Caffeic Acid Phenethyl Ester Inhibits Cell Proliferation and Induces Apoptosis in Human Ovarian Cancer Cells

  • The phenethyl ester of caffeic acid (CAPE), an active component of honeybee propolis extract, is shown to inhibit cancer growth previously. However, studies on human ovarian cancer are largely obscure. This study evaluated the effects of CAPE as a potential anti-proliferative and pro-apoptotic agent in the human ovarian cancer line, OVCAR-3. CAPE treated OVCAR-3 cells showed inhibition of cell viability and proliferation in a dose-dependent manner by WST-1 assay, LDH assay and bromodeoxyuridine (BrdU) incorporation assay. Furthermore, CAPE-mediated OVCAR-3 cell growth inhibition was associated with apoptotic changes as evident by cell cycle arrest and accumulation of cells in the apoptotic phase and DNA fragmentation. Taken together, CAPE inhibits cell proliferation via DNA synthesis reduction and induces apoptotic cell death via DNA damage, thus elucidating a novel, plausible mechanism of CAPE anti-tumorigenic property in OVCAR-3 cells.

Production and Characterization of Antihypertensive Angiotensin I-Converting Enzyme Inhibitor from Pholiota adiposa

  • Angiotensin I-converting enzyme (ACE) inhibitors have generally been very useful to remedy or prevent hypertension. This study describes the extraction and characterization of an ACE inhibitor from the fruiting body of Pholiota adiposa ASI 24012, which can be used as an antihypertensive drug. The maximal ACE inhibitory activity $(IC_{50};0.25mg)$ was obtained when the fruiting body of Pholiota adiposa ASI 24012 was extracted with distilled water at $30^{\circ}C$ for 12 h. After the purification of ACE inhibitor with ultrafiltration, Sephadex G-25 column chromatography, and reverse-phase HPLC, an active fraction with an $IC_{50}$ of 0.044 mg was obtained. The purified ACE inhibitory peptide was a novel pentapeptide, showing very little similarity to other ACE inhibitory peptide sequences. The molecular mass of the purified ACE inhibitor was estimated to be 414 daltons with a sequence of Gly-Glu-Gly-Gly-Pro, and showed a clear antihypertensive effect on spontaneously hypertensive rats (SHR) at a dosage of 1 mg/kg.

Evaluation on Anticancer Effect Against HL-60 Cells and Toxicity in vitro and in vivo of the Phenethyl Acetate Isolated from a Marine Bacterium Streptomyces griseus

  • We previously identified Streptomyces griseus as an anti-cancer agent (Kim et al., 2014). In this study, we isolated compounds from S. griseus and evaluated their anticancer effect and toxicity in vitro and in vivo. Preparative centrifugal partition chromatography (CPC) was used to obtain three compounds, cyclo($_{\small{L}}$-[4-hydroxyprolinyl]-$_{\small{L}}$-leucine], cyclo($_{\small{L}}$-Phe-trans-4-hydroxy-$_{\small{L}}$-Pro) and phenethyl acetate (PA). We chose PA, which had the highest anticancer activity, as a target compound for further experiments. PA induced the formation of apoptotic bodies, DNA fragmentation, DNA accumulation in $G_0/G_1$ phase, and reactive oxygen species (ROS) formation. Furthermore, PA treatment increased Bax/Bcl-xL expression, activated caspase-3, and cleaved poly-ADP-ribose polymerase (PARP) in HL-60 cells. Simultaneous evaluation in vitro and in vivo, revealed that PA exhibited no toxicity in Vero cells and zebrafish embryos. We revealed, for the first time, that PA generates ROS, and that this ROS accumulation induced the Bcl signaling pathway.

