Association between PCR-RFLP Polymorphisms of Five Gene Loci and Milk Traits in Chinese Holstein

  • The objective of this study was to assess the association of polymorphisms in ${\kappa}$-cn, ${\beta}$-lg, ${\beta}$-lg 5′ flanking region, CSN1S2, and IGFBP-3 genes with milk production traits and mastitis-related traits in Chinese Holstein. Traits analyzed were 305 day standard milk yield, protein percentage, fat percentage, the ratio of fat percentage and protein percentage, pre-somatic cell count, somatic cell count, and somatic cell score, respectively. CSN1S2 locus was uninformative because only one genotype BB was found in Chinese Holstein. Allele frequencies of A and B in IGFBP-3 gene were 0.5738 and 0.4262 in Chinese Holstein population, which was different from reported Qinchuan cattle population. The genotypes of animals at IGFBP-3 locus significantly affected 305 day standard milk yield, protein percentage, and somatic cell score. The ${\beta}$-lg genotypes had a significant effect on protein percentage and the ratio of fat percentage and protein percentage. Polymorphism in ${\beta}$-lg 5′ flanking region was associated with 305 day standard milk yield, protein percentage, fat percentage, pre-somatic cell count, and somatic cell count. No significant associations of the polymorphism in ${\kappa}$-cn gene were observed for any trait.

GPS TEC Fluctuations in the Low and High Latitudes During the 2015 St. Patrick's Day Storm

  • As a part of collaborative efforts to understand ionospheric irregularities, the Korea ionospheric scintillation sites (KISS) network has been built based on global positioning system (GPS) receivers with sampling rates higher than 1 Hz. We produce the rate of TEC index (ROTI) to represent GPS TEC fluctuations related to ionospheric irregularities. In the KISS network, two ground-based GPS sites at Kiruna (marker: KIRN; geographic: $67.9^{\circ}$ N, $21.4^{\circ}$ E; geomagnetic: $65.2^{\circ}$ N) and Chuuk (marker: CHUK; geographic: $7.5^{\circ}$ N, $151.9^{\circ}$ E; geomagnetic: $0.4^{\circ}$ N) were selected to evaluate the ROTI value for ionospheric irregularities during the occurrence of the 2015 St. Patrick's Day storm. The KIRN ROTI values in the aurora region appear to be generally much higher than the CHUK ROTI values in the EIA region. The CHUK ROTI values increased to ~0.5 TECU/min around UT=13:00 (LT=23:00) on March 16 in the quiet geomagnetic condition. On March 17, 2015, CHUK ROTI values more than 1.0 TECU/min were measured between UT=9:00 and 12:00 (LT=19:00 and 22:00) during the first main phase of the St. Patrick's Day storm. This may be due to ionospheric irregularities by increased pre-reversal enhancement (PRE) after sunset during the geomagnetic storm. Post-midnight, the CHUK ROTI showed two peaks of ~0.5 TECU/min and ~0.3 TECU/min near UT=15:00 (LT=01:00) and UT=18:00 (LT=04:00) at the second main phase. The KIRN site showed significant peaks of ROTI around geomagnetic latitude=$63.3^{\circ}$ N and MLT=15:40 on the same day. These can be explained by enhanced ionospheric irregularities in the auroral oval at the maximum of AE index

Azasugar-Containing Phosphorothioate Oligonucleotide (AZPSON) DBM-2198 Inhibits Human Immunodeficiency Virus Type 1 (HIV-1) Replication by Blocking HIV-1 gp120 without Affecting the V3 Region

