Effect of Substrates and Lyoprotectant on the Survival Ratio of Lyophilized Bacillus sp. SH1RP8 (동결건조 보호제와 기질이 동결건조된 Bacillus sp. SH1RP8의 생존율에 미치는 영향)

  • In order to develop an eco-friendly biofertilizer, a plant growth promoting rhizobacterium (PGPR), Bacillus sp., SH1RP8 was investigated. SH1RP8 was lyophilized via freeze-drying along with other protective agents that protect cells from lysis. The freezedried powder of Bacillus sp. SH1RP8, containing 5% skim milk (w/v), exhibited the highest survival rate of 30.6% among all the protective agents (skim milk, glucose, and peptone). The lyoprotective effect of the skim milk, mixture including 5% skim milk, and substrates on the survival of the test strain was examined. Control group was added only skim milk and test groups were added skim milk and other substrates. As a result, the group supplemented with both glycerol and 5% skim milk showed the protective effect much higher by 214.29% than the control group. Freeze-dried Bacillus sp. SH1RP8 could be a good candidate as a potential biofertilizer due to its effective PGPR activity.

  • Recently environmental pollution is serious and therefore, This study aims at reviewing individual mixing ratio and engineering characteristics of concrete due to mixture and mixing using fine powder and fly ash of blast furnace slag having effect on aspects of environmental preservation and resources recycling and performance increase of the concrete, and verifying possibility of application in the field. Test results are as follows: 1)As mixing quantity of the admixture has increased, performance of the slump has been improved, 2)As mixing quantity of the admixture has increased, there is a trend of delayed ending time, 3)As mixing quantity of admixture has increased, it has showed lower strength at short time age, however, as the age has elapsed and mixing quantity has increased, strength improvement has increased and the admixture has effect on the long term age. In this study, the characteristics and critical value of concrete contained blast furnace slag and fly ash are defined, and will be examined about the field applications.

  • The synthesis of SiC used for the parts of the gas turbine and the heat exchanger, was carried out. In this study, wire cutting slurry of silicon wafer and the graphite rod of spent zinc-carbon battery were applied to the starting materials for the synthesis. The powders of Si or Si+SiC were obtained from the waste material by filtration, gravity separation and magnetic separation. Graphite powder was produced by dismantling, grinding and gravity separation from spent zinc-carbon battery. The synthesis of SiC could be completed from the mixture powders of Si and C or Si+SiC and C at the condition of equivalent ratio of Si and C, atmosphere of Ar or vacuum, temperature of above 1$600^{\circ}C$ and 2 hours reactions. The purity of synthesized Si-C was above 99%.

  • A new process, gelcasting in aqueous medium, to fabricate complex-shaped ceramic core has investigated. The ceramic slurry, mixture of fused silica powder and additives such as zircon and cordierite, was electrosterically stabilizes. The slip was prepared by ball milling of polydispered ceramic suspension with monomer, dimer and dispersant. The rheological behavior of slip was evaluated by viscosity measurement. It was found that the high solid loading of polydispersed ceramic slip, which has low viscosity of 50vol%, is possible to obtained. The viscosity of the slip was significantly dependent upon the amount of polymer dispersant and the formulation of monomer and dimer. The green bodies were fabricated through casting and gelation at room temperature followed by drying at $25^{\circ}C$ for 48hrs under relative humidity of 80~85%. Crack-free green body was successfully fabricated through the above process.

  • The formation of high $T_c$ phase is very sluggish and c parameter of unit cell of high $T_c$ phase is about $37{\AA}$. High $T_c$ oxide superconductor with a $T_c$ above 100 K has been successfully prepared by solid state reaction method in BiSrCaCuO system by Pb adding. The microstructure related to the formation of the high $T_c$ phase has been investigated. As compared with YBCO compound, the formation reaction of the high $T_c$ requires long time heat treatment. It is due to the transformation from the low $T_c$ phase to high $T_c$ phase. The sintering just below the melting point of the calcined powder mixture is effective on the formation of the high $T_c$ phase in Pb-added BiSrCaCuO system. The growth of the high $T_c$ superconducting phase has a thin plate shape, which is characterized by the c parameter of $37{\AA}$. The formation of the high $T_c$ phases is delayed by the excessive addition of Pb. The lattice parameter(c) of the unit cell (both the low and high $T_c$ phases) is increased with increase of Pb.

Effect of Different Contents of Nuruk Extract on Fermentation Characteristics of Kwahaju (a Korean Traditional Alcoholic Beverage) (수국 첨가량이 과하주의 발효 특성에 미치는 영향)

  • Fermentation characteristics of kwahaju (a typical Korean traditional alcoholic beverage) base were investigated during fermentation with different contents of nuruk (Korean-style bran koji) extract. The nuruk extract which was prepared by incubating the mixture of nuruk powder and water at $25^{\circ}C$ overnight and by filtering it was used to be 0.6%, 2.7%, 5% and 10% (v/v). Total and reducing sugar contents as well as acidity of the kwahaju base with 0.6% nuruk extract were higher than those with 2.7%, 5% and 10% at the fermentation end. Final pH values of all the base samples were ranged from 3.3 to 4.1. Alcohol concentrations of the base samples with 2.7%, 5% and 10% nuruk extract were higher than those with 0.6%. Microbial growth rate was great and inner temperature was high in the sample with high content of nuruk extract, but fermentation period was short. Total sugar consumption and alcohol production increased as the content of nuruk extract increased, but total acid production decreased. The base sample with 10% nuruk extract showed the most excellent fermentation efficiency. Fusel oil content of the base sample with 2.7% nuruk extract was the highest (457.3 ppm), and those wity 5% and 10% nuruk extract were 438.9 ppm and 442.6 ppm, respectively. The sample with 0.6% nuruk extract had the lowest content (409.5 ppm). Sensory evaluation of both the kwahaju base and kwahaju mix with 25% and 40% alcohol by adding soju (Korean distilled liquor) showed that the base with 2.7% nuruk extract had the highest score, and that the kwahaju mix with 25% alcohol had higher score than that with 40%. The sensory results on overall desirability were consistent to those on color and alcohol concentration, and it turned out that the two factors were important to make kwahaju.

