For the elucidation of action mechanism on anti-HIV of natural resources, the extracts of $Campsis$$grandiflora$ were tested for their inhibitory effects on HIV-1 replication and its essential enzymes as the reverse transcriptase (RT), protease and ${\alpha}$-glucosidase. In the assay of HIV-1-infected human T-cell line, water extracts of stem inhibited the HIV-1-induced cytopathic effects with IC (inhibitory concentration) of 100 ${\mu}g$/ml. Moreover water extracts (100 ${\mu}g$/ml) of stem showed strong activity of 37.9% on anti-HIV-1 RT using Enzyme Linked Oligonucleotide Sorbent Assay (ELOSA) method. In the HIV-1 protease inhibition assay, methanol extracts of stem and leaf extract showed 33.6% and 31.5% inhibition of the enzyme activity to cleave an oligopeptide resembling one of the cleavage sites in the viral polyprotein which can only be processed by HIV-1 protease, but did not exhibited glucosidase inhibitory activities. From these results, it is suggested that the inhibition of the viral replication $in$$vitro$ is due to the inhibition of reverse transcriptase by water extracts of stem of $Campsis$$grandiflora$.
Objective of this study was to investigate effects of pre-processing method and number of sampling leaves on stability of the reflectance measurement for Chinese cabbage and kale leaves. Chinese cabbage and kale were transplanted and cultivated in a plant factory. Leaf samples of the kale and cabbage were collected at 4 weeks after transplanting of the seedlings. Spectra data were collected with an UV/VIS/NIR spectrometer in the wavelength region from 190 to 1130 nm. All leaves (mature and young leaves) were measured on 9 and 12 points in the blade part in the upper area for kale and cabbage leaves, respectively. To reduce the spectral noise, the raw spectral data were preprocessed by different methods: i) moving average, ii) Savitzky-Golay filter, iii) local regression using weighted linear least squares and a $1^{st}$ degree polynomial model (lowess), iv) local regression using weighted linear least squares and a $2^{nd}$ degree polynomial model (loess), v) a robust version of 'lowess', vi) a robust version of 'loess', with 7, 11, 15 smoothing points. Effects of number of sampling leaves were investigated by reflectance difference (RD) and cross-correlation (CC) methods. Results indicated that the contribution of the spectral data collected at 4 sampling leaves were good for both of the crops for reflectance measurement that does not change stability of measurement much. Furthermore, moving average method with 11 smoothing points was believed to provide reliable pre-processed data for further analysis.
Patient dose verification is one of the most Important responsibilities of the physician in the treatment delivery of radiation therapy. For the task, it is necessary to use an accurate dosimeter that can verify the patient dose profile, and it is also necessary to determine the physical characteristics of beams used in intensity modulated radiation therapy (IMRT) The Beam Intensity Scanner (BInS) System is presented for the dosimetric verification of the two dimensional photon beam. The BInS has a scintillator, made of phosphor Terbium-doped Gadolinium Oxysulphide (Gd$_2$O$_2$S:Tb), to produce fluorescence from the irradiation of photon and electron beams. These fluoroscopic signals are collected and digitized by a digital video camera (DVC) and then processed by custom made software to express the relative dose profile in a 3 dimensional (3D) plot. As an application of the BInS, measurements related to IWRT are made and presented in this work. Using a static multileaf collimator (SMLC) technique, the intensity modulated beam (IMB) is delivered via a sequence of static portals made by controlled leaves. Thus, when static subfields are generated by a sequence of abutting portals, the penumbras and scattered photons of the delivered beams overlap in abutting field regions and this results in the creation of “hot spots”. Using the BInS, inter-step “hot spots” inherent in SMLC are measured and an empirical method to remove them is proposed. Another major MLC technique in IMRT, the dynamic multileaf collimator (DMLC) technique, has different characteristics from SMLC due to a different leaf operation mechanism during the irradiation of photon and electron beams. By using the BInS, the actual delivered doses by SMLC and DMLC techniques are measured and compared. Even if the planned dose to a target volume is equal in our experimental setting, the actual delivered dose by DMLC technique is measured to be larger by 14.8% than that by SMLC, and this is due to scattered photons and contaminant electrons at d$_{max}$.

