Property and Surface Morphology of Copper Foil on the Current Density (구리 박막의 표면형상과 물성에 대한 전류밀도 영향)

  • This study examined the effect of current density on the surface morphology and physical properties of copper plated on a polyimide (PI) film. The morphology, crystal structure, and electric characteristics of the electrodeposited copper foil were examined by scanning electron microscopy, X-ray diffraction, and a four-point probe, respectively. The surface roughness, crystal growth orientation and resistivity was controlled using current density. Large particles were observed on the surface of the copper layer electroplated onto a current density of 25 mA/$cm^2$. However, a uniform surface and lower resistivity were obtained with a current density of 10 mA/$cm^2$. One of the important properties of FCCL is the flexibility of the copper foil. High flexibility of FCCL was obtained at a low current density rather than a high current density. Moreover, a reasonable current density is 20 mA/$cm^2$ considering the productivity and mechanical properties of copper foil.

High Level Production of Supra Molecular Weight Poly(3-Hydroxybutyrate) by Metabolically Engineered Escherichia coli

  • The supra molecular weight poly(〔R〕-3-hydroxybutyrate) (PH B), having a molecular weight greater than 2 million Da, has recently been found to possess improved mechanical properties compared with the normal molecular weight PHB, which has a molecular weight of less than 1 million Da. However, applications for this PHB have been hampered due to the difficulty of its production. Reported here, is the development of a new metabolically engineered Escherichia coli strain and its fermentation for high level production of supra molecular weight PHB. Recombinant E. coli strains, harboring plasm ids of different copy numbers containing the Alcaligenes latus PHB biosynthesis genes, were cultured and the molecular weights of the accumulated PHB were compared. When the recombinant E. coli XL1-Blue, harboring a medium-copy-number pJC2 containing the A. latus PHB biosynthesis genes, was cultivated by fed-batch culture at pH 6.0, supra molecular weight PHB could be produced at up to 89.8 g/L with a productivity of 2.07 g PHB/L-h. The molecular weight of PHB obtained under these conditions was as high as 22 MDa, exceeding by an order of magnitude the molecular weight of PHB typically produced in Ralstonia eutropha or recombinant E. coli.

Ethanol Fermentation of Corn Starch by a Recombinant Saccharomyces cerevisiae Having Glucoamylase and $\alpha$-Amylase Activities

  • Starch is an abundant resource in plant biomass, and it should be hydrolyzed enzymatically into fermentable sugars for ethanol fermentation. A genetic recombinant yeast, Saccharomyces cerevisiae GA-7458, was constructed by integrating the structural gene of both $\alpha$-amylase from Bacillus stearothermophilus and the gene (STA1) encoding glucoamylase from S. diastaticus into the chromosome of S. cerevisiae SH7458. The recombinant yeast showed active enzymatic activities of $\alpha$-amylase and glucoamylase. The productivity of ethanol fermentation from the pH-controlled batch culture (pH 5.5) was 2.6 times greater than that of the pH-uncontrolled batch culture. Moreover, in a fed-batch culture, more ethanol was produced (13.2 g/L), and the production yield was 0.38 with 2% of corn starch. Importantly, the integrated plasmids were fully maintained during ethanol fermentation.

Enhancing the Reliability of MODIS Gross Primary Productivity (GPP) by Improving Input Data (입력자료 개선에 의한 MODIS 총일차생산성의 신뢰도 향상)

  • The Moderate Resolution Imaging Spectroradiometer (MODIS) regularly provides the eight-day gross primary productivity (GPP) at 1 km resolution. In this study, we evaluated the uncertainties of MODIS GPP caused by errors associated with the Data Assimilation Office (DAO) meteorology and a biophysical variable (fraction of absorbed photosynthetically active radiation, FPAR). In order to recalculate the improved GPP estimate, we employed ground weather station data and reconstructed cloud-free FPAR. The official MODIS GPP was evaluated as +17% higher than the improved GPP. The error associated with DAO meteorology was identified as the primary and the error from the cloud-contaminated FPAR as the secondary constituent in the integrative uncertainty. Among various biome types, the highest relative error of the official MODIS GPP to the improved GPP was found in the mixed forest biome with RE of 20% and the smallest errors were shown in crop land cover at 11%. Our results indicated that the uncertainty embedded in the official MODIS GPP product was considerable, indicating that the MODIS GPP needs to be reconstructed with the improved input data of daily surface meteorology and cloud-free FPAR in order to accurately monitor vegetation productivity in Korea.

Effects of Low-Serum Medium and Various Culture Additives on Production of Recombinant Human Erythropoietin in CHO Cell Cultures (CHO 세포 배양을 통한 Recombinant Human Erythropoietin의 생산에서 저혈청 배지와 배양 첨가물질이 미치는 영향)

  • Mammalian cell cultures have been used extensively to produce proteins for therapeutic agent because of their ability to perform post-translational modification including glycosylation. To produce recombinant protein, many factors and parameter are considered such as media composition, host cell type, and culture process. In this study, recombinant human erythropoietin (rhEPO) producing cell line was established by using glutamine synthetase system. To reduce serum concentration in media, we compared direct adaptation with step adaptation. Cell growth was faster in step adaptation. In low-level serum media, there were insufficient glucose for cell growth. Thus, we added glucose in low-level serum media from 2 g/L to 4.5 g/L. Titer of rhEPO was higher than other conditions at 4.5 g/L of glucose. Additionally, N-methyl-D-aspartate (NMDA), 13-cis-retinal, and pluronic F-68 (PF-68) were added to enhance productivity in CHO cell cultures. In conclusion, we applied CHO cell producing rhEPO to low-level of serum in media using step-adaptation. Also, we confirmed positive effect of NMDA, 13-cis-retinal, and PF-68.

