Regulatory Mechanism of L-Alanine Dehydrogenase from Bacillus subtilis

  • L-alanine dehydrogenase from Bacillus subtilis exhibits allosteric kinetic properties in the presence of $ZN^{2+}$. $ZN^{2+}$ induces the binding of substrate (L-alanine) to be cooperative at pH 8.0. The effect of pH variation between pH 7.0 and pH 10.0 on the inhibition by $ZN^{2+}$ correlates with the pH effect on the $K_m$ values for L-alanine within these pH range indicating that $ZN^{2+}$ and substrate compete for the same site. No such cooperativity is induced by $ZN^{2+}$ when the reaction is carried out at pH 10. At this higher pH, $ZN^{2+}$ binds with the enzyme with lower affinity and noncompetitive with respect to L-alanine. Inhibition of L-alanine dehydrogenase by $ZN^{2+}$ depends on the ionic strength. Increase in KCI concentration reduced the inhibition, but allosteric property in $ZN^{2+}$ binding is conserved. A model for the regulatory mechanism of L-alanine dehydrogenase as a noncooperative substrate-cooperative cofactor allosteric enzyme, which is compatible in both concerted and the sequential allosteric mechanism, is proposed.

  • The exponential continuous time GARCH(p,q) model for financial assets suggested by Haug and Czado (2007) is considered, where the log volatility process is driven by a general L$\acute{e}$vy process and the price process is then obtained by using the same L$\acute{e}$vy process as driving noise. Uniform ergodicity and ${\beta}$-mixing property of the log volatility process is obtained by adopting an extended generator and drift condition.

난백분말과 유제품을 이용한 요구르트의 제조 (The Preparation of Yogurt from Egg White Powder and Milk Products)

  • 본 연구에서는 난백분말 3% (w/v)와 포도당 2% (w/v)에 4종의 유제품 4% (w/v)를 각각 첨가하여 기질을 만들고 젖산균(Lactobacillus)으로 발효하여 호상의 요구르트를 만든 후, 첨가된 유제품이 젖산균의 생육과 산생성 및 요구르트의 품질(관능성, 휘발성 향기 성분)에 미치는 영향을 조사하였다. L. acidophilus로 24시간 발효된 시료의 경우, 산도는 우유 시료(대조군)에 비하여 유제품 첨가 시료가 유의적으로 낮았으며(p<0.05), 유제품 첨가 시료 가운데는 카제인 시료의 산도가 가장 낮았다. 생균수는 우유 시료가 $2.0{\times}10^{9}/mL$인데 비하여 유제품 첨가 시료는 $5.0{\times}10^{8}{\sim}8.0{\times}10^{8}/mL$으로 현저하게 낮았다. L. acidophilus로 16.5시간 발효시켜 만든 호상요구르트의 관능성을 보면, 전반적인 기호도는 우유요구르트(표준시료)보다 유제품 첨가 시료가 저조하였으나 카제인 시료의 경우는 표준시료와 유의적인 차이를 보이지 않았으며(p<0.05), 유제품 첨가 시료 중에서는 유청 시료의 관능성이 가장 저조하였다. 본 실험에서 사용된 L. acidophilus (KCTC 2182)는 시료에 따라서 다소 차이가 있으나 발효 과정에 acetone, ethanol, diacetyl, acetoin을 생성하였다.

  • 케탈이나 아세탈반응에 의하여 이루어진 1, 3-디옥솔란 고리가 산수용액 중에서 불안정해 쉽게 가수분해되는 성질을 이용하여 소수부로는 지방산의 탄화수소사슬을 친수부로는 소디움 카르복시산(Soap)을 도입하여 분해 후 계면활성을 띠지 않는 분해성 계면활성제이다. 이 화합물의 수용액에 대한 표면장력은 31dyne/cm였고, 임계미셀농도는 $1.0{\times}10^{-3}mol/L$이었다. 기포력은 비교적 좋지 않았으며, 유화력은 콩기름보다 벤젠에서 양호하였다. 또한 이 화합물의 산분해 특성을 1wt% 수용액과 벤젠 사이의 계면장력 변화에 의하여 측정한 결과 pH 1 ${\sim}$ 4범위에서 약 300분 안에 모두 가수분해됨을 확인하였다.

Tetramethylpyrazine Protects Oxidative Stability and Gelation Property of Rabbit Myofibrillar Proteins

  • Tetramethylpyrazine (TMP), an alkaloid rich in Ligusticum wallichii and fermented products, possesses multiple pharmacological activities in antioxidant, antiinflammatory, and antibacterial. This study aimed to investigate the effect of TMP (15 mg/L) on the physicochemical and gelation properties of rabbit myofibrillar proteins (MPs) with/without oxidative stress. Results showed that compared to the control, oxidative stress to MPs decreased free thiol content, gel yield, whiteness, water-holding capacity, bounder water, immobilized water, and endogenous tryptophan fluorescence intensity, but increased surface hydrophobicity, dityrosine content, and free water content (p<0.01). Without oxidative stress, MPs treated with TMP increased free thiol content, whiteness, and bound water, but decreased dityrosine content and free water (p<0.05). Under oxidative conditions, all parameters were conversely affected by TMP (p<0.01). The results suggest that TMP can be an antioxidant to decrease the concern on oxidative deterioration during meat processing and storage by improving the oxidative stability, water retention, and gel forming property of rabbit MPs.

