Anti-Inflammatory Effect of Hexane Fraction from Eisenia bicyclis on Lipopolysaccharides-Treated RAW 264.7 Cells (LPS로 유도된 RAW 264.7 대식세포에 대한 대황(Eisenia bicyclis) 헥산 분획물의 항염증 효과)

  • Eisenia bicyclis is known to have secondary metabolites exhibiting various biological activities. In a preliminary study, the n-hexane fraction obtained from the ethanolic extract of E. bicyclis showed higher anti-inflammatory activity than the ethyl acetate and butyl alcohol fractions based on the inhibition of lipopolysaccharide (LPS)-stimulated nitric oxide (NO) production in RAW 264.7 cells. Using this fraction (E. bicyclis hexane fraction, EHF), we investigated the molecular mechanisms underlying its anti-inflammatory effect in LPS-stimulated RAW 264.7 cells. Pretreatment of the cells with up to 50 ㎍/mL EHF significantly inhibited NO and prostaglandin E2 production as well as their responsible enzyme proteins and mRNAs, in a dose-dependent manner (P<0.05). Similarly, EHF markedly reduced the production of pro-inflammatory cytokines, such as interleukin (IL)-1β, IL-6 and tumor necrosis factor (TNF)-α as well as their mRNA levels. Nuclear translocation of nuclear factor-kappa B (NF-κB) was strongly suppressed by EHF treatment. EHF significantly reduced the phosphorylation of mitogen-activated protein kinases and phosphatidylinositol 3-kinase/Akt in LPS-stimulated cells. Moreover, EHF reduced ear edema in phorbol myristate acetate (PMA)-induced mice. These results indicate that EHF contains potent anti-inflammatory compounds, which may be used as a dietary supplement for the prevention of inflammatory diseases.

Use of Weight-control or Health Functional Foods, Associated Weight-control Behavior and Perception among University Students in Cheongju (청주지역 일부 대학생의 체중조절 식품 및 건강기능식품 사용 실태와 관련 인식 연구)

  • Objectives: This study sought to assess the prevalence and duration of weight-control or health functional food use, associated weight-control behavior, perception, and knowledge among university students. Methods: The subjects were 442 university students in Cheonju, Korea, and data was collected by a self-administered questionnaire. Both χ2 and t-tests were conducted for categorical and mean comparisons. Results: An estimated 62.7% (female 69.0%, male 54.8%) had ever attempted weight-control. Among these, an estimated 59.2% of females and 38.9% of males had used weight-control or health functional foods with significant gender difference. The weight-control foods commonly used included chicken breast, protein powder, low-fat milk and soymilk, and meal replacement drinks, while garcinia cambogia extract and green tea extract were frequently used as health functional foods. One of 10 (10.3%) consumers of weight-control foods reported ≥ 7 months use, with less frequent long-term consumption of health functional foods (4.2%). The average degree of satisfaction was 3.24 ± 0.92 for weight-control foods and 2.72 ± 0.97 for health functional foods on a 5-point scale, meaning 'slightly satisfied' and 'slightly dissatisfied', respectively. Females or students with an experience of weight-control reported poorer perceptions of their health and body image as well as a higher need for weight control. Besides, both male and female subjects felt a high need for correct information regarding weight-control methods. Conclusions: Our results provide a better understanding of the characteristics associated with the use of weight-control or health functional foods among university students and will be useful in developing a nutrition education program by incorporating correct body image, knowledge, and practical yet desirable practices for weight control.

Characteristics of fermented milk containing Bifidobacterium growth promoter (BE0623) and dietary fiber