Most crop damages have been occurred by vermin(e.g., wild birds and herbivores) during the period between seeding and the cotyledon level. In this study, to minimize the damage by vermin and acquire the benefits such as protection against weeds and maintenance of water content in soil, immediately vinyl mulching after seeding was devised. Vinyl mulching has been generally covered with black color vinyl, that crop seeding locations cannot be detected by visible light range. Before punching vinyl, non-contact and non-destructive methods that can continuously determine the locations are necessary. In this study, a crop position detection method was studied that uses infrared thermal image sensor to determine the cotyledon position under vinyl mulch. The moving system for acquiring image arrays has been developed for continuously detecting crop locations under plastic mulching on the field. A sliding mechanical device was developed to move the sensor, which were arranged in the form of a linear array, perpendicular to the array using a micro-controller integrated with a stepping motor. The experiments were conducted while moving 4.00 cm/s speed of the IR sensor by the rotational speed of the stepping motor based on a digital pulse width modulation signal from the micro-controller. The acquired images were calibrated with the spatial image correlation. The collected data were processed using moving averaging on interpolation to determine the frame where the variance was the smallest in resolution units of 1.02 cm. For this study, the spline method was relatively faster than the other polynomial interpolation methods, because it has a lower maximum order of formulation when using a system such as the tridiagonal linear equation system which provided the capability of real-time processing. The temperature distribution corresponding to the distance between the crops was 10 cm, and the more clearly the leaf pattern of the crop was visually confirmed. The frequency difference was decreased, as the number of overlapped pixels was increased. Also the wave pattern of points where the crops were recognized were reduced.

We investigated the effect of harvesting time on the proximate compositions and biological activities of Lactuca indica L. Seonhyang and wild species. The highest moisture content (91%) was obtained from Seonhyang harvested in June. The wild species harvested in August showed the highest crude protein (4.8%) and crude fiber contents (2.7%). Potassium contents were 626~684 mg/100 g, and no significant difference in harvesting time was observed between Seonhyang and wild species. Calcium and phosphate contents of Seonhyang leaves harvested in August were highest (350 mg/100 g and 123 mg/100 g, respectively). The highest total polyphenol and total flavonoid contents were observed for wild species harvested in June (60 mg/g and 126 mg/g, respectively). Ethanol extracts of Seonhyang and wild species leaves harvested in June showed the highest DPPH radical scavenging activity (95%). ${\alpha}$-Amylase inhibitory activity was highest (94.8%) in water extract of Seonhyang leaves harvested in July. Nitric oxide production inhibitory activity was $14.3{\mu}M$ in ethanol extracts of Seonhyang and $16.8{\mu}M$ in ethanol extracts of wild species harvested in June. Calcium content and ${\alpha}$-amylase inhibitory activity of Seonhyang leaves were greater than those of wild species leaves. These results suggest that Seonhyang leaf can be used to develop processed foods.

A focused ion beam (FIB) system produces a beam of positive ions (usually gallium) which are heavier than electrons and can be focused by electrostatic lenses into a spot on the specimen. With its ability milling of the specimen material by 10 to 100 nm with each pass of the beam, FIB is widely adopted in materials science, semiconductor industry, and ceramics research. Recently, FIB has been increasingly employed in the field of biomedical sciences. Here we provide a brief introduction to FIB and its applications for a wide variety of biomedical research. The surface of specimen can be in situ processed and quasi-real time visualized by two beam combination of FIB and field emission scanning electron microscope (FESEM). Due to its milling process, internal structures can be exposed and analyzed: yeast cells, fungus-inoculated wheat leaf, mannitol particles in inhalation aerosols, and oyster shell. Serial blockface tomography with the system kindles 3-dimensional reconstruction researches in the realm of nervous system and life sciences. Two-beam system of FIB/FESEM is a versatile tool to be utilized in the biomedical sciences, especially in 3-dimensional reconstruction studies.
The phenolic substances contents of 45 plant foods in Korean diet were determined by different methods. Total phenolics contents by Folin-Denis method were $0.1{\sim}5.8%$ (dry matter basis), in which persimmon leaf, chestnut's inner skin, Chinese quince, walnut, sunflower seed and arrowroot exhibited the higher levels above 2%. Condensed tannin contents by vanillin method were $0{\sim}48%$, in which Chinese quince and chestnut's inner layer gave very high levels. Protein-precipitable phenolic substances ranged from 0.4% to 2.2%, in which chestnut's inner layer, walnut and Chinese quince had the highest content. The ability of phenolics to form precipitate was higher with pepsin and albumin than with trypsin. Among different phenolics content, total phenlolics correlated significantly with protein-precipitable phenolics (r=0.65) and condensed tannin (r=0.56). Chinese quince, chestnut's inner skin and sorghum showed a relatively lower degree of polymerization, as expressed by vanillin/FolinDenis ratio. Processed foods from buckwheat, acorn, mugwort and arrowroot showed a lower content of phenolic substances, suggesting a negligible adverse effect on the bioavailability of food proteins, if any.
