A Study on Accumulation of Cadmium and Induction of Metallothionein in Organs of Rats by Cadmium and Zinc (카드뮴 및 아연투여에 의한 흰쥐의 장기내 Cadmium 축적 및 Metallothionein 생성변화)

  • Tolerance to toxic effects of cadmium (Cd), including lethality has been shown following pretreatment with cadmium and zinc. This study was designed to determine if tolerance also develops to Cd-induced hepatotoxicity and renal toxicity. Three groups of rats (A, B, C), each consisting of 108 rats, were studied and each group was divided into three subgroups (1, 2, 3), 12 rats for each subgroup. Rats were subcutaneously pretreated with saline (A), CdCl$_2$ (0.5 mg/kg, B), and ZnCl$_2$ (13.0 mg/kg, C) during time periods of 5 days. At the end of the period, rats were challenged with CdCIa (3.0 and 6.0 mg/kg) by intraperitoneal injection. As for the cadmium levels in rat tissues after pretreatments, it was highest in the liver. Then kidney, heart, blood and muscle followed it in that order. After 24, 48 and 96 hours of intraperitoneal injection by challenge doses the concentration of cadmium in liver and kidney increased proportionally to the increase of challenge dosage. However metallothioneins in liver and kidney were increased by the pretreatment of cadmium and zinc. These data indicate the liver is a major target organ of acute Cd poisoning, and suggest that cadmium induced hepatic injury, via release of Cd-MT, may play and important role in the nephrotoxicity observed in response to short-term exposure to cadmium. This result suggests that increasing cadmium concentrations, gradually accumulating in liver and kidney as the result of the pretteatmerit, served to induced the synthesis of metallothionein, thus making them resistant to the challenge from cadmium.

Si-Containing Nanostructures for Energy-Storage, Sub-10 nm Lithography, and Nonvolatile Memory Applications

  • This talk will begin with the demonstration of facile synthesis of silicon nanostructures using the magnesiothermic reduction on silica nanostructures prepared via self-assembly, which will be followed by the characterization results of their performance for energy storage. This talk will also report the fabrication and characterization of highly porous, stretchable, and conductive polymer nanocomposites embedded with carbon nanotubes (CNTs) for application in flexible lithium-ion batteries. It will be presented that the porous CNT-embedded PDMS nanocomposites are capable of good electrochemical performance with mechanical flexibility, suggesting these nanocomposites could be outstanding anode candidates for use in flexible lithium-ion batteries. Directed self-assembly (DSA) of block copolymers (BCPs) can generate uniform and periodic patterns within guiding templates, and has been one of the promising nanofabrication methodologies for resolving the resolution limit of optical lithography. BCP self-assembly processing is scalable and of low cost, and is well-suited for integration with existing semiconductor manufacturing techniques. This talk will introduce recent research results (of my research group) on the self-assembly of Si-containing block copolymers for the achievement of sub-10 nm resolution, fast pattern generation, transfer-printing capability onto nonplanar substrates, and device applications for nonvolatile memories. An extraordinarily facile nanofabrication approach that enables sub-10 nm resolutions through the synergic combination of nanotransfer printing (nTP) and DSA of block copolymers is also introduced. This simple printing method can be applied on oxides, metals, polymers, and non-planar substrates without pretreatments. This talk will also report the direct formation of ordered memristor nanostructures on metal and graphene electrodes by the self-assembly of Si-containing BCPs. This approach offers a practical pathway to fabricate high-density resistive memory devices without using high-cost lithography and pattern-transfer processes. Finally, this talk will present a novel approach that can relieve the power consumption issue of phase-change memories by incorporating a thin $SiO_x$ layer formed by BCP self-assembly, which locally blocks the contact between a heater electrode and a phase-change material and reduces the phase-change volume. The writing current decreases by 5 times (corresponding to a power reduction of 1/20) as the occupying area fraction of $SiO_x$ nanostructures varies.

Pretreatment Effect on CO Oxidation over Highly Ordered Mesoporous Silver Catalyst

  • Highly ordered mesoporous silver material was successfully synthesized from a mesoporous silica template (KIT-6) with 3-D channel structure using the nano-replication method. The effects of $H_2$ or $O_2$ pretreatments on the catalytic performance of the mesoporous silver were investigated using a temperature programmed CO oxidation technique in a fixed bed reactor. The mesoporous silver material that was pretreated with $H_2$ exhibited an excellent catalytic activity compared to the as-prepared and $O_2$-pretreated catalysts. Moreover, this present mesoporous silver material showed good catalytic stability. For the CO oxidation, the apparent activation energy of the $H_2$-pretreated mesoporous silver catalyst was $61{\pm}0.5\;kJ\;mol^{-1}$, which was also much lower than the as-prepared ($132{\pm}1.5\;kJ\;mol^{-1}$) and $O_2$-pretreated ($124{\pm}1.4\;kJ\;mol^{-1}$) catalysts.

