Caffeic acid phenethyl ester protects against photothrombotic cortical ischemic injury in mice

  • In this study, we aimed to investigate the neuroprotective effects of caffeic acid phenethyl ester (CAPE), an active component of propolis purified from honeybee hives, on photothrombotic cortical ischemic injury in mice. Permanent focal ischemia was achieved in the medial frontal and somatosensory cortices of anesthetized male C57BL/6 mice by irradiation of the skull with cold light laser in combination with systemic administration of rose bengal. The animals were treated with CAPE (0.5-5 mg/kg, i.p.) twice 1 and 6 h after ischemic insult. CAPE significantly reduced the infarct size as well as the expression of tumor necrosis $factor-{\alpha}$, hypoxiainducible $factor-1{\alpha}$ monocyte chemoattractant protein-1, $interleukin-1{\alpha}$, and indoleamine 2,3-dioxygenase in the cerebral cortex ipsilateral to the photothrombosis. Moreover, it induced an increase in heme oxygenase-1 immunoreactivity and interleukin-10 expression. These results suggest that CAPE exerts a remarkable neuroprotective effect on ischemic brain injury via its anti-inflammatory properties, thereby providing a benefit to the therapy of cerebral infarction.

식물성 식품중 총플라보노이드 함량과 생리활성 탐색 (Contents of Total Flavonoid and Biological Activities of Edible Plants)

  • The contents of total flavonoids, total antioxidant status and superoxide scavenger activity of edible plants were measured. Among twenty one samples of leafy vegetables and teas, mugwort (Artemesia capillaris) and green teas gave high total flavonoids contents, 48.2 mg/g sample and 44.7 mg/g sample, respectively. The highest concentration of total flavonoids among eighty nine samples analyzed was 52.1 mg/g sample from propolis. When samples were tested for total antioxidant status, a kind of groundsel showed the highest Total Antioxidant Status(TAS) value, 13.4 mM among leafy vegetables and teas and green tea gave high TAS value, 11.84 mM. On the other hand, purslane gave the lowest TAS value, 0.01 mM. The superoxide scavenge activity was measured from the extracts of all edible plants. The superoxide scavenge activity of polygonatum(81.1%) and chrysantemum (78.5%) were the highest among leafy vegetables, teas and medicinalplants. On the other hand, ginseng, poria, cassiae semen and dioscoreae rhizoma showed no superoxide scavenge activities.

One-pot 합성을 통해 천연 화합물이 삽입된 아미노클레이 제조 및 항균성 연구 (Preparation of Natural Chemicals Intercalated Aminoclay via One-pot Synthesis and its Antimicrobial Property)

  • 본 연구에서는 마그네슘 이온 및 (3-aminopropyl)triethoxysilane (APTES)으로 표면에 amine group이 부착된 아미노클레이를 합성(AC)함과 동시에 천연 화합물인 프로폴리스 및 황련 추출물을 클레이 시트 사이에 삽입한 클레이를 합성(PIAC, CIAC)하였다. 주사전자현미경, 입도 분석법, FT-IR, TGA 및 XRD를 통해 클레이의 합성 및 천연 화합물의 삽입을 확인하였고, 특히 XRD data에서 천연 화합물의 삽입에 따른 층간거리(d-space)를 분석함으로써 명확하게 그 특성을 결정하였다. 또한 천연 화합물이 삽입된 클레이인 PIAC 및 CIAC의 경우 삽입되지 않은 아미노클레이(AC)에 비해 항균성이 향상된 것을 최소저해농도(MIC) 분석을 통해 확인하였다.

Use of natural ingredients in Japanese quail diet and their effect on carcass and meat quality - A review

  • The present paper reviews the findings of different research studies on the effect of natural ingredients in the Japanese quail (Coturnix coturnix japonica) diet on carcass characteristics and meat quality. The results show a relationship between the type and concentration of ingredients used in diets and carcass characteristics and meat quality. The inclusion of medicinal herbs (thyme, black seed, and mint), plants (canola), seeds (chickpea), spices (cinnamon and coriander), worms (earthworms), bee products (propolis), phytochemicals (lycopene), and edible fungi (common mushrooms) in the diet improved carcass quality characteristics compared to the control diets (basal diets). The inclusion of medicinal herbs (spearmint and green tea), spices (cinnamon), vegetables (tomato), plants (verbena and canola), seeds (marijuana), and edible fungi (oyster mushrooms) improved meat quality. In conclusion, the use of ingredients of natural origin in the diet of Japanese quail improves carcass quality characteristics and meat quality.

