유전자변형 생물체의 위해성평가 (Risk Assessment of Genetically Modified Organisms)

  • New breeding method by genetic engineering is expected as a key technology to solve food shortage due to the growing world population in the year 2000s. Many genetically modified organisms (GMOs) were already developed and the commercial cultivation had started. The first GMO, Flavr Savr tomato, which rotted at a much slower pace than ordinary ones, was developed in US in 1994. Since then, over than 70 different agricultural products including corn, cotton, soybean, papaya, potato, and squash made with genetically modified plants are reportedly on sale worldwide. Supporters favor the GMOs because they have greater yields, longer shelf lives and stronger resistance to disease and insects. On the other hand, opponents say that the supporters ignore a potential danger that they may damage the environment as well as human beings. To assure the safe development and use of GMOs as food and other biotech products, the possible risks on biological environment and human health should be throughly examined and regulated by developer and government. Because the biosafety problem is a global, environmental, and trade issue, a new international treaty is under development. The Cartagena Protocol on Biosafety was adopted at the 1 st Extraordinary Conference of Parties of the Convention on Biological Diversity which was held at Mont-real, Canada, Jan. 29th, 2000. The adoption of the Protocol is seen as a breakthrough in that it is based on the" Precautionary Principle" despite scientific uncertainties surrounding potential risks that GMOs may inflict on human health and the environment and that it has laid the ground for introduction of specific steps to handle international trading of GMOs. In this paper, the authors would like to introduce the current status and perspective of environmental and human risk assessment of GMOs.t of GMOs.

Botrytis cinerea에 의한 매실나무 잿빛곰팡이병 발생 (Occurrence of Gray Mold on Prunus mume Caused by Botrytis cinerea in Korea)

  • 2005년부터 2007년까지 경남농업기술원 과수포장에서 Botryitis cinerea에 의한 매실나무 잿빛곰팡이병이 발생하였다. 병징은 과실이 담갈색을 띄며 약간 함몰되면서 시간이 경과됨에 따라 대형으로 나타난다. 심하게 감염될 경우 부패되어지고 병반부위에 회색의 곰팡이가 많이 형성된다. 분생포자는 무색, 단포자이며 난형 또는 타원형으로 크기는 $6{\sim}17{\times}4{\sim}10{\mu}m$였고, 분생포자경 위에 분생포자가 많이 형성되었다. 분생포자경은 갈색으로 격막이 있고, 폭은 $15{\sim}30{\mu}m$였다. 균사생육과 균핵형성 적온은 $20^{\circ}C$였다. 병반에서 분리한 병원균의 균학적 특징을 조사한 결과 Botrytis cinerea Persoon: Fries에 의한 매실나무 잿빛곰팡이병으로 동정되었다.

Alternanthera mosaic virus - an alternative 'model' potexvirus of broad relevance

  • Alternanthera mosaic virus (AltMV) is a member of the genus Potexvirus which has been known for less than twenty years, and has been detected in Australasia, Europe, North and South America, and Asia. The natural host range to date includes species in at least twenty-four taxonomically diverse plant families, with species in at least four other families known to be infected experimentally. AltMV has been shown to differ from Potato virus X (PVX), the type member of the genus Potexvirus, in a number of ways, including the subcellular localization of the Triple Gene Block 3 (TGB3) protein and apparent absence of interactions between TGB3 and TGB2. Differences between AltMV variants have allowed identification of viral determinants of pathogenicity, and identification of residues involved in interactions with host proteins. Infectious clones of AltMV differing significantly in symptom severity and efficiency of RNA silencing suppression have been produced, suitable either for high level protein expression (with efficient RNA silencing suppression) or for Virus-Induced Gene Silencing (VIGS; with weaker RNA silencing suppression), demonstrating a range of utility not available with most other plant viral vectors. The difference in silencing suppression efficiency was shown to be due to a single amino acid residue substitution in TGB1, and to differences in subcellular localization of TGB1 to the nucleus and nucleolus. The current state of knowledge of AltMV biology, including host range, strain differentiation, host interactions, and utility as a plant viral vector for both protein expression and VIGS are summarized.