  • This study was carried out to investigate the effects of Bifidobacteria growth promoter BE0623 and a dietary fiber supplement, which included Bifidobacterium lactis BB12, Lactobacillus acidophilus, Streptococcus thermophilus, and Bifidobacterium lactis. In fermented milk containing BE0623, the viable cell count of Bifidobacteria significantly increased by about 45 to 75 times compared to the control, and the titratable acidity increased, whereas the pH decreased. All fractions obtained by isolating BE0623 had Bifidobacteria growth effect. Acacia dietary fiber is a pale yellow powder. It has a viscosity of 60 to 100 cPs and a pH between 4.1 and 5.0. Its general components are less than 10% moisture, more than 90% dietary fiber, and less than 4% ash. The optimal addition ratio of Bifidobacteria growth promoting material was determined to be 0.05%. The general components of the manufactured fermented milk were carbohydrate 17.85%, protein 3.63%, fat 3.00%, and dietary fiber 2.95%. During storage of the fermented milk for 24 days, its titratable acidity, viscosity, and sugar content all met the criteria. In addition, the viable cell counts of Bifidobacteria and lactic acid bacteria in the fermented milk were 1.7 × 108 CFU·mL-1 and 1.5 × 107 CFU·mL-1, respectively, and Escherichia coli was negative. There was no significant difference between the control group and the treatment group in the sensory evaluation of sweet, sour, weight, and flavor, and the preference for the treatment group was excellent. The acceptability of the fermented milk of the treated group according to the storage period was excellent in terms of color, flavor, and appearance.

The Daily Dose and Uses of Pinellia Tuber, Vinegar and Eggshell of Goju-tang in Treatise on Cold Damage Diseases (《상한론(傷寒論)》고주탕(苦酒湯)의 반하(半夏)와 고주(苦酒), 계자각(鷄子殼) 용량과 용도)

  • Objectives : The purpose of this study was to clarify the daily dose and uses of Goju-tang in Treatise on Cold Damage Diseases. Methods : We compared major editions of Treatise on Cold Damage Diseases with the original text of Supplement to the Essential Prescriptions Worth a Thousand Gold and chose the most appropriate text. If none of the existing texts seemed reasonable, We compared texts item by item. When there is nothing adequate even item by item, it was newly modified. These results were compared with currently distributed Pinellia Tuber and standards of eggs. Results : Goju-tang consisted of Goju(vinegar), Pinellia Tuber and an eggshell. Generally, a daily dose was extracted at once and taken 3 times a day, but in Goju-tang, 4 pieces of Pinellia Tuber and one-third Sheong of Goju(solvent) were used, which were one third of average volume. Pinellia Tuber was used cut in small pieces. And it was boiled gradually by low heat. Goju was mainly to reduce the pungency of Pinellia Tuber by cutting raphides of calcium oxalate. The Pot preparing medicines was eggshell which was chosen by its small size, because the volume of Pinellia Tuber and Goju was one-third of average volume. Since an egg shell is consisted of protein, it didn't react with vinegar(Goju). Conclusions : The 4 pieces of Pinellia Tuber and one-third Sheong of Goju(Vinegar) in the Goju-tang were one-third of average dose, and the eggshell was Pot preparing medicines.

Evaluation of standardized ileal digestibility of amino acids in fermented soybean meal for nursery pigs using direct and difference procedures

  • Objective: This study was to evaluate standardized ileal digestibility (SID) of amino acids (AA) in fermented soybean meal (FSBM) for nursery pigs using both direct procedure and difference procedure when FSBM was added at 20% in diets. Methods: Forty-eight pigs at 9.2±0.9 kg body weight (BW) were individually housed and allotted to 4 treatments. Treatments included NFD (a semi-purified N free diet), FSD (a diet with 20% FSBM), CBD (corn basal diet), and CFD (corn basal diet:FSBM at 80:20). The FSD was used to measure AA digestibility in FSBM using the direct procedure, whereas CBD and CFD were used in the difference procedure. Pigs were fed for 10 days (0.09×BW0.75 kg per day) and euthanized to collect ileal digesta for TiO2 and AA. Results: Total endogenous AA loss was 12.1 g/kg of dry matter intake. The apparent ileal digestibility (AID) Thr was greater (p<0.05) and AID His (p = 0.073) and Leu (p = 0.052) tended to be greater using the direct procedure compared with the difference procedure. The SID Thr were greater (p<0.05) in FSBM for nursery pigs calculated using a direct procedure compared with a difference procedure. In addition, SID Lys in FSBM was about 83% to 88% for nursery pigs higher than SID Lys described in National Research Council (2012). Conclusion: The SID of AA in FSBM when included at practical levels using the direct procedure were similar to those from the difference procedure. Considering the SID of AA obtained using both direct and difference procedures, FSBM is an effective protein supplement providing highly digestible AA to nursery pigs. The SID of AA from this study was considerably higher than those previous reported. This study also indicates the importance of including the test feedstuffs at practical levels when evaluating digestibility.

