Pharmacological Activity of Chaga Mushroom on Extraction Conditions and Immunostimulating Polysaccharide (추출조건에 따른 차가버섯 생리활성 및 면역활성 다당)

  • To investigate the pharmacological activity of chaga mushroom (Inonotus obliquus) on extraction conditions, chaga was extracted using water (reflux at $50^{\circ}C$, decoction over $90^{\circ}C$, pressure at $121^{\circ}C$) or ethanol (reflux at 50, 70, or $90^{\circ}C$). When water extract was further fractionated into crude polysaccharide (IO-CP), yields of IO-CP (4.8~16.8%) were higher than those of ethanolic extracts (IO-E, 1.9~2.7%) at increased temperature. For antioxidant activity, crude polysaccharide (IO-CP-121) obtained by pressurized extraction showed the highest polyphenolic and flavonoid contents (35.10 mg TAE/g and 18.48 mg QE/g, respectively) as well as DPPH and ABTS free radical scavenging activities (26.08 and 27.99 mg AEAC/100 mg, respectively). Meanwhile, IO-CP-D (decoction) and IO-CP-50 (reflux) had more potent mitogenic effects (2.10- and 1.95-fold of saline control at 100 ${\mu}g/mL$) as well as intestinal immune system modulating activities (6.30- and 5.74-fold) compared to IO-CP-121, whereas ethanolic extracts showed no activity. Although no IO-CP showed cytotoxicity against RAW 264.7 cells at 0.1 mg/mL, IO-CP-121 significantly inhibited TNF-${\alpha}$ and NO production as pro-inflammatory factors in LPS-stimulated RAW 264.7 cells (29.2 and 63.5%, respectively). Ethanolic extracts also showed no cytotoxicity at 0.1 mg/mL, whereas inhibition of TNF-${\alpha}$ and NO production was significantly low compared to that of IO-CP-121. In addition, active IO-CP-D was further fractionated into an unadsorbed (IO-CP-I) and seven adsorbed fractions (IO-CP-II~VIII) by DEAE-Sepharose CL-6B column chromatography in order to isolate immunostimulating polysaccharide. IO-CP-II showed the most potent mitogenic effect and macrophage stimulating activity (4.51- and 1.64-fold, respectively). IO-CP-II mainly contained neutral sugars (61.86%) in addition to a small amount of uronic acid (2.96%), and component sugar analysis showed that IO-CP-II consisted mainly of Glc, Gal, and Man (molar ratio of 1.00:0.55:0.31). Therefore, extraction conditions affect the physiological activity of chaga, and immunostimulating polysaccharide fractionated from chaga by decoction is composed mainly of neutral sugars.

Studies on the Effect of some Chemical Soil Components in relation to Rice Yield in Heavy Textured Paddy Soils developed on Alluvial Terrace (홍적대지에 발달된 중점질 논토양에서 벼 수량에 미치는 수종 화학성분의 영향에 관한 연구)

  • In order to study the effects of some chemical components of surface soils on the paddy yield in clayey Whadong series developed on old alluvium, NPK fertilizer experiments conducted from 1966 to 1969 were reviewed and discussed, and the results may be summarized as follows. 1. The paddy rice production of Whadong series without fertilizers varied from 156kg to 719kg per 10a. 2. The paddy yields in Whadong series were associated mainly with the contents of organic matter and available phosphorus in surface soils, but not with those of exchangeable K, Ca, and Mg and cation exchange capacity. 3. The contents of organic matter in these soils more effected in the paddy yield than those of phosphorus did. 4. In case less than 2.0% or more than 3.0% of organic matter in surface soils the effect of phosphate application was appreciable. The effect, however, was not recognized from 2.0 to 2.9% of organic matter. And the following suggestions were able to make. a. In case of less than 2.0% of organic matter, the effect was considered to be due to deficiency of available phosphorus in the soil. b. In case of more than 3.0% of organic matter, the greater effect was considered to be due to inhibition of P uptake even in higher P contents in soil. c. Consequently, correlation study of P testing in paddy soil should be limited to the soils which contains less than 2% of organic matter. d. If the contents of organic matter in paddy soils were above 3.0%, the effect of P application was considerable and considered to be due to inhibition of nutrient uptake. Accordingly, it is considered that only the increased application of P does not improve the production of paddy in such soils.