Environmental contamination and geochemical behaviour of heavy metals around the abandoned Songcheon Au-Ag mine, Korea

  • The objective of this study is to investigate the contamination levels and dispersion patterns of arsenic and heavy metals and to estimate the bioaccessible fraction of the metals in soil and plant samples in the vicinity of the abandoned Songcheon Au-Ag mine. Tailings, soils, plants (Chinese cabbage, red pepper, soybean, radish, sesame leaves, green onion, lettuce, potato leaves, angelica and groundsel) and waters were collected around the mine site. After appropriate preparation, all samples were analyzed for As, Cd, Cu, Pb and Zn by ICP-AES and ICP-MS. Elevated levels of As and heavy metals were found in tailings. Mean concentrations of As in agricultural soils were higher than the permissible level. Especially, maximum level of As in farmland soil was 513 mg/kg. The highest concentrations of As and Zn were found in Chinese cabbage (6.7 mg/kg and 359 mg/kg, respectively). Concentrations of As, Cd, and Zn in most stream waters which are used for drinking water around this mine area were higher than the permissible levels regulated in Korea. Maximum levels of As, Cd and Zn in stream waters were 0.78 mg/L, 0.19 mg/L and 5.4 mg/L, respectively. These results indicate that mine tailings can be the main contamination sources of As and heavy metals in the soil-water system in the mine area. The average of estimated bioaccessible fraction of As in farmland soils were $3.7\%$ (in simulated stomach) and $10.8\%$ (in simulated small intestine). The highest value of bioaccessible fraction of metal in farmland soils was $46.5\%$ for Cd.

Botrytis cinerea에 의한 스타티스 잿빛곰팡이병 (Gray Mold of Statice (Limonium shinuatum) Caused by Botrytis cinerea in Korea)

  • 2003년 4월 경남 함양군 서상면 고냉지 비닐하우스에서 Botrytis cinerea에 의한 스타티스 잿빛곰팡이병이 발생하였다. 병징은 꽃, 잎, 줄기에 발생하여 병든 부분이 수침상으로 물러지고 썩으면서 병반부위에 회색의 곰팡이가 많이 생기고, 심할 경우 감염된 부위는 결국 말라죽는다. 분생포자는 무색, 단포자이며 난형 또는 타원형으로 크기는 8∼20${\times}$6∼14 $\mu\textrm{m}$였고, 분생자경 위에 분생포자가 아주 많이 형성되었다. 분생자경은 갈색으로 격막이 있고, 폭은 14∼34 $\mu\textrm{m}$였다. 균사생육과 균핵형성 적온은 2$0^{\circ}C$였다. 병반에서 분리한 병원균은 Botrytis cinerea Persoon: Fries에 의한 스타티스 잿빛곰팡이병으로 동정되었다.

Weeding Efficacy of Sulfonylurea Resistance Weed, Monochoria (Monochoria vaginalis) with Brown Leaf Blight Caused by BWC01-54

  • A summer annual weed of monochoria (Monochoria vaginalis) grows in the edges of rice paddies, ditches, and moist upland throughout Korea. It is very difficult to control with herbicide because of its sulfonylurea resistance. It is very competitive with fast growing pattern, that can cause reducing yields of rice. Brown leaf blight of monochoria (Monochoria vaginalis) occurred naturally in rice paddy, is first reported in Korea. The fungal isolate BWC01-54 was successfully isolated from the diseased leaves of monochoria. The fungus BWC 01-54 was grown well at $25-28^{\circ}C$, conidia of the greysh black brown mycelia were abundant produced on PDA at 15 days. The fungus was grown well in potato dextrose broth at $28^{\circ}C$ and fully grown within 10 days in 250 ml of flask. In host and pathogenicity test, conidia suspension of BWC01-54 was the most effective to control of monochoria compare to others isolates. Typical symptoms having pin point brown lesions were formed on stem and leaf and which severely affected the whole plants ware blighted within two weeks, respectively. Under paddies field condition, conidial suspension of the fungus BWC01-54 gave around 90% control. Therefore, we conclude that the fungus may have a potential as a biological control agent against sulfonylurea resistance weed in rice paddy.

Coniella granati에 의한 석류 열매썩음병 (Fruit Rot of Pomegranate (Punica granatum) Caused by Coniella granati in Korea)

  • 2002년 7월 경남 통영시 광도면 석류재배 농가포장에서 열매가 부패되는 이상증상이 발생하였다. 분리된 병원균은 감자한천배지에서 균총의 색깔은 흰색이며 물결모양을 이루면서 왕성하게 자랐고, 배지상에서 분생포자각을 잘 형성하였다. 분생포자는 단세포이며 모양은 방추형이고, 색깔은 갈색 또는 올리브색이다. 크기는 10.3~17.4$\times$2.8~4.0 $\mu\textrm{m}$ 였다. 분생포자 형성 세포는 짧은 원통형이며 무색으로 크기는 12.4~1.4$\times$2.8~3.6 $\mu\textrm{m}$ 였다. 불생포자각의 모양은 구형이며 갈색으로, 각벽은 2~3세포이고, 크기는 124.6~228.4 $\mu\textrm{m}$ 였다. 균사생육 적온은 $25^{\circ}C$였다. 분리된 병원균을 건전한 석류에 유상, 무상접종한 결과 접종 3일 후 감염을 일으켰으며, 병원성이 강하였다. 이상과 같이 이 병원균을 Coniella granati에 의한 석류열매 썩음병으로 명명할 것을 제안한다.

