2,3-Dimethoxy-2′-hydroxychalcone ameliorates TNF-α-induced ICAM-1 expression and subsequent monocyte adhesiveness via NF-kappaB inhibition and HO-1 induction in HaCaT cells

  • Up-regulation of adhesion molecules plays an important role in the infiltration of leukocytes into the skin during the development of various inflammatory skin diseases, such as atopic dermatitis. In this study, we investigated the modulatory effects of 2,3-dimethoxy-2′-hydroxychalcone (DMHC) on tumor necrosis factor (TNF)-α-induced intercellular adhesion molecule-1 (ICAM-1) expression and monocyte adhesiveness, as well as the molecular mechanisms underlying its action in the HaCaT human keratinocyte cell line. Pre-treating HaCaT cells with DMHC significantly suppressed TNF-α-induced ICAM-1 expression and subsequent monocyte adhesiveness. DMHC inhibited TNF-α-induced activation of NF-ᴋB. In addition, DMHC induced HO-1 expression as well as NRF2 activation. Furthermore, HO-1 knockdown using siRNA reversed the inhibitory effect of DMHC on TNF-α-induced ICAM-1 expression and adhesion of monocytes to keratinocytes. These results suggest that DMHC may inhibit TNF-α-induced ICAM-1 expression and adhesion of monocytes to keratinocytes by suppressing the signaling cascades leading to NF-ᴋB activation and inducing HO-1 expression in keratinocytes. Effects of Progesterone and 17β-Estradiol under Presence or Absence of FBS on Plasminogen Activators Activity in Porcine Uterine Epithelial Cells

  • The present study was conducted to investigate the regulatory mechanism of plasminogen activators (PAs) activation by $17{\beta}$-estradiol ($E_2$) and progesterone ($P_4$) in porcine uterine epithelial cells (pUECs). pUECs were collected from porcine uterine horn and cultured at 80% confluence. Then, 0.1% (v/v) DMSO, 20 ng/mL $E_2$, and $P_4$ with or without fetal bovine serum (FBS) treated to cultured cells for 24 hours. The supernatants were used for measurement of PAs activity and expression of urokinase-type PA (uPA), tissue-type PA (tPA), uPA specific receptor (uPAR), and type-1 PA inhibitor (PAI-1) mRNA were analyzed by real-time PCR. The expression of PAs-related genes was not affect by steroid hormones in both of serum treatment groups. However, PAs activity was increased by treatment of $E_2$ compared to 0.1% DMSO treatment in serum-free group (p<0.05). Then, $E_2$ and $P_4$ were diluted with 0.002% (v/v) DMSO for reduction of its effect and treated to cultured cells without FBS. Only tPA mRNA was significantly increased by $E_2$ treatment (p<0.05). PAs activity was enhanced in $E_2$ treated group compared to control groups (p<0.05). These results indicate that serum-free condition is more proper to evaluate effect of steroid hormones and activation of PAs in pUECs was mainly regulated by estrogen. These regulation of PAs activation may be associated with uterine remodeling during pre-ovulatory phase in pigs, however, further studies are needed to investigate precise regulatory mechanism.

  • OWC-MIMO시스템에서 GSM변조기법은 송 수신기의 위치에 따라 상호 간섭이 큰 환경에서 성능 최적화를 위한 활성화 패턴집합을 선택하는 과정이 중요하다. 하지만, 높은 연산 복잡도로 인해 최적의 활성화 패턴집합을 선택하는 과정에서 높은 비용을 지불한다. 본 논문에서는 전송 활성화 패턴집합 선택을 위한 후보 활성화 패턴집합 생성 시 기본 활성화 패턴 집합을 미리 결정하는 방식을 통해 후보 활성화 패턴 집합의 수를 감소시키는 방법을 제안한다. 모의실험 결과, 제안된 방법은 높은 간섭 환경에서 일반적인 GSM변조기법과 비교할 때 동일한 BER성능과 약 90%정도 감소된 후보 활성화 패턴집합의 수를 가져 복잡도가 크게 감소함을 확인하였다.

TNF계 CD137L 및 RANKL의 파골세포와 T 세포에 대한 활성조절 (STUDY ON THE REGULATION OF OSTEOCLAST AND T CELL ACTIVATION VIA CELL MEMBRANE PROTEINS OF TNF FAMILY, CD137 LIGAND AND RANK LIGAND)

