Preparation and Characteristics of Bread by Medicinal Herb Composites with Cognitive Function (인지기능활성을 가진 생약복합물을 이용한 빵의 제조 및 특성)

  • In this study, the breads with medicinal herbs (MH) composites showing cognitive function were prepared and their characteristics were examined. Fifteen kinds of medicinal herbs were extracted with hot water and divided into 3 groups (MH-1, MH-2, MH-3) based on their contents. All groups showed cognitive function in terms of promoting muscarinic receptor, NMDA receptor and acetylcholinesterase inhibition. In the preparation of breads containing MH extracts of various contents (0, 10, 20, 30, and 50%), there were no significant difference among loaf weight of all groups, but loaf volume of all groups were significantly (p<0.05) decreased with increasing content of MH extracts compared to the control group. The "a" and "b" values of bread crumb increased with the content of MH extracts while "L" value decreased, but these values of bread crust were similar to the control group. The most improvements in hardness, fracturability, cohesiveness, gumminess and chewiness of bread were noticed by the addition of 10$\sim$20% MH extracts, while adhesiveness and springiness of bread were the most by the 30% addition ones. Through the sensory evaluation, it was revealed that appearance, mouth feeling, taste, overall preference and crumb texture of bread were not significantly different (p<0.05) between control and those with the 10% addition ones. Therefore the 10% addition ones, which got similar scores to control, will make the most desirable product.

Cordycepin inhibits lipopolysaccharide-induced cell migration and invasion in human colorectal carcinoma HCT-116 cells through down-regulation of prostaglandin E2 receptor EP4

  • Prostaglandin $E_2$ ($PGE_2$), a major product of cyclooxygenase-2 (COX-2), plays an important role in the carcinogenesis of many solid tumors, including colorectal cancer. Because $PGE_2$ functions by signaling through $PGE_2$ receptors (EPs), which regulate tumor cell growth, invasion, and migration, there has been a growing amount of interest in the therapeutic potential of targeting EPs. In the present study, we investigated the role of EP4 on the effectiveness of cordycepin in inhibiting the migration and invasion of HCT116 human colorectal carcinoma cells. Our data indicate that cordycepin suppressed lipopolysaccharide (LPS)-enhanced cell migration and invasion through the inactivation of matrix metalloproteinase (MMP)-9 as well as the down-regulation of COX-2 expression and $PGE_2$ production. These events were shown to be associated with the inactivation of EP4 and activation of AMP-activated protein kinase (AMPK). Moreover, the EP4 antagonist AH23848 prevented LPS-induced MMP-9 expression and cell invasion in HCT116 cells. However, the AMPK inhibitor, compound C, as well as AMPK knockdown via siRNA, attenuated the cordycepin-induced inhibition of EP4 expression. Cordycepin treatment also reduced the activation of CREB. These findings indicate that cordycepin suppresses the migration and invasion of HCT116 cells through modulating EP4 expression and the AMPK-CREB signaling pathway. Therefore, cordycepin has the potential to serve as a potent anti-cancer agent in therapeutic strategies against colorectal cancer metastasis.

Antimicrobial Activity of Extracts from Some Traditional Oriental Medicinal Plants against Dental Caries Bacteria (치아우식 원인균에 대한 한약재 추출물의 항균효과)

  • Antimicrobial activity of extract from some traditional oriental medicinal plants were evaluated for their antimicrobial activity against six oral pathogens, Streptococcus salivarius, Streptococcus oralis, Streptococcus mutans, Streptococcus sanguinis, Lactobacillus acidophilus and Lactobacillus casei, which are associated with caries disease. The antimicrobial activity was examined by determining the inhibition zone using the disc diffusion assay. In antibacterial activity test, extracts of Scutellaria baicalensis, Chrysanthemum indicum, Kochia scoparia, Hydnocarpus anthelmintica and Caesalpinia sappan showed inhibitory effects (40 mg/ml) against tested caries bacteria. Especially, the C. sappan extract showed the strongest activity on S. oralis (40 mm), L. casei (35 mm) and S. mutans (28 mm). Thus, this result suggests that C. sappan may be applicable to preventing dental caries.

