팥과 녹두단백질의 첨가수준과 가열처리가 흰쥐의 성장에 미치는 영향 (A Study of Elucidation of Protein Quality of Raw and Heated Legumes Fed by Three Different Dietary Levels on Rats)

  • Various kinds of legumes have included as essential foods in Korean diet. However, a little attention have been paid on variety of those beans other than soybean. Main purposes of this study is to evaluate the protein quality of raw and cooked, red mung bean. Eighty male, sprague-Dawely rats weighing 50 gram were devided into 16 groups, five rats each. Casein protein from red and mung bean were used was included to after true digestibility of the protein of legemes. After 4 weeks feeding period animals were sacrificed and following data: all determined food intake, body weight gain, F.E.R., pp.E.R. and weights of liver, kidney heart, spleen, testis, pancreas, skeletal muscles. Food intake, body weight gain, F.E.R., pp.E.R. of casein group were higher than those of experimental groups (red bean, mung bean) body weight gain was increased with protein increment in the diet in all groups. The weight of liver showed significant difference between standard group and experimental group. (red bean, mung bean) The nitro gen content of liver and muscles were increased with level of protein in the diets.

Review of Biological Network Data and Its Applications

  • Studying biological networks, such as protein-protein interactions, is key to understanding complex biological activities. Various types of large-scale biological datasets have been collected and analyzed with high-throughput technologies, including DNA microarray, next-generation sequencing, and the two-hybrid screening system, for this purpose. In this review, we focus on network-based approaches that help in understanding biological systems and identifying biological functions. Accordingly, this paper covers two major topics in network biology: reconstruction of gene regulatory networks and network-based applications, including protein function prediction, disease gene prioritization, and network-based genome-wide association study.

Dietary Protein Restriction Alters Lipid Metabolism and Insulin Sensitivity in Rats

  • Dietary protein restriction affects lipid metabolism in rats. This study was performed to determine the effect of a low protein diet on hepatic lipid metabolism and insulin sensitivity in growing male rats. Growing rats were fed either a control 20% protein diet or an 8% low protein diet. Feeding a low protein diet for four weeks from 8 weeks of age induced a fatty liver. Expression of acetyl-CoA carboxylase, a key lipogenic enzyme, was increased in rats fed a low protein diet. Feeding a low protein diet decreased very low density lipoprotein (VLDL) secretion without statistical significance. Feeding a low protein diet down-regulated protein expression of microsomal triglyceride transfer protein, an important enzyme of VLDL secretion. Feeding a low protein diet increased serum adiponectin levels. We performed glucose tolerance test (GTT) and insulin tolerance test (ITT). Both GTT and ITT were increased in protein-restricted growing rats. Our results demonstrate that dietary protein restriction increases insulin sensitivity and that this could be due to low-protein diet-mediated metabolic adaptation. In addition, increased adiponectin levels may influences insulin sensitivity. In conclusion, dietary protein restriction induces a fatty liver. Both increased lipogenesis and decreased VLDL secretion has contributed to this metabolic changes. In addition, insulin resistance was not associated with fatty liver induced by protein restriction.

최적 연관 속성 규칙을 이용한 비명시적 단백질 상호작용의 예측 (Prediction of Implicit Protein - Protein Interaction Using Optimal Associative Feature Rule)

  • 단백질들은 서로 다른 단백질들과 상호작용 하거나 복합물을 형성함으로써 생물학적으로 중요한 기능을 한다고 알려져 있다. 때문에 대부분의 세포작용에 있어 중요한 역할을 하는 단백질 상호작용의 분석 및 예측에 대한 연구는 여러 연구그룹으로부터 풍부한 데이타가 산출되고 있는 현(現) 게놈시대에서 또 하나의 중요한 이슈가 되고 있다. 본 논문에서는 효모(Saccharomyces cerevisiae)에 대해 공개되어있는 단백질 상호작용 데이타들에서 속성들 간의 연관을 통해 유추 가능한 잠재적 단백질 상호작용들을 예측하기 위한 연관속성 마이닝 방법을 제시한다. 단백질의 속성들 중 연속값을 가지는 속성값들은 최대상호 의존성에 기반을 두어 이산화 하였으며, 정보이론기반 속성선택 알고리즘을 사용하여 단백질들 간의 상호작용 예측을 위해 고려되는 단백질의 속성(attribute) 수 증가에 따른 속성차원문제를 극복하도록 하였다. 속성들 간의 연관성 발견은 데이타마이닝 분야에서 사용되는 연관규칙 발견(association rule discovery) 방법을 사용하였다 논문에서 제안한 방법은 발견된 연관규칙을 통한 단백질 상호작용 예측문제에 있어 최대 약 96.5%의 예측 정확도를 보였으며 속성필터링을 통하여 속성필터링을 하지 않는 기존의 방법에 비해 최대 약 29.4% 연관규칙 발견속도 향상을 보였다.

