Clinical Characteristics of Symptomatic Clostridium difficile Infection in Children: Conditions as Infection Risks and Whether Probiotics Is Effective

  • Purpose: This study investigated the clinical presentations of symptomatic Clostridium difficile infection (CDI) in children. Methods: We reviewed the medical records of 43 children aged <20 years who showed either positive C. difficile culture or C. difficile toxin test results between June 2010 and April 2014. Results: Of the 43 patients (mean age 6.7 years), 22 were boys. Sixteen patients (37.2%) showed both positive C. difficile culture and toxin test results. Seventeen out of 43 children (39.5%) had preexisting gastrointestinal diseases, and 26 children had other medical conditions that were risk factors for CDI. Twenty-eight children had a history of antibiotic treatment for >3 days, and the most frequently prescribed antibiotic was amoxicillin-clavulanate (35.7%). Twenty-eight patients were diagnosed with CDI despite taking probiotic supplements, most commonly Lactobacillus acidophilus (53.6%). The most common symptom was diarrhea (72.1%) at the time CDI was diagnosed. C. difficile was eradicated in 11 patients (25.6%) after treatment with oral metronidazole for 10-14 days, and in the two patients (4.6%) who required two courses of oral metronidazole. Sixteen patients (37.2%) showed clinical improvement without any treatment. Conclusion: This study showed the various clinical characteristics of CDI in children and that preexisting clinical conditions favored the development of CDI. In addition, CDI was found to occur in a number of patients even after probiotic prophylaxis given in conjunction with antibiotic therapy.

Bacterial Population in Intestines of Litopenaeus vannamei Fed Different Probiotics or Probiotic SupernatantS

  • The interactions of microbiota in the gut play an important role in promoting or maintaining the health of hosts. In this study, in order to investigate and compare the effects of dietary supplementation with Lactobacillus pentosus HC-2 (HC-2), Enterococcus faecium NRW-2, or the bacteria-free supernatant of a HC-2 culture on the bacterial composition of Litopenaeus vannamei, Illumina sequencing of the V1-V2 region of the 16S rRNA gene was used. The results showed that unique species exclusively existed in specific dietary groups, and the abundance of Actinobacteria was significantly increased in the intestinal bacterial community of shrimp fed with the bacteria-free supernatant of an HC-2 culture compared with the control. In addition, the histology of intestines of the shrimp from the four dietary groups was also described, but no obvious improvements in the intestinal histology were observed. The findings in this work will help to promote the understanding of the roles of intestinal bacteria in shrimps when fed with probiotics or probiotic supernatant.

Assessment of Characteristics and Functional Properties of Lactobacillus Species Isolated from Kimchi for Dairy Use

  • The objective of this study was to identify lactic acid bacteria (LAB) isolated from kimchi and to evaluate its characteristics and functional properties for application in fermented dairy products as a probiotic or commercial starter culture. Eight stains isolated from kimchi were selected through an investigation of phenotypic characteristics. Two strains (DK211 and DK303) were identified as Lactobacillus plantarum, another two (DK207 and DK215) as Lactobacillus paracasei, and one (DK301) as Lactobacillus sakei. The remaining three strains were identified as species of Weissella. All selected Lactobacillus strains had acid and bile tolerance, even though there was wide variation in the ability of each strain. DK303 showed a remarkably higher proteolytic activity. There were no significant differences in β-galactosidase activity among the tested strains, except that DK301 showed no activity. Auto-aggregation varied between 82.1 and 90.0%, and hydrophobicity values ranged from 0.5 to 51.6%.The strongest auto-aggregation and hydrophobicity were observed in DK211. All selected strains showed better 1,1-diphenyl-2-picrylhydrzyl (DPPH) scavenging activity than commercial strains. DK211, DK215, DK301, and DK303 had effective inhibitory activity against all pathogens tested except E. coli. When selected strains were used for yogurt preparation as a single starter culture, the time required to reach target titratable acidity (0.9) was 11-12 h. The yogurt fermented with DK211 had favorable panelists ratings for most sensory attributes, which were comparable with yogurt fermented with a commercial strain. The results suggest that strains isolated from kimchi could be potential probiotic and starter cultures for use in yogurt manufacturing.

