Effect of Naturally Derived Substances on Motion Parameters of In Vitro Non-Freezing Preserved Pig Sperm

  • Artificial insemination (AI) has been performed widely in swine industry using fresh liquid sperm instead of frozen type of sperm. However fresh sperm are not able to preserve more than three days with optimal motility and other sperm parameters for the successful fertilization, since in vitro stored sperm has an oxidative stress that resulted increase of abnormality and acrosome reation. To overcome these major problems, novel preservative formulation is needed to neutralize the oxidative stress and to provide suitable physiological environment for sperm in in vitro. In this study, naturally derived substances such as Poncirus trifoliate (Trifoliate orange), Garcinia mangostana (Mangosteen), pig placenta and testis extracts were tested as sperm preservative agents. Placenta extracts (PE), trifoliate orange extracts (TOE), testes extracts (TE) and mangosteen extracts (ME) were applied to analyze specific parameters for sperm motion characteristics individually and combinatorial. Each individual extract treatment can accelerate the sperm motility but noticeably TOE, TE and ME treatments exhibited the considerable and significant preservation of sperm motility. PE, TE and ME showed a significant (p<0.05) increase in ALH after one week. Further we evaluated the five different combinations of these extracts on sperm motility and its motion characteristics. Surprisingly even after one week ME, TOE and TE combination significantly preserved the sperm motility about 75%. It is noteworthy that unlike individual extract treatment, combination of ME, TOE and TE simultaneously protect the sperm motility and its motion characteristics. Taken together these data conclude that addition of ME, TOE and TE can be effective for preservation of pig sperm.

Gamma Knife Radiosurgery for Vestibular Schwannomas

  • Vestibular schwannoma (VS) is a benign tumor typically originated in the schwann cell of vestibular nerve and usually accompany hearing symptom. Microsurgical removal and radiosurgery have a great role for the treatment of VS. Recently radiosurgery has been considered as an alternative or primary treatment for VS with the tremendous increase of patients who were treated with gamma knife radiosurgery (GKS) though microsurgery still takes the premier. By many published results, it is proved that GKS is a effective and noninvasive technique for VS, especially small sized tumors with satisfactory tumor control rate. The authors assumed that GKS can be expected to achieve satisfactory tumor control rate for small VS under 5cc in volume. A major interest regarding radiosurgery nowadays is to determine the optimal radiation dose for hearing preservation to improve the quality of life of patients. The more high radiation dose are used for effective tumor growth control, the more radiation-related complications like as hearing deficit, the impairment of other cranial nerve function are increased. Since 1990's the mean radiation dose for tumor margin was more than 18 Gy, but there were high complication rate in spite of good tumor growth control. After the year of 2000, under the influence of advanced neuro-imaging techniques and radiosurgical planning system which enable clinicians to do more precise planning, marginal dose for VS has been decreased to 12-13 Gy and the radiation-related complications has been reduced. But because there may be a unexpected radiation induced complications as time goes by after the latency period, optimal radiation dose for VS should be established on the basis of more long term follow-up observation.

Studies on the High Temperature Induced Stress on the Biochemical Profile and Fecundity of Daba and Laria Ecoraces of Tropical Tasar Silkworm Antheraea mylitta Drury (Lepidoptera: Saturniidae)

  • Tropical tasar silkworm Antheraea mylitta Drury experiences extreme temperature stress conditions during its life cycle particularly during diapauses and first crop. The present study witnessed the impact of high temperature on some biochemical profiles and egg production (fecundity) of semi-domesticated Daba and Shorea robusta (Sal) based wild ecorace Laria during seed cocoon (pupa) preservation. Cocoons of Daba and Laria were treated with high temperature at $40^{\circ}C$ for 10 days in a BOD incubator. The protein profile and carbohydrate content in the hemolymph and fat body and total haemocyte count (THC) in the hemolymph of pupa were investigated. Further, the fecundity and fertility of egg was assessed. Significant increase in the protein concentration was observed in the hemolymph with reduction in the fat body (p<0.05). The difference in protein concentration was highly significant between the semidomesticated Daba and wild ecorace Laria (p<0.05). High pupal mortality (20%) and reduced fecundity (10-15%) in Daba was noticed compared to wild Laria. Also an increased THC (>28000) was recorded in Laria. The study infers the potentials of wild ecoraces in sustaining the extreme temperature conditions and need of adopting suitable package of practices for the preservation of diapause seed cocoons during extreme summer conditions. There is possibility to introgression thermal stress resistant traits in the semi-domesticated races of tasar silkworm by resorting to conventional breeding plans with wild races and keeping the thermal stress induced response as markers.