Neuroprotective effect of Korean Red Ginseng against single prolonged stress-induced memory impairments and inflammation in the rat brain associated with BDNF expression

  • Background: Post-traumatic stress disorder (PTSD) is a psychiatric disease that develops following exposure to a traumatic event and is a stress-associated mental disorder characterized by an imbalance of neuroinflammation. Korean Red Ginseng (KRG) is the herbal supplement that is known to be involved in a variety of pharmacological activities. We aimed to investigate the effects of KRG on neuroinflammation as a potential mechanism involved in single prolonged stress (SPS) that negatively influences memory formation and consolidation and leads to cognitive and spatial impairment by regulating BDNF signaling, synaptic proteins, and the activation of NF-κB. Methods: We analyzed the cognitive and spatial memory, and inflammatory cytokine levels during the SPS procedure. SPS model rats were injected intraperitoneally with 20, 50, or 100 mg/kg/day KRG for 14 days. Results: KRG administration significantly attenuated the cognitive and spatial memory deficits, as well as the inflammatory reaction in the hippocampus associated with activation of NF-κB in the hippocampus induced by SPS. Moreover, the effects of KRG were equivalent to those exerted by paroxetine. In addition, KRG improved the expression of BDNF mRNA and the synaptic protein PSD-95 in the hippocampus. Taken together, these findings demonstrate that KRG exerts memory-improving actions by regulating anti-inflammatory activities and the NF-κB and neurotrophic pathway. Conclusion: Our findings suggest that KRG is a potential functional ingredient for protecting against memory deficits in mental diseases, such as PTSD.

Lactobacillus rhamnosus JY02 Ameliorates Sarcopenia by Anti-Atrophic Effects in a Dexamethasone-Induced Cellular and Murine Model

  • Sarcopenia is defined as loss of muscle mass and strength due to aging. Recent studies show that sarcopenia may improve via the gut-muscle axis, suggesting that gut health may affect muscle phenotypes. In this study, we aimed to investigate the ability of Lactobacillus rhamnosus JY02 as a probiotic strain isolated from kimchi to alleviate sarcopenia. L. rhamnosus JY02-conditioned medium (CM) reduced dexamethasone (DEX)-induced myotube diameter atrophy and expression of muscle degradation markers (MuRF1 and atrogin-1) in C2C12 cells. The amelioration of sarcopenia was investigated by measuring body composition (lean mass), hand grip strength, myofibril size (using histological analysis), and mRNA and protein expression of muscle-related factors in a DEX-induced mouse model. The results of these analyses showed that L. rhamnosus JY02 supplementation promoted the production of muscle-enhancement markers (MHC Iβ, MHC IIα, and Myo-D) and reduced both the production of muscle degradation markers and the symptoms of muscle atrophy (loss of lean mass and muscle strength). We also found decreased levels of pro-inflammatory cytokines (IL-6, IFN- γ) and increased levels of anti-inflammatory cytokines (IL-10) in the serum of DEX+JY02-administered mice compared to those in DEX-treated mice. Overall, these results suggest that L. rhamnosus JY02 is a potent probiotic supplement that prevents sarcopenia by suppressing muscle atrophy.

In Vitro α-Amylase, α-Glucosidase, Pancreatic Lipase, Xanthine Oxidase Inhibiting Activity of Agaricus bisporus Extracts