94 KDa Glucose-regulated Protein의 BiP과의 결합 (Association of the 94 KDa Glucose-regulated Protein with Immunoglobulin Heavv Chain Binding Protein (BiP))

  • The 94 KDa glucose-resulsted Protein (SH 94), one of stress Proteins, is a Ca2+-binding protein in the endoplasmic reticulum (ER). In this study, the possible effect of Ca2+ on the native conformation of grp 94 was examined. When the purified grp 94 was analyzed by Sel filtration in the presence of either EGTA or CaCl2, it was eluted with apparent molecular weight (MW) of 100 KDa in both cases. When similarly analyzed with microtome or cell Ivsate, however, srp 94 was eluted with apparent IW of 200 KDa in the presence of E6TA, while with apparent MW of 100 KDa in the presence of CaCl2, indicating possible association of grp 94 with one or more other proteins in the absence of CaCl2. Consequently, immunoprecipitation with anti-grp 94 was carried out to determine which proteins specifically interact with grp 94. It is sho%un that srp 94 may interact, in a Ca2+_dependent manner. with other proteins including BiP (grp 78) which is also a stress protein in the ER.

Effects of Dietary Protein and Energy on Growth Performance and Muscle Composition in Broilers Treated with Clenbuterol

  • The present study was conducted to examine the effects of dietary protein (20, 22, 24%) with a constant protein-to-energy ratio on clenbuterol-induced performance in broilers. The protein-to-energy ratio was based on adequate level (22% protein, 3,100 kcal of energy). Female broiler chickens were used for a $3{\times}2$ factorial arrangement and fed diets with or without 1 ppm clenbuterol from 14- to 32-days of age. Feed efficiency improved with increasing dietary protein level, regardless of clenbuterol treatment. The dietary clenbuterol increased weights of breast and leg muscles (gastrocnemius and peroneus longus), and clenbuterol markedly reduced protein content of leg muscles in chickens fed the 20% protein diet, but did not in chickens fed the 22 and 24% protein diets. Feeding the 24% protein diet with clenbuterol improved the protien accretion (peroneus longus) by 8.4%. Clenbuterol decreased DNA content and increased the protein/DNA ratio in breast muscle regardless of dietary protein intake. Clenbuterol had no effect on RNA content in both breast and leg muscles. The present results demonstrated that various protein levels which retain the same protein-to-energy ratio in the diet markedly alter the protein accretion induced by ${\beta}$-agonist in broilers.

단백질 상호작용 네트워크에서 필수 단백질의 견고성 분석 (Analysis of Essential Proteins in Protein-Protein Interaction Networks)

  • 단백질 상호작용 네트워크는 허브(hub)라 할 수 있는 상호작용 수가 많은 소수의 단백질과 상호작용수가 적은 다수의 단백질들로 구성된다. 최근 들어 여러 연구들에서 허브 단백질이 비 허브(non-hub) 단백질보다 상호작용 네트워크에 필수적인 단백질일 가능성이 높다고 보고되고 있다. 이러한 현상을 중심-치명 룰(centrality-lethality rule)이라 하는데, 이는 복잡계 네트워크에서 허브단백질의 중요성 및 네트워크 구조의 중요성을 설명하기 위한 방법으로 폭넓게 신뢰받고 있다. 이에 본 논문에서는 중심-치명 룰이 항상 옳게 적용되는지를 확인하기 위해 Uetz, Ito, MIPS, DIP, SGD, BioGRID와 같은 효모에 관한 공개된 모든 단백질 상호작용 데이터베이스들을 분석하였다. 흥미롭게도, 상호작용 데이터가 적은 데이터베이스들(Uetz, Ito, DIP)에서는 중심-치명 룰을 잘 나타냈지만 상호작용 데이터가 대용량인 데이터 베이스들(SGD, BioGRID)에서는 중심-치명 룰이 잘 맞지 않음을 확인하였다. 이에 따라 SGD와 BioGRID 데이터베이스로 부터 얻은 상호작용 네트워크의 특징을 분석하고 DIP 데이터베이스의 상호작용 네트워크와 비교하였다.