Assessment of Productivity and Vulnerability of Climate Impacts of Forage Corn (Kwangpyeongok) Due to Climate Change in Central Korea (국내 중부지역에 있어서 기후변화에 따른 사료용 옥수수의 생산성 및 기후영향취약성 평가)

  • A two-year study was conducted from 2017 to 2018 by the establishment of a test field at Chungju-si and Cheongyang-gun. Plant height, number of leaves, insects and diseases, and fresh and dry matter yields for corn hybrid('Kwangpyeongok') were investigated. Daily average, maximum, and minimum temperature, monthly average temperature, daily precipitation, and sunshine duration during the growing season were investigated. We selected climate-critical factors to corn productivity and conducted an evaluation of vulnerability to climate change from 1999 to 2018 for both regions. In 2018, the dry matter yield of forage corn was 6,475 and 7,511 kg/ha in Chungju and Cheongyang, respectively, which was half of that in 2017. The high temperature and drought phenomenon in the 2018 summer caused the corn yield to be low. As well as temperature, precipitation is an important climatic factor in corn production. As a result of climate impact vulnerability assessment, the vulnerability has increased recently compared to the past. It is anticipated that if the high temperature phenomenon and drought caused by climate change continues, a damage in corn production will occur.

Application Effects of Organic Fertilizer Utilizing Livestock Horn Meal as Domestic Organic Resource on the Growth and Crop Yields (국내산 유기자원 우각을 활용한 유기질비료의 작물 생육 및 수량에 미치는 영향)

  • Objective of this study was to develop an organic fertilizer utilizing domestic livestock horn meal and to investigate the application effect of rice and eggplant. The possibility of utilization of livestock horn meal as an organic resource to replace imported expeller cake fertilizer was examined. In order to select domestic organic resources with high nitrogen content, 8 kinds of organic matter such as chicken manure, fish meal, soybean meal, sesame meal, perilla meal, blood meal, livestock horn meal, and beer sludge were analyzed and organic resources with high nitrogen content were selected. In addition, the conditions for the production of organic fertilizers that can be used in organic agriculture were established by mixing of the rice husk biochar and the rice bran as the supplements with the raw materials for mixing ratios. The content of total nitrogen (T-N) in the livestock horn meal was 12.0 %, which was the next low in 13.5 % blood meal. The content of total nitrogen was 5.9 ~ 7.9 % in fish meal and oil cakes. Total nitrogen content of non-antibiotic chicken manure for organic farming was 3 % and nitrogen content in beer sludge was 3.5 %. Organic fertilizer was produced by using biochar, rice bran as a main ingredient of non-antibiotic chicken manure, livestock horn meal and beer sludge. Compared to nitrogen content (4.0 to 4.2 %) of imported expeller cake fertilizer (ECF), the nitrogen content of organic fertilizer utilizing domestic livestock horn meal is as high as 7.5 %. The developed organic fertilizer is met as Zn 400 mg/kg, Cu 120 mg/kg the quality of organic agricultural materials such as or less. To investigate the effect of fertilizer application on the crops, prototypes of developed organic fertilizer were used for the experiment under selected conditions. As a result of application the developed organic livestock horn meal fertilizer (LHMF) for cultivation of the rice and eggplant, the application quantity of the developed organic LHMF 100 % was decreased by 40 % compared to that of the mixed expeller cake fertilizer (MECF). The application of LHMF, which refers to the application rate corresponding to the nitrogen fertilization recommended by the soil test, was reduced by 40% compared to the application rate of MECF, but the same results were obtained in crop growth and yield. The selection of a new high concentration nitrogen source utilizing domestic organic resources and the development of organic fertilizer is the starting point of the research for substitution of imported ECF using domestic local resources at the present time that the spread of eco-friendly agriculture is becoming increasingly important. If it is expanded in the future, it is expected to contribute to the stable production of eco-friendly agricultural products.