발효 소시지 제조를 위한 기능성 유산균의 선발 (Screening of Lactic Acid Bacteria as Starter Culture for Making Fermented Sausage)

  • 발효 소시지의 제조에 starter로 사용한 균주를 선발하기 위해 각 유산균 균주들의 ACE 저해 활성, 콜레스테롤 흡착능력, 발효 육제품 저장 중 S. aureus에 대한 저해 황성을 평가하였다. ACE 저해 활성은 L. plantarum L167이 가장 우수하였고(58.75%), 콜레스테롤 흡착 능력이 우수한 균주로는 L. plantarum L155를 선발하였다. 박테리오신을 생산하는 P. damnosus를 starter로 이용하여 제조한 발효 소시지에 S. aureus를 접종하여 상온 저장 중 생균수를 측정한 결과 저장 35일째에 대조구에 비하여 1 log의 균수 감소 효과가 나타났다.

김치 젖산균의 In Vitro에서 콜레스테롤 저하 효과 (In Vitro Evaluation of Cholesterol Reduction by Lactic Acid Bacteria Extracted from Kimchi)

  • 본 연구는 김치에 분포하는 것으로 알려져 있는 대표적인 젖산균에 관한 콜레스테롤 저하 효과에 관하여 연구하였다. Lactobacillus 1종, Leuconostoc 2종과 Weissella 2종을 대상으로 콜레스테롤 저하 효과를 측정하였다. 김치 초기 발효균인 Leu. citreum은 55.46% 콜레스테롤 감소율을 보였고, 대표적인 산패균인 Lac. plantarum은 55.91%, Leu. mesenteroides은 56.37%였으며 W. kimchii,, W. confusa 등에서도 55% 이상의 콜레스테롤 저하 효과가 나타났다. 김치의 발효 숙성 과정에 나타나는 젖산균들이 시험관 내에서 콜레스테롤 저하 효과가 확인되어, 향후 in vivo 조건하에서 콜레스테롤 저하에 어느 정도 영향을 미치는지 검토해볼 필요가 있다고 사료된다.

Bifidobacterium bifidum DS0908 and Bifidobacterium longum DS0950 Culture-Supernatants Ameliorate Obesity-Related Characteristics in Mice with High-Fat Diet-Induced Obesity

  • Probiotic supplements have promising therapeutic effects on chronic diseases. In this study, we demonstrated the anti-obesity effects of two potential probiotics, Bifidobacterium bifidum DS0908 (DS0908) and Bifidobacterium longum DS0950 (DS0950). Treatment with DS0908 and DS0950 postbiotics significantly induced the expression of the brown adipocyte-specific markers UCP1, PPARγ, PGC1α, PRDM16 and beige adipocyte-specific markers CD137, FGF21, P2RX5, and COX2 in C3H10T1/2 mesenchymal stem cells (MSCs). In mice with high-fat diet (HFD)-induced obesity, both potential probiotics and postbiotics noticeably reduced body weight and epididymal fat accumulation without affecting food intake. DS0908 and DS0950 also improved insulin sensitivity and glucose use in mice with HFD-induced obesity. In addition, DS0908 and DS0950 improved the plasma lipid profile, proved by reduced triglyceride, low-density lipoprotein, and cholesterol levels. Furthermore, DS0908 and DS0950 improved mitochondrial respiratory function, confirmed by the high expression of oxidative phosphorylation proteins, during thermogenesis induction in the visceral and epididymal fat in mice with HFD-induced obesity. Notably, the physiological and metabolic changes were more significant after treatment with potential probiotic culture-supernatants than those with the bacterial pellet. Finally, gene knockdown and co-treatment with inhibitor-mediated mechanistic analyses showed that both DS0908 and DS0950 exerted anti-obesity-related effects via the PKA/p38 MAPK signaling activation in C3H10T1/2 MSCs. Our observations suggest that DS0908 and DS0950 could potentially alleviate obesity as dietary supplements.

