Monocyte chemoattractant protein-1(MCP-1) is released from the macrophages and endothelial cells, regulated luteotropic and luteolytic actions of macrophages and induced luteolysis. However, the mechanisms of MCP-1 on the development and maintenance of pregnant corpora lutea are thoroughly unknown. In this experiment, TUNEL stain, ED1, ED2, and MCP-1 immunohistochemistry on the corpora lutea of pregnant rats were carried out to reveal the role of macrophages in the developing corpora lutea. In the postpartum corpora lutea, the number of macrophages was increased significantly, and the intensity of ED1 and ED2 immunoreactivity in macrophages were increased moderately, and MCP-1 immunoreactivity was also increased. In conclusion, macrophages in the postpartum corpora lutea may exert phagocytic action mainly, and the macrophages in the pregnant corpora lutea maintain the structure and function of lutein cells.
Limb bud (LB) and central nerve system (CNS) cells were prepared from 12.5 day old pregnant female Crj:CD (SD) rats and treated with olaquindox and vitamin A. Cytotoxicity and inhibition on differentiation were measured in each cell. Three doses of olaquindox (4, 21 and 100 mgkg), and 0.2 and 75 mg/kg of vitamin A were administered to pregnant rat for 11 days from $6^{th}$ to $16^{th}$ of pregnancy. $IC_{50}$ values of olaquindox for proliferation and differentiation in CNS cell were 22.74 and $28.32\;{\mu}g/ml$ and 79.34 and $23.29\;{\mu}g/ml$ in LB cell and those values of vitamin A were 8.13 and $5.94\;{\mu}g/ml$ in CNS cell and 0.81 and $0.05\;{\mu}g/ml$ in LB cell, respectively. Mean body weights of pregnant rats were decreased at high dose of olaquindox (110 mg/kg) but relative ovary weight, number of corpus lutea, and number of implantation were not changed. Resorption and dead fetus were increased at high dose of olaquindox, and relative ovary weight, the number of corpus lutea and implantation, and sex ratio of male to female were not significantly changed in all dose of olaquindox. Mean fetal and placenta weights were significantly (p < 0.01) decreased in rats of high group. Seven fetuses out of 103 showed external anomaly like bent tail, and 10 out of 114 fetuses showed visceral anomalies at high group. The ossification of sternebrae and metacarpals were significantly (p < 0.01) increased by low and middle dose of olaquindox but it was significantly (p < 0.01) prohibited by high dose of olaquindox. In rats treated with vitamin A, the resorption and dead fetus were increased by high dose. Mean fetal weights were significantly (p < 0.01) increased by low dose but significantly (p < 0.01) decreased by high dose. Thirty four fetuses out of 52 showed external anomaly; bent tail (1), cranioarchschisis (14), exencephaly (14), dome shaped head (22), anophthalmia (15), brcahynathia (10) and others (19). Forty five fetuses out of 52 showed soft tissue anomaly; cleft palate (42/52) and anophthalmia (22/52) by high dose of vitamin A. Sixty one fetuses out of 61 (85.2%) showed skull anomaly; defect of frontal, partial and occipital bone (21/61), defect of palatine bone (52/61) and others (50/61). In summary, we support that vitamin A is strong teratogen based on our micromass and in vivo data, and olaquindox has a weak teratogenic potential in LB cell but not in CNS cell. We provide the in vivo evidence that a high dose of olaquindox could have weak embryotoxic potential in rats.

The properties of cytosolic $Ca^{2+}$ level$([Ca^{2+}]_i)$ movement of high KCl, carbachol and oxytocin were examined with myometrium isolated from non-pregnant rat(estrus cycle). High concentration of KCl$({\leq}23.3mM)$ induced rhythmic increases in $[Ca^{2+}]_i$ and muscle contraction. However, sustained $[Ca^{2+}]_i$ and contracion were obtained at higher KCl concentration $({\geq}30.3mM)$ The rhythmic and sustained contraction closely associated with changes in $[Ca^{2+}]_i$ induced by high KCl. Carbachol $(3{\sim}30{\mu}M$ generated rhythmic increases with tonic component in $[Ca^{2+}]_i$ and muscle contraction. Myometrial contraction stimulated by carbachol was also closely correlated with change in $[Ca^{2+}]_i$. And the $[Ca^{2+}]_i/contraction$ relationships were similar when muscle strips were stimulated by high KCl and carbachol. Maximal concentration of carbachol $(10{\mu}M)$ and oxytocin(100 nM) increased $[Ca^{2+}]_i$ and contraction which were slightly greater than that of high KCl in non-pregnant myometrium, respectively. However, the $[Ca^{2+}]_i$ and contraction were strongly inhibited by verapamil $(10{\mu}M)$, a 1-type $Ca^{2+}$ channel blocker, as in the case of high KCl. Additionally, although carbachol further increased $[Ca^{2+}]_i$ and contraction induced by high KCl, these changes also strongly inhibited by application of verapamil. These results suggest that uterotonic agents, carbachol and oxytocin, induced contraction by increase in $[Ca^{2+}]_i$ through $Ca^{2+}$ influx than by a regulation of $Ca^{2+}-sensitization$ in non-pregnant myometrium.