  • With a view of improving the quality of sardine sausage, the processing conditions of sardine sausage used raw sardine as materials and the effects of adding soybean protein and smoke flavor on the quality of product were investigated. Optimal amounts of additives in processing sardine sausage were 1.5% of salt, 1.5% of sugar, 0.2% of monosodium glutamate, 0.2% of white pepper, 0.2% of garlic powder, 0.2% of nutmeg, 0.4% of beef extract, 0.05% of food color solution(10% mixture solution of Red 40 and Yellow 5) and 0.1~0.2% of smoke flavor(Smok-EZ, Alpha Foods Co., Ltd.) based on washed sardine meat. The results showed that the benificial effects of adding corn starch(5%), ${\alpha}$-starch(2%), soybean protein(3%) to the washed sardine meat were exhibited in the improvement of texture and acceptability.

  • A feasible way to fabricate in-situ Al/TiC composites was investigated. An elemental mixture of $Al-TiO_2-C$ pellet was directly added into an Al melt at $800-920^{\circ}C$ to form TiC by self-combustion reaction. The addition of CuO initiates the self-combustion reaction to form TiC in $1-2{\mu}m$ at the melt temperature above $850^{\circ}C$. Besides the CuO addition, a diluent element of excess Al plays a significant role in the TiC formation by forming a precursor phase, $Al_3Ti$. Processing parameters such as CuO content, the amount of excess Al and the melt temperature, have affected the combustion reaction and formation of TiC, and their influences on the microstructures of in-situ Al/TiC composites are examined.

Effect of the Crucible Cover on the Distillation of Cadmium

  • The distillation of liquid cathode is necessary to separate cadmium from the actinide elements in the pyroprocessing since the actinide deposits are dissolved or precipitated in a liquid cathode. It is very important to avoid a splattering of cadmium during evaporation due to the high vapor pressure. Several methods have been proposed to lower the splattering of cadmium during distillation. One of the important methods is an installation of crucible cover on the distillation crucible. A multi-layer porous round cover was proposed to avoid a cadmium splattering in our previous study. In this study, the effect of crucible cover on the cadmium distillation was examined to develop a splatter shield. Various surrogates were used for the actinides in the cadmium. The surrogates such as bismuth, zirconia, and tungsten don't evaporate at the operational temperature of the Cd distiller due to their low vapor pressures. The distillation experiments were carried out in a crucible equipped with cover and in a crucible without cover. About 40 grams of Cd was distilled at a reduced pressure for two hours at various temperatures. The mixture of the cadmium and the surrogate was heated at $470{\sim}620^{\circ}C$. Most of the bismuth remained in the crucible equipped with cover after distillation under $580^{\circ}C$ for two hours, whereas small amount of bismuth decreased in the crucible without cover above $580^{\circ}C$. The liquid bismuth escaped with liquid cadmium drop from the crucible without cover. It seems that the crucible cover played a role to prevent the splash of the liquid cadmium drop. The effect of the cover was not clear for the tungsten or zirconia surrogate since the surrogates remained as a solid powder at the experimental temperature. From the results of this work, it can be concluded that the crucible cover can be used to minimize the deposit loss by prevention of escape of liquid drop from the crucible during distillation of liquid cathode.

Distillation of Cd- ZrO2 and Cd- Bi in Crucible With Splatter Shield

  • The liquid cathode processing is necessary to separate cadmium from the actinide elements in the pyroprocessing since the actinide deposits are dissolved or precipitated in a liquid cathode. Distillation process was employed for the cathode processing owing to the compactness. It is very important to avoid a splattering of cadmium during evaporation due to the high vapor pressure. Several methods have been proposed to lower the splattering of cadmium during distillation. A multi-layer porous round cover was proposed to avoid a cadmium splattering in our previous study. In this study, distillation behavior of $Cd-ZrO_2$ and Cd - Bi systems were investigated to examine a multi-layer porous round cover for the development of the cadmium splatter shield of distillation crucible. It was designed that the cadmium vapor can be released through the holes of the shield, whereas liquid drops can be collected in the multiple hemisphere. The cover was made with three stainless steel round plates with a diameter of 33.50 mm. The distance between the hemispheres and the diameter of the holes are 10 and 1 mm, respectively. Bismuth or zirconium oxide powder was used as a surrogate for the actinide elements. About 40 grams of Cd was distilled at a reduced pressure for two hours at various temperatures. The mixture of the cadmium and the surrogate was distilled at 470, 570 and $620^{\circ}C$ in the crucible with the cover. Most of the bismuth or zirconia remained in the crucible after distillation at 470 and $570^{\circ}C$ for two hours. It was considered that the crucible cover hindered the splattering of the liquid cadmium from the distillation crucible. A considerable amount of the surrogate material reduced after distillation at $620^{\circ}C$ due to the splattering of the liquid cadmium. The low temperature is favorable to avoid a liquid cadmium splattering during distillation. However, the optimum temperature for the cadmium distillation should be decided further, since the evaporation rate decreases with a decreasing temperature.
