Inactivation of Escherichia coli O157:H7, Salmonella and Listeria monocytogenes by Organic Acid (유기산이 Escherichia coli O157:H7, Salmonella 및 Listeria monocytogenes의 증식에 미치는 영향)

  • The inhibitory effect of the food processing agent on growth of Escherichia coli O157:H7, Salmonella Enteritidis, and Listeria monocytogenes was performed with organic acid, and combination of citric acid, acetic acid, propionic acid and vanillic acid. The minimun inhibitory concentration(MIC) of propionic acid was 5,000 ppm in E. coli O157:H7, 2,500 ppm in Salmonella Enteritidis and Listeria monocytogenes. MIC of citric acid was 10,000 ppm in E. coli O157:H7 and Salmonella Enteritidis, 2,500 ppm in Listeria monocytogenes. MIC of acetic acid was 2,500ppm, while in vanillic acid was 5,000 ppm in Escherichia coli O157:H7, Salmonella Enteritidis, and Listeria monocytogenes. MIC of combined organc acid in E. coli O157:H7 were 2,500ppm in PC, 1,250 ppm in PA, PV, CA, CV and AV. MIC of combined organc acid in Salmonella Enteritidis were 2,500 ppm in PC, PA, PV, CA, and CV, 1,250 ppm in AV. MIC of combined organc acid in Listeria monocytogenes were 1,250 ppm in all treatment group. MIC of combined treatment of three organc acid in E. coli O157:H7, S. Enteritidis and L. monocytogenes were 1,250 ppm in PCA, PCV, PAV and CAV. The inhibitory effect of organc acid in E. coli O157:H7, S. Enteritidis and L. monocytogenes could be confirmed from the result of this experiment. Therefore, it was expected that the food process would increase or maintain by using organic acid.

  • Chitin, the natural polymer has been known as harmless and innoxious material to human and has been also known to be biodegradable. Chitosan which was prepared by the deacetylation of chitin, was propionylated to obtain porous bead shaped propionyl chitosan and its possibility as a water-absorbant polymer was investigated. Propionyl chitosan porous bead was synthesized by acylation reaction using emulsion method of acetic acid solution and propionyl chitosan was partially crosslinked using ethyleneglycol diglycidyl ether. Through the experiment varying the moles of propionic anhydride, reaction time and reaction temperature, best results for water-absorption ability was obtained at reaction condition of 5 moles of propionic anhydride, 10 hours of reaction time and $22^{\circ}C$ of reaction temperature. The absorption ability to the distilled water, various salt solutions, artificial urine and artificial blood, absorption time and retention of water of synthesized porous bead were investigated and also mechanical strength after crosslinking was determined.

Quality Characteristics of Makgeolli Supplemented with Pineapple (파인애플 첨가 막걸리의 품질특성)

  • Pineapple flesh and core were added during fermentation to develop a new style of fruit makgeolli. Physicochemical characteristics were measured during fermentation, and sensory evaluation was performed after fermentation to estimate the quality of pineapple makgeolli. No significant differences were observed for pH, $^{\circ}Bx$, and alcohol contents between makgeolli with and without pineapple after fermentation. Total acidity of the pineapple makgeolli increased due to considerable elevation of citric acid, tartaric acid, malic acid, and pyroglutamic acid contents as well as slight elevation of lactic acid and propionic acid contents. On the other hand, the total amino acid contents of pineapple makgeolli were lower than those of control. The sensory preferences for pineapple makgeolli were higher overall than those for control, which can be attributed to the preferences for fruit flavor and sour taste in pineapple makgeolli.

  • The ripening of cheese allows for the development of characteristic taste and flavour, nutritional substances, bio-active components and texture, helping to improve quality. Many different microbiological, biochemical and nutritional changes occur during the process depending on the quality of raw milk, added cultures and enzymes, as well as specific processing and ripening conditions. During the ripening lactose is hydrolyzed to lactic, propionic and acetic acid, helping to reduce potential effects of the problem of lactose intolerance. Fat is hydrolyzed to butyric, propionic and conjugated linoleic acid, which function as bio-active substances. Protein is hydrolyzed to different peptides and amino acids which all show various bio-activities. However, errors of cheese ripening can happen and affect the quality of the product. To guarantee good quality cheese the process needs to be managed carefully with the right microbes used and ensuring cleanliness of processing facilities, staff, ventilation and hazard analysis and critical control points (HACCP). Research into and controlling of ripening technology is crucial for producing high quality cheeses.