천연소독제 Clean Natural의 Salmonella typhimurium에 대한 복귀돌연변이시험 (Bacterial Reverse Mutation Test of Clean Natural using Salmonella typhimurium)

  • Clean Watural은 주성분이 참나무로부터 추출정제한 목초액과 프로폴리스로 구성된 새로운 천연 살균소독제이다. Salmonella typhimurium TA98, TA100, TA102, TA1535 및 TA1537 등의 5종의 균주를 이용한 복귀돌연변이성 시험을 수행한 결과, 모든 균주에서 대조군과 비교하여 시험용량군 사이에 돌연변이 유발성이 관찰되지 않았다. 그러나, 앙성대조물질에서는 대조군과 비교하여 통계학적으로 유의한 균주의 증가가 관찰되었다. 따라서, 시험물질인 천연 살균소독제, Clean Natulat은 돌연변이 유발 가능성이 없는 것으로 평가되었다.

  • Clean Natural is a new disinfectant of which main components are propolis and wood vinegar from Quercus mongolica. The mutagenicity of Clean Natural was studied by a micronucleus test in male ICR mice. The maximally tolerated dose (MTI) of Clean Natural was determined to >2.0 g/kg body weight. Therefore, the doses adopted for the micronucleus test was 2.0 g/kg as a high dose, 1.0 g/kg as a medium and 0.5 g/kg as a low of dose, respectively. Each of group was consisted of three doses of Clean Natural, positive control 2 mg/kg of mitomycin C and negative control 20 ml/kg of saline. A slide preparation was made at 24 hours following administration. No significant induction of micronuclei was observed in any of the three doses of Clean Natural orally administered. No cytotoxicity such as inhibition of hemopoiesis was observed in any group of test agent as the rate of polychromatic erythrocytes to total erythrocytes was over 40%. These results indicate that Clean Natural is not capable of inducing micronuclei in vivo mouse cells and thus has no genotoxicity in micronucleus test.

노봉방에서 추출된 카페인산 페네틸 에스테르가 함유된 한방화장품의 미백 개선에 관한 임상적 연구 (A Clinical Research about Herbal Cosmetics Containing Caffeic acid phenethyl ester Isolated from Vespae Nidus on the Whitening Effects)

  • Objectives: The purpose of this clinical research was to investigate the effects of herbal cosmetics containing Caffeic acid phenethyl ester (CAPE), an ingredient compound of various herbs including Vespae Nidus, on the Whitening Effects. Methods: A total of 20 subjects who visited Pusan National University Hospital from March 2nd, 2017 to April 11th, 2017 were included. In this study, we evaluated the visual evaluation, the melanin index (M) and erythema index (E) through skin analysis equipment, the subjective whitening improvement analysis, and the adverse reaction according to product use. Statistical analysis was performed with independent t-test and Mann-Whitney's U test. Statistical significance was achieved if the probability was less than 5% (p<0.05) Results: As a result of the visual evaluation, it was effective in improving skin whitening. The subjective whitening improvement analysis also showed positive results. However, no statistically significant differences were found between the test and control cosmetics in the melanin index (M) and erythema index (E) through skin analysis equipment. In the safety evaluation, some adverse reactions were reported, but no significant were observed directly from cosmetics. Conclusions: Considering the above results, we have confirmed the possibility of herbal cosmetics containing containing CAPE of Vespae Nidus Extracts.

각질형성세포에서 Chrysin이 Vitamin D Receptor의 전사 활성화에 미치는 영향 (The Effect of Chrysin on the Transcriptional Activity of Vitamin D Receptor in Human Keratinocytes)

  • Chrysin (5,7-dihydroxyflavone)은 프로폴리스, 꿀 같은 음식과 다양한 식물에 존재하는 천연 플라보노이드이다. Chrysin은 항산화, 항노화, 항염, 항암 효과 등 다양한 생물학적 효과를 가진다고 알려져 있다. 이 연구에서, 우리는 사람의 각질형성세포에서 chrysin이 VDR을 통한 transcriptional activity에 미치는 영향을 dual-luciferase assay을 통하여 살펴보았다. Chrysin은 농도 의존적으로 VDR을 통한 transcriptional activity를 증가시켰다. Quantitative real time PCR을 통해 chrysin이 사람의 각질형성세포에서 VDR mRNA의 발현을 증가시킴을 확인하였다. 또한, chrysin이 각질형성세포의 분화 마커인 keratin 10, involucrin 그리고 filaggrin의 mRNA 발현을 증가시킴을 확인하였다. 이러한 연구 결과는 chrysin이 VDR을 통한 transcriptional activity를 조절하여 각질형성세포의 분화를 촉진시킬 수 있다는 것을 시사한다.

