Effect of Testosterone Propionate and Estradiol -l7$\beta$ on the Biochemical Changes in the Fat Body and Haemolymph of the Bivoltine Silkworm Bombyx mori L.

  • Effect of topical application with 10, 20 and 30 ${\mu}g$/ml testosterone propionate and estradiol -17${\beta}$ on the fourth and fifth instar bivoltine NB18 silkworm larvae Bombyx mori, on the glycogen and protein contents of the Fat body and trehalose and protein contents of the haemolymph has been studied. Glycogen content of the fat body was significantly decreased in both testosterone propionate and estradiol -17${\beta}$ treatment groups except in the group treated with 30 ${\mu}g$ testosterone propionate where the increase was not significant when compared with those of carrier controls. The increase/decrease in haemolymph trehalose content did not show any significant difference in all the treated groups. Protein content of the fat body significantly increased in 10 and 20 mg testosterone propionate and estradiol -l7${\beta}$ treated groups but in 30 mg treated groups the increase was not significant when compared with those of carrier controls. There was no significant change in the haemolymph protein content in all the testosterone propionate and estradiol -17${\beta}$ treated groups except in group treated with 10 ${\mu}g$ estradiol -17${\beta}$ where it showed a significant decrease when compared with that of carrier control.

The Effect of Microwave Diathermy against the Body Composition of Obese Women (비만 여성들의 체성분에 대한 극초단파 심부투열치료의 효과)

  • Purpose: This study investigated the effect of microwave diathermy against the body composition of obese women with protein content, muscle quantity, and the change of body weight for three weeks by microwave diathermy. Methods: we performed the change of body weight for three weeks using the method of 100,000 Hz microwave diathermy against obese women. The obese women were examined the change of body weight by pre-test using microwave diathermy. Results: The effect of microwave diathermy in protein content, muscle quantity, and body weight did not show any statistically significant in an experimental group of eight female university students for three weeks, but the change of body weight was statistically significant (p>0.05) for three weeks to eight students who were examined by microwave diathermy. The control group did not show any statistically significant in protein content, muscle quantity, body fat, and the change of body weight for three weeks.s). Conclusion: These results suggest that microwave diathermy may be an effective method for the management of the body and the diet in obese women.

  • In this paper, we adopt a position specific scoring matrix as an abstraction of amino acid type to derive two new statistical potentials for protein structure prediction, and investigated its effect on the quality of the potentials compared to that derived using residue specific amino acid identity. For stringent test of the potential quality, we carried out folding simulations of 91 residue A chain of protein 2gpi, and found unexpectedly that the abstract amino acid type improved the quality of the one-body type statistical potential, but not for the two-body type statistical potential which describes long range interactions. This observation could be effectively used when one develops more accurate potentials for structure prediction, which are usually involved in merging various one-body and many-body potentials.

  • Due to the fourth-instar larvae of C. riparius have a sensitive to ecdysteroidal molting hormones for the life cycle developments, accordingly the emerged adult affected corresponding to larval phase's environments. The emerged female from larval phase exposure to DEHP observed a fact body and clumsy fling behavior in females. The body volume of treated female groups was clearly larger than that of control fe- males. In the 2D/E gel 1108 protein spots were identified. The visualized protein spots allowed extraction of 27 protein spots differed more than 3 fold in DEHP treated animals, which was approximately 2.4% of the total protein spots. In this view, the body volume (or morphological characters) was well observed and detected faster than physiological detection for various EDCs. In this study, the body volume as a detecting po-int for EDCs suggested a bio-marker in individual levels.

  • A large portion of total energy expenditure associated with ruminant livestock production goes towards maintenance. Approximately 55% of whole body energy use is consumed by visceral tissues (including internal organs) with the majority of this going to the liver and gastrointestinal tract. Muscle and adipose tissues consume about 27% of total body energy expenditure. Metabolic components within the viscera responsible for the majority of energy consumption include ion transport, protein turnover, substrate cycling, and urea synthesis (liver). Within muscle tissue of growing animals ion transport and protein turnover account for most of the energy expenditure. Protein synthesis consumes approximately 23% of whole body energy use and visceral tissues account for proportionally more of whole body protein synthesis than skeletal muscle. Research efforts focused on improving energetic efficiency of the tissues and metabolic mechanisms responsible for the majority of whole animal energy expenditure should provide information leading to more efficient production of an edible product.

The Effect of Protein and Carbohydrate on the Recovery Rate of Restricted Rats (탄수화물 및 단백질이 식이 제한을 받은 흰쥐의 회복에 미치는 영향)

  • This study was designed to compare the effect of protein with that of calorie, both of which were supplemented by separate feeding, on the recovery rate and metabolic change of undernourished rats. During the two weeks of food restriction, the weight of body and some major internal organs was reduced, compared with normal growing rats, but the extent of reduction was various. After that, recovery food was supplemented for two weeks. The amount of body nitrogen retention and its -percentage were lower in unsupplemented and sugar supplemented groups. Among the supplemented groups, its amount was increased according as the protein intake was higher, while its percentage was decreased. Body and internal organs weight change showed a similar tendency. Interrelation between calorie intake and body fat retention (liver fat content and epididymal fat pad weight) was not found regularly. Consequently, the recovery rate from restriction was higher in protein supplemented group than calorie supplemented group. But no significant difference could be found between the groups.