Effect of Medium Composition on Cell Growth and Bioethanol Production in Clostridium ljungdahlii Culture (Clostridium ljungdahlii 배양에서 배지 조성에 따른 균주 성장과 바이오에탄올 생산에 대한 영향)

  • In this work, effect of the culture medium composition on the fermentation process of Clostridium ljungdahlii, which is acetogenic bacteria to product ethanol from synthesis gas, was examined to improve the microbial growth and ethanol production. Components of the culture medium such as yeast extract, fructose, $NH_4Cl$, and $K_2HPO_4$ were selected as influence factors for the cell growth and ethanol production. As the concentration of yeast extract increased, both of the cell growth and ethanol production increased. And the ethanol productivity was the highest at an yeast extract of 0.05 g/L, which is lower than that of base medium. As the concentration of fructose increased, the cell growth increased, but the ethanol production decreased when the concentration of fructose was higher than that of base medium (5 g/L). In an experiment with the yeast extract of 5 g/L, produced ethanol concentration was the highest (0.297 g/L) when fructose concentration was 5 g/L, however, the specific ethanol productivity was higher (0.281 g/g DCW) when the fructose was not added due to very low cell mass. The cell growth and ethanol production were not significantly influenced by $NH_4Cl$ concentration, however the growth inhibition was observed at a 30 g/L of $NH_4Cl$. When the concentration of $K_2HPO_4$ increased, both of the cell growth and ethanol production increased. In experiments with $NH_4Cl$ and $K_2HPO_4$, specific ethanol productivities were higher when the low concentration of yeast extract was used.

Involvement of p53-Mediated Mitochondrial Stress in the Apoptosis Induced by Flavonoids Purified from Rhus verniciflua Stokes in Human Osteosarcoma Cells

  • Dietary flavonoids have antioxidant and antitumor promoting effects. Rhus verniciflua Stokes (RVS) is a flavonoid-rich herbal medicine and has long been used as a food additive and an antitumor agent in Korea. Previous study demonstrated that a purified flavonoid fraction prepared from RVS, herein named RCMF (the RVS chloroform-methanol fraction), exhibited growth inhibition and induced apoptosis in human osteosarcoma(HOS) cells. This study evaluated if p53-mediated pathway is associated with the RCMF-induced apoptosis in HOS cells. RCMF was shown to be capable of inducing apoptosis of the cells, as expected, and transparently increased p53 expression in the cells. However, the RCMF-induced cytotoxicity was suppressed by transfecting the cells with antisense p53 oligonucleotide, which also inhibited the decrease of Bcl-2 and the increase of Bax in mitochondria, and the release of cytochrome c into cytosol. This finding suggests that p53-mediated mitochondrial stress is required for RCMF-induced apoptosis in HOS cells.

Effects of Combined Treatment of Gamma Irradiation and Addition of Fucoidan/laminarin on Ready-to-eat Pork Patty (감마선 조사와 푸코이단/라미나린 병용처리가 돈육 패티의 품질에 미치는 영향)

  • The objective of this study was investigated to apply the fucoidan and laminarin extract from low-grade Undaria pinnatifida to real food product, a pork patty. Pork patties added with fucoidan and laminarin showed lower lipid oxidation and the inhibition against lipid oxidation was shown to be dependent on the irradiation doses. The Hunter color values of pork patty added with fucoidan increased significantly with an increase of irradiation dose. The hardness profiles of the patties with fucoidan and laminarin was decreased, but the amount of water in the patties was increased. Also, the combination of gamma irradiation and addition of fucoidan and laminarin was shown to be effective for the microbiological control. These results suggested that gamma irradiation and fucoidan and laminarin treatment showed the positive effect on microbial stability and quality of the pork patty.

Functional Activities of Makgeolli By-products as Cosmetic Materials (막걸리 부산물의 미용 소재로서의 기능성 분석)

  • To investigate the potential use of makgeolli by-products as cosmetic materials, their phenolic and kojic acid contents, antioxidant activity, whitening effect, and anti-wrinkle activity were evaluated. Extracts were obtained with five different solvents (containing 0, 25, 50, 75, and 100% ethanol) from nuruk lees (NL), rice lees (RL), raw makgeolli (RM), and commercial makgeolli (CM) at 20 and $50^{\circ}C$. NL and CM extracts prepared with 75% ethanol had the highest phenolic contents (13.26 and 16.66 mg gallic acid equivalents/g, respectively) at 20 and $50^{\circ}C$, respectively. The highest kojic acid content was found in NL extracts with 0% ethanol at $20^{\circ}C$, while kojic acid was not detected in extracts prepared at $50^{\circ}C$. NL and RL extracts at $20^{\circ}C$ showed significant antioxidant activity. Whitening effects, determined by tyrosinase inhibitory activity, were highest for the NL extract prepared with 75% ethanol at $50^{\circ}C$. Noticeable anti-wrinkling effects, estimated by elastase inhibition activity, were also found in NL and RL extracts. These results suggest that makgeolli by-products could be valuable cosmetic materials with antioxidant, whitening, and anti-wrinkle activities.