  • DBM-2198, a six-membered azasugar nucleotide (6-AZN)-containing phosphorothioate (P = S) oligonucleotide (AZPSON), was described in our previous publication [Lee et al. (2005)] with regard to its antiviral activity against a broad spectrum of HIV-1 variants. This report describes the mechanisms underlying the anti-HIV-1 properties of DBM-2198. The LTR-mediated reporter assay indicated that the anti-HIV-1 activity of DBM-2198 is attributed to an extracellular mode of action rather than intracellular sequence-specific antisense activity. Nevertheless, the antiviral properties of DBM-2198 and other AZPSONs were highly restricted to HIV-1. Unlike other P = S oligonucleotides, DBM-2198 caused no host cell activation upon administration to cultures. HIV-1 that was pre-incubated with DBM-2198 did not show any infectivity towards host cells whereas host cells pre-incubated with DBM-2198 remained susceptible to HIV-1 infection, suggesting that DBM-2198 acts on the virus particle rather than cell surface molecules in the inhibition of HIV-1 infection. Competition assays for binding to HIV-1 envelope protein with anti-gp120 and anti-V3 antibodies revealed that DBM-2198 acts on the viral attachment site of HIV-1 gp120, but not on the V3 region. This report provides a better understanding of the antiviral mechanism of DBM-2198 and may contribute to the development of a potential therapeutic drug against a broad spectrum of HIV-1 variants.

Study on Evaluation of Plastic Deformation Zone at Crack Tip for the Multi-Passed Weld Region of the Pressure Vessel Steel Using Nondestructive Method (비파괴법에 의한 압력용기 강 다층용접부의 균열선단에서 소성변형 역성장거동 평가에 관한 연구)

  • The purpose of this study is to evaluate the behaviour of the plastic deformed zone at crack tip on the standard Charpy specimens which were taken from the multi-passed weld block of the pressure vessel steel. Notch was machined on the standard Charpy test specimens and pre-crack which was located around the fusion line was made under the repeat load. Four point bend and acoustic emission tests were carried out simultaneously. The size of plastic region at crack tip was calculated using stress intensity factor. Relationships between characteristics of acoustic emission and plastic zone size at crack tip were discussed through the cumulative AE energy. Regardless of the specimens, AE signals were absent within the elastic region almost and most of AE signals were produced at the plastic deformation region from yield point to the mid-point between yield and maximum load. More AE signals for the weldment were produced compared with the base-metal and PWHT specimen. Relations between plastic deformed zones at crack tip and cumulative AE energy for the weldment and PWHT specimen were different quietly from the base-metal. Besides, number of AE counts for the weldment was the larger than those of the base-metal and PWHT specimen.

  • In order to investigate effects of grid resolution on large-eddy simulation of flow over a heavy vehicle, large-eddy simulations over the vehicle with coarse grid and fine grid are conducted. In addition, comparison of drag coefficients with the experimental data obtained by a wind tunnel experiment is conducted. Both of the drag coefficients of coarse grid and fine grid large-eddy simulation show good agreement with the experimental data. Flow fields obtained by the coarse and the fine grid large-eddy simulation are compared in the vehicle frontal-face region, the vehicle rear wheel region, and the vehicle base region. Coarse grid large-eddy simulation shows good agreement with the fine grid large-eddy simulation in the vehicle front face region and the vehicle rear wheel region, since the flow over the present vehicle is dominated by flow separation which is geometrically pre-determined, not by the skin friction which is known to be sensitive to grid resolution.

  • Estimation of carbon in the natural forest regions is a pre-requisite for carbon management. In the light of increasing carbon dioxide concentration in the atmosphere, the amount of carbon present in the plants and soils are very much needed to estimate the sequestered carbons stock of any region. Carbon stock estimation studies are limited in India, especially in the natural forest regions of Eastern ghats of Tamil Nadu. Remote sensing, Geographical Information System (GIS) and global positioning system (GPS) were used along with extensive field and laboratory works to estimate the carbon stock in the living biomass and soil. About five forest types were identified and mapped using satellite data. The total biomass carbon including above and below ground were 2.74 Tg and the total soil organic carbon was 3.48 Tg. This study has yielded significant information about the carbon stock in a natural forest region and it could be used for future comparative studies.

  • This paper presents a pre-processing method to facilitate the container's BIC-code recognition. We propose a network that can find ROI(Region Of Interests) containing a BIC-code region and estimate a homography matrix for warping. Taking the structure of STN(Spatial Transformer Networks), the proposed network consists of next 3 steps, ROI detection, homography matrix estimation, and warping using the homography estimated in the previous step. It contributes to improving the accuracy of BIC-code recognition by estimating ROI and matrix using the proposed network and correcting perspective distortion of ROI using the estimated matrix. For performance evaluation, five evaluators evaluated the output image as a perfect score of 5 and received an average of 4.25 points, and when visually checked, 224 out of 312 photos are accurately and perfectly corrected, containing ROI.