Pretreatment and enzymatic saccharification process of rapeseed straw for production of bioethanol

  • This study was conducted to evaluate the yield of bio-ethanol produced by separate hydrolysis and fermentation (SHF) with the pretreated rapeseed straw (RS) using crude enzyme of Cellulomonas flavigena and Saccharomyces cereviase. Crude enzyme of C. flavigena showed enzymatic activity of 14.02 U/mL for CMC 133.40 U/mL, for xylan 15.21 U/mL, for locust gum and 15.73 U/mL for rapeseed straw at pH 5.0 and $40^{\circ}C$, respectively. The hemicellulose contents of RS was estimated to compromise 36.62% of glucan, 43.20% of XMG (xylan + mannan + galactan), and 2.73% of arabinan by HPLC analysis. The recovering ratio of rapeseed straw were investigated to remain only glucan 75.2% after 1% $H_2SO_4$ pretreatment, glucan 45.44% and XMG 32.13% after NaOH, glucan 44.75% and XMG 5.47% after $NH_4OH$, and glucan 41.29% and XMG 41.04% after hot water. Glucan in the pretreatments of RS was saccharified to glucose of 45.42 - 64.81% by crude enzyme of C. flavigena while XMG was made into to xylose + mannose + galactose of 58.46 - 78.59%. Moreover, about 52.88 - 58.06 % of bio-ethanol were obtained from four kinds of saccharified solutions by SHF using S. cerevisiae. Furthermore, NaOH pretreatment was determined to show the highest mass balance, in which 21.22 g of bio-ethanol was produced from 100 g of RS. Conclusively, the utilization of NaOH pretreatment and crude enzyme of Cellulomonas flavigena was estimated to be the best efficient saccharification process for the production of bio-ethanol with rapeseed straw by SHF.

Fermentative Hydrogen Production from the Pretreated Food-Processing Waste and Sewage Sludge using Chemical/Ultra-Sonication (두부제조폐기물과 하수슬러지의 화학/초음파 전처리에 의한 가용화 및 혐기발효 수소생산)

  • Acid and alkali pretreatments were applied to tofu processing waste (TPW) to increase the solubility of ingredients in TPW. Pretreatment at 1.0% of HCl and 2.5% of NaOH condition resulted in the increase of SCOD concentration from 3.2 g COD/L to 27 g COD/L and 33 g COD/L, respectively. The acid and alkali-pretreated TPW was studied for its fermentative $H_2$ production capacity in batch mode using a thermophillic mixed culture. Alkali pretreatment on presence of 2.5% NaOH exhibited more soluble portion released compared to acid pretreatment using HCl, however the $H_2$ production from acid pretreated TPW was better than alkali-pretreated TPW probably due to the sodium inhibition on microbial activity. In addition, sewage sludge was externally added to the acid-pretreated (1.0% HCl) TPW by 20% (on volume basis). Average H2 production rate was increased from 31 to 78 ml/L-broth/hr, and it was attributed to the high buffer capacity and abundant nutrients especially divalent cation in sewage sludge.

Effect of Packaging Methods on the Storage of Banana (포장방법이 바나나 저장성에 미치는 영향)

  • The storage stabilities of philipines' bananas were investigated according to the storage pretreatments and packaging methods during 60 days storage at $13^{\circ}C$. The better storage stability was got in bananas packaged with 60{\mu}m low density polyethylene (PE) film inside the carton box than those without inside pack. Radiation gave the adverse effects on bananas' storage stabilities. Shelf-life of bananas in PE package was 60 days, but that of radiated bananas was only 20 days. And shelf-life of bananas in no inner pack was 20 days. Soluble solid content of bananas packaged with PE film was 17 Brix degree after 60 days storage, but that with no inner package was around 20 Brix after 20 days. Color was below 3 in color chart after 60 days storage and hardness was rapidly decreased from 40 days in bananas packaged with PE film, but color was became to 3 color after 30 days and hardness was rapidly decreased after 20 days in bananas without inner package. Percentage of deteriorated bananas were below 3% in PE films after 60 days storage, but 100% after 40 days in bananas without inner package.

