Bronchoalveolar lavage (BAL) is a useful tool in researches and in clinical medicine of lung diseases because the BAL fluid contains biochemical and cytological indicators of the cellular response to infection, drugs, or toxicants. However, the variability among laboratories regarding the technique and the processing of the BAL material limits clinical research. The aim of this study was to determine the suction frequency and lavage fraction number necessary to reduce the variability in lavage using male Sprague-Dawley rats. We compared the total cell number and protein level of each lavage fraction and concluded that more cells and protein can be obtained by repetitive lavage with a suction frequency of 2 or 3 than by lavage with a single suction. On the basis of total cell recovery, approximately 70% of cells were obtained from fractions 1~3. The first lavage fraction should be used for evaluation of protein concentration because fractions 2~5 of lavage fluid were diluted in manifolds. These observations were confirmed in bleomycin-induced inflamed lungs of rats. We further compared the BAL data from the whole lobes with data from the right lobes and concluded that BAL data of the right lobes represented data of the whole lobes. However, this conclusion can only be applied to general lung diseases. At the end, this study provides an insight into the technical or analytical problems of lavage study in vivo.
0.5 mg of natural ginsenoside mixture and 0.8 $\mu$Ci of synthesized 14C-ginsenosides were administered orally to a rat and killed at one hour after the ginsenoside administration and the liver was fractionated into nuclear fraction, mitrochondria microsomes and cytosol fraction. Radioactivity distribu lion in subcellular fractions of the liver showed that 32o1c of total radioactivity absorbed in the liver was in cytosol fraction but a significant portion of the radioactivity was also found in mitochondria (26.6%) and microsomal fraction (18.l%). 5.8% of the total radioactivity was recovered from the nuclear fraction as well. This suggested that ginsenosides might be distributed into all subcellular fractions. Activities of mitochondrial aldehyde dehydrogenase, lactate dehydrogenase and malate dehydrogenase of the liver of rat at two hours after the ginsenoside administraion were found appreciably stimulated, suggesting that the ginsenoside concentration in the liver might be around 10-5%, since optimum concentrations for most enzyme catalyzed reactions in vitro were known to be 10-6% 10-4%. A significant portion of the radioactivity recovered from subcellular fractions of the liver was found in protein fractions, suggesting that proteins might interact with ginsenosides. Examination of protein-ginsenoside interation by gel filtration, equilibrium dialysis and amonium sulfate precipitation technique suggesting that proteins and ginsenosides do not bound covalently but weakl\ulcorner combined. When purified ginsenoside Rbl and Rgl were incubated with rat liver cytosolic enzymes for 20 min, the above ginsenosides were hydrolyzed quickly, suggesting that ginsenosides might be rapidly hydrolyzed and metabolized in the liver. It was also observed in vitro that the ginsenosides such as Rbl and Rgl were easily hydrolyzed by rat liver cytosol preparation suggesting that absorbed ginsenosides might be quickly hydrolyzed and metabolized in the liver.
Twelve dietary combinations were prepared using 70 parts of fermented wheat straw (FWS) as the sole roughage supplemented with 30 parts of either the low protein concentrate mixture (Conc.-I), high protein concentrate mixture (conc.-II), maize grains (M), solvent extracted mustard cake (DMC), deoiled rice bran (DRB), uromol bran mixture (UBM), deep stacked poultry litter (DSPL), dried poultry droppings (DPD), M-DMC mixture (50:50), M-UBM mixture (50:50), M-DPD mixture (50:50) or M-UBM-DPD mixture (50:25:25) and evaluated by in-sacco technique. The above dietary combinations were also evaluated by changing the roughage to concentrate ratio to 60:40. The digestion kinetics for DM and CP revealed that FWS:DPD had the highest, whereas, the FWS:M-DMC had the lowest rapidly soluble fraction. The potentially degradable fraction was found to be maximum in FWS:M and minimum in FWS:DPD dietary combinations. The higher degradation rate of FWS:DRB and FWS:UBM combinations was responsible for their significantly (p<0.05) higher effective degradability as compared to other combinations. The highest undegradable fraction noted in FWS:M-UBM-DPD followed by FWS:DMC was responsible for high rumen fill values. The FWS:DRB, FWS:UBM and FWS:DPD combinations had higher potential for DM intake. The dietary combination with higher concentrate level (60:40) was responsible for higher potentially degradable fraction, which was degraded at a faster rate resulting in significantly higher effective degradability as compared to the corresponding dietary combination with low concentrate level (70:30). The low undegradable fraction in the high concentrate diet was responsible for low rumen fill values, which predicted of high potential for DM intake. Out of 24 dietary combinations, FWS with either of UBM, DRB, DMC, Maize, M-DMC or DPD in 70:30 ratio supplemented with minerals and vitamin A in comparison to conventional feeding practice (roughage and concentrate mixture) could be exploited as complete feed for different categories of ruminants.
