Nitric Oxide Inhibition and Procollagen Type I Peptide Synthesis Activities of a Phenolic Amide Identified from the Stem of Lycium chinense Miller

  • The bioactivities of boxthron fruits, a source of oriental medicine, are well known, whereas phytochemical studies of the boxthorn stem are rare. In this study, the stem extract of boxthorn (Lycium chinense Miller) and its subfractions were evaluated for their effects on nitric oxide (NO) inhibition and procollagen type I peptide (PIP) synthesis. A phenolic amide isolated from the stem extract was also assayed for these effects. The compound, N-trans-feruloyltyramine, was identified by $^1H$, $^{13}C$, and 2D-nuclear magnetic resonance analyses. In NO inhibition, the chloroform fraction (CF) exhibited the strongest inhibitory activity ($MIC_{50}=24.69{\mu}g/ml$) among the subfractions of the ethanol extract (EE). N-trans-feruloyltyramine isolated from the CF showed strong NO inhibitory activity, presenting with an $MIC_{50}$ of $31.36{\mu}g/ml$. The EE, CF, and N-trans-feruloyltyramine shown to have NO inhibition activity were assayed for the activity of PIP synthesis. The EE and CF showed relatively high PIP values of 38.8% and 24.21% at $100{\mu}g/ml$, respectively. The PIP value for $20{\mu}g/ml$ N-trans-feruloyltyramine showed a 36% increase compared with the non-treated control, whereas that treated with $20{\mu}g/ml$ ascorbic acid as a positive control showed a 13% increase. The results suggest that the proper stem extract of boxthorn stem could be efficiently used to produce good cosmetic effects.

Anti-Aging Effect of Ligustrum japonicum Extract in the Human Fibroblast Cells (피부 섬유아세포에서 광나무 추출물의 항 노화 효과)

  • To develop wrinkle care cosmetic ingredients, various species of plant extracts were investigated. Accordingly, Ligustrum japonicum was selected as a candidate for developing cosmetic ingredient. By high performance liquid chromatography, 31.06 % of oleanolic acid and 8.92 % of ursolic acid which are well-known for anti-wrinkle effect were analyzed. The possibility of Ligustrum japonicum fruits extract (LJE) as a cosmetic ingredient was investigated using several biomarkers related to anti-aging, including anti-wrinkle, moisturizing and anti-inflammation. Procollagen type I and hyaluronan synthase-3 gene expression were increased by LJE in a concentration-dependent manner, whereas elastase activity and matrix metalloproteinase (MMP)-1, MMP-2 and cyclooxygenase-2 gene expression were inhibited. As the results LJE is applicable for a potential cosmetic ingredient focused on anti-aging effect.

Constituents of Collagen Synthesis Activation from the Extracts of Gynostemma pentaphyllum Leaves (돌외 잎 추출물의 콜라겐 합성 증진 성분 규명)

  • In order to discover ingredients for wrinkle-care cosmetics, we prepared 70% ethanol extract from Gynostemma pentaphyllum and examined its activity on collagen synthesis using fibroblast HDFn cells. The G. pentaphyllum extract induced the production of type I procollagen in a dose-dependent manner without showing cell toxicity. The active constituents were isolated from the extract by solvent fractionation and chromatographic purification procedures. NMR data and literature studies led to determine the two isolated compounds as the flavonoid glycosides such as ombuine 3-O-rutinoside (1) and quercetin 3-O-rutinoside (2). The activity screening tests showed that the isolates 1 and 2 induced the production of type I procollagen in a dose-dependent manner. These results suggested that G. pentaphyllum extract containing the flavonoids 1 and 2 could be useful as an active ingredient for wrinkle-care cosmetics.

Effect of Ganoderma lucidum Solid-state Fermented Salix gracilistyla Extract on Type I Procollagen Biosynthesis in HDFn Cells (불로초 균사배양 갯버들 추출물이 인간 피부 섬유아세포의 제1형 프로콜라겐 생성에 미치는 영향)

  • This study was carried out to investigate the feasibility of Salix gracilistyla production for cosmetic use through mycelial fermentation. The efficacy of this method was confirmed by fermentation using the mycelia of Ganoderma lucidum (a representative medicinal mushroom). Total polyphenol and flavonoid content and DPPH radical scavenging activity of S. gracilistyla extract (SGE) were found to be higher than those of G. lucidum fermented S. gracilistyla extract (GLSGE). GLSGE had relatively lower collagenase activity than SGE. However, GLSGE increased HDFn cell viability more potently than SGE, and increased the biosynthesis of type I procollagen. Thus, GLSGE could be used as an anti-aging cosmetic active ingredient. These results indicate that extract fermentation using G. lucidum mycelia can effectively enhance some beneficial effects of functional materials.