Purpose: Damage to pulse crops by wild birds is a serious problem. The damage is to such an extent that the rate of damage during the period between seeding and cotyledon stages reaches 54.6% on an average. In this study, a crop-position detection method was developed wherein infrared (IR) sensors were used to determine the cotyledon position under a vinyl mulch. Methods: IR sensors that helped measure the temperature were used to locate the cotyledons below the vinyl mulch. A single IR sensor module was installed at three locations of the crops (peanut, red lettuce, and crown daisy) in the cotyledon stage. The representative thermal response of a $16{\times}4$ pixel area was detected using this sensor in the case where the distance from the target was 25 cm. A spatial image was applied to the two-dimensional temperature distribution using a non-integral moving-average method. The collected data were first processed by taking the moving average via interpolation to determine the frame where the variance was the lowest for a resolution unit of 1.02 cm. Results: The temperature distribution was plotted corresponding to a distance of 10 cm between the crops. A clear leaf pattern of the crop was visually confirmed. However, the temperature distribution after the normalization was unclear. The image conversion and frequency-conversion graphs were obtained based on the moving average by averaging the points corresponding to a frequency of 40 Hz for 8 pixels. The most optimized resolutions at locations 1, 2, and 3 were found on 3.4, 4.1, and 5.6 Pixels, respectively. Conclusions: In this study, to solve the problem of damage caused by birds to crops in the cotyledon stage after seeding, the vinyl mulch is punched after seeding. The crops in the cotyledon stage could be accurately located using the proposed method. By conducting the experiments using the single IR sensor and a sliding mechanical device with the help of a non-integral interpolation method, the crops in the cotyledon stage could be precisely located.

This study examined changes in antioxidant activity as well as contents of rosmarinic acid, homoplantaginin, and luteolin, which is the main substance of Salvia plebeia R. Br. (SPR) known to have anti-inflammatory efficacy, according to drying, blanching, and fermentation conditions. Rosmarinic acid content was 16.42 mg/g upon hot-air drying and 10.19 mg/g upon hot-air drying after blanching, and there was no significant difference in the case of leaf and root freezing or cold-air drying. Rosmarinic acid content was 8.69 and 8.15 mg/g in the case of air-drying in the shade and freeze-drying, respectively, and decreased to 0.05 mg/g or undetected after fermentation. SPR processed by freeze-drying, cold-air drying in the shade, and hot-air drying showed ABTS radical scavenging ability over 98.5% at a concentration over $500{\mu}g/mL$ as well as excellent radical scavenging ability of 87.3% in the case of hot-air drying after blanching. Root showed lower ABTS radical scavenging ability than leaves. SOD-like activity was measured to be 6.1~27.8% at a concentration of $1,000{\mu}g/mL$, which was significantly difference from ABTS radical scavenging ability. As rosmarinic acid and homoplantagine, an anti-inflammatory material contained in SPR, are almost undetectable after oxidation fermentation during processing, hot-air drying after blanching or drying seems to be suitable to develop SPR as a functional substance.
In this study, we conducted the anti-HIV-1 enzymes assay in vitro and its active components were predicted by QSAR in silico for the elucidation of action mechanism on anti-HIV of natural resources. The extracts of Gardenia jasminoides var. radicans Makino were tested for their inhibitory effects on the reverse transcriptase (RT), protease and ${\alpha}$-glucosidase. In the enzyme inhibition assay, the methanol extracts of Gardenia jasminoides var. radicans Makino stem showed a strong activity of 32.5% on the enzyme activity to cleave an oligopeptide, resembling one of the cleavage sites in the viral polyprotein which can only be processed by HIV-1 protease. Moreover the methanol extracts of stem exhibited alpha-glucosidase inhibitory activities of 26.1%. The methanol extracts ($100{\mu}g/ml$) of stem showed a weak activity of 13.4% on anti-HIV-1 RT using Enzyme Linked Oligonucleotide Sorbent Assay (ELOSA) method. However, all extracts of leaf and stem didn't exhibit the HIV-1-induced cytopathic effects with IC (inhibitory concentration) of $100{\mu}g/ml$ in HIV-1-infected human T-cell line. From these results, it is suggested that Gardenia jasminoides var. radicans Makino extracts may possibly be involved in the inhibition of reverse transcriptase, protease and alpha-glucosidase but can't vitally concerned with the viral replication in vitro.