High-Dose Nicotinamide Suppresses ROS Generation and Augments Population Expansion during CD8+ T Cell Activation

  • During T cell activation, mitochondrial content increases to meet the high energy demand of rapid cell proliferation. With this increase, the level of reactive oxygen species (ROS) also increases and causes the rapid apoptotic death of activated cells, thereby facilitating T cell homeostasis. Nicotinamide (NAM) has previously been shown to enhance mitochondria quality and extend the replicative life span of human fibroblasts. In this study, we examined the effect of NAM on $CD8^+$ T cell activation. NAM treatment attenuated the increase of mitochondrial content and ROS in T cells activated by CD3/CD28 antibodies. This was accompanied by an accelerated and higher-level clonal expansion resulting from attenuated apoptotic death but not increased division of the activated cells. Attenuation of ROS-triggered pro-apoptotic events and upregulation of Bcl-2 expression appeared to be involved. Although cells activated in the presence of NAM exhibited compromised cytokine gene expression, our results suggest a means to augment the size of T cell expansion during activation without consuming their limited replicative potential.

Purification and Characterization of a Major Fibrinolytic Enzyme from Bacillus amyloliquefaciens MJ5-41 Isolated from Meju

  • Meju is a traditional Korean fermented soy product used as a key element for soy sauce and doenjang. Bacilli with antimicrobial activity were isolated from meju prepared by traditional methods at Sunchang county, Jeollabukdo, Korea. Six isolates were identified as Bacillus amyloliquefaciens by recA gene sequencing and RAPD-PCR. One isolate, B. amyloliquefaciens MJ5-41, showed the strongest fibrinolytic activity. A 27 kDa active fibrinolytic enzyme, AprE5-41, was purified from the culture supernatant of MJ5-41 grown on LB by chromatographic methods. The optimum pH and temperature for purified AprE5-41 were 7.0 and $45^{\circ}C$, respectively. AprE5-41 quickly degraded $A{\alpha}$ and $B{\beta}$ chains but not the ${\gamma}$-chain of fibrinogen. AprE5-41 exhibited the highest specificity for N-succinyl-Ala-Ala-Pro-Phe p-nitroanilide, a known substrate for ${\alpha}$-chymotrypsin, cathepsin G, and subtilisin BPN'. The structural gene, aprE5-41, was cloned by PCR and successfully expressed in B. subtilis.

Pro-Apoptotic Role of the Human YPEL5 Gene Identified by Functional Complementation of a Yeast moh1Δ Mutation

  • To examine the pro-apoptotic role of the human ortholog (YPEL5) of the Drosophila Yippee protein, the cell viability of Saccharomyces cerevisiae mutant strain with deleted MOH1, the yeast ortholog, was compared with that of the wild-type (WT)-MOH1 strain after exposure to different apoptogenic stimulants, including UV irradiation, methyl methanesulfonate (MMS), camptothecin (CPT), heat shock, and hyperosmotic shock. The $moh1{\Delta}$ mutant exhibited enhanced cell viability compared with the WT-MOH1 strain when treated with lethal UV irradiation, 1.8 mM MMS, $100{\mu}M$ CPT, heat shock at $50^{\circ}C$, or 1.2 M KCl. At the same time, the level of Moh1 protein was commonly up-regulated in the WT-MOH1 strain as was that of Ynk1 protein, which is known as a marker for DNA damage. Although the enhanced UV resistance of the $moh1{\Delta}$ mutant largely disappeared following transformation with the yeast MOH1 gene or one of the human YPEL1-YPEL5 genes, the transformant bearing pYES2-YPEL5 was more sensitive to lethal UV irradiation and its UV sensitivity was similar to that of the WT-MOH1 strain. Under these conditions, the UV irradiation-induced apoptotic events, such as FITC-Annexin V stainability, mitochondrial membrane potential (${\Delta}{\psi}m$) loss, and metacaspase activation, occurred to a much lesser extent in the $moh1{\Delta}$ mutant compared with the WT-MOH1 strain and the mutant strain bearing pYES2-MOH1 or pYES2-YPEL5. These results demonstrate the functional conservation between yeast Moh1 and human YPEL5, and their involvement in mitochondria-dependent apoptosis induced by DNA damage.

