The Expression of DNA Polymerase-$\beta$ and DNA Damage in Jurkat Cells Exposed to Hydrogen Peroxide under Hyperbaric Pressure

  • Long term exposure of Jurkat cells to 2 ATA pressure resulted in the inhibition of cell growth. Under a 2 ATA pressure, the morphological changes in the cells were visualized by electron microscopy. The cells exhibited significant inhibitory responses after three passages. However, short-term exposure study was carried out, 2 ATA pressure may have beneficial effects. The Jurkat cells were exposed to $H_2O_2$ (25 and $50{\mu}M$) in order to induce DNA damage, and then incubated under at either normal pressure or 2 ATA for 1 or 2 hours in order to recover the DNA damage. The extent of DNA damage was determined via Comet assay. More recovery from DNA damage was observed at 2 ATA than at normal pressure. The activity of the DNA repair enzymes, DNA polymerase-$\beta$, was also evaluated at both normal pressure and 2 ATA. The activity of DNA polymerase-$\beta$ was observed to have increased significantly at the 2 ATA than at normal pressure. In conclusion, the effects of hyperbaric pressure from 1 ATA to 2 ATA on biochemical systems can be either beneficial or harmful. Long term exposure to hyperbaric pressure clearly inhibited cell proliferation and caused genotoxic effects, but short-term exposure to hyperbaric pressure proved to be beneficial in terms of bolstering the DNA repair system. The results of the present study have clinical therapeutic application, and might prove to be an useful tool in the study of genotoxicity in the future.

The Characteristics of Solid Mixing in a Vibrating Type Feeder and Pressure Fluctuation of Packing Materials for a Fluidized Bed Combustor (유동층 연소로에서 진동형 공급기의 고체혼합 및 충전물에 대한 압력요동 특성)

  • This study attempts to analyse the solid mixing in the feeder and the packing effect for pressure fluctuations in the fluidized bed. To study the mixing characteristics of solid in vibrating feeder for the stable operations of fluidized combustion, the system consisted of two groups of particles such that fine particles were located on the top of the coarse particles before vibratory mixing had started. The effects of packing materials on the pressure fluctuations in a fluidized bed were analysed by using a statistical method to interpret the behavior of fluidized bed. The experiments were carried out in a fluidized bed of 6.7cm-ID, and the experimental variables were particle sizes, of 115 to 1,015$\mu\textrm{m}$ in diameter and the multi-sized particles haying Rosin-Rammler and Gaussian distributions. The settled bed heights of particles to diameter ratios (L/D) were ranged from 0.5 to 2.0. And fluidizing of particles was carried out by air. The packing materials used were screen packing, and the properties of the pressure fluctuations in the fluidized bed were measured by a differential pressure transducer. The properties of the pressure fluctuations calculated were the mean, the standard deviation, and the major frequency of the power spectral density functions. From the characteristics of fluidizing, it was found that the standard deviation of pressure fluctuations could be effectively used to explain the fluidized phenomena, and the packing materials affected severely the properties of the pressure fluctuations. As a result, from the interpretation by spectral analysis, the effects of measuring radius of pressure fluctuations on standard deviation were constant in the case of the fluidized bed with and without packing materials. However, the effects of measuring the height of pressure fluctuations on standard deviations were linear increasing for the fluidized bed with packing materials, but were constant for the fluidized bed without packing materials at 4.5cm above the gas distributor. The major frequency of pressure fluctuations was found to be nearly independent of fluidized system. Also, the major frequency of pressure fluctuations decreased with increasing packing size, and it had maximum value at 10% of the packing amount.