  • 본 연구는 TNFR family인 CD137 및 RANK, 파골세포의 CD137L와 T 세포의 RANKL 간의 역신호에 의한 이들 세포 의 역할을 알아보고자 하였다. 이에 RANKL 및 CD137L 자극으로 유도되는 역신호 전달에 의한 T 세포 활성과 파골세포분 화에 미치는 영향을 규명하고자 웅성 생쥐의 골수세포와 T 세포를 공동배양하여 다음과 같은 결과를 얻었다. 1. 생쥐 단핵세포주 및 골수유도 단핵전구세포에서 CD137L이 발현되며, CD137L 단클론 항체로 자극을 주었을 경우 파 골세포 표지단백질인 TRAP 양성 파골세포의 형성이 억제되었다. 2. 활성화된 $CD4^+$$CD8^+$ T 세포에서 RANKL을 발현하였으며 RANKL의 유사 수용체인 OPG 재조합 단백질을 처리 하여 $CD4^+$$CD8^+$ T 세포의 세포증식이 억제되었다. 이 연구의 결과는 CD137 자극에 의한 T 세포활성 및 RANK 자극에 의한 파골세포분화 및 활성이 각각 수용체에 결합하 는 라이겐드의 역신호에 의해 억제되었는데, 이는 파골세포와 T 세포의 과도한 활성을 제어하는 생체의 항상성조절에 관여하 는 기전으로 생각된다.

Optimization of Post-Activation Systems to Improve the Embryonic Development in Porcine Parthenogenesis and Somatic Cell Nuclear Transfer

  • This study was conducted to establish the optimal chemical post-activation conditions in porcine embryonic development after parthenogenesis (PA) and somatic cell nuclear transfer (SCNT) using 4 different chemical compositions (cytochalasin B (CB), cyclohexamide (CHX), demecolcine (DC), 6-dimethylaminopurine (DMAP). Porcine embryos were produced by PA and SCNT and then, cultured for post-activation with CB ($7.5{\mu}g/mL$), CB ($7.5{\mu}g/mL$) + CHX ($10{\mu}g/mL$), CB ($7.5{\mu}g/mL$) +DC ($0.4{\mu}g/mL$), and CB ($7.5{\mu}g/mL$) + DMAP (2 mM). In PA embryonic development, cleavage rates have been significantly higher in CB group (94.7%) and CB+DMAP group (94.1%) than that of CB+CHX and CB+DC group (88.1 and 84.3%, respectively). There have been no significant differences in blastocyst formation rates among the four groups. In cell number of blastocyst was shown in CB group (42.3%) significantly higher than CB+CHX and CB+DC group (40.6 and 40.6%, respectively). In SCNT embryonic development, CB+DMAP group (89.7%) significant differences were found on embryo cleavage rates when compared with other three groups. Blastocyst formation rates in CB+DMAP group (26.9%) were significantly higher when compared with CB, CB+CHX, and CB+DC groups (25.5, 20.2, and 22.1%, respectively). In blastocyst cell number, CB+DMAP group (41.4%) was found higher significant difference compared with other three groups. Additionally, we have investigated survivin expression in early development stages of porcine SCNT embryos for more confirmation. Our results establish that CB group and CB+DMAP group for 4 h during post-activation improves pre-implantation improvement of PA and SCNT embryos.

Neuronal Nitric Oxide-mediated Cytotoxicity in Trophoblast Cells Induced by Increase of Intracellular Calcium

  • Cell death of trophoblast, particularly by abnormal release of physiological nitric oxide (NO) has been known to be a causative factor of pre-eclampsia. In the present study, effects of intracellular calcium increase enhancing the activity of NO synthases (neuronal NO synthase, nNOS in this trophoblast cells) on the cell death were examined in a human placental full-term cell line (HT-1). Furthermore, we analyzed the possible mechanisms underlying the augmentation of $Ca^{++}$-mediated NOS activity mediated by protein kinases like PKC, PKA, or CaM-KII. In experiments for cell toxicity, a calcium ionophore (ionomycin $10{\mu}M$) enhanced cell death confirmed by MTT assay, and increased significantly nNOS activity determined with a hemoglobin oxidation assay. This cell death was partially protected by pre-treatment of 7-nitroindazole (7-NI, $10{\mu}M$ and $100{\mu}M$), a nNOS-specific inhibitor. Additionally, $Ca^{++}$-ionophore -induced increase of nNOS activity also was partially normalized by pre-treatment of specific inhibitors of protein kinases, PKC, PKA or CaM-KII. Therefore, we suggest that an increase of calcium influx, leading to the activation of nNOS activity, which in turn may result in the death of trophoblast cells by involvement of signaling mechanisms of protein kinases.

미량 귀금속 첨가에 의한 Ni-계열 메탄 수증기 개질 촉매의 반응 활성 향상에 관한 연구 (A Study of Reactivity Improvement of Ni-based Methane Steam Reforming Catalysts by Small Addition of Noble Metals)

  • The promotion effects of noble metals upon the activity and reducibility in steam methane reforming over $Ni/MgAl_2O_4$ catalysts were investigated. While $Ni/MgAl_2O_4$ catalysts require the pre-reduction by $H_2$, the noble metal-added catalysts show high catalytic activities without pre-treatment. According to $CH_4$-TPR, the addition of noble metal makes the $Ni/MgAl_2O_4$ catalyst easily reducible. The reduction degree of NiO in the noble metal-added catalysts after using at $650^{\circ}C$ without pre-reduction was $15{\sim}20%$, and was lower than that in the $H_2$-reduced $Ni/MgAl_2O_4$ catalyst(reduction degree=27%). The enhancement of the catalytic activity over noble metal-added catalysts results from easier reducibility by addition of noble metal and the synergy effect between noble metal and Ni.