PERFORMANCE OF RAINBOW TROUT (Oncorhynchus mykiss) FED ON DIFFERENT DIETARY PROTEIN WITH FIXED ENERGY RATIO

  • An experiment was conducted on rainbow trout(Oncorhynclus mykiss) for eight weeks to investigate the growth performance of the fish fed with different dietary protein with constant diet energy of $20kJg^{-1}$. Four diets containing 25, 30, 35 and 40% crude protein were used. The highest mean final weight was obtained for the fish fed with diet having 35% protein. Growth performance in terms of Specific Growth Rate (SGR), Food Conversion Ratio (FCR) and Protein Efficiency Ratio (PER) were calculated for each diet. There were no significant differences in SGR but the highest value was exhibited by fish fed with 35% protein diet. Significant differences were found among FCR of different diets. Diets with 35 and 40% crude protein gave better FCR value than that of 25 and 30% crude protein. Although significant differences were not found between PER of different diets but PER of diet with 35% protein was found to be better than PER of both high and low protein diets (diets of 40 and 30% crude protein). It is concluded that diet having 35% protein with protein energy ratio of $17.53mgkJ^{-1}$ was suitable for rainbow trout (O. mykiss) among the protein spectrum used.

EFFECTS OF DIETARY CELLULOSE AND PROTEIN LEVELS ON NUTRIENT UTILIZATION IN CHICKENS

  • Effects of dietary cellulose and protein levels on nutrient utilization in chickens were investigated. Four experimental diets containing 5% (low cellulose) or 20% (high cellulose) cellulose in combination with 10% (low protein) or 20% (high protein) protein of 70 g/day were alternatively forced-fed to eight colostomized White Leghorn cockerels once a day to make $4{\times}4$ Latin-square design. The digestibilities of DM and energy decreased with the increase in cellulose level, but not affected by dietary protein level. Ether extract digestibility was higher in the high cellulose diets than in the low cellulose protein level. Ether extract digestibility was higher in the high cellulose diets than in the low cellulose diets. The digestibility of nitrogen free extract had the same trend with the digestibility of DM and energy. The digestibility of acid detergent fiber was not so much different among the diets, but the NDF digestibility was lower in the high cellulose diets than in the low cellulose diets, due to the low hemicellulose digestibility. The true digestibility of protein was influenced by both of the dietary protein and cellulose levels, and their interaction was found. The dietary protein level affected the biological value of protein but the dietary cellulose level did not, and consequently the biological value of protein in the low protein diets was lower than in the high protein diets.

50세 이상 한국인의 식물성 단백질 섭취와 건강 관련 삶의 질과의 연관성: 국민건강영양조사 2016-2018 (Plant Proteins in Relation to Health-related Quality of Life in South Korean Individuals Aged 50 Years or Older: Korea National Health and Nutrition Examination Survey 2016-2018)

  • 본 연구에서는 50세 이상 한국인을 대상으로 식물성 단백질 섭취와 건강관련 삶의 질과의 연관성을 분석하였다. 2016~2018년 국민건강영양조사 자료를 이용하여 7,956명(남성 3,434명, 여성 4,522명)이 연구에 포함하였다. 건강관련 삷의 질은 신체 기능, 자기 관리, 일상 활동, 통증/불쾌감, 불안/우울 문제로 구성된 삶의 질 5차원 지표(EQ5D)를 사용하여 측정하였다. 식물성 단백질 일일 섭취량과 건강관련 삶의 질 사이의 연관성은 회귀 분석을 사용하여 평가하였다. 총 식물성 단백질, 콩과 견과류 종자(LNS) 및 발효 콩(FERMB) 단백질의 섭취는 남성과 여성 모두에서 건강관련 삷의 질과 건강관련 삷의 질과 유의미한 연관이 있었다(p < 0.01). EQ5D의 지표 중 신체 기능과 일상 활동이 식물성 단백질 섭취와 가장 밀접한 관련이 있었다. LNS 단백질이나 FERMB 단백질 섭취에 비해 총 식물성 단백질 섭취가 건강관련 삷의 질과 더 큰 연관성을 보였다. 결론적으로, 식물성 단백질 섭취와 건강관련 삷의 질 사이에는 유의미한 연관성이 있었다. 이는 식물성 단백질의 적절한 섭취가 한국 성인의 건강관련 삷의 질 유지 또는 개선에 도움이 될 수 있음을 시사한다.