낙농산업에 필요한 미생물 검사방법과 전망: 총설 (Perspective on Rapid and Selective Method for Detecting Microbiology in Dairy Industry: A Review)

  • 낙농 미생물학에서 사용하는 주요 분석 방법들에 대한 지속적인 개정이 요구되어지고 있는 것이 사실이다. 현재 이용 가능한 자료들을 바탕으로 균주의 생리 및 대사 특성을 더 깊이 이해할 수 있으며, LAB와 probiotic 생리적 상태를 측정할 수 있는 기술을 개발할 수 있다. 예를 들어, 고유 균주의 생화학 반응으로부터 얻은 생물학적 생리학적 지식을 바탕으로 발색형광배지(chromogenic media)의 개발이 가능하다. 이 기술은 오랫동안 사용되어온 한천 배지보다 사용법이 간단하며, 미생물을 제어할 수 있다. 유망한 유동세포계수법(flow cytometry, FC) 기술은 우유, 요구르트, 그 외 발효 유제품의 혼합 배양의 분석에 뛰어나다. 이 기술은 공정 과정 중에 또는 생산품의 유통 기한 관리에 사용될 수 있지만, 이 기술을 사용하기 위해서는 우유 단백질에 의해서 생성되는 인공 산물을 방지하기 위해 최적화 되어야 한다. 또한 유동세포계수법(flow cytometry, FC)은 정량 한계가 높기 때문에, 현재 사용하고 있는 탐침(probe)은 종 또는 속이 아닌 표적 세포 구성을 분석한다. 따라서 유동세포계수법(flow cytometry, FC)은 특정 물질의 계수보다는 생리적 상태를 측정에 더 적합한 기술이라고 볼 수 있다. Omics 자료를 통해 생리적 상태를 더 신속하게 측정할 수 있는 핵심적인 생체지표 확인이 가능해졌다. PCR은 이미 식품매개 병원균 및 부패균의 검출에 일상적으로 사용되고 있다. 이 기술은 더욱 나아가 발효균 및 probiotic 박테리아 검출에도 사용되어 핵산 추출, PCR 과정, 자료 해석을 표준화 시킬 수 있으며, 또한 다른 기술에 비해 PCR은 표적 발효 균주를 더 신속하게 선택하며, 공정 과정을 최적상태로 유지시킬 수 있을 것이다. LAB와 probiotic의 적응성 및 생리적 상태의 연구에 필요한 기준, 특허품, 상업화된 신속분석장치(kit)는 아직까지 개발되지 않았다. 본 총설 논문에서는 여러 가지 기술을 검토함으로써 이 모든 기술에는 2가지 주요한 요인이 있다. 첫 번째는 낙농업 산업의 특정 요구에 따른 방법의 평가이며, 두 번째는 공식적인 기준에 따른 검증이다. 사실상 식품매개 병원균의 특성 분석에 사용될 수 있는 표준법이 개발되고 검증되었지만, LAB, probiotic 등의 박테리아 분석에 있어서 표준화된 방법과 진단산업과의 연관성이 부족한 것이 사실이다. 낙농 업계에 의해서 수행되어진 품질관리의 양을 고려해 볼 때 이것은 매우 중요하게 인식되어야 할 부분이다.