Cloning and sequence analysis of Wild Argali short palate, lung and nasal epithelium clone 1 cDNA

  • Objective: Experiments were conducted to clone the sequence of Wild Argali short palate, lung and nasal epithelium clone 1 (SPLUNC1) cDNA, and to lay the foundation for further study the biological function of Wild Argali SPLUNC1. Methods: The complete sequence of Wild Argali SPLUNC1 cDNA was generated by rapid amplification of cDNA ends. The entire coding sequence was inserted into the pPIC9K vector and expressed in Pichia pastoris (P. pastoris) GS115. The recombinant SPLUNC1 protein was detected by Western blot and purified by $Ni^{2+}$ chelate affinity chromatography. The test of effect of the protein on Mycoplasma ovipneumoniae (MO) was performed with real-time polymerase chain reaction. Results: The Wild Argali SPLUNC1 cDNA was 1,076 bp with an open reading frame of 768 bp, which encoded a 26.49 kDa protein composed of 255 amino acids. Its amino acid sequence shared 98.4%, 96.9%, 94.5%, 90.2%, 80.8%, 78.4%, 78.3%, 72.5%, 72.3%, 68.8% identity with those of SPLUNC1 cDNA from Ovis aries (accession no. NP_001288334.1), Capra hircus (accession no. XP_005688516.1), Pantholops hodgsonii (accession no. XP_005979709.1), Bos taurus (accession no. NP_776851.1), Felis catus (accession no. XP_006929910.1), Homo sapiens (accession no. NP_001230122.1), Sus scrofa (accession no. NP_001005727.1), Chinchilla lanigera (accession no. NP_001269294.1), Mus musculus (accession no. NP_035256.2), and Rattus norvegicus (accession no. NP_742028.1), respectively. The recombinant protein corresponded to the expected molecular mass of 25.47 kDa as judged by sodium dodecyl sulfate-polyacrylamide gel electrophoresis, and it was detected in the supernatant of P. pastoris, and it could be purified. The results from the test of inhibition effect of argali recombinant SPLUNC1 protein on MO showed that the product could inhibit MO very well (p<0.01). Conclusion: The amino acid sequence of Wild Argali SPLUNC1 was different from other organisms. The recombinant SPLUNC1 protein has good biological activity.

Anti-oxidative and Anti-inflammatory Constituents from the Extracts of Hydrangea macrophylla Flowers (수국 꽃 추출물 유래 항산화 및 항염 활성 성분)

  • In this study, the extracts of Hydrangea macrophylla (H. macrophylla) flowers were investigated for the anti-oxidative and anti-inflammatory activities, and their active constituents were identified. The anti-oxidative effects were tested by DPPH and ABTS+ assays. To evaluate anti-inflammatory activities, LPS-induced RAW264.7 cells were examined. Among the extracts, the ethyl acetate fraction showed potent radical scavenging activities and inhibition of nitric oxide (NO) production. Chromatographic purification of the extract led to isolation of the compounds; hydrangenol (1), prunin (2) and astragalin (3). The chemical structures of the constituents were elucidated based on spectroscopic data including NMR spectra, as well as comparison of the data in the literature values. Quantitative analysis by high pressure liquid chromatography (HPLC) determined hydrangenol (1) as the major constituent. Isolated compounds 1-3 decreased the NO level without causing cell toxicities. Based on these results, it was suggested that the extract from H. macrophylla flowers could be potentially applicable as an anti-oxidative and/or anti-inflammatory ingredients.

Citric acid Fermentation by Mutant Strain of Candida lipolytica (Candida lipolytica 변이주에 의한 구연산발효)

  • In order to increase citric acid productivity. several attempts were made; isolation and characterization of the mutant strain produced citric acid in a high yield, citric acid fermentation in a medium containing relatively higher amount of glucose and citric acid production by the use of semicontinuous ceil recycle system. By the treatment of Candide lipolytica S-109 with NTG, a mutant J-24 was selected as the highest producer of citric acid among the strains formed larger CaCO$_3$ lytic zone. it produced 72g/1 citric acid in 10% glucose medium. Because mutant J-24 produced 85g/l citric acid and showed 53% yield in 16% glucose medium, several factors were adjusted to increase the yield in 16% glucose medium. 0.8-1.0$\times$10$^{-3}$ P/C ratio, 0.15% urea, 0.25% yeast extract were suitable at citric acid production in 16% glucose medium. Under this condition, J-24 strain produced 93g/l citric acid and showed 58% yield. Semicontinuous cell recycle system was used to protons the effective production phase, to minimize the product inhibition and to shorten the lag phase. The productivity of semicontinuous cell recycle system was 0.79g/l h while that of batch system was 0.53g/l.h