Automatic Blood Pressure Control Using PI Controller with $H_{\infty}$ Loop-Shaping

  • In this paper, we show a new form of blood pressure controller combined PI control with $H_{\infty}$ loop-shaping. Hypertensive patients or post-operative patients need to maintain normally blood pressure. Exact regulation of blood pressure is needed for maintaining variable blood pressure of preventing complications. The regulation of blood pressure is achieved by injecting drugs, and usually sodium nitroprusside is used as those kinds of drugs. It is necessary to control the infusion rate sodium-nitroprusside carefully to achieve the desired blood pressure. It has been known that regulation of blood pressure by automatic controller is more effective than regulation of blood pressure by human operators. The control of blood pressure has many constraints and uncertainties. Most of biological system has the time-varying variables and the side effects such as increased risk of sepsis and organ failure. To solve such a problem, we design a new robust PI controller using $H_{\infty}$ loop-shaping to decrease noise effects that come out from human body and errors for time delay. The system with designed controller shows more stable control of mean blood pressure and more robust performance for uncertainties. Validation methods for the control performance are confirmed to computer simulations.

Experimental study on the effects of Guibitanggamibang on blood pressure and hyperlipidemia (귀비탕가미방(歸脾湯加味方)이 고혈압(高血壓) 및 고지혈증(高脂血症)에 미치는 영향(影響))

  • In order to study the effects of Guibitanggamibang on blood pressure and hyperlipidemia, experimental study were performed on hypertension in normal and SHR rats, and on hyperlipidemia induced by Triton WR-1339 in normal rats. Also the level of total cholesterol, triglyceride, LDL-cholesterol, HDL-cholesterol, phospholipid and transaminase(GOT, GPT) were measured. The results are summarized as follows: 1. After Guibitanggamibang was given to normal rats, the results showed that inhibitory effects on blood pressure and heart beat were not statistically significant. 2. After Guibitanggamibang was given to SHR rats, the results showed that inhibitory effects on blood pressure were statistically significant. 3. In the model of hyperlipidemia induced by 2% cholesterol food, Guibitanggarrubang had significantly-decreasing effects on total cholesterol, triglyceride, HDL-cholesterol, LDL-choleste rol, Transaminase(GOT, GPT) level in serum. 4. In the model of hyperlipidemia induced by Triton WR-1339, Guibitanggamibang had significantly-decreasing effects on total cholesterol, triglyceride, LDL-cholesterol, phospholipid, Transaminase(GOT, GPT) level in serum. 5. In the model of hyperlipidemia induced by Triton WR-1339, Guibitanggamibang had no significant effects on HDL-cholesterol level in serum. These results show that Guibitanggamibang(歸脾湯加味方) has significant inhibitory effects on blood pressure and hyperlipidemia and that it could be clinically applied for hypertension and hyperlipidemia.

Effects of the curing pressure on the torsional fatigue characteristics of adhesively bonded joints (경화 압력이 접착 조인트의 비틀림 피로 특성에 미치는 영향)

  • Adhesive joints have been widely used for fastening thin adherends because they can distribute the load over a larger area than mechanical joints, require no hole, add very little weight to the structure and have superior fatigue resistance. However, the fatigue characteristics of adhesive joints are much affected by applied pressure during curing operation because actual curing temperature is changed by applied pressure and the adhesion characteristics of adhesives are very sensitive to manufacturing conditions. In this study, cure monitoring and torsional fatigue tests of adhesive joints with an epoxy adhesive were performed in order to investigate the effects of the applied pressure during curing operation. From the experiments, it was found that the actual curing temperature increased as the applied pressure increased, which increased residual thermal stress in the adhesive layer. Therefore, the fatigue life decreased as the applied pressure increased because the mean stress during fatigue tests increased due to the residual thermal stress.

Synthesis and pressure effects on the La doped CaFe2As2

  • We have synthesized La doped $CaFe_2As_2$ single crystals with Sn flux in an evacuated quartz ampule. Doping and pressure effects on the magnetic and superconducting properties of the under-doped $Ca_{1-x}La_xFe_2As_2$ (x=0.08, 0.1) were studied by measuring electrical resistivity under quasi-hydrostatic pressure up to 21 kbar. Magnetic transition temperatures for all studied concentrations were sharply suppressed with slight amplitude of pressure, less than 3 kbar, while superconducting transition temperatures were robust against pressure. In this communication, we report temperature-pressure phase diagram for the La-doped $CaFe_2As_2$ single crystals.

