  • The AFM is an imaging tool or a profiler with unprecedented 3-D resolution for various surface types. The AFM technology, however, leaves a lot of room for improvement due to its delicate and fragile probing mechanism. The distance between probe tip and sample surface must be maintained in below the nano meter level in order to measure the sample surface in Angstrom resolution. In this paper, the mode analysis of AFM system, modification based on the mode analysis are performed and finally the sample surface is measured by the home-built AFM.

A Study on Course-Embedded Assessment for Program Outcomes of Information Security Program for Engineering Education Accreditation (공학교육인증을 위한 정보보호학 프로그램의 교과 기반 학습성과 평가에 관한 연구)

  • This paper evaluates the achievement of program outcomes using course-embedded assessment for information security programs. In the case study concerning the development of course-embedded assessment used in this paper, we evaluate program outcomes using various evaluation tools including the course evaluation data. In addition, we consider the evaluation rubric and evaluation procedure for course-embedded program outcomes, set performance criteria, select the probe course with high contribution to the program outcomes according to curriculum flowchart, set course objectives for the probe course, and explain the rubric for each assessment tool to measure achievement level. Subsequently, we perform course-embedded program outcomes assessment, based on tests and projectevaluation tool for each program objectives, and present the achievement levels based analysis results. These results prove the effectiveness of the proposed course-embedded assessment for program outcomes.

Optical Spectroscopic Analysis of Muscle Spasticity for Low-Level Laser Therapy (LLLT)

  • Current therapeutic methods for suppressing muscle spasticity are intensive functional training, surgery, or pharmacological interventions. However, these methods have not been fully supported by confirmed efficacy due to the aggravation of the muscle spasticity in some patients. In this study, a combined system was developed to treat with a low-level laser and to monitor the region of the treatment using an optical spectroscopic probe that measures oxygen saturation and deoxygenation during low-level laser therapy (LLLT). The evaluation of the wavelength dependence for LLLT was performed using a Monte Carlo simulation and the results showed that the greatest amount of heat generation was seen in the deep tissue at ${\lambda}$ = 830 nm. In the oxy- and deoxygenation measurements during and after the treatment, oxygen-Hb concentration was significantly increased in the laser-irradiated group when compared to the control group. These findings suggest that LLLT using ${\lambda}$ = 830 nm may be of benefit in accelerating recovery of muscle spasticity. The combined system that we have developed can monitor the physiological condition of muscle spasticity during the laser treatment in real time and may also be applied to various myotonia conditions such as muscle fatigue, back-pain treatment/monitoring, and ulcer due to paralysis.

Research on the Contents Construction for the Information-oriented Family Life and Development of Professional Occupations : Based on the Analysis of the Present Condition of Information Needs among the Human Ecology Majors (가정생활 정보화 콘텐츠 구성과 전문 직업 개발을 위한 연구 : 생활과학 전공자의 정보화 요구 실태분석을 통하여)

  • This study is aimed at sowing two purposes. The first is to analyze the real condition and demand of profession development from the human ecology majors. The second is to develop necessary methods in which ordinary people can collect necessary information about family life. Related questionnaire was distributed to 147 the human ecology majors, and analysis was performed on the several web sites providing with information related to family life. Web sites such as“www.yahoo.com”and“www.naver.com”were included in the analysis. Questionnaire consisted of questions about whether the students majoring in the human ecology were familiar with the information-related terms or futuristics-related books, and whether they have ever taken the related courses in college. The results of analysis are as follows: First, analysis shows that the present level of the human ecology majors' information orientation and networking experiences is extremely low. Secondly, according to the analysis on whether they have optimistic or pessimistic attitude toward the contemporay informatized society, the human ecology majors have rather optimistic attitude in group while having pessimistic one individually. As to their response to the question about whether informatized society is connected with industrial society or not, the human ecology majors are divided in neatly equal ratio. Thirdly, analysis of the human ecology majors' understanding of profession relevant to their major indicates that they have high level of perception and information about the professions of fashion designing, traditional garment designing, nutrition counselling or consumer counselling. On the other hand, they are not familiar with the information about professions such as professional QR Programming, eating habit-related information business and family welfare. Lastly, level of web sites supplying information about family life is fragmentary. Especially, probe into the directories providing with necessary information of family life subdivided into special areal of life shows that they lack systematic organization making more convenient consumer use.

A Prediction of Specific Heat Capacity for Compacted Bentonite Buffer (압축 벤토나이트 완충재의 비열 추정)

  • A geological repository for the disposal of high-level radioactive waste is generally constructed in host rock at depths of 500~1,000 meters below the ground surface. A geological repository system consists of a disposal canister with packed spent fuel, buffer material, backfill material, and intact rock. The buffer is indispensable to assure the disposal safety of high-level radioactive waste, and it can restrain the release of radionuclides and protect the canister from the inflow of groundwater. Since high temperature in a disposal canister is released to the surrounding buffer material, the thermal properties of the buffer material are very important in determining the entire disposal safety. Even though there have been many studies on thermal conductivity, there have been only few studies that have investigates the specific heat capacity of the bentonite buffer. Therefore, this paper presents a specific heat capacity prediction model for compacted Gyeongju bentonite buffer material, which is a Ca-bentonite produced in Korea. Specific heat capacity of the compacted bentonite buffer was measured using a dual probe method according to various degrees of saturation and dry density. A regression model to predict the specific heat capacity of the compacted bentonite buffer was suggested and fitted using 33 sets of data obtained by the dual probe method.

