In order to induce somatic embryogenesis from the stem explants and anther of Kalanchoe daigremontiana, the explants were cultured on MS medium supplemented with auxin (2,4-D, IAA, NAA) and/or cytokinin (BAP) for 8 weeks. Callus from explants was induced most efficiently on MS medium containing. 2.0mg/L NAA and 0.2mg/L BAP. Somatic embryogenesis in stem callus was formed by transfering embryogenic callus from induction media containing growth regulators to medium without growth regulators and then to the medium containing auxin and cytokinin (0.1 mg/L IAA and 1.0mg/L BAP). Callus formation occurred actively in the anthers at early uninucleate stage, and by low temperature pretreatment at $4^{\circ}C$ for 3days. Somatic embryogenesis from the anther callus was induced on MS medium containing 1.0mg/L NAA and 1.0mg/L BAP, 2.0mg/L NAA and 0.2mg/L BAP. The tetraploid of 5.4% was obtained among plants regenerated from anthers.

Renewable Portfolio Standards(RPS) is a regulation that requires a renewable energy generated from eco-friendly energy sources such as biomass, wind, solar, and geothermal. The RPS mechanism generally is an obligatory policy that places on electricity supply companies to produce a designated fraction of their electricity from renewable energies. The domestic companies to supply electricity largely rely on wood pellets in order to implement the RPS in spite of undesirable situation of lack of wood resources in Korea. This means that the electricity supply companies in Korea must explore new biomass as an alternative to wood. Palm kernel shell (PKS) and empty fruit bunch (EFB) as oil palm wastes can be used as raw materials used for making pellets after their thermochemical treatment like torrefaction. Torrefaction is a pretreatment process which serves to improve the properties including heating value and energy densification of these oil palm wastes through a mild pyrolysis at temperature typically ranging between 200 and $300^{\circ}C$ in the absence of oxygen under atmospheric pressure. Torrefaction of oil palms wastes at above $200^{\circ}C$ contributed to the increase of fixed carbon with the decrease of volatile matters, leading to the improvement of their calorific values over 20.9 MJ/kg (=5,000 kcal/kg) up to 25.1 MJ/kg (=6,000 kcal/kg). In particular, EFB sensitively responded to torrefaction because of its physical properties like fiber bundles, compared to PKS and hardwood chips. In conclusion, torrefaction treatment of PKS and EFB can greatly contribute to the implement of RPS of the electricity supply companies in Korea through the increased co-firing biomass with coal.

In order to dissolve Au, Ag, and other valuable metals from gold ore concentrate, raw gold concentrate was pre-treated by roasting and salt-roasting at $750^{\circ}C$. The roasted concentrate was treated with aqua regia digestion to dissolve the valuable metals and higher amount of Au, Ag, and valuable metals were extracted from the roasted concentrates than from the raw concentrate. Higher amount of these metals were also extracted from the salt-roasted concentrate than from the roasted concentrate. The results of the gold dissolution experiments showed that the gold dissolution was most efficient when particle size, roasting temperature, and the percentage of added salt in salt roasting were about $181{\sim}127{\mu}m$, $750^{\circ}C$, and was 20.0%, respectively. The XRD analysis suggests that quartz and pyrite were not destroyed even through roasting at $750^{\circ}C$ and decomposition with aqua regia. However, through salt roasting, pyrite was completely decomposed, whereas quartz could not be destroyed through salt-roasting at $750^{\circ}C$ and aqua regia digestion. Accordingly, it was expected that the gold contained in quartz can not be dissolved through salt-roasting and treatment with aqua regia.
An ubiquitous bacterium, Pseudomonar cepacia KH410 was isolated from fresh water plant root and identified. Adsorption of heavy metals of lead, cadmium and copper by this strain was investigated. Optimal conditions foradsorption was 1.0 dry g-biomass, at pH 4.0 and temperature of $40^{\circ}C$. Adsorption equilibrium reached max-imum after 120 min in 1000 mg/l metal solutions. The adsorption capacity (K) of lead was 5.6 times higher thancadmium and 4.0 times higher than that of copper. Adsorption of lead was applicable for Langmuir modelwhereas Freundlich model for cadmium and copper, respectively. Adsorption strength (1/n) of heavy metal ionswere in the order of lead>copper>cadmium. Uptake capacity of lead, cadmium and copper by dried cell was83.2,42.0,65.2 mg/g-biomass, respectively. Effective desorption was induced 0.1 M HCI for lead and 0.1 $HNO_3$ for cadmium and copper. Pretreatment to increase ion strength was the most effective with 0.1 M KOH.Uptake by immobilized cell was 77.8,58.5,71.2 mg/g-biomass for lead, cadmium and copper, respectively. Theimmobilized cell was more effective than ion exchange resin on removal of heavy metals in solution containinglight metals.