  • The purpose of this study is to reduce the costs required to proactively prevent the turnover intention by the results of personality type of locus of control. which is the behavioral outcome of the members of the organization. Based on this problem consciousness, the present study focused on the personal characteristics of the members of the organization in the enterprise. Among the individual characteristics, the intentions, which are distinct from others, are called personality, Focusing on the locus of control among these types of characteristics of the organization members, the study about how these control positions affect their job involvement and their turnover intention. The results validated in this study are summarized as follows. Among the personality types of the organization's members, internals are found to have higher job involvement than externals. In addition, it was found that internals among the personality types of the members of the organization lower the meaning of turnover intention than external member. In order to overcome uncertain management environment and create and sustain a competitive advantage in the organization, management has a weight-oriented trend and Increasing job involvement in organizational members and lowering their turnover intention will lead to continued productivity, among other things, important management items that managers and managers should pursue. Given these aspects, it is deemed necessary to analyze the personality aspects of the members of the organization and to transform and manage a new management mindset that leads to the enhancement of the personality as an internal rather than an external one and the task immersion of the members of the organization based on it.

Assessment on Nitrous Oxide (N2O) Emissions different Nitrogen Application Rates during the Red Pepper Cultivation in Flat Upland

  • Generally, nitrogen (N) fertilization higher than the recommended dose is applied during vegetable cultivation for increasing in productivity. However, excessive N application rate beyond plant requirement could cause adverse environmental impact such as nitrate leaching and nitrous oxide emission. In this experiment, the impacts of N fertilization was studied on nitrous oxide ($N_2O$) emission to standardize the optimum fertilization level for minimizing of $N_2O$ emission as well as most of the crop productivity. Herein, we assessed the $N_2O$ emission in the flat upland soil which was cultivated with different N application rates on red pepper for 3 years (2010~2012). $N_2O$ emission was measured in chemical N fertilizer amounts 0 (N 0), 95 (N 0.5), 190 (N 1.0), $380(N_2.0)kgha^{-1}$ by using the abnormal shape chamber closed repeating three times. In average for 3 years, the total $N_2O$ emissions of each treatment in field of soybean were 2.110 (N 0), 3.165 (N 0.5), 5.039 (N 1.0), and $7.228(N_2.0)kgN_2Oha^{-1}yr^{-1}$, respectively. And then the primary regression between nitrogen fertilizer amount and the total $N_2O$ emission was showed as y = 0.0138x + 2.0942 ($r^2=0.9885$), and an average of the emission factor was $EF_1$ 0.0148(0.0118~0.0191) $N_2O-NkgN^{-1}kg^{-1}$ from 2010 to 2012. The result was a little higher than the emission default of the IPCC 1996 Guideline ($EF_1$ 0.0125) when the results are converted into $N_2O$ emission factor.

  • The effects of glucose concentration on the production of PHB by fed-batch culture of Ralstonia eutropha were investigated. In the range of glucose concentration of $2.5\;{\sim}\;40\;g/l$, it was found that the high glucose concentration was not favorable for the PHB formation after the phosphate limitation. It was further confirmed by the specific PHB synthesis rates and yields. The PHB concentration decreased much with the increase of glucose concentration. But if the glucose concentration was very low, e.g. 2.5 g/l, the cell growth and PHB synthesis also could be limited because of inadequate glucose supply. Itwould be better to maintain the glucose concentration at about 9.0 g/l to obtain high DCW, PHB concentration and productivity.

  • As enterprises, based on the forecast of the customer's demand and collaboration with the suppliers, establish the integrated system directing supply, production, and distribution for the increase of productivity, Thus, this study intends to find the most urgent and critical factors for the improvement of the information system by externalizing factors affecting the operation of information system, suggest the process to improve the relevant functions of information system, and design the process. As a result of the analysis of the previous studies on the improvement of the information system, many studies were conducted on the improvement of ERP and SCM, yet there was no study conducted targeting about APS (Advanced Planning & Scheduling). Thus, this study chose APS as the subject for the design of the process for the improvement for the information system.

Effect of Acetic Acid Pretreatment on Drought Stressed Alfalfa Plants

  • Drought stress is one of the major factors that reduce plant growth and productivity. This study was conducted to investigate the effect of exogenous acetic acid pretreatment on drought stress tolerance response in plants. Fourteen-day-old alfalfa plants were pretreated with 15 mM acetic acid, and then subsequently subjected to drought stress for 6 days. The fresh weight and relative water content in the leaves of acetic acid pretreated alfalfa plants were increased compared to the control group. The chlorophyll and carotenoid contents were slightly decreased in the acetic acid treatment. The H2O2 and proline contents were also significantly decreased in the acetic acid treatment. These results suggest that the scavenging mechanism of reactive oxygen species in alfalfa activated by acetic acid pretreatment is involved in conferring tolerance to drought stress.