Comparative ileal amino acid digestibility and growth performance in growing pigs fed different level of canola meal

  • The digestibility of different vegetable protein sources were investigated and the effects of supplementing canola meal (CM) as partial inclusions were studied in growing pigs, to determine the performance parameters and its economic importance. In Exp. 1, four pigs (average initial $BW=15.4{\pm}0.35kg$, 5 weeks of age) fitted with simple T-cannula at terminal ileum, were fed four diets following repeated $4{\times}4$ Latin square design having adoption period of 7 days. Diet 1 was Nitrogen free diet containing corn starch. Diets 2, 3, and 4 were the basal diet supplemented with soybean meal (SBM), rapeseed meal (RSM), and domestic CM respectively. The AID of crude protein was decrease in RSM in comparison to SBM supplementation. The AID of Dietary indispensable amino acids (DIAA) such as Lys, Meth, Pha, and dispensable amino acid Ala, Pro, Asp were decreased (P < 0.05) in RSM supplemented diets. The SID of DIAA does not differ but the SID of Asp was higher (P < 0.05) in RSM and CM diets while SID of Pro was lower (P < 0.05) in RSM in comparison to SBM supplemented diets. In Exp. 2, 192 growing pigs (average initial BW $24.76{\pm}2.55kg$) were randomly allotted to four dietary treatments with increasing levels of CM i.e. 0, 3.75, 7.50, and 11.25 % respectively. Diets were fed in meal form for 35 days. Increasing CM levels in diets had no effects (P > 0.05) on growth performance and apparent total tract digestibility (ATTD) of nutrients and energy. Total weight gain, total feed intake, and feed cost per kg weight gain were not affected by increasing levels of CM in diets but total feed cost (TFC) per pigs was linearly reduced (26.463 to 25.674; P < 0.05). Broadly, the AID, and SID of amino acid was reduced in RSM but was not effected in CM in comparison to SBM supplemented pigs. Moreover, increasing levels of CM in pigs diet had no effect on the ATTD and performance but TFC per pig was reduced. Thus CM inclusion of up to 11.25 % in diets can be used for reducing the production cost in growing pigs without any negative effect.

A Study on Self-concept, Shopping Orientation and Store Patronage Behavior of College Women's Fashion Life-style Segments (여대생의 패션 라이프 스타일에 따른 자아개념, 쇼핑성향, 상점애고에 관한 연구)

  • The purposes of the study were (1) to seg-ment the female college apparel market based on fashion life style and (2) to develop a pro-file of each segment regard to self-concept, shopping orientation and patronage behavior. The data were collected through questionnaire by random sample of 526 female college students. By cluster analysis of lifestyle factors, three groups were identified, (fashion leaders, fashion followers and fashion aversion) Three groups were then compared through multivariate analysis of variance and chi-square statistics on 10 self-concept variables, 6 shopping statistics on 10 self-concept variables, 6 shopping orientation factors and 1 patronage behavior variable. Significant difference were found among the three groups on all these variables which indi-cate that fashion lifestyle can be a useful base for segmenting female apparel market and these groups are unique in terms of self-con-cept, shopping orientation and patronage behavior.

Changes in Functional Properties of Casein by Different Chemical Modifications

  • Casein was chemically modified with acetic, succinic, and maleic anhydride and changes in functional pro-perties were evaluated as affected by the degree of modification. Chemical modification resulted in casein with unique functional properties depending upon the type of anhydrid used and the degree of modification. It was possible to control heat coagulation, calcium precipitability, forming and emulsion capacity and stability. At pH 4.5 heat coagulation was 0% in the case 74.1% acetylated casein; on the contrary, succinylation and maleyation resulted in highly heat sensitive protein. Foaming properties were improved markedly by suc-cinylation and maleylation at pH 4.5. However, emulsifying properties were enhanced only by maleylation.

  • The effect of cations on the formation of protein aggregates was examined by in vitro refolding of mutant tryptophan synthase $\alpha$-subunit in which Pro 24 was replaced by Leu. $NH^{4+},; K{^+}; and; Na^{+}$ and no effect, but $Mg^{2+}; and; Ca^{2+}$stimulated the formation of protein aggregates in dose-dependent manner. It is suggested that $Mg^{2+} and Ca^{2+}$ may be implicated in the formation of protein aggregates in vivo.