The purpose of this study was to investigate the aflatoxin $B_1$ binding of lactic acid bacteria (LAB) isolated from Korean traditional soybean paste and to evaluate the effect of incubation conditions and physico-chemical factors on the binding ability of LAB to this mutagen. The amount of aflatoxin $B_1$ bound by Enterococcus faecium DJ22, Lactobacillus fermentum DJ35, Lactobacillus rhamnosus DJ42, and Lactobacillus pentosus DJ47 was strain specific with the percent bound ranging from 19.3% to 52.1%. However, Enterococcus faecalis DJ14, Lactobacillus panis DJ29, and Pediococcus halophilus DJ50 strains did not exhibit any of the binding ability to aflatoxin $B_1$. For most strains, the binding ability was significantly affected by the environmental conditions such as the aflatoxin $B_1$ level, incubation time and temperature, and the initial cell count of LAB. The stability of the aflatoxin $B_1$-bacteria complexes was significantly more unstable after washing. In addition, the binding stability between viable and nonviable cells was not statistically significant. Treatment with heating, acidic pH, ${\alpha}$-amylase, protease, lysozyme, or sodium metaperiodate caused a significant (P<0.05) decrease in aflatoxin $B_1$ binding for the tested strains, suggesting that carbohydrates or proteins in the cell walls may be involved in aflatoxin $B_1$ binding ability. Since the aflatoxin $B_1$ binding of LAB was significantly reduced (P<0.05) by the pretreatment of the urea, the binding force observed in this study may have resulted from hydrophobic interaction.
Studies were made to optimize the simultaneous hydrolysis-fermentation (SSF) process for the production of ethanol from rice straw. Trichoderma sp. KI 7-2 was selected to produced cellulase by solid culture for SSF. Ethanol production was highest when the SSF process utilized koji culture of the fungus grown on a medium of wheat bran-rice straw 3 : 2 mixture with moisture content of 50% adjusted to pH 4.5 for 7 days as the enzyme source. It was found that pretreatment of the substrate is not necessary. To ferment 1g of rice straw by SSF 2.47 units of cellulase were required, and the initial yeast concentration of 2.5$\times$10$^{7}$ cell/$m\ell$ was found to be sufficient. Optimum pH and temperature for the process were 4.5 and 4$0^{\circ}C$, respectively. It was also found that higher ethanol concentration in the broth can be obtained by the addition of substrate or substrate and enzyme to SSF broth.
In this thesis, both soap and enzymatic degumming method were adopted and the optimum degumming conditions were obtained. Difference between the two degumming methods in silk fiber state was investigated and analyzed on the basis of the results of physical testings, polarizing microscopy, scanning electron microscopy, viscosity measurement, (${\alpha}$+$\varepsilon$) amino group contents measurement, birefringence measurement, amino acid analysis, thermal analysis, infrared spectroscopy and x-ray diffraction analysis. The results obtained were summarized as follows; Physical test results of the degummed silk fiber showed that the tenacity and the elongation of enzymatic degummed silk fiber were lower than those of soap degummed fiber. But SEM observation and amino acid analysis showed almost the same tendency in the two degumming methods. The viscosity of enzymatic degummed silk fiber was lower than that of soap degummed fiber, but (${\alpha}$+$\varepsilon$) amino group contents was higher in the enzymatic degummed fiber. It can be suggested that the enzymatic degummed silk fibroin was more degraded than the soap degummed fibroin. The birefringence, endothermic temperature of DSC spectrum, IR crystallinity and X-ray lateral order factor of enzymatic degummed silk fiber were higher than those of soap degummed fiber. It seems that the enzymatic degummed silk fiber has the higher crystallinity than that of soap degummed one according to the above results. However, it can be inferred that these differences between soap and enzymatic degummed fiber would be lessened if pretreatment and aftertreatment were included in the enzymatic degumming process.
