Ultrastructural Study on Induced Resistance of Cucumber Plants against Sphaerotheca fuliginea by Oligochitosan

  • The induced resistance of cucumber leaves treated with oligochitosan to the infection of the cucumber powdery mildew, Sphaerotheca fuliginea, was investigated using transmission electron microscopy. The results showed that when the plants were treated with oligochitosan and challenged with inoculum, a significant decrease of the disease occurred. The mycelial development in the treated leaves was markedly inhibited. The cytoplasm of the powdery mildew mycelium was aggregated, with its organelles disintegrated and the cytoplasm collapsed. The protoplasm in haustoria became electron-dense. Haustoria became malformed, their organelles disintegrated, the hausterial wall thickened and eventually the whole complex necrotized. The host cells produced defence structures and materials associated with infection and a hypersensitive response. The host cell wall was thickened and deeply stained; several layers of papilla structure were produced under the cell wall; dark materials were deposited between the cell wall and plasmalemma; extrahaustorial plasmalemma was deeply stained and extrahaustorial matrix appositions had large deposits of electron-dense material; the cytoplasm was disordered, host organelles disintegrated and eventually the whole host cell disintegrated and necrotized.

Histological Observation of Embryogenic and Non-embryogenic Callus in Long-term Subculture of Wild Viola (Viola patrinii DC.) (흰제비꽃 배양세포에 있어서 분화세포와 미분화세포 조직의 비교 관찰)

  • To obtain a basic information of the development of Genus Viola, morphological and histological observation of in vitro calli and cells in Viola culture cells were investigated. There were two callus types obtained by long term subculture of wild viola (Viola partrinii DC. ) petiole callus. One was friable callus - soft and pale green in color and small cells in size, and the other was compact callus - compact and deep bluish green in color, large cells in size. In scanning electron microscopic observation, friable callus was composed of voculated cell around small. cell clump, while compact callus was composed of cells filled with protoplasm Somatic embryogenesis was observed from suspension culture of the compact callus.

  • A mushroom as a functional food material is being increasingly used in these days. It has been that about 1,500 species of mushrooms grow naturally in Asia. Among them about 700 species are edible, but about 30-50 species are poisonous. Even a poisonous mushroom is a very interesting subject for the dietary life research from biochemical viewpoint due to the fact that any mushroom can be the physiological active material. Not a few identified poisonous materials are being used as research reagents thus far. It is suggested that more useful materials could be obtained by progresses in future researches. The identification of the poisonous ingredient can open up a route for mushroom intoxication cure. It is known that cell destruction and troubles on skin, liver, and kidney troubles can be occurred from mushroom intoxication. Recently the author reported as a collection of treatises regarding anti-tumor substances of mushroom (2005), antibiotic substances of mushroom (2006), living-body functional and control substance (2007), and medicinal mushroom commonly used in Korean herbal medicines (2008). In this sequel, published research data on protoplasm, nerve, stomach and intestines, malodor, and acute pain poison of mushroom are collected and reported.

Morphological Changes of Mouse Ovary by X-Ray Irradiation (방사선 조사선량에 따른 생쥐 난소의 형태학적 변화)

