effects of Fiber Source on Organ Weight, Digesta pH, Specific Activities of Digestive Enzymes and Bacterial Activity in the Gastrointestinal Tract of Piglets

  • The aim of this study was to explore the effects of fiber sources on gut development and bacterial activity in the gastrointestinal tract of piglets. Eighteen crossbred (Duroc${\times}$Landrace${\times}$Yorkshire) barrows were fed a basal diet based on corn plus soybean meal or similar diets in which a portion of the corn and soybean was replaced by 5% wheat bran or 5% sugar beet pulp. The results indicate that pigs fed diets containing 5% wheat bran or 5% sugar beet pulp had lower liver weights than control pigs (p<0.01). The relative weight of the pancreas in pigs fed diets containing 5% sugar beet pulp was greater than that of control pigs or pigs fed diets containing 5% wheat bran (p<0.05). The pH of the ileal digesta of pigs fed the diet containing 5% wheat bran was higher than that of control pigs or pigs fed the diet containing 5% sugar beet pulp (p<0.05). The lipase activity in the distal jejunum, proximal, and distal ileum of pigs fed the control diet was higher than that of pigs fed the diets containing 5% wheat bran or 5% sugar beet pulp (p<0.05). The concentration of volatile fatty acids anterior to the caecum was greater for the pigs fed the diet containing 5% sugar beet pulp, while the concentration of volatile fatty acids posterior to the ileum was greater for the pigs fed the diet containing 5% wheat bran. This means that sugar beet pulp increased the bacterial fermentation precaecum, while wheat bran increased the bacterial fermentation post-ileum. The concentration of bacterial nitrogen and bacterial protein/total protein in ileal digesta of pigs fed the control diet was higher (p<0.05) than that of pigs fed the diets contained either fiber source. Bacterial protein/total protein in the feces of pigs fed the diet containing 5% sugar beet pulp was higher than that of pigs fed the control diet. This means that inclusion of 5% wheat bran or sugar beet pulp in diets influenced the development of the digestive tract of piglet. The mechanism by which dietary fiber reduced specific activity of lipase needs further consideration. Dietary fiber influenced the bacterial activity in the digestive tract of piglets, sugar beet pulp increased the fermentation in the upper gastrointestinal tract, and while wheat bran increased the fermentation in the lower gastrointestinal tract.

방사선조사가 당뇨 백서의 치수조직에 미치는 영향 (Effect of irradiation on the dental pulp tissues in streptozotocin-induced diabetic rats)

  • Purpose : To observe the histopathological changes in the pulp tissues of mandibular molars in streptozotocin-induced diabetic rats after irradiation. Materials and Methods : The male Sprague-Dawley rats weighing approximately 250 gm were divided into four groups: control, diabetes, irradiation, and diabetes-irradiation groups. Diabetes mellitus was induced in the rats by injecting streptozotocin. Rats in control and irradiation groups were injected with citrate buffer only. After 5 days, the head and neck region of the rats in irradiation and diabetes-irradiation groups were irradiated with a single absorbed dose of 10 Gy. All the rats were sacrificed at 3, 7, 14, 21, and 28 days after irradiation. The specimen including the mandibular molars were sectioned and observed using a histopathological method. Results : In the diabetes group, capillary dilatation was observed. However, there was no obvious morphologic alteration of the odontoblasts. In the irradiation group, generalized necrosis of the dental pulp tissues was observed. Vacuolation of the odontoblasts and dilatation of the capillaries were noted in the early experimental phases. In the diabetes-irradiation group, generalized degeneration of the dental pulp tissues was observed. Vacuolation of the dental pulp cells and the odontoblasts was noted in the late experimental phases. Conclusion : This experiment suggests that dilatation of the capillaries in the dental pulp tissues is induced by diabetic state, and generalized degeneration of the dental pulp tissues is induced by irradiation of the diabetic group.