Pilot system을 이용한 감자의 알콜발효중 성분 변화 (Changes in the Components during Alcohol Fermentation of Potatoes Using Pilot System)

  • To proceed mass production and improve its quality, we fermented potatoes using pilot system and investigated the changes in components during fermentation. After liquefaction and saccharification of potatoes by Nuruk(group I), crude enzyme(group II) and glucoamylase(group III), sugar contents in all groups were 18brix equally. However sugar contents in group(II) and group(III) after 24hrs decreased deeply to 7.2 and 8.8 % respectively, after 24hrs. Alcohol content in group(I) increased slowly and was the highest such as 6.8% after 48hrs. Fusel oils in all groups were n-propanol, isobutanol and isoamylalcohol. The major fusel oil in all groups was isoamylalcohol. At the early stage of fermentation, free sugars were glucose, maltose and lactose. Glucose decreased deeply during fermentation and at latter of the fermentation, galactose was detected in all groups. The contents of total free amino acid were 516.57~569.98 mg% in group(I), 193.97~292.11 mg% in group (II) and 186.31~270.53 mg% in group(III). The contents of aspartic acid, serine, glutamic acid, alanine, arginine, and histidine were high in all groups.

저장온도에 따른 '쓰가루' 사과의 호흡 및 품질특성 변화 (Changes in Respiratory and Quality Characteristics of 'Tsugaru' Apple by Storage Temperature)

  • The research was conducted to measure the effect of temperature on respiratory and quality characteristics of 'Tsugaru' apple(Malus domestica Borkh) during storage at 4$^{\circ}$C, 10$^{\circ}$C and 20$^{\circ}$C. Respiration rate of the apple just after harvest was 7.57 mL CO$_2$/kg/h at 20$^{\circ}$C. It was decreased rapidly and was 2.86 mL CO$_2$/kg/h after storage for 45 days at 20$^{\circ}$C. The apples showed the highest ethylene production rate at 10$^{\circ}$C and 20$^{\circ}$C after 14 days, and at 4$^{\circ}$C after 28 days during storage. Weight of the apple was reduced by 2.9% after 120 days at 4$^{\circ}$C and by 6.1% after 45 days at 20$^{\circ}$C. Color difference of peel and pH increased with the temperatures but titratable acidity and flesh firmness showed the reverse trends during storage. According to the subjective quality test, it could be suggested that the shelf-life of the apple is 86 days at 4$^{\circ}$C, 65 days at 10$^{\circ}$C and 37 days at 20$^{\circ}$C, respectively.

배추의 저온유통용 골판지포장상자 개발 (Development of the Corrugated fiberboard Box for Cold-chain Distribution of Chinese Cabbage)

  • 예냉$.$저온유통에 적합한 배추포장상자를 개발하기 위하여 3∼4포기를 담을 수 있는 골판지 포장상자를 T11형 KS 표준 팔레트에 적재율이 96%이상이 되도록 제작하여 상자 구조별로 수직압축강도, 배추의 냉각속도 및 냉각균일도, 배추의 품질변화를 측정하고 비교하였으며 그 결과를 요약하면 다음과 같다. 가. 배추의 저온유통에 사용하는 골판지포장상자는 개공율을 5.4%로하고, 포장상자 형태를 접음식 형태로 하는 것이 예냉속도 및 예냉균일도에서 유리하였고, 저장후 수직압축강도에서는 브리스 상자에 비하여 낮게 나타났지만, 안전압축강도 이상의 수직압축강도를 나타내어 실제 사용에 문제가 없었다. 나. 예냉후 저장중의 배추 줄기의 절단력을 측정한 결과 기존상자에 비해 개선된 포장상자에서 유통기한을 2∼3일정도 연장시킬 수 있는 것으로 나타났다.

예냉처리 및 포장 방법 개선에 따른 양송이의 선도 연장 효과 (Effect of Prolongation by Precooling Treatment and Improved Packing of Mushrooms (Agaricus bisporus))

  • 양송이의 유통과정중 선도연장을 위해 수확 후 예냉처리와 포장개선 시도가 행해졌다. 기존의 관행적인 골판지상자에 담아 무예냉 상온유통하는 방법과 예냉처리후 스티로폼 상자에 담아 냉장수송한 양송이에 대하여 $10^{\circ}C$$25^{\circ}C$에 보관하면서 품질 변화를 조사하였다. 예냉처리에 의해 양송이의 호흡속도는 2-3배 억제되었으며 감소율은 $25^{\circ}C$에서는 2일후 무예냉구가 15%, 예냉구가 7% 정도, $10^{\circ}C$에서는 3일후에 무예냉구가 7%, 예냉구는 0.3% 정도로 나타났다. 표면색을 나타내는 L-value는 예냉처리구가 높게 나타났으며 저장중 생육정도를 나타내는 갓과 줄기의 길이도 예냉처리에 의해 더 작게 나타났다. 전반적으로 예냉처리와 보냉 유통을 통해 여름철 양송이의 선도는 기존 1-2일 정도에서 3-4일 정도로 연장되었다.