To investigate antitumor components of Korean higher fungi, the carpophores of Ramaria formosa $(Fr.)Qu\acute{e}l$. were collected in Gang Won Province and extracted with hot water. The extract was concentrated and precipitated by four volumes of ethanol. The precipitate was centrifugated and purified by dialyzing through Visking tube and a protein-bound polysaccharide fraction was obtained. The fraction was tested for antitumor activity against sarcoma 180 implanted in mice. The tumor inhibition ratio of the fraction was 66% in the dose of 50mg/kg/day for the period of ten days. The tumor in two of the eight mice was completely regressed. The components of the fraction were found to be a polysaccharide and a protein. The chemical analysis of the fraction showed that the polysaccharide moiety consisted of four monosaccharides and that the protein moiety contained fourteen amino acids.
Among proteins separated from methanol extract of follicular fluid with superose column, the components inducing sperm swim-up separation through sucrose layer were analysed with superose column in Smart system and SDS-PAGE. And the results obtained were as follows; The fractions of retention volume (RV) 0.83ml and RV 1.36ml separated with superose column should stimulate sperm migration and movement. However, RV 0.83 fraction was consisted of complex materials containing RV 1.36 component. RV 1.36 fraction contained a BSA analogue of 67 kilodaltons (Kd) and showed identical peak pattern with BSA fraction V. In conclusion, the protein of 67 Kd in follicular fluid should stimulate sperm migration and movement.

Addition of AAF to murine splenocytes culture produced a dose-related suppression of lymphoproliferative response to lipopolysaccharide (LPS). The time course of the suppression showed that a significant inhibition was occured after a 18 hr AAF treatment. Total protein kinase C activity in splenocytes was decreased to 72% of control level after a 18 hr AAF treatment. Phosphorylation of a PKC specific 80 kDa protein was increased by LPS and AAF down-regulated LPS-induced PKC activity. LPS-induced phosphorylation of overall proteins in membrane and cytosolic fraction were also decreased by the treatment of AAF. A significant increase of PKC activity in membrane fraction was noticed within 10 min of AAF treatment compared to LPS alone and then gradually decreased to LPS level in 60 min. Meanwhile, PKC activity in cytosolic fraction was increased slightly in 10 min by the treatment of AAF and then decrease to 80% LPS level in 30 min. These results suggested that suppressive effect of AAF on LPS-induced lymphoproliferative response may be associated with the down-regulation of PKC and other susceptible kinases in spleen cells.

The main protein peak was observed in fraction No.9 and 109 when cellulase seperation was conducted by use of DEAE-Sephadex. The protein obtained from fraction No.9 has the characteristics of Cx component and that from fraction No.109 characteristics similiar to $C_1$ component. The effective reaction condition of the ensyme used was $40^{\circ}C$ in temperature. pH 5.0 and 90 minutes in treatment time. For the case of $C_1$ pH 5.5 in temperature range of $30^{\circ}C 50^^{\circ}C$, 4.0 5.5 in pH, and over 30 minutes of treatment time, the reaction was in the range of 80% of the maximum. Affinity of enzyme increased as freeness, increased, and this effect was more visible in fiber than in fines.