  • Ovarian Yolk protein (YP2) synthesis has been investigated in mosquito, Culex pipiens pallens. Yolk protein amount which was syntheized in fat body, accumulated into ovary were analyzed by Rocket immunoelectrophoresis and in vitro organ culture. The result was that yolk protein synthesis began to occur at 6hrs after blood meal, reached at maximum level by 24hrs, and was completed within 48hrs. Yolk protein accmulation into the ovary began to start at 6hrs and coutinued for up to 60hrs after blood meal. Extract from 0, 24, 48, 72hrs ovaries after blood meal were analyzed by electrophoresis and Western blotting. The result was that 24hrs ovary contain one yolk protein(YP1), and 48, 72hrs ovaries contain two kinds of yolk proteins(YPl and YP2). When 48hr ovaries and fat bodies were incubated in $^3$H-leucine contained medium, protein synthesis was not occurred in fat body, but ovary synthesized much protein contained yolk protein (YP2). The result of crossed immunoelectrophoresis represented the same immunity between YPl and YP2. The present data suggest that ovary synthesize yolk protein(YP2) in mosquito, Culex pipiens pallens.

The Change of Electrolytes Composition in Skeletal Muscles by Protein Restriction and Rehabilitation (단백질의 제한과 회복으로 인한 골격 근육내의 성분변화)

  • The objective of this study was to compare the electrolytes contents in skeletal muscles of two different groups of rats, one for control fed 20% casein diet, and the other experimental group recovered from 7% casein diet as protein restriction feeding for two weeks at the beginning of this experiment. Two different comparison criteria were used in this study, one for comparison between animals at the same age groups and the other between groups weighing same body weight as the control groups. Food consumption of animals for experimental and control groups were ad libitum. Following measurement and samples were collected: body weight, five different muscles-anterior tibialis (A.T.) extensor digitorium longus (E.D.L.) soleus, plantaris, gastrocnemius-for electrolytes and protein analysis. The experimental groups showed lower body weight than that of control group. Same trend was showen in protein content in five different muscles. Magnesium and potassium content in four muscles except gastrocnemius showed lower and sodium higher in experimental groups than those in control group at fourteenth day, after recovery from protein restriction, electrolytes content change showed differently in four muscles. Magnesium and potassium contents were increased as protein content in diet. Sodium content decreased as one week intake of recovery diet started, thereafter it was rather slowly increased. Phosphorus content in gastrocnemius at the second week in experimental group was slightly lower than that of control group, and it recovered rapider while at the same body weight group it was higher in experimental group.

  • This study was carried out to investigate the development of endoplasmic reticulum and the formation of Protein body in the endosperm cell during seed formation of Panax ginseng C. A. Meyer with electron microscope. In the endosperm cell of early developmental process after pollination, vesicles that contain storage materials produced in rough endoplasmic reticulum incorporated into central vacuole. The central vacuole is gradually subdivided into several small-sized vacuoles and increased in number. Amorphous proteinaceous materials of high electron density are produced in rough endoplasmic reticulum. Rough endoplasmic reticulum increase in number and surround the protein body and vesicles circularly. Spherical proteinaceous granules with limited membrane appeared from the amorphous granules at the peripheral region of the rough endoplasmic reticulum. Gradually, storage materials are accumulated within the vacuole surrounded by spherosomes. Protein bodies are formed by interfusing between vacuoles and vesicles derived from rough endoplasmic reticulum which contained the amorphous protein of high electron density.

Studies on the Trypsin Inhibitor in Raw Beans of Korea (한국산 두류의 Trypsin Inhibitor에 관한 생화학적 연구)

  • This study was undertaken establish the relationship between trypsin inhibitor in raw soybean and antinutritional effect of raw legumes. 1) Among legumes produced in Korea, Glycine max contains a relatively high amount of protein(higher than 40%) compared with kindey bean, sword bean and mung bean and, furthermore, soybean which contains a high amount of protein possesses high trypsin inhibitory activity. 2) Disc electrophoretic pattern exhibited pattern exhibited that the crude protein preparation from Glycine max produced about 9-12 protein bands, and the pattern of electrophoretic mobility was very similar to each other. However, only a few protein bands were observed from the crude protein preparation of yard long bean, sword bean, adzuki bean, mung bean and rice adzuki. From the eluate of the sliced gel, it was confirmed that among those bands, only the fastest moving band contains trypsin inhibitory activity. 3) In chicks fed the normal diet the body weight was increased steady from one week and reached to 40% increase for three weeks but in chick fed raw bean diet, there was no body weight gain until two weeks feeding and only 10-20% of body weight gain was observed at the end of three week feeding. On the other hand, in chicks fed raw bean diet the weight of pancreatic tissue per 100g body weight was increased to about two-fold for two or three weeks but there was no change in liver weight. 4) In the case of amylase secretion from the pancreatic fragment, very strong stimulation on amylase secretion from pancreatic tissue of chicks fed a normal diet was produced by one unit of cholecystokin-pancreozymin. However, no stimulation was observed from pancreatic fragment of chick fed raw bean diet.