근적외 분광법을 응용한 휴대용 벌꿀 품질 평가 장치 개발 (Development of a Portable Quality Evaluation System for Bee-honeys by Using Near Infrared Spectroscopy)

  • This study was conducted to develop a portable quality evaluation system of bee-honey by near infrared spectroscopic technique. Two kinds of bee-honeys from acacia and polyflower sources were tested in this study. The system consists of power supply, tungsten-halogen lamp, detector, and optical fiber probe. Performance of the system was analyzed by comparing the prediction accuracy of the laboratory spectrophotometer. Total of 346 spectra was divided into a calibration set and a prediction set. The PLS (Partial Least Squares) models were developed to predict the quality parameters of bee-honeys. Reflectance spectra, moisture contents, ash, invert sugar, sucrose, F/G ratio, HMF(hydroxy methyl furfural), and $C^{12}/C^{13}$ ratio of honeys were measured. The PLS models of the laboratory spectrophotometer showed good relationships between predicted and measured quality parameters of honeys in the wavelength range of 1.100~2.200 nm. The PLS analysis of the portable quality evaluation system showed good relationships between predicted and measured quality parameters of honeys in the wavelength range of 1.100~1.300 nm and 1.400~1.700 nm. The results showed the feasibility of the portable quality evaluation system to determine the quality parameters of bee-honey in the field during harvesting.

유기농 및 저농약 재배한 참다래의 저장 중 품질변화 (Changes in the Fruit Quality of Organic and Low-level Agrochemical-grown Kiwifruit during Storage)

  • 관행, 저농약 및 유기농 재배한 참다래 과실의 저온저장 중 품질변화를 조사하였다. 경도는 저장초기 심하게 감소한 다음 연화되는 경향을 보였는데 연화 속도는 유기농 과실에서 다소 빨랐다. 당도는 저장 중 증가하는 경향을 보인 반면, 산함량은 감소하였고, pH는 변화가 없었다. 환원당 함량은 전분함량이 감소하면서 저장초기 크게 증가하는 경향을 보였다. 유기농 과실은 저장 중 부패율이 관행재배나 저 농약재배에 비해 유의하게 높았고, 병을 유기하는 병원균은 잿빛곰팡이, 연부병, 꼭지썩음병 순으로 높았다. 호흡량과 함께 에틸렌발생량이 증가하면서 과실이 연화되었는데, 호흡량과 에틸렌함량은 유기농 과실에서 다소 높은 경향을 나타내었다. 유기농 과실은 연화와 함께 부패과 증가로 관행재배나 저 농약재배에 비해 저장력이 감소되었다.

포도박 농축액을 이용한 분말 제조 (Production of Powder Using Concentrated By-products of Grape Processing)

  • 포도주와 포도즙 등 가공품의 생산 후 대량 발생되는 포도 부산물(포도박)의 이용도를 제고하기 위하여 포도박을 조건을 달리하여 추출 농축하여 분말을 제조하여 그 특성을 조사하였다. 물은 용매로 추출한 농축 분말의 경우 가용성 고형물의 함량이나 기능성 성분의 함량이 낮게 나타났으며 카테킨의 경우에서는 에탄올 추출물에서는 검출되지 않은 EGCg를 함유하였다. 에탄올 추출 농축 분말은 가용성 고형물의 함량이 높으며 기능성 성분인 카테킨과 레스베라트롤의 함량이 높게 나타났다. 카테킨은 포도박을 분쇄 후 추출한 경우가 분쇄하지 않았을 때보다 약 1.5배의 함량 증가를 나타냈다. 그러나 레스베라트롤은 분쇄하지 않고 추출한 시료가 더 높게 나타났다. 건본 연구의 결과 포도박 추출 분말은 에탄올로 추출하고 추출 전 분쇄한 것은 카테킨의 함량이 높고 분쇄하지 않은 것은 레스베라트롤의 함량이 높았으며 5% 덱스트린을 첨가하여 건조 한 것이 기능성 성분을 가장 많이 포함하였다.
