제올라이트에 담지된 전이금속 촉매상에서 메탄의 이산화탄소 개질반응에 관한 연구 (A study on the Reforming of Methane by Carbon Dioxide on the Transition Metal Catalysts Supported Zeolite)

  • Nickel catalyst has been used for natural gas reforming with carbon dioxide, In this study, catalyst support used was HY zeolite. The optimum loading of Ni in the catalysts was 13 wt%. The effect of promoters, such as Mg, Mn, and K, was also studied. The addition of promoters to Ni catalyst improved the stability of catalysts and carbon deposition on Ni catalyst was suppressed. The reforming reactivity of promoter-added Ni catalyst was higher than that of Ni catalyst without any promoters. SEM, XRD, BET, TGA and FTIR tests were tried to characterize the catalyst structure before and after reaction.

합성유전자를 이용한 식물단백질의 향상 (Plant Protein Improvement by Synthetic Gene)

  • 식량으로 쓰이는 식물 단백질은 공통적으로 Isoleucine, Lysine, Methionine, Threonine, Tryptoplan등 5가지 필수아미노산이 결핍되어있다. 본 연구에서는 이러한 필수아미노산을 다량 함유한 단백질을 발현시킬 수 있는 합성유전자를 fekaqo에서 높은 수준으로 발현시키고자 강한 식물 promoter로 알려진 CaMV 35S, CaMV duplicate 35S promoters를 사용하였다. 형질 전환 및 재분화된 식물을 분석한 결과 본 합성 유전자가 식물 nuclear genome 안으로 도입을 안정하여 잘되었고 mRNA수준까지는 유의적인 증가를 보였으나 단백질 수준에서는 유의적 수준의 증가를 관찰할 수 없었다.

Optimization of the Viability of Probiotics in a Fermented Milk Drink by the Response Surface Method

  • Growth promoters were added to skim milk to retain the viability of Lactobacillus acidophilus and Bifidobacterium longum to help the product meet the "therapeutic minimum" at the time of consumption. The experiments were divided into two parts. The first part of the study used chicory inulin, isomalto-oligosaccharides and sucrose to investigate the effects of sugars on the activity of L. acidophilus and B. longum. The results indicated that the addition of isomalto-oligosaccharides stimulated growth of L. acidophilus and B. longum, resulting in a higher level of the probiotics after one month storage and yielded better $\beta$-galactosidase activity during fermentation. The second part studied the effects of three growth promoters on the viability of the probiotic cultures and the response surface method was employed to find the optimal ratio for addition of the growth promoters. The optimal ratio for added calcium gluconate, sodium gluconate and N-acetylglucosamine in fermented milk drinks were established. The response surface method proved to be a very effective way of optimizing the activity of probiotic cultures when developing a new fermented milk drink.

Current Progress in the Analysis of Transcriptional Regulation in the Industrially Valuable Microorganism Aspergillus oryzae

  • Aspergillus is considered to be an attractive host for heterologous protein production because of its safety and ability to secrete large amounts of proteins. In order to obtain high productivity, thus far promoters of amylases have been most widely used in A. oryzae. Recent progress in cloning and expression analysis, including EST sequencing, revealed that glycolytic genes represent some of those most strongly expressed in A. oryzae. Therefore, promoters of glycolytic genes could be important alternatives to promoters of amylases because lower amounts of proteases are produced in the presence of glucose. Several A. oryzae transcription factors responsible for the induction and/or maximum expression of many industrially important genes encoding amylases and proteases have been cloned and characterized. In addition to the transcriptional regulatory factors, the gene encoding the largest subunit of RNa polymerase II, constituting the basic transcription machinery, has also been cloned from A. oryzae. This recently acquired understanding of the details of transcriptional regulatory mechanisms and factors will facilitate engineering flexible controls for the expression of proteins important for the fermentation industries.

공여 균주인 알카리 내성 Bacillus속에 도입된 Promoter 의 특성 (Properties of Promoters Transferred to the Donor Strain, Alkali-tolerant Bacillus sp. YA-14.)

  • 토양에서 분리된 알카리 내성 Bacillus sp. YA-14의 chromosomal DNA로부터 B. subtilis 207-25 에 cloning된 promoter 유전자들을 그 공여 균주로 재형질전환시킬 수 있었다. 그 결과 promoter 유전자들은 공여 균주내에서 대부분의 특성들이 B. subtilis 207-25 내에서와 유사하였다. 재조합 plasmid p-12, p-l2Bl 및 p-l2B2의 CAT 비활성은 B. subtilis 207-25의 형질전환체보다 약간 높게 나타났으며 반면 pPL708의 CAT 비활성은 Bacillus sp. YA-14에서 오히려 상대적으로. 낮게 나타났다. Chloramphenicol에 의한 induction 정도도 월등하게 높은 것으로 나타나 이를 이용한 expression vector의 유용성이 높은 것으로 생각되었다.

사료 첨가 항생제 금지 전후 돼지 설사증 유래 대장균의 병원성 인자 및 항생제 내성 유전자 (Virulence and antimicrobial resistance genes of pathogenic Escherichia coli from piglets showing diarrhea before and after ban on antibiotic growth promoters in feed)

  • This study examined the prevalence of adherence factors, toxin genes, antimicrobial resistance phenotypes, and resistance genes in Escherichia coli (E. coli) isolated from piglets with diarrhea before and after the ban on antibiotic growth promoters (AGPs) in Korea from 2007 to 2018. In this period, pathogenic 474 E. coli isolates were obtained from diarrheic piglets. The virulence factors and antimicrobial resistance genes were assayed using a polymerase chain reaction, and the susceptibility to antibiotics was tested according to the Clinical and Laboratory Standards Institute guidelines. After the ban on AGPs, the frequency of F4 (12.5% to 32.7%) increased significantly, and LT (31.9% to 20.3%) and EAST-I (46.5% to 35.2%) decreased significantly. In addition, the resistance to streptomycin (45.8% to 67.9%), cephalothin (34.0% to 59.4%), and cefazlin (10.4% to 28.8%) increased significantly. Colistin resistance plasmid-mediated genes, mcr-1 and mcr-3, were detected after the ban on AGPs. The results of this study can provide useful data for analyzing the impact of the ban on AGPs on the virulence profiles and antimicrobial resistance of E. coli isolated from piglets with diarrhea in Korea.