Methanol extracts of Commelina communis, a Korean wild edible plant showing hypoglycemic effect on diebetic rats were fractionated systematically by hexane, chloroform, butanol, and water, and these four fractions were administered orally to streptozotocin-induced diabetic rats to evaluate the hypoglycemic effect. The butanol and water fractions of Commelina communis lowered blood glucose levels of diebetic rats at a dose of 500 mg/kg/day. Administration of each fraction except that of chloroform increased the plasma protein levels. Liver protein levels were significantly higher in the butanol- and water fraction-fed groups. The results suggest that the butanol fraction of Commelina communis has hypoglycemic effect on diabetic rats and the intake of Commelina communis may be beneficial for the management of diabetes mellitus.
Whole faba beans (WFB) were dry roasted at different temperatures (110, 130, $150^{\circ}C$) for 15, 30, 45 minutes to determine the optimal heating conditions of time and temperature to increase nutritional value. Ruminant degradation characteristics of crude protein (CP) of WFB were determined by the nylon bag incubation technique in dairy cows fed 60% hay and 40% concentrate. Measured characteristics of crude protein (CP) were soluble (washable) fraction (S), undegradable fraction (U), lag time (T0), potentially degradable fraction (D) and the rate of degradation (Kd) of insoluble but degradable fraction. Based on measured characteristics, percentage bypass crude protein (%BCP) and bypass crude protein (BCP in g/kg) were calculated. Degradability of CP was reduced by dry roasting (p < 0.01). S was reduced rapidly with increasing time and temperature, from 49.0% in the raw WFB (RWFB) to 26.3% in $150^{\circ}C/45$ min. D varied from 50.7% in RWFB to 73.7% in $150^{\circ}C/45^{\prime}$. U varied from 0% in $130^{\circ}C/45^{\prime}$, $150^{\circ}/30^{\prime}$ and $150^{\circ}/45^{\prime}$ to 0.66% in $110^{\circ}/45^{\prime}$ (0.24% for the RWFB). Lag time (T0) varied from 1.58 h in $130^{\circ}C/30^{\prime}$ to 2.40 h in $150^{\circ}C/45^{\prime}$ (1.87 h for RWFB). Kd varied from 24.2% in the $110^{\circ}C/30^{\prime}$ to 4.3% in $150^{\circ}C/45^{\prime}$ (21.4% for the RWFB). Kd was significantly reduced with time and temperature. All these effects resulted in increasing % BCP from 8.9% in the $110^{\circ}C/45^{\prime}$, 11.3% in the RWFB to 43.1% in the $150^{\circ}C/45$. Therefore BCP increased from 31.3 and 39.9 to 148.4 g/kg respectively. Both %BCP and BCP at $150^{\circ}C/45$ increased nearly 4 times over the raw faba beans. The effects of dry roasting temperature and time on %BCP and BCP seemed to be linear up to the highest values tested. Therefore no optimal dry roasting conditions of time and temperature could be determined at this stage. It was concluded that dry roasting was effective in shifting crude protein degradation from rumen to intestine to reduce unnecessary nitrogen (N) loss in the rumen. To determine the optimal treatment, the digestibility of each treatment should be measured in the next trial using mobile bags technique.

Mackerel water-soluble protein fraction produced by washing the mackerel meat were concentrated by isoelectric point shifting precipitation process, and the concentrates were utilized as the alternative feed of fish meal. In the 1st aquaculture diet experiment for Israel common carp, the feed conversion ratio decreased in proportion to the rise in the percentage of the recovered protein containing a residual lipid, which was added to the fish meal. It was supposed that the low feed efficiency was because of lipid oxidation in the recovered protein fraction. In addition, 2nd aquaculture diet experiment for Israel common carp was conducted after removing the oxidized lipid in the recovered protein fish meal. When a portion of the fish meal was substituted by the recovered protein devoid of the residual lipid, the feed conversion ratio increased in proportion to the amount of the substitute being added to the fish meal. Therefore, the recovered protein fraction of the mackerel washing wastewater from mackerel processing factory could be used as the alternative feed of fish meal.