Application Effects of Biochar Derived from Pruned Stems of Pear Tree on Growth of Crops and Soil Physico-chemical Properties (배 전정지 바이오차 시용이 작물 생육 및 토양이화학성에 미치는 영향)

  • This study was conducted to develop the manufacturing method of biochar using pruned stems of pear tree and its application effect on the crop growth and soil physico-chemical properties. In this study, biochar derived from pruned stems of pear tree at heating temperature of $300^{\circ}C$, $500^{\circ}C$ and $700^{\circ}C$ in heating times of 2, 3 and 4 hours, were tested in the changes of their chemical properties during biochar processing. The pH, Exch. K, Exch. Mg and cation exchange capacity (CEC) increased as the pyrolysis temperature increased during the production of biochar, and the change of these properties rapidly occurred at $500^{\circ}C$. However, as the pyrolysis temperature increased, ash content increased and total carbon (T-C), yield decreased. And the change of the properties in response to the heating time was not shown. It was thought that it would be desirable to set the production conditions of biochar at $500^{\circ}C$ for 2 hours in consideration of the change of chemical properties and the ash content and yield. And also, were conducted the experiments to establish manufacturing method of farm-made biochar using drum biochar manufacturing machine and investigate the application effects of biochar on the cultivation of chinese cabbage and tomato. Application of biochar derived from pruned stems of pear tree could enhance pH, organic matter (OM), total carbon (T-C) of soil. On the other hand, soil electrical conductivity (EC), NO3-N were lowered compared to the control which has no application. The bulk density, porosity and aggregate formation of soil were improved by biochar application. The fresh matter yields of chinese cabbage and tomato were significantly increased in proportion to the application rate of biochar. This study demonstrated the effect of the biochar derived from agricultural byproduct to be as a low cost potential soil ameliorant by physico-chemical properties in eco-friendly greenhouse cultivation.

Analysis of Physicochemical Properties of Red Ginseng Powder Based on Particle Size (홍삼분말 입자크기에 따른 이화학적 특성 분석)

  • Most of the red ginseng (RG) products contain active substances derived from hot water or alcohol extraction. Since active substances of RG are divided into two types - water-soluble and liposoluble - water or alcohol is needed as an extraction solvent and this leads the different extraction yields and components of the active substances. To overcome the limit, whole red ginseng powder can be used and consumed by consumers. In this study, the physicochemical properties and extractable active substance contents of variable-sized RG powder ($158.00{\mu}m$, $8.45{\mu}m$, and $6.33{\mu}m$) were analyzed, and dispersion stability was measured to investigate the suitable size of RG powder for industrial processing. In the results, no significant difference was found from the changes in color intensity and thiobarbutric acid tests at $4^{\circ}C$, $25^{\circ}C$, and $40^{\circ}C$ for 4 weeks. There was no significant difference on the production of antioxidants and ginsenoside among the samples (p>0.05). In dispersion stability, $RG-158.00{\mu}m$ was precipitated immediately, and the dispersion stabilities between $RG-8.45{\mu}m$ and $RG-6.33{\mu}m$ showed no significant difference. It implies that fine RG is suitable for the production process. With further study, it seemed that the physicochemical effects of RG particle sizes can be clearly revealed.

An Optimum Summer Cultivation Sowing Date for Seed Production of Oats (Avena sativa L.) (귀리 종자 생산을 위한 여름 재배의 적정 파종 시기 구명)