형질전환된 담배 어린 싹의 녹화과정 중 Arabidopsis Cab 프로모터의 활성 차이 (Differential Activation of Arabidopsis Cab Promoters during Greening of Transgenic Tobacco Shoots)

  • Promoters of the chlorophyll a/b bidning protein genes, cab1, and cab2, of Arabidopsis thaliana were studied for their functions in differential expression during greening of etiolated shoots. The etiolated shoots were derived from leaves of transgenic tobacco plants with the cab-CAT (chloramphenicol acetyltransferase) translational fusions, and CAT activity was measured to monitor the activities of the cab promoters. Cab1 promoter activity increased rapidly and showed saturation after about 24 hours of greening, but that of cab2 increased with about 2 day-lag period and showed saturation after 6 days. Cab1 promoter activity was more sensitive to levulinic acid (LA) compared with cab2 activity. Cab2 promoter activity was inhibited more sensitively by chloramphynicol (CAP) than by inhibitors of Chl formation. Cab1 promoter activity was, however, inhibited less sensitively by CAP than by LA. The treatment of abscisic acid (ABA) did not block Chl synthesis so significantly as LA treatment did, and cab2 promoter activity was much less sensitive to ABA compared with that of cab1. These results suggest that cab1 expression is strongly related with Chl formation, possibly with $\delta$-aminolevulinic acid accumulation, and cab2 expression is suppressed more by the blockage of translation of Chl a-apoproteins than by the blockage of Chl a accumulation.

Structural Characteristics of Two Wheat Histone H2A Genes Encoding Distinct Types of Variants and Functional Differences in Their Promoter Activity

  • To investigate the regulation of plant histone H2A gene expression, we isolated two H2A genes (TH254 and TH274) from wheat, which encode different types of variants. Both genes had an intron in the coding region. In the promoters, some characteristics sequences, such as Oct and Nona motifs, which are conserved among plant histone genes were also found, and they were located in a short region (about 120 bp) upstream from the putative TATA box. Analyses of promoter activity with H2A-GUS fusion genes in the transient system using tobacco protoplasts revealed novel types of positive cis-acting sequences in the TH254 promoter: a direct repeat of a 13-bp sequence (AGTTACATTATTG) and a stretch composed of an AT-rich sequence (ATATAGAAAATTAAAA) and a G-box (CACGTG). A quantitative S1 assay of the mRNA amounts from the TH254/GUS and TH274/GUG chimeric genes in stably transformed and cell cycle-synchronized tobacco cell lines showed that the promoters of both genes contained at least one cis-acting element responsible for S phase-specific expression. Histochemical analysis of transgenic tobacco plants carrying the chimeric genes showed that the promoters of the two H2A genes were both active in developing seedlings and flower organs but regulated in different manner.

Association between expression levels and growth trait-related SNPs located in promoters of the MC4R and MSTN genes in Spinibarbus hollandi

  • Melanocortin 4 receptor: (MC4R) and Myostatin (MSTN) are two important growth trait-related genes in animals. In this study, we showed that two SNPs, MC4R-719A>G and MSTN-519C>T, found in the promoters of the MC4R and MSTN genes, respectively, are both associated with growth traits in Spinibarbus hollandi. Furthermore, we observed that there were significant associations between the expression levels of the MC4R and MSTN genes and these two growth trait-related SNPs. The expression level of MC4R gene in brain was lower in GG genotype fish with extremely high growth performance than that in AA genotype fish with extremely low growth performance. Expression level of the MSTN gene in muscle was lower in TT genotype fish with extremely high growth performance than that in CC and CT genotype fish with lower growth performance. The results indicated that these SNPs located in the promoters of MC4R and MSTN are associated with growth-related traits through modification of gene expression levels. The MSTN and MC4R SNPs may have useful application in effective marker-assisted selection aimed to increase output in S. hollandi.

Novel sinIR promoter for Bacillus subtilis DB104 recombinant protein expression system

  • Transcriptome analysis revealed that the sinR gene encoding a transition-state regulator of Bacillus pumilus, genetically close to B. subtilis, was expressed at high levels during growth. The sinR gene is the second gene of the sinIR operon consisting of three promoters and two structural genes in B. subtilis. This study used the sinIR promoter of B. subtilis DB104 to construct a recombinant protein expression system. First, the expression ability depending on the number of sinIR promoter was investigated using enhanced green fluorescent protein (eGFP). The expression level of eGFP was slightly higher when using two promoters (Psin2) than using original promoters. The Psin2 promoter was further engineered by modifying the repressor binding site and -35 and -10 regions. Shine-Dalgarno (SD) sequence of the sinI gene was modified to the consensus sequence. Finally, combining the engineered Psin2 promoter with the modified SD sequence increased the expression level of eGFP by about 13.4-fold over the original promoter. Our results suggest that the optimized sinIR promoter could be used as a novel tool for recombinant protein expression in B. subtilis.
