Bacillus substilits에 의한 치자황색소 부산물로부터 치자청색소의 생산 (Production of Gardenia Blue Color from Gardenia Waste by the Bacillus subtilis)

  • For waste recyle, we were investigated on Gardenia blue color production using Gardenia by-product by Bacillus subtilits. Optimum conditions for producing blue pigment were found to be 30$^{\circ}C$, initial pH 6.5, glucose as a carbon source 3% and yeast extract as a nitrogen source 0.5%, respectively. Optimum conditions for fermentor culture were agitation speed 400rpm, aeration 2 vvm and inoculum 5%. The optimum perculture time for inoculum was 20 hrs for blue pigment production.

부영양화해역의 내부생산효율에 대한 계절변동예측 (Prediction of Seasonal Variations on Primary Production Efficiency in a Eutrophicated Bay)

  • The Primary Production of phytoplanktons produces organic matter in high concentration in eutrophicated Hakata Bay, Japan, even during the winter season in spite of low water temperature. Phytoplanktons are considered to have any biological capabilities to keep activities of photosynthesis under the unfavorable conditions, and this affects water quality of the bay. In this study, seasonal variations in primary production efficiency were predicted by using a simple box-type ecosystem model, which introduced the concept of efficiency for absorption of solar radiation energy in relation to growth of phytoplanktons under the low solar radiation intensity. According to the simulation result of primary production, it was organic pollution comes from dissolved organic carbon (DOC) throughout the year, DOC of which is originated from the primary production of phytoplanktons on biological response of the seasonal variation of ambient conditions.

Porphyridium cruentum의 성장 및 당질 생산에 미치는 배양 조건의 영향 (Effect of Culture Conditions on Production of Polysaccharides and Growth Rate of Porphyridium cruentum)

  • The growth of Porphyridium cruentum and its porphyran polysaccharide production were measured as functions of light intensity, temperature, light quality (fluorescent, blue, red, and green) and nitrate concentration. The optimum light intensity, temperature, and nitrate concentration for the growth of Porphyridium cruentum and for its polysaccharide production were 1,400 lx, $25^{\circ}C$, and 0.03%, respectively. The maximum cell concentration and polysaccharide content under the optimum conditions were 1.95 and 0.23 mg/mL, respectively. Light quality did not influence growth or polysaccharide production. The best results for growth and polysaccharide production were obtained using fluorescent light.

  • For polyhydroxyalkanoic acid (PHA) production, several microorganisms were isolated from sewage sludge. One of them, GB-77 strain, was chosen from its PHB/HV copolymer production on only fructose without cosubstrate. The isolated strain GB-77 was identified as the genus Alcaligenes. Optimal temperature and pH for cell growth were 36C and 6.8. Optimal medium composition was 10 g/l of fructose and 5 g/l of polypeptone, 1 $\times$ 10$^{-2}$M Na$^{2}$HP0$^{4}$, 1.3 $\times$ 10$^{-2}$M KH$^{2}$PO$^{4}$. To investigate the optimal condition for polyhydroxyalkanoic acid production two-stage culture technique was used; first stage for cell growth and second stage for PHA production on unbalanced growth conditions. Optimal conditions for high PHA production were C/N ratio 50, temperature 36$\circ$C and pH 6.8. To overcome fructose inhibition on cell growth, intermittent feeding fed-batch culture technique was used. Total cell concentration was 17.4 g/l with 9.1 g/l of PHA. The purified PHA was identified PHB/HV copolymer by NMR analysis.

  • The aim of the current study is to evaluate the effects of medium compositions on the production of protease inhibitor in Streptomyces sp. SMF13. The production of protease inhibitor was counter-currently linked to extra-cellular protease, which were regulated by the culture conditions. Nitrogen source was the most critical ingredient affecting the production of protease inhibitor and protease. Carbon source was an important factor to determine the culture pH which affected very clearly the formation of protease and protease inhibitor. Inorganic phosphate inhibited the protease inhibitor production which was linked to the cell growth rate, although the optimal conditions for the production of protease inhibitor were not favouring to the cell growth.

  • This paper describes a procedure for optimizing the route selection and production conditions in alternate process plans under a cellular manufacturing environment. The type of production is mainly production-to-order which deals with unexpected products as the changes factor. The flexible cellular manufacturing can be viewed as a complete unification of both flexible manufacturing process and flexible production management. The integrated problem for designing flexible cellular manufacturing associated with determining the optimal values of the machining speeds, overtime, and intercell flow is formulated as Nonlinear Mixed Integer Programming(NMIP) in order to minimize total production change cost. This is achieved by introducing the marginal cost analysis into the NMIP, which will compute the optimal machining speed, overtime, intercell flow, and routing. The application of this procedure offers greater flexibility to take advantage of the cellular manufacturing due to the optimum use of resources. A solution procedure for this problem was developed and a numerical example is included.