The Effect of Caffeic Acid Phenethyl Ester (CAPE) on Phagocytic activity of septic Neutrophil in vitro

  • Caffeic acid phenethyl ester (CAPE) is an active component of propolis obtained from honeybee hives. CAPE possesses anti-mitogenic, anti-carcinogenic, anti-inflammatory, and immunomodulatory activities in diverse systems, which know as displays antioxidant activity and inhibits lipoxygenase activities, protein tyrosine kinase, and nuclear factor kappa B (NF-κB) activation. This study aimed to investigate the effect of CAPE on lipopolysaccharide (LPS)-induced human neutrophil phagocytosis. Human neutrophils were cultured with various concentrations of CAPE (1, 10, and 100 µM) with or without LPS. The pro-inflammatory proteins (tumor necrosis factor-alpha [TNF-α], interleukin [IL]-6 and IL-8) levels were measured after 4 h incubation. To investigate the intracellular signaling pathway, we measured the levels of mitogen-activated protein kinases (MAPK), including phosphorylation of p38, extracellular signal-regulated protein kinases 1 and 2 (ERK1/2) and c-Jun N-terminal kinase (JNK). Next, to evaluate the potential phagocytosis, neutrophils were labeled with iron particles of superparamagnetic iron oxide nanoparticles (SPIONs, 40 nm) for 1 h in culture medium containing 5 mg/mL of iron. The labeling efficiency was determined by Prussian blue staining for intracellular iron and 3T-wighted magnetic resonance imaging. CAPE decreased the activation of intracellular signaling pathways, including ERK1/2 and c-Jun, and expression of pro-inflammatory cytokines, including TNF-α and IL-6, but had no effect on the signaling pathways of p38 and cytokine IL-8. Furthermore, images obtained after mannan-coated SPION treatment suggested that CAPE induced significantly higher signal intensities than the control or LPS group. Together, these results suggest that CAPE regulates LPS-mediated activation of human neutrophils to reduce phagocytosis.

아저해농도(亞沮害濃度)의 항균물질이 mutans streptococci의 세포표면성질과 독력인자에 미치는 영향 (EFFECTS OF SUBINHIBITORY CONCENTRATIONS OF ANTIMICROBIAL AGENTS ON CELL SURFACE PROPERTIES AND VIRULENCE FACTORS OF MUTANS STREPTOCOCCI)

  • 아저해농도(亞沮害濃度)란 세포의 성장을 저해하는 최소 저해 농도(minimum inhibitory concentrations; MIC)이하의 농도를 말하는데, 적용된 화학물질의 동력학에 따라 항균물질은 반드시 이러한 농도로 존재하는 시기를 거치게 되며, 아저해농도의 항균물질은 세포의 성장을 저해하지는 않지만 세균의 대사와 성장에 스트레스로 작용하게 되어 세포의 성질과 독력인자 발현에 영향을 줄 수 있을 것이다. 본 연구는 여러 항균물질의 최소저해농도를 결정하고 아저해농도의 항균물질이 존재 시 세포 성질과 독력 인자에 미치는 영향을 알아보고, 치아우식증의 예방에 대한 화학적 접근법의 효용성을 조사하기 위해 계획되었다. 연구대상 세균으로는 mutans그룹 streptococci 중 두 가지 대표적인 세균종인 S. mutans (S. mutans Ingbritt)와 S. sobrinus (S. sobrinus 6715-7)를 사용하였고 화학물질 8종 (Sanguinaria extract; SG. Chlorhexidine digluconate; CHX, Fluoride; F, Propolis; PP, Hydrogen peroxide HP, Triclosan; TC, Sodium dodecyl sulfate: SDS, Cetylpyridinium chloride; CC) 을 단계 희석하여 배지에 첨가하여 최소저해농도를 구하였으며 아저해농도의 항균물질을 배지에 첨가하여 5% $CO_2$ 존재 하에 $37^{\circ}C$에서 배양하여 이러한 항균물질을 첨가하지 않은 배지에서 배양한 세균과 성장속도, 산생성능, 체표소수성, 수산화인회석에의 부착능, glucan생성능, 세균응집능 등을 비교 관찰하였다. 아저해농도의 항균물질은 세포의 성장과 체표소수성, 타액으로 처리한 수산화인회석에 대한 세균의 부착과 glucan합성에 영향을 주는 것이 관찰되었다. 또한 12시간 배양 후 S. mutans와 S. sobrinus 모두에서 대조군에 비해 통계적으로 유의하게 최종 pH가 높게 나타나 산생성능이 억제되는 것이 관찰되었다 (p<0.05). 세균을 proteinase K로 전처리한 경우 처리하지 않았을 때 보다 세균응집역가가 증가하거나 응집이 관찰되지 않았다. 본 연구의 결과에 따르면, 각각의 항균물질은 아저해농도에서도 알려진 작용기전과 유사하게 세균의 성질에 영향을 주었고 이러한 농도에서도 지속적으로 세균을 억제할 수 있었다. 따라서 이러한 항균물질의 사용은 치아우식증의 효과적 예방법의 하나가 될 수 있다