  • We focus on the interations of the weighted Berezin transform Tα on Lp(τ), where τ is the invariant measure on the complex unit ball Bn. Iterations of Tα on L1R(τ) the space of radial integrable functions played important roles in proving 𝓜-harmonicity of bounded functions with invariant mean value property. Here, we introduce more properties on iterations of Tα on L1R(τ) and observe differences between the iterations of Tα on L1(τ) and Lp(τ) for 1 < p < ∞.

Effects of Ultra-high Pressure Homogenization on the Emulsifying Properties of Whey Protein Isolates under Various pH

  • The effect of ultra-high pressure homogenization on the emulsifying properties of whey protein was investigated in a model emulsion made with whey protein isolate and soya oil under various pH. The emulsifying properties, the average diameter of the oil droplets ($d_{vs}$), and the protein load, were measured for each emulsion produced at different homogenization pressures (50 to 200 MPa) and pH values (4.6 to 8.0). According to the results of variance analysis and response surface, the pH had more influence on oil droplet size and protein load than homogenization pressure. The model equations, which were obtained by response surface analysis, show that pH and homogenization pressure had the major effect on oil droplet size and protein load. Higher homogenization pressure decreased the average droplet size and the protein load. Homogenization at high pressure, as opposed to low pressure, causes no overprocessing, but the effect was pH-dependent. The average diameter of the oil droplets increased slightly by decreasing the pH from 8.0 to 6.5 and then increased dramatically toward the isoelectric point of whey protein (i.e., at pH 4.6). Moreover associated droplets were found at acidic pH and their size was increased at high temperature.

  • The glass structure and antibacterial properties of $(65-x)ZnO-xB_2O_3-35P_2O_5$ glasses were investigated. Zinc borophosphate glasses were prepared using a conventional melt-quenching technique at $1000^{\circ}C$. Glass transition temperature and CTE were studied and the structure of zinc borophosphate glasses was evaluated by FTIR. The $Zn^{2+}$ state increase with increasing ZnO content was investigated by XPS and a single sharp Zn $2P_{3/2}$ peak was confirmed, showing that Zn $2P_{3/2}$ exists as $Zn^{2+}$. In order to to evaluate the antimicrobial activity, Escherichia coli (E. coli) was used following the Japanese Industrial Standard JIS Z 2801; the E. coli death rate was found to increase with increasing $Zn^{2+}$ content of glasses.

활엽수림, 침엽수림 및 혼효림 지역의 강우시 비점오염물질 유출특성 (Runoff Characteristics of Non-point Source Pollutants from Different Forest Types During Rainfall Events)

  • Long-term monitoring was conducted to identify the runoff characteristics of non-point source according to the three forest types (deciduous forest, coniferous forest and mixed forest) in this study. Rainfall events of each deciduous forest, coniferous forest, and mixed forest were 10, 8, 12, respectively. Average runoff depth and coefficients of each forest type were founded to be coniferous forest and were followed by others in turns : deciduous forest, and mixed forest because various conditions (i.e., rainfall property, Antecedent Precipitation Index (API), soil property, slope, and forest management) could change runoff characteristics. In the analysis of the first flush phenomenon, it showed that SS and T-P were sensitive for the first flush phenomenon. The first flush phenomenon of them were showed differently by rainfall intensity, rainfall duration, and amount of rainfall. The research results indicated that range of the Event Mean Concentration (EMC) values in deciduous forest were 0.8~2.4 mg/L for $BOD_5$, 2.0~13.4 mg/L for $COD_{Mn}$, 1.3~2.9 mg/L for DOC, 1.150~3.913 mg/L for T-N, 0.010~0.350 mg/L for T-P and 3.1~291.8 mg/L for SS and in coniferous forest were 0.8~2.2 mg/L for $BOD_5$, 1.9~3.6 mg/L for $COD_{Mn}$, 1.0~2.0 mg/L for DOC, 1.025~2.957 mg/L for T-N, 0.002~0.084 mg/L for T-P and 0.8~5.4 mg/L for SS. Also, range of the EMC values in mixed forest were 1.3~2.3 mg/L for $BOD_5$, 2.4~4.8 mg/L for $COD_{Mn}$, 1.1~2.1 mg/L for DOC, 0.385~2.703 mg/L for T-N, 0.016~0.080 mg/L for T-P and 2.3~30.0 mg/L for SS.

  • 다변량 정규모집단에서 공분산행렬의 m.l.e.를 유도하기 위하여 m.l.e.의 불변성에 기초를 둔 Anderson의 방법은 그 절차의 단순성이라는 측면에서 일반적으로 선호되고 있다. 본고는 공통요인분석모형이나 Simplex 구조모형과 같은 다변량구조모형에서 이 접근방법을 적용하여 요구되는 m.l.e.를 구하는 과정을 보이고 있다.
