Pharmacological and preventive properties of Petroselinum sativum seed extracts are well known, but the anticancer activity of alcoholic extracts and oil of Petroselinum sativum seeds on human breast cancer cells have not been explored so far. Therefore, the present study was designed to investigate the cytotoxic activities of these extracts against MCF-7 cells. Cells were exposed to 10 to $1000{\mu}g/ml$ of alcoholic seed extract (PSA) and seed oil (PSO) of Petroselinum sativum for 24 h. Post-treatment, percent cell viability was studied by 3-(4, 5-dimethylthiazol-2yl)-2, 5-biphenyl tetrazolium bromide (MTT) and neutral red uptake (NRU) assays, and cellular morphology by phase contrast inverted microscopy. The results showed that PSA and PSO significantly reduced cell viability, and altered the cellular morphology of MCF-7 cells in a concentration dependent manner. Concentrations of $50{\mu}g/ml$ and above of PSA and $100{\mu}g/ml$ and above of PSO were found to be cytotoxic in MCF-7 cells. Cell viability at 50, 100, 250, 500 and $1000{\mu}g/ml$ of PSA was recorded as 81%, 57%, 33%, 8% and 5%, respectively, whereas at 100, 250, 500, and $1000{\mu}g/ml$ of PSO values were 90%, 78%, 62%, and 8%, respectively by MTT assay. MCF-7 cells exposed to 250, 500 and $1000{\mu}g/ml$ of PSA and PSO lost their typical morphology and appeared smaller in size. The data revealed that the treatment with PSA and PSO of Petroselinum sativum induced cell death in MCF-7 cells.
Objective: This study was performed to assess prostate biomarkers with reference to body mass index and duration of prostate cancer. Materials and Methods: A hospital based retrospective study was undertaken using data retrieved from the register maintained in the Department of Biochemistry of Manipal Teaching Hospital, Pokhara, Nepal between $1^{st}$ January, 2009 and $28^{th}$ February, 2012. Biomarkers studied were prostate specific antigen (PSA), acid phosphatase (ACP) and prostatic acid phosphatase (PAP), alkaline phosphatase (ALP) and gamma glutamyl transpeptidase (${\gamma}GT$). Demographic data including age, duration of disease, body weight, height and body mass index (BMI) were also collected. Duration of disease was categorized into three groups: <1 year, 1-2years and >2 years. Similarly, BMI ($kg/m^2$) was categorized into three groups: <23 $kg/m^2$, 23-25 $kg/m^2$ and >25 $kg/m^2$. Descriptive statistics and testing of hypothesis were used for the analysis using EPI INFO and SPSS 16 software. Results: Out of 57 prostate cancers, serum level of PSA, ACP and PAP were increased above the cut-off point in 50 (87.5%), 30 (52.63%) and 40 (70.18%) respectively. Serum levels of PSA, ACP and PAP significantly declined with the duration of disease after diagnosis. We observed significant and inverse relation between PSA and BMI. Similar non-signficiant tendencies were apparent for ACP and PAP. Conclusions: Decreasing levels of prostate biomarkers were found with the duration of prostate cancer and with increased BMI. Out of prostate biomarkers, PSA was found to be significantly decreased with the duration of disease and BMI.
Objective : This study was undertaken to determine if Plantaginis Semen herbal acupuncture(PSA) has a protective effect on glycerol-induced acute renal failure in rats. Methods : Rats were dehydrated for 24hr and then injected with 4 ml/kg of 50% glycerol in each hindlimb muscle. In experiments for PSA effect, rats were received 0.1 ml of PSA extraction in both sides of corresponding $Sh\grave{e}nsh\bar{u}(BL_{23})$ of human body for 3 days after injection of glycerol. The experimental group were divided into the normal group, the control group, the sample 1,2,3 group. Results : Glycerol injection decreased glomerular filtration rate and increased urine volume, serum creatinine, BUN level and fractional excretion of $Na^+,\;K^+\;and\;Cl^-$. These results show that glycerol injection bring about acute renal failure. PSA significantly increased glomerular filtration rate and significantly decreased serum creatinine, BUN level and fractional excretion of $Na^+,\;and\;Cl^-$ as compared with control group. Conclusion : These results suggest that PSA can be used in prevention and treatment for acute renal failure. However, the precise mechanisms of PSA protection remain to be determined.
We demonstrate the possible application of the sandwich type surface-enhanced Raman scattering (SERS) immunoassay using antigen-antibody binding for detection of prostate-specific antigen (PSA) in cancer cells. In this sandwich type of SERS immunoassay, to capture antigens onto the immobilized layer of antibodies on the gold substrate we prepared the monolayer of gold nanoparticles on the APTMS-derivatized surface of a glass slide by using the SAM technique. This sandwich type of SERS immunoassay in which antigens on the substrate specifically capture antibodies on a Raman reporter (DSNB coated gold nanoparticles with R6G) could successfully detect PSA at low levels. A strong SERS spectrum of Raman reporter was observed only with a substrate in which PSA is present.
According to the results of Probabilistic Safety Assessment(PSA) for a Nuclear Power Plant(NPP), an Emergency Power Supply(EPS) system has been considered as one of the most important safety system. Especially, the interests in the reliability of the EPS system have been increased after the severe accidents of Fukushima Daiichi. Firstly, we performed the risk assessment and the importance analysis of the EPS system based on the PSA models of the reference plant, which is the Korean standard NPP type. Considering a portable Diesel Generator(DG) system as the reliability reinforcement of the EPS system, we modified the PSA models and performed the risk impact assessment and the importance analysis. Although the reliability of the potable DG could be about 20% of the reliability of the alternative AC DG, we identified that Core Damage Frequency(CDF) was decreased by at least 4.6%. In addition, the risk impacts due to the unavailability of the EPS system on CDF were decreased.
