A New large-scale Pre-purification for Peroxidase from Plant Cell Cultures (식물세포 배양으로부터 Peroxidase 대량 정제를 위한 전처리 공정 개발)

  • A novel pre-purification method was developed for producing peroxidase to guarantee high purity and yield from plant cell cultures in large-scale process. This method was a simple and efficient procedure for the isolation and pre-purification of peroxidase from the biomass consisting of active clay treatment followed by cationic exchange chromatography. The use of active clay in the pre-purification process allows for rapid and efficient separation of peroxidase from interfering compounds and dramatically increases yield and purity of crude peroxidase for purification steps compared to alternative processes. This pre-purification process serves to minimize the buffer usage size and complexity of the HPLC operations for peroxidase purification. This process is readily scalable to a pilot plant and eventually to a production environment where mass production of material are expected to be produced.

  • This paper describes a fuzzy controlled pre-chlorination technique for purifying the pulluted raw water in water purification lants. For the purpose of obtaining the high quality water, the appropriate pre-chlorine dosage rate has to be continuously adjusted according to a change in quality of a intake raw water, weather, solar nergy mount, temperature and etc. Therefore, the method of expressing an expert's empirical knowledge cumulated from his past carrier by fuzzy reasoning and the fuzzy controller design technique is necessary.In this paper fuzzy membership functions and rules accordingto emprircal knowledge and experimental field data were obtained, And also fuzzy cintriller design using four feedforward components for the determination of pre-chlorine dosage rate and four feedback ones for the compensation of its dosage rate with residual chlorine and its change rate, was executed.

A Novel Purification Process for Homoharringtonine from Celphalotaxus koreana

  • An effective purification method was developed for producing Homoharringtonine (HHT), to guarantee high purity and yield from Cephalotaxus koreana. This process was a simple and efficient procedure, for the isolation and purification of HHT form the biomass of Cephalotaxus koreana, consisting of extract, adsorbent treatment, precipitation and followed by a chromatography. The extraction, adsorbent treatment and precipitation in pre-purification process allows for rapid and efficient separation of HHT from many compound and dramatically increases the yield and purity of crude HHT for HPLC purification steps compared to alternative processes. This purification processes serves to minimize solvent usage, size, and complexity of the operations for HHT purification.

  • The recovery and purification of a paclitaxel from plant cell cultures is essential to commercial process. This review describes a large-scale recovery and purification method for producing paclitaxel, to guarantee high purity and yield from plant cell cultures. Also, the process of separation and purification is optimized in conjunction with a extraction step, pre-purification, purification, and polishing (drying) as an integrated process to meet final product quality requirements such as purity, residual solvents, product morphologies, impurities, bacterial endotoxin, etc. This information is very useful for production and quality control of pharmaceuticals in commercialization step.

  • The optimal operation of the water purification plant can be carried out only when the required flow rate is supplied in a timely manner using the minimum electrical energy by accurately predicting the pattern and amount of tap water used in the consumer. In order to ensure the stability of tap water production and supply, a system that can be pre-verified before applying AI-based composite sensors to the water purification plant was established to derive complementary matters through the pre-verification model for each composite sensor and improve the quality and operation stability of the composite sensor data.

  • PreS domain of Hepatitis B virus (HBV) surface antigen is a good candidate for an effective vaccine as it activates both B and T cells besides binding to hepatocytes. This report deals with overexpression and purification of adr subtype of surface antigen that is more prevalent in Pakistan. PreS region, comprising 119 aa preS1 region plus a 55 aa preS2 region plus 11 aa from the N-terminal S region, was inserted in pET21a+ vector, cloned in E. coli $DH5\alpha$ cells and expressed in E. coli BL21 codon+ cells. The conditions for over expression were optimized using different concentrations of IPTG (0.01-5 mM), and incubating the cells at different temperatures (23-$41^{\circ}C$) for different durations (0-6 h). The cells were grown under the given optimized conditions (0.5 mM IPTG concentration at $37^{\circ}C$ for 4 h), lysed by sonication and the protein was purified by ion exchange chromatography. On the average, 24.5 mg of recombinant protein was purified per liter of culture. The purified protein was later lyophilized and stored at $-80^{\circ}C$.

Identification and Characterization of Homoharringtonine from Cephalotaxus koreana

  • A novel purification method was developed for producing homoharringtonine from Cephalotaxus koreana, to guarantee high purity and yield. Our simple, efficient procedure for isolating and purifying homoharringtonine from C. koreanabiomass consisted of solvent extraction, synthetic adsorbent treatment, low-pressure chromatography, followed by high performance liquid chromatography (HPLC). The use of active clay treatment and silica gel low-pressure chromatography in the pre-purification process allowed for the rapid, efficient separation of homoharringtonine from interfering compounds and dramatically increased the yield and purity of crude homoharringtonine for high-performance liquid chromatography (HPLC) purification steps compared with alternative processes. Homoharringtonine could be obtained simply with high yield and purity from biomass using this purification method, while minimizing solvent use and the scale and complexity of HPLC operations for homoharringtonine purification. Purified homoharringtonine was identified and characterized.

Application of membrane distillation process for tap water purification

  • Membrane distillation process was used for purification of pre-treated natural water (tap water). The rejection of inorganic and organic compounds in this process was investigated. The obtained rejection of inorganic solutes was closed to 100%, but the volatile organic compounds (VOCs) diffused through the membrane together with water vapour. The content of trihalomethanes (THMs) in the obtained distillate was two-three fold higher than that in the feed, therefore, the rejection of the total organic compounds present in the tap water was reduced to a level of 98%. The intensive membranes scaling was observed during the water separation. The morphology and composition of the fouling layer was studied using scanning electron microscopy coupled with energy dispersion spectrometry. The influence of thermal water pre-treatment performed in a heat exchanger followed by filtration on the MD process effectiveness was evaluated. This procedure caused that significantly smaller amounts of $CaCO_3$ crystallites were deposited on the membrane surface, and a high permeate flux was maintained over a period of 160 h.