Purpose: To elucidate the effects of the irradiation and calcium-deficient diet on expression of interleukin (IL)-1 during tooth formation of rat molar. Materials and Methods: The pregnant three-week-old Spague-Dawley rats were used for the study. The control group was non-irradiation/normal diet group, and the experimental groups were irradiation/normal diet group and irradiation/calcium-deficient diet group. The abdomen of the rats on the 9th day of pregnancy were irradiated with single dose of 350 cGy, The rat pups were sacrificed on the 14th day after delivery and the maxillae tooth germs were taken. The specimen were prepared to make sections for light microscopy, and some of tissue sections were stained immunohistochemically with anti-IL-l antibody. Results: In the irradiation/normal diet group, dental follicle showed fewer blood vessels, mononuclear cells, and fusions of mononuclear cells than in non-irradiation/normal diet group. Alveolar bone showed a few osteoblasts and osteoclasts. Periodontal ligament showed collagen fibers and fibroblasts with irregularity. Weak immunoreactivity for IL-l was shown in dental follicle, alveolar bone, and periodontal ligament. In the irradiation/calcium-deficient diet group, dental follicle showed sparse cellularity. Alveolar bone showed diminished number of osteoblasts. Periodontal ligament showed irregular collagen fibers and atrophy of cementoblasts and fibroblasts. No immunoreactivity for IL-1 was shown in dental follicle, alveolar bone, and periodontal ligament. Conclusion: Irradiation and calcium-deficient diet seems to cause disturbance of the expression of interleukin-l during tooth formation of rat molar.
The absorption, distribution and excretion of $^{14}C$ labeled YH1885 {5,6-Dimethyl-2(4-fluorophenylamino)-4-(1-methyl-1,2,3,4-tetrahydroisoquinolin-2-yl)pyrimidine hydroc hloride), a new proton pumpinhibitor, were investigated in rats after a single administration of $^{14}C$-YH1885. 1. After intravenous administration of 5mg/kg, the blood level of radioactivity declined in a biphasic fashion with the mean terminal elimination half-life of 12.4hr. 2. After oral administration of 20mg/kg, the maximum blood level of radioactirity was reached at 4.0hr in female rats. The blood level of radioactivity-time profiles in male and female rats were similar, and the absorptionof $^{14}C$-YH1885 was not affected by food. 3. Appproximately 89% and 1% of radioactivity of the total dose were excreted in feces and urine, respectively. 4. Biliary excretion of radioactivity was 47.9% of the dose. Enterohepatic circulation of radioactivity was 49.6%. 5. Radioactivity was excreted maily into feces via bile. 6. The concentration of radioactivity in most tissues reached the peak level at 4.0hr after dosing, and then declined. Autoradiograms of male rats showed that the radioactivity levlels in the fat, harder's gland, liver and G-Itract were higher than those in the other tissues and the elimination of radioactivity from fat and liver was slow. 7. Autoradiograms of a pregnant rat showed that radioactivity was transferred to mammary gland, placenta and fetus. The radioactivity level in the mammary gland was higher than that in the blood.
The present study was performed to analyze the gene expressions of pituitary adenylate cyclase-activating polypeptide(PACAP) and its receptor in the rat uterus, a candidate for novel extrahypothalamic source and target. The PACAP cDNA fragments corresponding to the common exon region which is found in both the rat hypothalamus and testis were produced from all tissue samples including the rat uterus by reverse transcriptionpolymerase chain reaction (RT-PCR). No PCR product was amplified from the rat hypothalamic, pituitary, ovarian and uterine samples when the 5' primer corresponding to the testis-specific exon 1 region was used, while the predicted size of product was detected from the testis sample. RT-PCR using the uterine RNA and specific primers for the PACAP receptor yielded products with predicted sizes. Transcripts for the rat uterine PACAP receptor were identified as type I isoforms with hip-hop and hip- or hop-type inserts. After pregnant mare's serum gonadotropin (15 IU) treatment of immature rats (day 25), the level of PACAP mRNA was increased in 24 h and 48 h group, and was declined to the lowest in 72 h group. The present study shows the presence of transcripts for PACAP and its receptor isoform in the rat uterus. These finding ssuggest that the uterine PACAP ight act as a novel autocrine and/or paracrine factor via its specific receptors on the reglulation of rat uterine function and physiology during the reproductive cycle.