The PSA cycle was performed for the separation of binary gas mixture $H_2/Ar$ (80%/20%) using the six-step two-bed process. Adsorption equilibrium contains a LRC model for equilibrium adsorption isotherms and a LDF model for mass transfer. Aspen ADSIM, simulator was applied to predict the separation performance. The effect of cycle parameters such as feed rate, adsorption pressure and P/F ratio on the separation of hydrogen has been studied in experiment and simulation. In the results, maximize the recovery of hydrogen as a high purity was 13LPM feed flowrate, 120sec adsorption time, 11atm adsorption pressure and 0.1 P/F ratio in a cyclic steady-state come out since 10th cycle.
The Advanced Power Reactor Plus(APR+), which is a GEN III+ reactor based on the APR1400, is being developed in Korea. In order to enhance the safety of the APR+, a passive auxiliary feedwater system(PAFS) has been adopted in the APR+. The PAFS replaces the conventional active auxiliary feedwater system(AFWS) by introducing a natural driving force mechanism while maintaining the system function of cooling the primary side and removing the decay heat. As the PAFS completely replaces the conventional AFWS, it is required to verify the cooling capacity of PAFS for the core damage frequency(CDF) evaluation. For this reason, this paper discusses the cooling performance of the PAFS during transient accidents. The test case and scenarios were picked from the result of the sensitivity analysis in APR+ Probabilistic Safety Assessment(PSA). The analysis was performed by the best estimate thermal-hydraulic code, RELAP5/.MOD3.3. This study shows that the plant maintains the stable state without the core damages under the given test scenarios. The results of PSA considering this analysis' results shows that the CDF values are decreased. The analysis results can be used for more realistic and accurate performance of a PSA.
Fuel cell systems are being supplied to households and buildings to reduce greenhouse gases. The fuel cell systems have problems of high cost and slow startup due to fuel processors. Greenhouse gas reduction of the fuel cell systems is also limited by using natural gas. The problems can be solved by using a hydrogen generator consisting of a reformer and pressure swing adsorption (PSA). However, part load operation of the hydrogen generator is required depending on the hydrogen consumption. In this paper, PSA operation strategies are investigated for part load of the hydrogen generator. Adsorption and purge time were changed in the range of part load ratio between from 0.5 to 1.0. As adsorption time increased, hydrogen recovery increased from 29.09% to 48.34% at 0.5 of part load ratio. Hydrogen recovery and hydrogen purity were also improved by increasing adsorption and purge time. However, hydrogen recovery dramatically decreased to 35.01% at 0.5 of part load ratio.
To maintain safety and reliability of reactors, redundant sensors are usually used to measure critical variables and estimate their averaged time-dependency. Nonhealthy sensors can badly influence the estimation result of the process variable. Since online condition monitoring was introduced, the online cross-calibration method has been widely used to detect any anomaly of sensor readings among the redundant group. The cross-calibration method has four main averaging techniques: simple averaging, band averaging, weighted averaging, and parity space averaging (PSA). PSA is used to weigh redundant signals based on their error bounds and their band consistency. Using the consistency weighting factor (C), PSA assigns more weight to consistent signals that have shared bands, based on how many bands they share, and gives inconsistent signals of very low weight. In this article, three approaches are introduced for improving the PSA technique: the first is to add another consistency factor, so called trend consistency (TC), to include a consideration of the preserving of any characteristic edge that reflects the behavior of equipment/component measured by the process parameter; the second approach proposes replacing the error bound/accuracy based weighting factor ($W^a$) with a weighting factor based on the Euclidean distance ($W^d$), and the third approach proposes applying $W^d$, TC, and C, all together. Cold neutron source data sets of four redundant hydrogen pressure transmitters from a research reactor were used to perform the validation and verification. Results showed that the second and third modified approaches lead to reasonable improvement of the PSA technique. All approaches implemented in this study were similar in that they have the capability to (1) identify and isolate a drifted sensor that should undergo calibration, (2) identify a faulty sensor/s due to long and continuous missing data range, and (3) identify a healthy sensor.
Following the accident at Fukushima, the true impact of multi-unit accidents came to light. Accordingly, research related to multi-unit accident effect analysis, risk evaluation, and accident prevention/prevention technology has been conducted. Specific examples are mobile/fixed equipment such as multi-barrier accident coping strategy (MACST) and diverse and flexible coping strategies (FLEX), which have been introduced and installed in multi-units for preventing and mitigating multi-unit accidents. These strategies are useful for enhancing the safety of nuclear power plants (NPPs); however, a more efficient strategy is required in terms of the costs of physical and human resources. To effectively and efficiently mitigate an increase in multi-unit accidents, it is necessary to not only to utilize mobile/fixed equipment but to also use crosstie options with resources that already exist at NPPs. Therefore, we analyzed the current international and domestic status of crosstie systems technology and propose a method to evaluate feasibility alongside risk based on a multi-unit probabilistic safety assessment (PSA). To analyze the international and domestic status of crosstie systems technology, actual cases and related research were studied, and a list of potential crosstie safety resources was derived. Additionally, a case study was performed on crosstie cases of two systems within the assumed six units on-site under a multi-unit accident, and a multi-unit PSA-based risk evaluation method is proposed.