Effect of Biosolids on Heavy Metal Bioavailability and Organic Acid Production in Rhizosphere of Zea mays L. (Biosolid 처리가 옥수수 근권의 유기산 생성과 중금속 유효도에 미치는 영향)

  • A greenhouse experiment was conducted to evaluate the composition of organic acids on the bioavailability of heavy metals in the rhizosphere of corn (Zea mays L.) over 12 weeks after treatment of biosolids. After planting the plants were sampled at 12th week and analyzed for heavy metals in shoot and root and organic acids in the rhizosphere. Results showed that biomass yields of Zea mays L. grown on biosolids-treated media were significantly higher than those grown on standard media. The effect of biosolids on biomass yields of Zea mays L. was in order of Riverside ${\approx}$ Los Angeles ${\approx}$ MWRDGC > Milorganite > Nu-earth > standard media. Metal uptake by Zea mays L. was closely related with the contents contained on biosolids treated. In the plot treated with Nu-earth the uptake of Cd and Zn by shoot was significantly higher than those at the plots treated with other biosolids. The uptake patterns of Cd and Zn by root were similar to those of shoot. The uptake of Cr and Ni was significantly higher with application of Nu-earth over other biosolids. In all cases, the major organic acids in the rhizoshpere were lactic, acetic, propionic, butyric, and oxalic acids. Other organic acids were glutaric and succinic and occasionally, pyruvic and tartaric acids were also found. And the organic acid compositions did not vary significantly with the treatments of biosolids. Butyric and acetic acids were distinctively dominant both in the standard and the biosolids treatments.

THE EFFECT OF FOUR KINDS OF ACID AND CONCENTRATION ON THE FORMATION OF ARTIFICIAL CARIOUS LESION IN HUMAN TOOTH ENAMEL (수종의 유기산이 법랑질 인공우식의 형성에 미치는 영향)

  • The end products of the metabolism of the oral microorganism, organic acids, are an element that produces dental caries. Four organic acids in plaque fluid, lactic acid, acetic acid, succinic acid, propionic acid which take the important role in producing dental caries, were chosen to evaluate the effect of acid type and concentration. The subject, $100{\mu}m$ in thickness, were immersed in acid-buffer solution which has the different acid concentration of 10mM, 25mM, 50mM, 100mM and pH 4.3 and degree of saturation was $0.153{\pm}0.003$ kept in constant and were operated to produce artificial caries under different demineralization time (1, 2, 3 day) at x25. The results were obtained by observing under polarizing microscope at x25. 1. The subsurface lesion, specific finding of incipient enamel caries, showed positive birefringence. but surface zone and sound enamel showed negative birefringence. 2. The demineralization rate of enamel was increased as the acid concentration increased. 3. The subsurface lesion showed increasing depth in the order of lactic, acetic, propionic acid, succinic acid. 4. The concentration of organic acid in artificial caries system had an independent effect on demineralization rate in enamel under the constant pH and degree of saturation. The result of this study showed that not only pH and the acid strength but the concentration of organic acid had an independent effect on demineralization rate in early enamel caries. And through the further research on the factors influencing enamel demineralization, it will be necessary to develop an effective caries preventive therapy.

  • Propionic acidemia (PA) is a rare autosomal recessive metabolic disease caused by the deficiency of propionyl-CoA carboxylase (PCC). PA affects the catabolism of branched chain amino acid and oddchain fatty acid then results in accumulation of propionic acid and other metabolites in plasma and urine. Catabolic stress such as infection, illness or any stress can precipitate cause acute metabolic decompensation, especially in the first years of life. Acute metabolic decompensation commonly calls for emergency treatment or admission and if the patient is in a serious condition, it can lead to coma or death. But frequent admissions or visiting the emergency room are much burden to the patients and their kins. And we experienced the propionic academia with a confirmed novel mutation and the patient suffered from frequent admission and visiting the emergency room. So, we tried the regular home carebased fluid therapy after securing a central venous line. Finally, we succeeded in preventing frequent admissions resulted from acute metabolic decompensation and could contribute to relieving the burden to the patient and their family.

The Effect of Feeding Order on Dry Matter Intake and Ruminal Characteristics Chage of Daily Cattle (젖소에 급여하는 사료의 급여 순서가 사료섭취량 및 반추위내 생리적 변화에 미치는 영향)

  • This experiment was carried out to investigate the effect of feeding order bay-concentrate-silage, concentratehay- silage, silage-concentrate-hay, silage-concentrate-hay, concentrate-silage-hay) on ruminal characteristics change with 3 fistulated dry Holstein cows in a Latin square design. The main results obtained were as follows : DM intake in forage hayconcentrate-silage feeding order was 1.65% of body weight, which is the highest of all treatments, but that in concentrate-hay-silage 1.4%, which is the lowest. The difference between max and min rumen pH in hayconcentrate-silage feeding order was lowest as 0.55, but there is no significant. The mean rumen $NH_3N$ content in silage-concentrate-hay feeding order was highest as 6.12mg/100ml, but that in silageconcentrate-hay feeding order lowest as 4.82mg/100ml. Acetic acid and propionic acid content was highest in forage hay-concentrate-silage feeding order. but there is no significant. The ratio of acetic acid to propionic acid averaged 3.47~3.69(NS). In conclusion, the best feeding order fitted in ruminal physiology was forage hay-concentrate-silage.

  • The electrochemical approach to the characteristics of black chrome solar selective coatings has been reviewed. Based on the formula of the ChromOnyx bath solution manufactured by Harshaw Company, the chromic acid-propionic acid bath solutions have been prepared by KIER. Comprehensive review of the black chrome electrodeposition mechanism also has been performed on the role played by constituents in the bath solution during electrodeposition. The optical properties such as absorption and emittance of electrodeposited black chrome coatings obtained from the new formula and their surface analysis will be reported in detail.