  • This study was conducted to determine the optimal sowing date for seed yield of summer oat (Avena sativa L.) cultivars "Darkhorse (DH)" and "Highspeed (HS)" in Wanju, Jeonbuk province between 2017 and 2018. We investigated seed yield from 4 sowing dates: July 15, July 30, August 15, and August 30. We evaluated the agronomic characteristics of summer oats (DH and HS). We found the heading date of all cultivars to be within 50 days. Delayed sowing resulted in significantly increased plant height for both years and cultivars. There was no significant difference in spike length of DH and HS which ranged from 12.8 to 17.8 cm. The sowing date of July 30 produced a higher number of grains per spike, but this yield differed significantly by year and cultivars. In 2017, the first sowing resulted in the lowest DH yield at 132 kg per 10a, while the second sowing had the highest yield at 227 kg. HS yield was the lowest in the first sowing at 126 kg and the highest in the third sowing at 219 kg. In 2018, DH had the lowest yield from the first sowing at 184 kg per 10a, and the highest from the second sowing at 240 kg, but there was no significant difference between these yields. The first sowing for HS gave the lowest yield at 160 kg, and the second sowing produced the highest at 258 kg. The germination rate of harvested seeds from each sowing date in 2017 and 2018 was found to be higher than 85% and there was no significant difference between the two cultivars in the 2018 germination rate test. Thus, we found the optimal sowing date for summer cultivation of oats for the highest seed yield to be between July 30 (second sowing) and August 15 (third sowing).

Ethanol extract of Aster glehni exhibits anti-inflammatory and anti-oxidant effects in RAW 264.7 cells and Caenorhabditis elegans (섬쑥부쟁이 에탄올 추출물이 대식세포와 예쁜꼬마선충에서의 항염증 및 항산화 효과 )

  • This study investigated the anti-oxidative and anti-inflammatory effects of Aster glehni (AG) extract in RAW 264.7 cells and Caenorhabditis elegans. The total polyphenol and flavonoid contents were higher in the ethanol extracts than in the hot water extracts. As a result of measuring the moisture contents (%) and extraction yields (%) of AG and drying A. glehni for processing (DAG), 70% ethanol, which has the highest percentage of extraction yield, was selected as the final solvent. DPPH radical scavenging activity showed higher antioxidant activity of ethanol extracts of DAG than AG. The cytotoxicity assay of the AG or DAG ethanol extracts was treated at different concentrations (25, 50, and 100 ㎍/mL), and cell viability rates were higher than 80% at all concentrations. The LPS-stimulated nitric oxide (NO) production in RAW 264.7 was significantly reduced at all concentrations of AG and DAG groups. As a result of measuring the gene expression of iNOS, which induces NO production, the AG or DAG group decreased by 33% and 32%, compared with the phosphate buffer saline (PBS) group. Under inflammatory stress conditions, the survival rate of C. elegans treated with AG or DAG ethanol extract with LPS showed concentration-dependent improvement in survival rate compared with the PBS group. Considering these results, AG could potentially be developed as an antioxidant and anti-inflammatory functional food material.

Effect of Calving Season on Postpartum Milk Production and Persistency of TMR Fed Holstein Heifers (분만계절이 TMR 급여 홀스타인 육성우의 분만 후 유생산과 비유지속성에 미치는 영향)