  • In this study, the α-amylase inhibitory activity, α-glucosidase inhibitory activity, pancreatic lipase inhibitory activity, and Xanthine Oxidase inhibitory activity of the fruiting body extracts of 5 varieties of Agaricus bisporus (AB) were confirmed. First, the α-amylase inhibitory activity of AB12, AB13, AB18, AB34, and AB40 methanol extracts was lower than that of acarbose, a positive control, in all concentration ranges. The α-glucosidase inhibitory activity of the AB40, AB13, and AB12 methanol extracts at the extract concentration of 1.0 mg/mL was 80.5%, 81.3%, and 78.5%, respectively, similar to that of acarbose, a positive control. The pancreatic lipase inhibitory activity of the methanol extract of Agaricus bisporus fruiting body was significantly lower than that of the positive control orlistat in the concentration range of 50~1.000 (mg/mL). The Xanthine Oxidase inhibitory activity was 0.5~8.0 mg/mL of each extract, which was significantly lower than that of the positive control allopurinol in the same concentration range. However, the Xanthine Oxidase inhibitory activity of AB13 and AB40 at 8.0 mg/mL was about 70%, which was higher than that of other mushrooms. In conclusion, five kinds of Agaricus bisporus fruiting bodies seem to have inhibitory effects on enzymes such as α-amylase, α-glucosidase, pancreatic lipase, and Xanthine Oxidase that degrade starch and protein. In particular, it has an inhibitory effect and a reduction effect on xanthine oxidase that causes gout, so it is expected that it can be developed and used as a food or health supplement with health functional properties through future research.

Comparison of the metabolic profile of the mycelia and fruiting bodies of artificially cultured Cordyceps militaris

  • Cordyceps militaris, a well-known traditional Chinese medicine, has multiple health-promoting effects. It is used as a herbal remedy and health food in Asian countries. Cultured mycelia are often used as a substitute for natural C. militaris. In the present study, the mycelia and fruiting bodies of artificially cultured C. militaris were analyzed using a metabolomics approach. The protein and crude fat contents of the mycelia were substantially higher than those of the fruiting bodies. The top three abundant amino acids in the mycelia were proline (3.9 g/100 g), aspartic acid (2.9 g/100 g), and glutamic acid (2.7 g/100 g). The carbohydrate content was similar in the fruiting bodies and mycelia. Analysis revealed that both the fruiting bodies and mycelia are rich in phenolic compounds and exhibit antioxidant activity. Further, six metabolites were significantly different between the mycelia and fruiting bodies. The levels of Ca, glucose, Mg, and Se were higher in the mycelia than in the fruiting bodies. In contrast, mannitol and Zn were more abundant in the fruiting bodies. The current study provides a comprehensive metabolic profile of the mycelia and fruiting bodies of artificially cultured C. militaris. Such an exercise is potentially important for understanding the metabolism of C. militaris and facilitating the use of cultured mycelia as a supplement to C. militaris fruiting bodies in traditional Chinese medicine.

Effect of temporary weaning and creep feeding on calf growth and the reproductive efficiency of their Hereford dams

  • Objective: The objective was to test if creep feeding (CF) improves the average daily gain (ADG) and weaning weight of calves submitted to temporary weaning (TW) and if the combination of CF and TW improves conception and pregnancy rates of cows. Methods: Primiparous (n = 74) and primiparous and multiparous (n = 104) cows grazing native grasslands were used in experiment 1 and 2; respectively. The experimental design was in plots divided into complete random blocks with two replications. The CF was the big plot and TW the small plot, thus four experimental groups were formed: i) -CF-TW (n = 21 and 27); ii) -CF+TW (n = 16 and 24); iii) +CF-TW (n = 20 and 26); iv) +CF+TW (n = 17 and 27) with cow-calf pairs for experiments 1 and 2; respectively. Nose plate application for TW had a duration of 14 and 15 days for experiment 1 and 2: respectively. In experiment 1, calves were fed at 1% of live weight for 112 days using a commercial supplement with 18.4% crude protein. In experiment 2, the supplementation lasted 98 days, and was carried out with corn dried distillers grains with soluble (DDGS) at 40% of the potential intake on a daily basis. Results: The TW reduced ADG during the TW period and the following 14 days, but the negative effect of TW was maintained until the final weaning only in experiment 2. The CF increased ADG during TW period in both experiments. The TW promoted an earlier conception of the dams (12 days in -CF treatment and 19 days in +CF treatment, p<0.01) and CF increased pregnancy rate in experiment 1, being the effects not consistent between experiments. Conclusion: The CF consistently promoted an increase in ADG during the period of TW and increased final weaning weight of calves, therefore it is economically viable.