Pseudoisodicentric X chromosome in a female with primary amenorrhea (원발성 무월경 여성에서 관찰된 Pseudoisodicentric X 염색체)

  • A 24-year-old female with primary amenorrhea was referred for a chromosome study. The karyotype of the patient was 46,X,der(X) under initial GTG-banding analysis. Fluorescence in situ hybridization (FISH) analysis with an LSI Kallmann (KAL) region probe [probes for Xp22.3(KAL) and CEP(X) for control] was carried out. The abnormal chromosome was KAL- and CEP(X)${\times}2$. In addition, interphase FISH analysis revealed the patient to be mosaic for two different cell lines: 90% of cells had three signals and 10% of the cells had only one signal for CEP(X). Based on these results, the karyotype of the patient was 45,X/46,X,psu idic(X)(p22.1), which is partial trisomy for Xqter${\rightarrow}$Xp22.1 and partial monosomy for Xpter${\rightarrow}$Xp22.1. This karyotype was considered a variant of Turner syndrome. In summary, Idic(X) and low-level mosaicism was successfully characterized by FISH analysis with a CEP(X) probe.

Construction of a Reporter Strain Pseudomonas putida for the Detection of Oxidative Stress Caused by Environmental Pollutants

  • A green fluorescent protein-based Pseudomonas putida reporter was successfully constructed and shown to be capable of detecting oxidative stress. In this whole-cell reporter, the promoter of the paraquat-inducible ferredoxin-$NADP^+$ reductase (fpr) was fused to a promoterless gfp gene on a broad-host-range promoter probe vector. Pseudomonas putida KT2440 harboring this reporter plasmid exhibited an increased level of gfp expression in the presence of redox-cycling agents (paraquat and menadione), hydrogen peroxide, and potential environmental pollutant chemicals such as toluene, paint thinner, gasoline, and diesel. Induction of fpr in the presence of these chemicals was confirmed using Northern blot analysis.

독성물질 대사효소 조절기전에 관한 연구

  • 약물, hormone, 독성물질등의 대사능과 발암 가능성등이 간장 장해시 및 ketosis시에 달라지는 원인과 기전, 독성물질 대사효소의 변동과 그 작용기전을 규명하고자, 대표적인 간장장해 물질인 carbon tetrachloride를 rat에 투여하여 간장 장해를 일으키고, 당뇨병, starvation, high-fat diet처리하여 ketosls상태를 만든 후에, specific cytochrome P45O polyclonal antibodies와 cDNA probes를 사용하여, enzyme activitieg, Western immunoblot analysis와 mRNA Northern blot analysis 등을 실험하여, 간장 장해와 ketosis시 cytochrome P45O의 변동과 그 작용기전, regulation을 규명하고자 하였다. 실험 결과, $CCl_4$투여후 P450IIE enzyme (aniline hydroxylase) 활성이 시간 의존적으로 급격히 떨어졌고, P450IIE protein양이 똑같은 방식으로 감소되었으나 mRNA level은 변화가 없었다. $CCl_4$에 의해서 P450IIE는 protein의 특이적인 파괴에 의한 post-translational reduction됨을 알 수 있었다. 반면에 당뇨병, starvation, high-fat diet등 ketosis시에는 P450IIE 효소활성이 2-3배 증가되었고, P450IIE protein양도 같은 수준으로 증가되었으며, mRNA도 증가 되었다. Ketosis시에는 P450IIE가 pretranslational activation됨을 알 수 있었다.

  • The relationship between Schwanniomyces occidentalis CBS 2863 (formerly castellii) and CBS 1153 (formerly alluvius), and their variety persoonii was examined at alpha-amylase gene level. Using Sch. occidentalis alpha-amylase gene as probe, Sch. occidentalis alpha-amylase gene homologues were obtained from Sch. occidentalis CBS 1153 and Sch. occidentalis var. persoonii. The restriction analysis of these homologues showed that the restriction enzyme sites between Sch. occidentalis CBS 2863 and CBS 1153 was identical but different between these strains and Sch. occidentalis var. persoonii.

High-Resolution Microarrays for Mapping Promoter Binding sites and Copy Number Variation in the Human Genome

  • NimbleGen has developed strategies to use its high-density oligonucleotide microarray platform (385,000 probes per array) to map both promoter binding sites and copy number variation at very high-resolution in the human genome. Here we describe a genome-wide map of active promoters determined by experimentally locating the sites of transcription imitation complex binding throughout the human genome using microarrays combined with chromatin immunoprecipitation. This map defines 10,567 active promoters corresponding to 6,763 known genes and at least 1,196 un-annotated transcriptional units. Microarray-based comparative genomic hybridisation (CGH) is animportant research tool for investigating chromosomal aberrations frequently associated with complex diseases such as cancer, neuropsychiatric disorders, and congenital developmental disorders. NimbleGen array CGH is an ultra-high resolution (0.5-50 Kb) oligo array platform that can be used to detect amplifications and deletions and map the associated breakpoints on the whole-genome level or with custom fine-tiling arrays. For whole-genome array CGH, probes are tiled through genic and intergenic regions with a median probe spacing of 6 Kb, which provides a comprehensive, unbiased analysis of the genome.