  • This research was performed to investigate the morphological changes of folliculus ovary according to the radiation dose. The whole body radiation of 200 cGy, 400 cGy, and 600 cGy was given to the each groups of 5 months-aged female mouse. Various staining methods used in this research are: Hematosylin-Eosin method, and immunohistochemistrical methods using BrdU, TUNEL, p53, p21, PCNA and inhibin. The minute structural changes of folliculus ovary were observed through an electron microscope with high magnification. The morphological changes of growing folliculus ovary became distinct as the dose of X-rays increased. Especially, the nuclei of granular cells showed manifest condensation and the changes of the transparent zone were distinct. As a result of histochemical reaction according to Masson's trichrome method and reticular fiber method, the changed granular cells, the deformed basilar membrane of folliculus ovary and the abnormal arrangement of the reticular fiber were observed. In the reaction of BrdU, the granular cells of normal folliculus ovary with positive reaction rapidly decreased according to the increase of the dose of X-rays. In TUNEL study, granular cells showing positive reaction in retarded folliculus ovary were expanded to growing folliculus ovary and primordial folliculus ovary according to the increase of the dose of X-rays. In case of 600 cGy of X-rays, oocyte underwent apoptosis. In p53 immunohistochemistry, p53 manifested to be stronger as the dose of X-rays increased. p53 reactivity was manifested distinctively in all cells comprising folliculus ovary following irradiation of 600 cGy. p21 was manifested in granular cells of folliculus ovary and showed very positive reaction around follicular antrum according to the increase of the dose of X-rays. In PCNA, positive reaction was manifested in growing folliculus ovary, mature folliculus ovary and primordial folliculus ovary, but the extent of the reaction decreased as the dose of the X-rays decreased. The finding that the reaction of granular cells around folliculus ovary was stronger than that near follicular membrane indicates that what was damaged first by X-ray was the cells near folliculus ovary and follicular antrum. The reactivity of $inhibin-{\alpha}$ showed difference according to the growing stage of folliculus ovary: $inhibin-{\alpha}$ showed the most strong reaction in mature folliculus ovary with follicular antrum. There was strong reaction in granular cells around follicular membrane but $inhibin-{\alpha}$ did not occur at all in theca cells comprising follicular membrane. $Inhibin-{\alpha}$ in ovary tissue exposed to 400 cGy of X-rays was manifested more strongly than in ovary tissue exposed to 600 cGy of X-rays, which was related to the phenomenon that granular cells of mature folliculus ovary underwent necrosis or apoptosis increasingly due to X-rays. In an electron microscope with high magnification, nuclei and protoplasm of granular cells in growing folliculus ovary abruptly underwent minute structural changes according to the increase of dose of X-rays. Cell residue, by-product of cell decease, neutrophil and macrophage around follicular antrum were observed. The minute structural changes in granular cells showed typical characteristics of apoptosis: the increase of electronic density due to nuclear condensation, fragmentation of nuclei and atrophy of protoplasm. Necrosis of cells was identified but it was not so remarkable. Macrophage with apoptotic bodies was scattered. Proportional to the radiation dose, we found that the generation of heterogeneous substance of normal ovary texture's follicular fluid, the emergence of dyeing characteristic in the basilar membrane of folicle, the generation of apoptosis, and the transformation of macrophages, etc. From this results, we can infer the possible radiation hazard on the ovary of cervix cancer patient with radiation therapy.

Ultrastructural antigenic localization in Paragonimus iloktsuenesis during developmental stage by immunogold labeling method (면역황금표지법에 의한 일록춘폐흡충의 발육단계별 항원성부위)

  • Antigenic localization in Parofonimn iloktsuenensis worm tissues (tegument, intestine and vitelline gland) in different developmental stages of 2 weeks, 3 weeks, 4 weeks, 5 weeks and 33 weeks from albino rats (Sprague-Dawley) infected with P iloktsuenensis was observed by electron microscopy. These worm tissues of different developmental stage of R iloktsuenensis was observed on electromicrograph by immunogold labeling method using R iLoktsuenensis infected rat serum of 10 weeks. Antigenic localization was demonstrated as labeling of gold particles in tissues on electronmicrograph. In tegument, gold particles were labeled on tegumental tissue, generally more numerous on secretory granules in tegumental syncytium 2 weeks than those on the other elder developmental stages, but there was a little variation in antigenicity according to individual worm tissue. In general, antigenicity in tegumental tissue was not strong (gold particles: 0.1-5/1 Mm2). In intestine, a large number of gold particles (15-18/1 Mm2) were labeled in intestinal epithelium. Gold particles were concentrated especially on secretory granules in cytoplasm, and gold particles were labeled not only in cytoplasmic protrusions, but also in intestinal luminal contents. Intencity of labeling of gold particles was not correlated with developmental stage of worms. In vitelline gland, a large number of gold particles were labeled on vitelline globules. The gold particles in vitelline globules (8- 11/1 Mm2) were concentrated in protoplasm among segmental globules . Seasonal Variation of Picoplankton Community in Lake Juam (주암호에서 미세조류의 계절적 군집 변화)

  • The purpose of this study is to investigate the seasonal variation of picoplankton community in Lake Juam depending on the change of physico-chemical factors such as rainfall, water depth, DO and pH. The concentration of chlorophyll-a was most high as 18.03 mg/$m^3$ in July when the rainfall and water temperature were highest. The concentration was gradually decreased in October, April and that of January was decreased most low as 1.86 mg/$m^3$. The highest concentration of the Chl-a was shown at 2 and 5 m of water depth than surface, and the concentration was gradually decreased when the water depth becomes deep. Overall, microplankton was the highest rate as 33.9~54.2%, nanoplankton was 24.3~30.5% and picoplankton was 21.6~41.2%. Picoplankton was included as considerable concentration in the water of Juam lake. Therefore it is necessary to remove thoroughly the picoplankton in the water treatment processes such coagulation·sedimentation and sand filtration. The protoplasm released from destruction of picoplankton by chlorine has high possibility to cause regrowth of bacteria and pathogenic microorganism in the distribution system by playing the role of the assimilable organic carbon.