Effects of Polygoni Multiflori Radix on the Elastase, and Collagenase Activities and the Procollagen Synthesis in Hs68 Human Fibroblasts

  • Objectives : Polygoni Multiflori Radix (PMR), the roots of Polygonum multiflorum Thunberg, is used to nourish the blood and yin and used for preventing premature greying of the hair. There are some articles on its preventing effects on the melanogenesis. However, there is no report about its effects on the collagen and elastin. The present study was designed to investigate its effects on collagen metabolism and elastase activity. Methods : The effects of PMR on type I procollagen production and collagenase activity in human normal fibroblasts Hs68 after UVB (312 nm) irradiation were measured by ELISA method. Cells were pretreated with the PMR for 24 hours prior to UVB irradiation. After UVB irradiation, cells were retreated with the sample and incubated for additional 24 hours. The amount of collagen type I was measured with a procollagen type I C-peptide assay kit. The activity of collagenase was measured with a MMP-1 human biotrak ELISA system. The elastase activities after treatment of PMR were measured as well. Results : In the present study, the collagen production was not increased. However, the increased collagenase activity after UVB damage was significantly recovered to $50.2{\pm}14.5%$, $8.2{\pm}3.1%$, and $10.0{\pm}3.3%$ (10, 30, and $100{\mu}g/ml$). The elastase activities (10, 100, and $1000{\mu}g/ml$) significantly reduced to $75.2{\pm}5.2%$, $40.3{\pm}1.2%$, and $27.0{\pm}1.9%$, respectively. Conclusion : PMR showed the inhibitory effects on collagenase and elastase activity. These results suggest that PMR may have potential as an anti-aging ingredient in cosmetic herbal treatment.

Inhibitory Effect of Angelica Gigantis Radix on Fibrogenesis in Rat Hepatic Stellate Cells (당귀(當歸)가 간성상세포의 섬유화 기전 억제에 미치는 영향)

  • Objectives : This study was performed to investigate the anti-fibrogenic effect of Angelica Gigantis Radix on cultured rat hepatic stellate cells. Materials and Methods : Hepatic stellate cells(HSC-T6) were treated with various concentrations of Angelica Gigantis Radix extract for both 24 and 48 hours. The extraction was done either with distilled water or 80% EtOH. After the treatment, cell viability, cell proliferation, procollagen production and the mRNA expression of the ASMA, TIMP1, TIMP2, procollagen Type 1a2, and Cytokine IL-6 production were measured by using MTT assay, BrdU assay, RT-PCR, procollagen Type I C-peptide EIA and IL-6 ELISA assay. Results : The cell viability treated with water extraction was significantly increased, but there were no significant changes treated with 80% EtOH extraction. The cell proliferation treated with water extraction decreased only in the 24 hours group, while there were significant decreases either in the 24 and 48 hours groups treated with 80% EtOH extraction. The mRNA expressions of the ASMA, TIMP2 and procollagen 1a2 decreased in a concentration-dependent manner in the 48 hours group. Procollagen production decreased in a concentration-dependent manner in both the 24 and 48 hours groups. Cytokine IL-6 production increased in a concentration-dependent manner in both the 24 and 48 hours groups. Conclusion : These results suggest that Angelica Gigantis Radix is beneficial in the treatment of cirrhotic patients as well as for patients with chronic hepatitis.

The Effect of Hydrolyzed Jeju Ulva pertusa on the Proliferation and Type I Collagen Synthesis in Replicative Senescent Fibroblasts (제주 구멍갈파래 가수분해물에 의한 노화된 섬유아세포 증식 및 콜라겐 합성증진 효과)

  • Skin dermal fibroblast is the major collagen-producing cell type in human skin. As aging process continues in human skin, collagen production is reduced and fragmentation is increased, which is initiated by matrix metalloproteinase-1 (MMP-1). This imbalance of collagen homeostasis impairs the structure and function of dermal collagenous extracellular matrix (ECM), thereby promoting skin aging. Cysteine-rich protein 61 (CCN1), a member of the CCN family, negatively regulates collagen homeostasis in primary human skin dermal fibroblast cells. It is known in aging fibroblast cells that elevated CCN1 expression substantially reduces type I procollagen and concurrently increases MMP-1, which initiates fibrillar collagen degradation. And proliferation rate of aging fibroblast cells is reduced compared to the pre-aging fibroblast cells. In this study, we confirmed that the replicative senescence dermal fibroblast cells increased the expression levels of MMP-1 and decreased the production of type I procollagen. Our results also showed that the replicative senescence dermal fibroblast cells increased in the expression of CCN1 and decreased in the proliferation rate. Hydrolyzed Ulva pertusa extracts are the materials to improve photo-aging by reducing the expression of MMP-1 that was increased by ultraviolet and by promoting the synthesis of new collagen from fibroblast cells. In this study, we also investigated the hydrolyzed U. pertusa extract to see whether it inhibits CCN1 protein expression in the senescence fibroblasts. Results showed that the hydrolyzed U. pertusa extract inhibited the expression of MMP-1 and increased the production of type I procollagen in the aging skin fibroblast cells cultured. In addition, the proteins that regulate collagen homeostasis CCN1 expression were greatly reduced. The hydrolyzed U. pertusa extract increased the proliferation rate of the aging fibroblast cells. These results suggest that replicative senescent fibroblast cells may be used in the study of cosmetic ingredients as a model of the natural aging. In conclusion, the hydrolyzed U. pertusa extract can be used in anti-wrinkle functional cosmetic material to improve the natural aging skin care as well as photo-aging.