PNF의 CI기법이 편마비 환자의 근활성도, 근피로도 및 균형에 미치는 효과 (The Effects of CI Technique in PNF on the Muscle Activity, Fatigue, and Balance in Hemiplegic Patients)

  • Purpose: The purpose of this study was to investigate the effects of a combination of isotonic (CI) technique on the balance, muscle fatigue and muscle activities of the quadriceps muscle in the paretic leg of hemiplegic patients. Method: All the subjects received a combined isotonic technique of proprioceptive neuromuscular facilitation (PNF) for about 30 minutes 3 times per week for 6 weeks. The root mean square (RMS) and mid frequency (MF) were recorded by surface electromyography (EMG)(EMG/EP system, OXFORD Medelec, UK) during maximal voluntary isometric contractions (MVIC). The balance ability was measured by a balance system (BIODEX, USA). Result: The RMS of the quadriceps muscle at post-treatment was significantly increased compared to that at pretreatment (p<0.05). Yet there was no significant difference among the three muscles. There were no significant difference in the MF of the vastus lateralis and rectus femoris between pre- and post-treatment, but the MF of the vastus medialis was significantly decreased (p<0.05). There was a significant difference in the vastus medialis and lateralis between pre- and post-treatment (p<0.05). Moreover, the overall stability index at post-treatment was significantly decreased compared to that at pre-treatment (p<0.05). But there was no significant difference between the anterior/posterior and medial/lateral stability indexes. Conclusion: The results of this study indicated that a CI technique of PNF had an effect to improve the balance and muscle activity in hemiplegic patients.

표면매립된 철계-형상기억합금 스트립으로 휨 보강된 RC보의 장기 휨거동 (Long-term Flexural Behavior of RC Beams Strengthened in Flexure with NSM Fe-SMA Strips)

  • 표면매립공법으로 매립한 철계-형상기억합금으로 보강한 보의 휨 거동을 장기 하중 재하실험을 통해 평가하였다. 철계-형상기억합금 길이대비 2%와 4%의 사전변형 및 형상기억효과 활성화에 의한 프리스트레스 하중 도입을 실험변수로 설정하였다. 1 tonf의 콘크리트 추를 보 중앙에 거치한 후 6개월간의 보 중앙부의 장기 처짐을 측정하였다. 실험결과, 철계-형상기억합금으로 보강한 보의 휨 강성이 증대되었으며, 사전변형이 증가할수록 보강재의 강성감소로 인한 처짐이 증가하는 것으로 나타났다. 프리스트레스 하중 도입에 따른 처짐을 비교했을 때, 프리스트레스 하중을 도입하지 않은 실험체에 비해, 프리스트레스 하중을 도입한 실험체는 약 30%의 처짐 감소 효과를 보이는 것으로 나타났다.

Calcium Ions are Involved in Modulation of Melittin-induced Nociception in Rat: I. Effect of Voltage-gated Calcium Channel Antagonist

  • Melittin-induced nociceptive responses are mediated by selective activation of capsaicin-sensitive primary afferent fibers and are modulated by excitatory amino acid receptor, cyclooxygenase, protein kinase C and serotonin receptor. The present study was undertaken to investigate the peripheral and spinal actions of voltage-gated calcium channel antagonists on melittin-induced nociceptive responses. Changes in mechanical threshold and number of flinchings were measured after intraplantar (i.pl.) injection of melittin $(30\;{\mu}g/paw)$ into mid-plantar area of hindpaw. L-type calcium channel antagonists, verapamil [intrathecal (i.t.), 6 or $12\;{\mu}g$; i.pl.,100 & $200\;{\mu}g$; i.p., 10 or 30 mg], N-type calcium channel blocker, ${\omega}-conotoxin$ GVIA (i.t., 0.1 or $0.5\;{\mu}g$; i.pl., $5\;{\mu}g$) and P-type calcium channel antagonist, ${\omega}-agatoxin$ IVA (i.t., $0.5\;{\mu}g$; i.pl., $5\;{\mu}g$) were administered 20 min before or 60 min after i.pl. injection of melittin. Intraplantar pre-treatment and i.t. pre- or post-treatment of verapamil and ${\omega}-conotoxin$ GVIA dose-dependently attenuated the reduction of mechanical threshold, and melittin-induced flinchings were inhibited by i.pl. or i.t. pre-treatment of both antagonists. P-type calcium channel blocker, ${\omega}-agatoxin$ IVA, had significant inhibitory action on flinching behaviors, but had a limited effect on melittin-induced decrease in mechanical threshold. These experimental findings suggest that verapamil and ${\omega}-conotoxin$ GVIA can inhibit the development and maintenance of melittin-induced nociceptive responses.