Morphology of Ectomycorrhizae of Pinus rigida × P. taeda Seedlings Inoculated with Pisolthus tinctorius, Rhizopogon spp. and Suillus pictus (Pisolthus tinctorius, Rhizopogon spp. 및 Suillus pictus균(菌)의 접종에 의한 리기테다소나무 묘목에 형성된 외생균근의 형태)

  • The objectives of this study were to isolate ectomycorrhizal fungi associated with roots of Pinus rigida ${\times}$ P. taeda trees planted in Chonbuk and Daejon locations and to identify them, and to observe the morphology of mycorrhizae formed by the isolated fungi. Pinus rigida ${\times}$ P. taeda seedlings formed ectomycorrhizae with Rhizopogon rubescens, Rhizopogon sp. and Suillus pictus isolated from the roots of mature Pinus rigida ${\times}$ P. taeda trees, Pinus rigida ${\times}$ P. taeda seedlings formed ectomycorrhizae with Pisolithus tinetorius too. The root surface of inoculated seedlings was covered with mycelia, and Hartig net were well developed in those roots. The size of vacuole was small, and the protoplasm was planty in ectomycorrhizal cell, compared to nonmycorrhizal cell. But the size of nuclei are not changed even in ectomycorrhizal cell.

Reproductive Cycle of Small Filefish, Rudarius ercodes (그물코쥐치, Rudarius ercodes의 생식주기)

  • The reproductive cycle of the small filefish, Rudarius ercodes was investigated based on the annual variations of gonadosomatic index(GSI) and hepatosomatic index(HSI) by electronic and photic microscophy. The specimens used were collected at the coastal area of Benden island, Sizuokagen, Japan, from September 1982 to August 1983. GSI began to increase from March, starting season of longer daylength and higher water temperature, and reached the maximum value between June and August. It began to decrease from September with the lowest value appearing between November and February without any evident variation. The annual variations of HSI were not distinct in male filefish and were negatively related to GSI in female : HSI decreased in the summer season when the ovary was getting mature and reached the maximum in the winter season when the ovary was getting retrogressive. The ovary consisted of a pair of saccular structure with numerous ovarian sacs branched toward the median cavity. Oogonia divided and proliferated along the germinal epithelium of the ovarian sac. Young oocytes with basophile cytoplasm showed several scattering nucleoli along the nuclear membrane. when the oocytes growing to about 300 ${\mu}m$, nuclear membrane to disappear with nucleus migrating toward the animal pole. The regions of protoplasm were extremely confined within the animal hemisphere in which most of cytoplasms were filled with yolk materials and oil drops. After ovulation, residual follicles and growing oocytes remaining in the ovarian sacs degenerated. But perinucleatic young oocytes without follicles formed were not degenerated, and growing continuously still in the next year. Mitochondria and endoplasmic reticula in the cytoplasm remarkably increased with oocytes maturing and yolk accumulating. Those were considered to be functionally related to the yolk accumulation. Five or six layers of possible vitellogenin, oval-shaped disc structures with high electron density, appeared in the apex of follicular processes stretching to the microvilli pits of mature oocytes. Testis consisting of a pair of lobular structures in the right and left were united in the posterior seminal vesicle, Cortex of testis was composed of several seminiferous tubules, and medulla consisting of many sperm ducts connected with tubules. Steroid hormone-secreting cells with numerous endoplasmic reticula and large mitochondria of well developed cristae were recognized in the interstitial cells of the growing testis. Axial filament of spermatozoon invaginated deeply in the central cavity of the nucleus and the head formed U-shape with acrosome severely lacking, mitochondria formed large globular paranuclei at the posterior head, and microtubular axoneme of the tail represented 9+9+2 type. The annual reproductive cycles could be divided into five successive stages : growth(March to July), maturation(May to September), Spawning(mid May to early October) and resting stages(October to February). The spawning peak occurred from June to August.