Investigating the future changes of extreme precipitation indices in Asian regions dominated by south Asian summer monsoon

  • The impact of global warming on the south Asian summer monsoon is of critical importance for the large population of this region. This study aims to investigate the future changes of the precipitation extremes during pre-monsoon and monsoon, across this region in a more organized regional structure. The study area is divided into six major divisions based on the Köppen-Geiger's climate structure and 10 sub-divisions considering the geographical locations. The future changes of extreme precipitation indices are analyzed for each zone separately using five indices from ETCCDI (Expert Team on Climate Change Detection and Indices); R10mm, Rx1day, Rx5day, R95pTOT and PRCPTOT. 10 global climate model (GCM) outputs from the latest CMIP6 under four combinations of SSP-RCP scenarios (SSP1-2.6, SSP2-4.5, SSP3-7.0, and SSP5-8.5) are used. The GCMs are bias corrected using nonparametric quantile transformation based on the smoothing spline method. The future period is divided into near future (2031-2065) and far future (2066-2100) and then the changes are compared based on the historical period (1980-2014). The analysis is carried out separately for pre-monsoon (March, April, May) and monsoon (June, July, August, September). The methodology used to compare the changes is probability distribution functions (PDF). Kernel density estimation is used to plot the PDFs. For this study we did not use a multi-model ensemble output and the changes in each extreme precipitation index are analyzed GCM wise. From the results it can be observed that the performance of the GCMs vary depending on the sub-zone as well as on the precipitation index. Final conclusions are made by removing the poor performing GCMs and by analyzing the overall changes in the PDFs of the remaining GCMs.

  • L1251B is an excellent example of a small group of pre- and protostellar objects in low-mass star-forming region. Previous interferometer data with a high angular resolution resolved the outflows associated with L1251B into a few components. To understand the physical conditions of the protostellar outflows in L1251B, we mapped this region spectroscopically with Spitzer/IRS and obtained spectral line data from Herschel/PACS. Spitzer/IRS provides the S(0)-S(7) pure rotational lines of H2 as well as fine-structure emissions produced in shocks such as S, [Si II], and [Fe II] and it is a powerful tool for studying shocked interstellar gas. In addition, [O I] lines observed with Herschel/PACS are described well by J-type shock models expected in the outflows from protostars. We will present an analysis of the L1251B protostellar outflow in the H2 pure rotational lines and fine-structure emissions.

Local Structure Refinement of the $BaFe_{1-x}Sn_xO_{3-y}$ System with Fe K-Edge X-Ray Absorption (XANES/EXAFS) Spectroscopy

  • Local structure refinement of the BaFe1-xSnxO3-y system (x=0.00-0.50) has been carried out with Fe K-edge x-ray absorpion spectroscopic studies. It is found out that the Fe ions are placed in two different symmetric sites such as tetrahedral and octahedral sites in the compounds by comparison with Fe K-edge x-ray absorption near edge structure (XANES) spectrum of the γ-Fe2O3 compound as a reference. Small absorption peaks of dipole-forbiden transitions appear at a pre-edge region of 7111 eV due to the existence of Fe ions in the tetrahedral and octahedral sites. The peak intensity decreases with the substitution amount of Sn ion. Three different absorption peaks of 1s→4p dipole-allowed transition appear on the energy region between 7123 and 7131 eV. The peaks correspond to 1s→4p main transition of Fe ions in tetrahedral and octahedral sites and 1s→4p transition followed by the shakedown process of ligand to metal charge transfer. The bond distances between Fe ions in the tetrahedral site and nearest neighboring oxygen atom (Fe-4O), and those in octahedral site (Fe-6O) are determined with the extended x-ray absorption fine structure (EXAFS) analysis. Two different interatomic distances increase with the substitution amount of Sn ion and also the bond lengths of Fe-4O are shorter than those of Fe-6O in all compounds.