The purpose of this study was to investigate the effect of hydrocortisone in the fusion mechanism of the secondary palatine shelves in the rat. Pregnant rats were injected with 5.0 mg/100 gm body weight of hydrocortisone for 3 days between the 7th day and the 9th day of pregnancy. Between the 14th day md the 18 day of pregnancy, the fetuses were removed and decapitated to be immersed in $10\%$ formalin and Carney's solution. Preparations were stained with alizarin red S, hematoxylin-eosin and alcian blue respectively, and partly were treated for Periodic acid-Schiff reaction. The results were as followings: 1. The incomplete fusion of the palatal epithelium took place though the polarity of epithelial cells. 2. At the edge of shelves the differentiation of mesenchymal cells was observed, but the inter penetration of mesenchyme was not shown. 3. It was considered that the phenomenon of hypocalcification in matrix had relation to the decrease stainability to alizarin red S in shelves. 4. It might be concluded that the connective tissue under epithelium showed the decrease tendency to alcian blue and PAS reaction due to the inhibition of the synthesis of the mucopolysaccharide.

The author observed morphological change in palate development of rat embryo after irradiation of x-ray on the one side of the duplex uterus. The time-matings occured between 6 p.m. and 8 p.m. and all females with copulation plugs at 8 a.m. were isolated and properly marked for evidence of copulation. The lower left abdomen of mothers were exposed to x-radiation on the 7 1/2th, 9 1/2th, 11 1/2th day of gestation, respectively 150, 250, 350, 500rads. At 18 1/2th day of post-conception, the pregnant females were dissected and the contents of the two uteri examined. The translucent sample by Alizarin red S stain were prepared. The results were as follows; 1. The result that groups irradiated by 250rads and 350rads made marked difference in comparison with the control group suggests the x-ray to be a inducing factor of cleft palate. 2. At 11 1/2th day of gestation, incidence of cleft palate induced by x-irradiation was highest. 3. Mortality showed the highest frequency at 7 1/2th day of gestation and tended to decrease in according to increasing of age. 4. Morphology of cleft palate induced by x-irradiation showed similarity in comparison with those induced by other factors having reported ever.
Our previous study demonstrated that 2.3', 4.4'. 5- Pentachlorobiphenyl (PCB 118) showed an antiestrogenic activity in vitro and in vivo. In the present study. we examined the effect of PCB 118 on postnatal reproductive development in female rats. PCB 118 (0.001. 0.01 or 0.1 mg/kg/day) was administered to pregnant female SD rats from gestation day (GO) 6 to 18 via subcutaneous injection. and developmental parameters such as vaginal opening were determined. PCB 118 significantly delayed vaginal opening of female offsprings at dose of 0.1 ${\mu}g$/kg/day. whereas had no effects on body weights. In addition. in utero treatment of PCB 118 caused significant decreases in serum levels of E2, T3 and T4 in female oftsprings at certain doses on postnatal day (PND) 22. Our data of results indicate that in utero exposure to PCB 118 may postnatal reproductive development in female rat through its antiestrogenic activity.
This study was carried out to determine effectively the sex of rabbit embryos using H-Y antiserum. H-Y antiserum was obtained from inbred SD strain female rat which was immunized by injection of testis cell of inbred SD strain male rate into its spleen. The titer of antiserum was identified by sperm cytotoxicity test and culture of rabbit embryos with antiserum. The developed or undeveloped embryos were separated by exposure the embryos to the antiserum with H-Y antibody. Developed embryo were transferred to the recipients and sex of offspring were examined. 1. In the sperm cytotoxicity test, the rate of dead sperm showed no difference between two antisera from spleen and testis cell as antigens. It is confirmed that H-Y antibody in antiserum was absorbed by H-Y antigen in male rat spleen cells. 2. When rabbit morulae were exposed to antiserum and complement, the rate of embryos developed or arrested was 51 and 49% respectively and the rate was closely same as natural sex ratio of 50:50%. 3. When rabbit morulae were cultured for 12h in the medium containing antiserum produced by antigen of testis cell, the rate of embryos developed or arrested was 48 and 52% respectively and the rate was closely same as natural sex ratio of 50:50%. 4. Eighty rabbit embryos which were not affected by H-Y antiserum were transferred to four recipients. Two recipients were pregnant and born 13 pups among which 2 (14%) were male and 11 (86%) were female. In conclusion, existence of H-Y antibody in the serum from female rat immunized by injecting testis cell from newborn male rat to the spleen of the female rat was confirmed. When rabbitmorulae were exposed to H-Y antiserum and complement, about a half of embryos were developed to blastocysts. When the rabbit embryos not affected by H-Y antiserum were transferred, the rate of female offspring was 86%. Therefore, it was identified that most of embryos which were not affected by H-Y antiserum were female.