  • Total of 20 Holstein calves of 10 calves (3.90±0.26 month of age) born in spring (S) and 10 calves (4.10±0.30 month of age) born in fall (F) were reared in this study for 24 months and diets were divided into separate feeding of forage and concentrates (C) and TMR (T). Therefore, 4 treatments in this study were composed of CS, CF, TS and TF with the factors of diets and calving season. After parturition of heifers, all animals were fed the same diet and milk production was recorded monthly. DM intakes in growing period were influenced by calving season, and those of the animals calved in fall were higher than in those calved in spring (P<0.01), but there were no significant differences by feeding method. CP intakes and TDN intakes were significantly influenced by calving season (P<0.05) and feeding method (P<0.001), and the animals calved in fall were about 1.2% higher than those calved in spring, and the animals fed TMR were about 4.7% higher than those fed concentrates and forage separately. Average, 9th and 10th months' milk yields were significantly influenced by feeding method in which those in the treatments fed TMR (TS, TF) were higher than in separate feeding of concentrates and forage (CS, CF; average P<0.05; 9th and 10th months P<0.01). Average milk persistency was also significantly influenced by calving season (P<0.05) and feeding method (P<0.01) and those in the animals calved in fall were higher than in spring and those of the TMR fed animals were also higher than in separate feeding of concentrates and forage. Milk persistency was similar to the results of milk yield, showing statistically significant differences affected by the feeding method at 9th and 10th months of late lactation (P<0.01), and it was about 8% higher in the animals fed TMR, showing higher tendency at 7th (P=0.12) and 8th months of late lactation (P=0.09). Therefore, it is expected that postpartum milk yield and milk persistency would be higher when the hiefers are fed TMR in growing period and calved in fall. Average milk fat content was influenced by feeding method. Milk fat content of the animals fed TMR during growing period were 7.8% higher than those fed concentrates and forage separately (P<0.01). This suggests that feeding TMR during growing period influenced first postpartum eating behavior, which stabilized the rumen and resulted in the increased milk fat. At 3rd month after calving, milk fat content was lower in the animals calved in spring than in those calved in fall, suggesting that it might have been influenced by the seasonal differences. MUN showed significant differences by feeding method in which those in separate feeding of concentrates and forages were higher especially in average, 4th, 5th and 6th months (average and 4th P<0.01; 5th and 6th months P<0.05). SCC was higher in the animals fed TMR than in those fed concentrates and forage separately especially in average, 3rd and 4th months after calving (P<0.01). In conclusion, when feeding TMR during growing period and calving in fall, it was not influenced by the high temperature in summer, and it resulted in the improved milk yield, milk persistency and milk fat content.

Application Effects of Some Nitrogen Fertilizers Forms for the Growth and Yield of Rice Plant (몇가지 형태(形態)의 질소비료시비(窒素肥料施肥)가 수도(水稻)의 생육(生育) 및 수량(收量)에 미치는 영향(影響))

  • Nitrogen fertilizer effectiveness on rice production was studied to evaluate the different forms and sources. Seven kind of nitrogen fertilizers were applied in two levels, 15 and 30 kg per 10a on Jinjubyeo (Japonica type) in silt loam paddy soil of pot cultivation. The results were summerized as follows; 1. pH in soils was decreased with enhancement of ammonium sulfate application in $NH_4-N$, but it was increased with times after nitric-acid application and PH change in soil was not remarkable when $NO_3-N$ with accessory component was applied. 2. $NH_4-N$ contents in soil were the lowest at 2 weeks after application in N 15kg/10a regardless of different sources of nitrogen fertilizer. $NO_3-N$, in N 30kg/10a, was decreased continuously until 4 weeks, while $NH_4-N$, Urea-N were at minimum during 2-3 weeks. 3. Growth of culm length and straw weight applied with AN (Ammonium Nitrate), AS (Ammonium Sulfate) and urea were superior to the form of nitrate. While NA (Nitric Acid), PN (Potassium Nitrate) and CN (Calcium Nitrate) plot of the $NO_3-N$ was the dominant fertilizers for root elongation. 4. Brown rice yields were increased dominantly by $NH_4-N$ application such as AS or AP than $NO_3-N$ pot. But the yields in case of $NO_3-N$ application CN, PN and NA were decreased. 5. N, P, Mg and Mn content of straw ranked the effectiveness of nitrogen forms as $NH_4-N$, Urea-N and $NH_4-N+NO_3-N$, while K, Ca and $SiO_2$ content of straw in $NO_3-N$ fertilizer plot were high while N, P, Mg, Mn, Fe and Mg were low. 6. Increament of nitrogen absorption in straw was stimulated by enhancement of phosphorous absorption and the growth and yield of rice plant were increased. Absorption of N, P, Ca and Mg was decreased by CN application. Absorption of N, P and Mg also was decreased by $NO_3-N$ application and N, P, Mg or Ca content were seemed to simulated the growth and yield of rice plant. 7. $SiO_2$, Zn and Fe contents of the root at harvest stage were higher than those of the straw. N, P, Mg, Mn, Zn and Fe contents were high in $NH_4-N$ and Urea treatment. While K, ca and $SiO_2$ contents, however, were high in $NO_3-N$ treatment.