Effects of Parsley Extract on Skin Anti-aging and Anti-irritation (파슬리추출물의 피부 노화 방지와 자극 완화에 대한 효과)

  • In order to investigate the beneficial effects of parsely (Petroselinurn sativum) extract on skin, we measured the synthesis of total collagen and type I procollagen in cultured normal human fibroblast (NHF), the synthesis of prostaglandin E$_2$(PCE$_2$), interleukin 1 ${\alpha}$(IL -1 ${\alpha}$) and tumor necrosis factor ${\alpha}$ (TNF ${\alpha}$) in HaCaT cell and we also measured dermal thickness and density in hairless mouse (Female albino hairless mice, Skh:hr-1). As the results, the synthesis of total collagen and type I procollagen were increased 23% and 18% respectively, after 1 $\mu\textrm{g}$/mL parsley extract treatment. The producions of PGE$_2$ induced by UVB irradiation were decreased 60% after 1 $\mu\textrm{g}$/mL parsley extract treatment. The treatment with 1 $\mu\textrm{g}$/mL parsley extract also decreased the synthesis of IL -1 ${\alpha}$ and TNF ${\alpha}$ induced by 10 uM RA, 100 $\mu\textrm{g}$/mL SLS and 30 mJ/$\textrm{cm}^2$ UVB irradiation, After 4 days treatment with 1% parsley extract, the dermal thickness of hairless mouse was increased 1.5 times and the density of dermis was tighter than control. These results indicate that parsley extract have anti-aging and anti-irritation effects on skin.

Inhibitory Effect of Artemisiae Capillaris Herba on Fibrogenesis in Primary Cultured Rat Hepatic Stellate Cells (인진이 간성상세포의 섬유화 억제에 미치는 영향에 대한 연구)

  • Object : This study was performed to investigate the anti-fibrogenic effect of Artemisiae Capillaris Herba(ACH) on cultured rat hepatic stellate cells. Methods : Hepatic Stellate Cells were obtained from a 350gm Sprague-Dawley rat by tissue perfusion system, and the cells for the study were selected after 3 passages of culture on non-coated plastic culture dishes which enable the cells to activate, thus producing collagens in the cell media. Cells were treated with various concentrations of Artemisia Capillaris Herba(ACH) extract powder for 24 or 48 hours. After the treatment, Soluble collagen, procollagen levels and the mRNA of the procollagen type I C were measured by using assay kit and RT-PCR method. Results : Procollagen production by the hepatic stellate cells decreased after the treatment in a time-dependent dose-dependent manner. The mRNA expression decreased consistently with the volume of the secreted procollagen which indicates the herb hat inhibitory effect on fibrogenesis of the liver by regulating one of the fibrosis associated genes in transcription. Conclusion : These results suggest that ACH is beneficial in the treatment of cirrhotic patients as well as for the patients with chronic hepatitis.

Age-related Reference Intervals for Total Collagen-I-N-terminal Propeptide in Healthy Korean Population

  • Background: Procollagen type I N-terminal propeptide (PINP) is one of the most clinically useful bone formation biomarkers. Therefore, the purpose of this study was to independently evaluate the performance of automated total PINP assay and established age- and gender- specific reference intervals for PINP in healthy Korean population. Methods: The imprecision, linearity, and detection capability of Elecsys total PINP assay was determined and reference interval was established using 599 serums from Korean population with normal bone mineral densities based on bone densitometry. Age groups were divided into 20s, 30s, 40s, 50s, 60s and over. Results: Elecsys total PINP had excellent performance in imprecision, linearity, and detection capability. When partitioning age groups in Korean male and female populations, there was significant difference in total PINP between different age groups. In male populations, PINP level was decreased with increasing age, then it remained steady after middle-age. In female populations, there was a decreasing tendency similar to that in the male population with a sharp increase in the 50 to 59 age group. Conclusions: Elecsys total PINP assay showed precise and reliable performance in our study. We established age-related PINP reference intervals for Korean male and female population with normal bone mineral densities.