고추탄저병(Colletotrichum gloeosporioides) 방제를 위한 엽권 길항미생물의 탐색 (Screening for Phyllospheral Antagonistic Microorganisms for Control of Red-pepper Anthracnose (Collectotrichum gloeosporioides))

  • 고추탄저병(Colletotrichum gloeosporioides)에 대한 생물학적방제 능력을 가지는 엽권미생물을 선발하기 위하여 38과 87종의 약용식물로부터 243균주의 세균과 51균주의 사상균 그리고 30균주의 효모를 분리하여 이들에 대하여 항진균활성검정을 in vitro 및 in vivo에서 실시한 결과는 다음과 같다. 분리된 엽권미생물 중에서 PDA 배지 대치배양에서 10 mm 이상의 생육저지대를 형성하는 4개의 엽권미생물이 선발되었다. 선발된 4개의 세균균주 KB6, KB12, KB13 및 KB14의 PDB 배양여액 중에서 KB12 균주가 균사생장 억제율이 가장 높았다. 액체배양여액 처리에서 푸른 고추보다 붉은 고추에서 방제효과가 컸다. Bacterial cell 처리에서는 푸른 고추나 붉은 고추에서도 높은 방제효과가 있었다. 선발된 4개의 세균균주들은 모두 Bacillus subtilis로 동정되었다.

Population Dynamics and Fitness Comparison of Sensitive and Resistant Phenotypes of Botrytis cinerea to Benzimidazole, Dicarboximide, and N-phenylar-bamate Fungicides

  • A total of 2109 isolates of Botrytis cinerea were collected from infected plants fo strawberry, tomato, and cucumber in Korea from 1994 to 1996. Based on in virtotests for mycelial growth on potato-dextrose agar containing fungicides, the esolates were classified into six phenotypic groups : SSR, SRR, RSS, RRS, RSR, and RRR, representing sensitivity (S) or resistance (R) to carbendazim, procymidone, and diethofencarb. In that order the isolation frequencies of the SSR, SRR, RSS, RRS, RSR, and RRR phenotypes were 28.7, 1.1, 28.8, 39.4, 1.0, and 0.9%, respectively. Three isolates from each SSR, SRR, RSS, RRS, and RSR and an isolate of RRR phenotype were selected and evaluated for their fitness-related characteristics such as pathogenic aggressiveness, mycelial growth rate, sporulation, and sclerotial formation. Competitive abilities of the SSR, SRR, RSS, RRS, and RSR phenotypes were also compared by inculating mixtures of conidial suspensions of two phenotypes to cucumber plant, and then determining re-isolation frequencies from lesions. In general, significant differences in fitness-related characteristics, except pathogenic aggressiveness, were found not only between but also within phenotype groups. In the competitiveness tests, carbendazim-sinsitive phenotypes (SSR and SRR) were found to be more competitive than the resistant ones (RSS and RSR), whereas, the procymidone-resistant phenotypes (SRR and RRS) appeared to be more competitive than the sensitive ones (SSR, RSS, and RSR). There was no consistent dominance in competitiveness between the diethofencarb-resistant and sensitive phenotypes. The RSR phenotype was the least competitive among the five phenotypes.

Pathogenicity and Host Range of a Potential Mycoherbicide, Isolate BWC98-105, Causing White Root Rot on Trifoliorum repens

  • White root rot of wild white clover (Trifoliorum repens) caused by isolate BWC98-105 has been first reported in Korea. Typical symptoms on root include water-soaked and dark-brown rot, resulting in complete blight of the whole plant. The fungus grew well at $20-28^{\circ}C$ and produced abundant sclerotia at 10-15 days after full mycelial growth on potato dextrose agar. Sclerotia were brown to dark-brown in color and 1-3 mm in length. When white clover plants were inoculated with mycelial suspension ($10^5$ cfu/ml) of isolate BWC98-105, the plant shoots were killed within 4-6 days and the roots were completely blighted. Sclerotia were also formed on the surface of the root covered with whitish mycelia within 10-15 days in the field. All nine isolates developed high incidences of white root rot disease on white clover seedlings, of which the symptoms were similar to those observed in the fields. Hence, their pathogenicity was confirmed on white clover. The infection rate of the fungal isolates varied from 78.5% to 95.2%, among which BWC98-105 was the most virulent isolate. The weeding efficacy of the fungus was maintained until the following year, leading to a significant reduction of reshooting. The fungus was specifically parasitic to white clover, but not to four lawn species including zoysiagrass (Zoysia japonica) under greenhouse test. The fungus also had no response to some Gramineae species including rice, but caused little damage to five species of Leguminosae.