발효사료의 생산에 관한 연구 2 (Studies on the Production of Fermented Feed (2))

  • In solid culture of Endomycopsis fibuliger No.55, Eudomuopsis javanensis No.112 and Candida tropicalis No.340, the conditions of enzyme (protease, anylase and cellulase) production and the influence of addition of $(NH_4)_2;SO_4$ were examined, and the results obtained were as follows. 10 Wheat bran medium is found to be the best on the enzyme production in case of simple material. The optimum conditions ; are water content added 100 to 120%, temperature 25 to $80^{\circ}C$ and incubation times 2 to 3 days. 2) The cellulase production was scarely produced in the case of Endomyopsis fibuliger No.55, as well as, the amylase production was scarely producted in the case of Endomycopsis javanensis No.112 and Candida tropicalis No.340. 3) The enzyme production was remarkably increased when 5% of$(NH_4)_2;SO_4$ as inorganic nitrogen sources was admixed to wheat bran. 4) When 5% of $(NH_4)_2;SO_4$ was admixed to medium, the ratio of protein increase was 10.2 to 17.7% in wheat bran medium and 10.6 to 17.9% in sweet potato cake medium.

수소생산을 위한 Rhodopseudomonas sphaeroides의 최적 배양조건 (Optimum Culture Conditions for Hydrogen Production of Rhodopseudomonas sphaeroides)

  • We examined optimum culture conditions of Rhodopseudomonas sphaeroides B5 for effective utilization of substrate and sunlight for hydrogen production. The optimum concentration range of DL-lactate as electron donor for hydrogen production by resting cells was from 5 to 50mM, and optimun CN ratio (lactate/glutamat) for maintenence of hydrogen production activity by growing cultures was from 5 to 6. Hydrogen production by the cultures of low cell density (0.36mg/ml dry cells) was saturated with 10 Klux light intensity. Under constant illumination of 50Klux which was set up as the average medium value of annual variation of sunlight intensity, hydrogen production with various cell densities in the culture resulted in highest production rate (132${\mu}$l/hr/mg dry cells) up to 0.64mg/ml dry cells. However, the amount of total hydrogen production was saturated with cell density of 2.1mg/ml dry cells. In addition to these, the optimum inner thickness pervious to light of the culture vessel for hydrogen production which was measured under sunlight was 5 cm.

Enhanced Production of Epothilone by Immobilized Sorangium cellulosum in Porous Ceramics

  • Epothilone, which is produced by the myxobacterium Sorangium cellulosum, contributes significant value in medicinal development. However, under submerged culture conditions, S. cellulosum will accumulate to form bacterial clumps, which hinder nutrient and metabolite transportation. Therefore, the production of epothilone by liquid fermentation is limited. In this study, diatomite-based porous ceramics were made from diatomite, paraffin, and poremaking agent (saw dust). Appropriate methods to modify the porous ceramics were also identified. After optimizing the preparation and modification conditions, we determined the optimal prescription to prepare high-performance porous ceramics. The structure of porous ceramics can provide a solid surface area where S. cellulosum can grow and metabolize to prevent the formation of bacterial clumps. S. cellulosum cells that do not form clumps will change their erratic metabolic behavior under submerged culture conditions. As a result, the unstable production of epothilone by this strain can be changed in the fermentation process, and the purpose of increasing epothilone production can be achieved. After 8 days of fermentation under optimized conditions, the epothilone yield reached 90.2 mg/l, which was increased four times compared with the fermentation without porous ceramics.

모바일 스마트 기기 덮개 유리 순차이송형 성형기기의 가열시스템 최적화에 관한 연구 (Study on Optimization for Heating System of Sequential Feed-Type Mobile Smart Device Cover Glass Molding Machine)

  • Nowadays, flat-shaped cover glass is widely used for mobile devices. However, for its good design and convenience of use, curved cover glass has been demanded. Thus, many companies have tried to produce curved cover glass through the shaving technique, but the production efficiency is very low. Therefore, the molding technique has been adopted to increase the efficiency for the curved-glass production system. For a glass-molding system, several heating blocks are installed, and the flat cover glass is sequentially heated and molded. The production time for the cover glass is very different depending on the heating conditions; thus, the prediction of the production time for different heating conditions should be needed. Therefore, in this study, the computations were performed with different heating conditions (uniform and non-uniform) in the present cover glass-molding machine. For uniform and non-uniform heating conditions, the simple correlation between the heating time and the heater capacity and the heating time to achieve higher durability can be suggested, respectively.