Effects of boundary layer and liquid viscosity and compressible air on sloshing characteristics

  • In this paper, numerical investigations for tank sloshing, based on commercial CFD package FLUENT, are performed to study effects of boundary layer grid, liquid viscosity and compressible air on sloshing pressure, wave height and rising time of impact pressure. Also, sloshing experiments for liquids of different viscosity are carried out to validate the numerical results. Through comparison of numerical and experimental results, a computational model including boundary layer grid can predict the sloshing pressure more accurately. Energy dissipation due to viscous friction leads to reduction of sloshing pressure and wave elevation. Sloshing pressure is also reduced because of cushion effect of compressible air. Due to high viscosity damping effect and compressible air effect, the rising time of impact pressure becomes longer. It is also found that liquid viscosity and compressible air influence distribution of dynamic pressure along the vertical tank wall.

Dynamic Response of Pneumatic Transmission Lines (공압 전달관의 동적 응답)

  • Transient analysis for compressible fluid flow has been performed experimentally and analytically to study the dynamic characteristics of the end volume transmission lines following a sudden pressure change a its entrance. The numerical method was developed based on the method of characteristics. The sudden pressure at its entrance was generated by rupture of diaphragm in a shock tube. The sudden pressure was used to obtain the response, as input signal for the numerical analysis. The response to the sudden pressure at the end volume was measured using a pressure transducer. The experimental result shows good agreements with the numerical result. The effects of tube length, its diameter and end volume magnitude are evaluated on the responses of the pressure and on the damping factor. It is found that the viscous damping effects on the response through the transmission pipeline becomes larger with increasing pi;eline length and decreasing diameter of the pipe and the fluid-elastic stiffness decreases with increasing the terminal volume. The numerical approach presented in this paper can be very useful in designing the instrument and control system.

Numerical Study on the Characteristics of Pressure Pulsations according to Design Factors of Fuel Rail with Self Damping Effect (자체 맥동 감쇠 효과를 갖는 연료레일의 설계 변수별 압력맥동 특성에 관한 수치적 연구)

  • In general, pulsation damper is installed in fuel rail for conventional MPI engine to decrease undesirable noise in vehicle cabin room. However, pulsation damper is so expensive that there are prevailing studies to reduce fuel pressure pulsations with integrated damping effect. This paper is one of basic studies for development of fuel rail to abate pulsations with self-damping effect. Primarily, the pressure pulsation characteristics was investigated with aspect ratio of cross section, wall thickness, and materials of fuel rail. A high aspect ratio or thin wall was found to absorb the pressure pulsations effectively. But volume effects on the fuel pressure pulsation reductions were not especially significant than cross section effects because volume increment rate is larger than pressure pulsation reduction rate. The fuel rail made of aluminum is effective for reduction of pressure pulsation than that of low-carbon steel. Pressure change period increases on the basis of same lengths of supply line and fuel rail as the volume is enlarged and/or the thickness of wall is thinned.

The Effects of the Arc Pressure Variation on the Penetration by the filler Wire Feed Rate in Pulsed TIG Welding (펄스 TIG용접에서 필러 와이어 송급속도에 따른 아크압력 변동이 용입에 미치는 영향)

  • In the standpoint of the arc pressure, the effects of the filler wire feed rate on the penetration was investigated in this study. The pure Ar gas was used as a shield gas and the parameters investigated were the welding current and the filler wire feed rate. By making the experiment on the arc pressure, we could know that the arc pressure was fluctuated as the depth-into-arc of the tungsten rod. Instead of the filler wire, the tungsten rod was supplied into the molten pool to make this experiment. Because the filler wire melted in arc and then we couldn't measure the arc pressure. So, the tungsten rod - the highest melting point - was used. According to the depth-into-arc of the tungsten rod, the arc pressure could be measured by using the manometer. It was proved that the arc pressure got higher as the wire feed rate was slow. It is reported the arc pressure is proportion to welding voltage and the square of welding current. But, in the filler wire TIG welding, we could blow that arc pressure was fluctuated as the depth-into-arc of filler wire was changed. We could measure the arc pressure by the variation of the filler wire feed rate and could verify that it affected bead shape and penetration.