Naesohwangryeon-tang Induced Apoptosis and Autophagy in A549 Human Lung Cancer Cells

  • Objectives: Naesohwangryeon-tang (NHT) is a type of traditional herbal formula, however, little is known about its antitumor activity. In this study, the antitumor properties of NHT was evaluated in human lung adenocarcinoma cells. Methods: To check the inhibitory effect of NHT, MTT assay was performed. Cell cycle analysis and detection of ROS production were conducted by flow cytometry. To evaluate the signaling pathway, Western blotting was conducted. Results: Our results showed that the decrease of cell proliferation by NHT stimulation occurred more significantly in A549 cells than in NCI-H460 cells. In addition, NHT-induced apoptosis was associated with the activation of caspases and production of reactive oxygen species (ROS). NHT-induced apoptosis was attenuated after pretreatments with z-VAD-fmk or N-acetylcysteine, suggesting that NHT-induced apoptosis was caspaseand ROS-dependent. Interestingly, NHT treatment led to the development of autophagic vesicular organelles and upregulation of several autophagy-related genes. The pretreatment of bafilomycin A1 decreased apoptosis slightly but increased cell viability in the presence of NHT. Conclusion: These findings indicated that NHT induces both apoptosis and cell-protective autophagy in human lung cancer cells. This data suggests that NHT might be a novel herbal drug for lung cancer.

Advanced Wastewater Treatment using Bioreactor Combined with Alternative Membrane (하수의 고도처리를 위한 저비용 저에너지의 대체 막을 조합한 생물반응기의 개발)

  • In order to decrease the high costs of membrane process, we have tried to develop two alternatives to membrane; a cartridge type filter and a metal membrane were tested for the high permeation flux with low cost and low energy. This research mainly focused on three points; 1) operation with high permeation flux by using of a cartridge type filter and a metal membrane, 2) removals of the filterable organic materials (FOC) by pretreatments for the membrane fouling control, and 3) advanced wastewater treatment by SMBR process with intermittent aeration and high MLSS. An Intermittently aerated membrane bioreactor using a submerged micro filter (cartridge type) was applied in laboratory scale for the advanced wastewater treatment. To minimize membrane fouling, intermittent aeration was applied inside of the filter with $3.0kg_f/cm^2$. The experiments was conducted for 6 months with three different HRTs (8, 10, 12 hr) and high MLSS of 6,000 and 10,000mg/L. The filtration process could be operated up to 50 days with permeation flux of 500LMH. Regardless of the operating conditions, more than 95% of COD, BOD and SS were removed. Fast and complete nitrification was accomplished, and denitrification was appeared to be the rate-limiting step. More than 75% T-N could be removed due to the endogenous denitrification. T-P removal efficiency was increased to 80% under the condition of MLSS 10,000mg/L.

The Effect of Pretreatment of Veneer on the Improvement of Plywood Quality (단판(單板)의 약제처리(藥劑處理)가 합판성질개선(合板性質改善)에 미치는 영향(影響))

  • 1. In order to make the improvement of plywood quality, this study has been made. The pretreatments applied to the veneers are as follows. a) The soaking treatment of the veneer in 30 percent methanol solution for 24 hours was applied to remove some resinous materials which may cause many stain discolouring on the face of finished plywood. b) The preservative treatment of the veneer in 2 percent malenit solution for 2 to 3 minutes was adopted to make resistance against decay and insect damages. c) The fire retardant treatment of the veneer in 40 percent ammonium phosphate solution for 2 hours was applied to give retardation effect against fire burning. 2. The results summarized in this study are as follows. a) One percent resinous materials was extracted, after the soaking of the veneers in 30 percent diluted methanol. b) No marks of the dirty stains of resinous materials on the face of the treated plywood was shown, although many quite dirty stains on the face of untreated plywood have contrary seen. c) However, the strip shear test strength of the treated plywood was not decreased. It means that there is no difference in the strength between the treated plywood and the untreated plywood. The strength values were 25.08 kg/$cm^2$ and 24.98 kg/$cm^2$, respectively. d) The strip shear test strength of plywood made of the treated veneers in 2 percent malenit solution was not decreased. e) The slight decrease of the strip shear test strength of the treated plywood made of the treated veneer in 40 percent ammonuim phosphate solution was shown. However, the remarkable difference of the fire retardation activities between the treated specimens and the untreated specimens has seen as in Table 10, that is, the fire proofed specimens had taken about 28 seconds to start to burn, while the untreated specimens had taken 15 seconds to reach to burning. This means that the fire retardation effect of the fire proofed plywood was greater than that of the unproofed plywood.