Sabaiksan has been prescribed to treat various allergic diseases in herbal medicine which were induced by various vasoactive amine released from the mast cells. The constituents of Sabaiksan are Mori Cortex Radices(MCR), Lycii Cortex Radicis(LCR) and Glycyrrhizae Radix(GR). Recently, simple models of compound 48/80 induced anaphylactic shock and cutaneous reaction in vivo were developed to test various agents employed in the field of allergy and toxicology research. The purpose of this study is to evaluate the effects of Sabaiksan on compound 48/80 induced anaphylactic stock, cutaneous reaction and mesenteric mast cell degranulation rate in ICR mice, and on compound 48/80 induced peritoneal mast cell degranulation and histamine release in vitro. Groups of ICR mice were intraperitoneally pretreated with $100{\mu}{\ell}$ of saline, $MCR(2g/m{\ell}),\;LCR(2g/m{\ell}),\;GR(g/m{\ell})$ or Sabaiksan itself(MCR+LCR+GR) at 24, 12 and 1 hour before compound 48/80 solution ($10{\mu}{\ell}/gm$ B. W) were peritoneally given into them, and then mortality within 72 hours after the compound 48/80 injection, and mesenteric mast cell degranulation rate at 15 minutes after compound 48/80 injection were calculated. In vitro experiment, $400{\mu}{\ell}$ of rat peritoneal mast cell suspension$(10^6cell/m{\ell})$ were pretreated with $50{\mu}{\ell}$ of saline, $MCR(2g/m{\ell}),\;LCR(2g/m{\ell}),\;GR(g/m{\ell})$ or Sabaiksan itself at room temperature for 30 minutes, and then $50{\mu}{\ell}$ of compound 48/80 solution $(100{\mu}g/m{\ell})$ were added into it. 30 minutes after the addition of compound 48/80 solution, histamine release assay in the supernatant of peritoneal mast cell suspension were performed employing radioisotope enzymatic assay and morphologic changes of mast cells in each regular time point were photographed. Compared with controls, compound 48/80 induced anaphylactic shock was significantly inhibited by MCR and GR pretreatment into the ICR mice. Significant inhibition of compound 48/80 induced cutaneous reaction, mesenteric mast cell degranulation rate in vivo and histamine release from the rat peritoneal mast cells in vitro was observed only in MCR pretreated group. From the above results, it is suggested that MCR component of Sabaiksan may playa key role to suppress mast cell function since it has been applied to various allergic diseases.

This study was measured and analyzed from January to November 2019 to confirm the effect that Abietic acid, an asthma-causing substance, which can be exposed to workers in the electronics industry, is removed by plasma treatment. The experiment was carried out using a solder wire and natural rosin. Air at temperatures of 250℃, 300℃, and 350℃ was collected with a glass fiber filter paper using an air sampler for 10 minutes at a flow rate of 2ℓ/min. An analysis of the collected samples was performed by pretreatment with methyl alcohol and quantitative analysis by high performance liquid chromatography (HPLC). This procedure confirmed that abietic acid was generated in both natural rosin and solder wires, and the quantum of abietic acid increased as the treatment temperature increased. The amount of abietic acid was higher in natural rosin than solder wire. As a result of plasma treatment, a removal efficiency of about 92% or more was confirmed in natural rosin. A peak of abietic acid was not detected in the solder wire. Therefore, a removal efficiency of 100% was confirmed. This study, confirmed that abietic acid, an asthma-trigger can be generated in solder wire and natural rosin, and can be removed by plasma treatment.

In this research, we synthesized potassium ferrate(VI) acting as an oxidant, disinfectant, and coagulant, and used it to treat natural organic matter(NOM, HA and FA) in river water. The removal efficiencies obtained by $UV_{254}$ ranged from 20.7 to 73.6% for 10 mg/L HA and from 52.6 to 77.5% for 10 mg/L FA in Nakdong river sample as the ferrate dose varied from 2 to 46 mg/L(as Fe). However, the removal efficiencies by TOC analysis ranged from 0 to 20.3% for HA and from 0 to 26.6% for FA at the same ferrate doses. The removal efficiencies of NOM increased either with decreasing pH or with increasing temperature. The removal efficiency of HA by ferrate was comparable to those by traditional coagulants such as $Al_2(SO_4)_3{\cdot}18H_2O$, $FeSO_4{\cdot}7H_2O$, and FeO(OH). The reaction between ferrate and HA reached a steady state within 60 seconds, showing first-order with respect to the reaction time. The removal efficiencies of HA by traditional coagulants were improved by pretreatment of HA using a small amount of ferrate.