새로운 생균제 CS61 배양액의 사료 내 급여가 육계의 생산성에 미치는 영향 (Effects of Dietary Supplementation of a New Probiotic CS61 Culture on Performance in Broiler Chickens)

  • 가금류 제품에서 항생제의 잔류와 내성균의 출현으로 인해 항생제 대체물질에 대한 연구가 활발히 진행되고 있으며, 특히 생균제와 활성촉진제 또는 이들을 조합한 신바이오틱의 사용이 권장되고 있다. 본 연구는 새로운 생균제 CS61 배양액의 사료첨가 급여가 육계의 성장 및 사료효율에 미치는 영향과 안전성을 평가하여 항생제를 대체할 수 있는 사료첨가제로서의 개발가능성을 알아보기 위해 수행하였다. CS61 배양액은 0, 0.1 및 1%의 용량으로 28일간 사료에 혼합하여 육계에게 급여하였다. 시험결과, 시험물질 처치군에서 부검 시의 체중과 일당증체량이 대조군에 비해 용량의존적으로 증가하였다. CS61 배양액의 사료 내 첨가는 대조군 동물에 비해 사료효율도 개선하는 것으로 나타났다. 반면, 일반증상과 사망률, 부검소견, 혈액학치 및 혈청생화학치에서는 시험물질의 처치와 관련된 독성소견이 관찰되지 않았다. RAW 264.7 세포를 이용한 일산화질소 시험에서 정제된 CS61 펩타이드는 lipopolysaccharide에 유도된 일산화질소 생성을 용량의존적으로 억제하였다. 본 시험결과는 육계에 CS61 배양액의 사료첨가 급여는 항염증효과를 통해 성장과 사료효율을 개선할 수 있음을 보여주며, 사료첨가제로서 CS61 배양액의 유용성과 개발가능성을 시사해 주고 있다.

감귤 진피 추출물을 이용한 발효액 제조 및 Campylobacter jejuni 에 대한 항균 활성 (Preparation of Fermented Citrus Peels Extracts for Their Antimicrobial Activity against Campylobacter jejuni)

  • Jeju citrus, which contains an abundance of calcium and vitamin, was used to develop fermented citrus peel extract. A total of seven probiotic strains were applied to tangerine dermis to select the best growing bacteria in citrus peel extracts. B. longum, B. bifidum, and L. mesenteroides were found to grow best in citrus peel extract culture containing glucose, yeast extracts, peptone, and potassium phosphate. Citrus peel extract culture consisting of 1% yeast extract, 5% peptone, and 0.1% phosphate was the best environment for growth of probiotics. The pH, acidity, and viable cell numbers of these fermented extracts were measured. The initial pH level of fermented extracts with nutrients was 5.25 and dropped rapidly to 3.39 after 72 hours of fermentation. The acidity of fermented extracts increased to 4.08 % after 72 hours of fermentation, and the viable cell number in fermented extracts after refrigeration for 2 weeks was $1.3{\times}10^{10}CFU/mL$. The antimicrobial activity of citrus peel fermented extracts against Campylobacter jejuni was determined, and concentrations more than 25,000 ppm showed antimicrobial activity.

Probiotics에 의한 해수양식어의 성장 촉진 및 항균효과 (Antibacterial Activity and Growth Promotion in Aquacultured Fish by Probiotics)

  • For the development of probiotics in aquaculture of marine organisms, three strains having psychrophilic and salt tolerant characteristics were isolated from Kimchi. Amng the isolated strains. MG19, MG89 and MG208 were identified as Lactobacillus brevis, Enterococcus faecium and Lactobacillus plantarum, respectively. The neutralized culture broth of isolated strains were tested in order to evaluate the antibactrial activity, 조초 showed high antibacterial activity against Edwardsiella tarda, Vibrio cholerae andPseudomonas fluorescens. In mixed culture of pathogens and isolated strain,pathogens were signifi-cantly inhibited after 2 days of cultivation but the isolated strains showed normal growth. When the Edwardsiella tarda was cultured with three isolated strains, its growth was completely inhibited after 254 hours of cultivation. The effect of isolated three strains as probiotics was investigated based on the changes in body weight of aquacultured flounder. After 50 days feeding trial, it was found that the mean body weight gain of the tested group fed freeze-dried isolated cells was significantly greater than that of the control group. These results suggest that these isolated strains can play an important role as probiotics in aquaculture.
