Reducing Characteristics of Potassium Tri-sec-butylborohydride

  • The approximate rates and stoichiometry of the reaction of excess potassium tri-sec-butylborohydride ($K_s-Bu_3BH$) with selected organic compounds containing representative functional groups were determined under the standard conditions (0$^{\circ}C$, THF) in order to define the characteristics of the reagent for selective reductions. Primary alcohols evolve hydrogen in 1 h, but secondary and tertiary alcohols and amines are inert to this reagent. On the other hand, phenols and thiols evolve hydrogen rapidly. Aldehydes and ketones are reduced rapidly and quantitatively to the corresponding alcohols. Reduction of norcamphor gives 99.3% endo- and 0.7% exo-isomer of norboneols. The reagent rapidly reduces cinnamaldehyde to the cinamyl alcohol stage and shows no further uptake of hydride. p-Benzoquinone takes up one hydride rapidly with 0.32 equiv hydrogen evolution and anthraquinone is cleanly reduced to the 9,10-dihydoxyanthracene stage. Carboxylic acids liberate hydrogen rapidly and quantitatively, however further reduction does not occur. Anhydrides utilize 2 equiv of hydride and acyl chlorides are reduced to the corresponding alcohols rapidly. Lactones are reduced to the diol stage rapidly, whereas esters are reduced moderately (3-6 h). Terminal epoxides are rapidly reduced to the more substituted alcohols, but internal epoxides are reduced slowly. Primary and tertiary amides are inert to this reagent and nitriles are reduced very slowly. 1-Nitropropane evolves hydrogen rapidly without reduction and nitrobenzene is reduced to the azoxybenzene stage, whereas azobenzene and azoxybenzene are inert. Cyclohexanone oxime evolves hydrogen without reduction. Phenyl isocyanate utilizes 1 equiv of hydride to proceed to formanilide stage. Pyridine and quinoline are reduced slowly, however pyridine N-oxide takes up 1.5 equiv of hydride in 1 hr. Disulfides are rapidly reduced to the thiol stage, whereas sulfide, sulfoxide, sulfonic acid and sulfone are practically inert to this reagent. Primary alkyl bromide and iodide are reduced rapidly, but primary alkyl chloride, cyclohexyl bromide and cyclohexyl tosylate are reduced slowly.

  • The approximate rates, stoichiometries and products of the reaction of potassium triethylborohydride $(KEt_3BH)$ with selected organic compounds containing representative functional groups under the standard condition $(0^{\circ}C,$ THF) were examined in order to explore the reducing characteristics of this reagent as a selective reducing agent. Primary alcohols, phenols and thiols evolve hydrogen rapidly whereas secondary and tertiary alcohols evolve very slowly. n-Hexylamine is inert to this reagent. Aldehydes and ketones are reduced rapidly and quantitatively to the corresponding alcohols. Reduction of noncamphor gives 3% exo- and 97% endo-norboneol. Anthraquinone is cleanly reduced to 9,10-dihydro-9,10-dihydroxyanthracene stage. Carboxylic acids liberate hydrogen rapidly and quantitatively but further reduction does not occur. Anhydrides utilize 2 equiv of hydride to give an equimolar mixture of acid and alcohol. Acid chlorides, esters and lactones are rapidly and quantitatively reduced to the corresponding alcohols. Epoxides are reduced at moderate rates with Markovnikov ring opening to give the more substituted alcohols. Primary amides liberate 1 equiv of hydrogen rapidly. Further reduction of caproamide is slow whereas benzamide is not reduced. Tertiary amides are reduced slowly. Benzonitrile utilizes 2 equiv of hydride in 3 h to go to the amine stage whereas capronitrile takes only 1 equiv. The reaction of nitro compounds undergo rapidly whereas azobenzene and azoxybenzene are reduced slowly. Cyclohexanone oxime rapidly evolves hydrogen without reduction. Phenyl isocyanate utilizes 1 equiv of hydride to proceed to formanilide stage. Pyridine N-oxide and pyridine is reduced rapidly. Disulfides are rapidly reduced to the thiol stage whereas sulfoxide, sulfonic acid are practically inert to this reagent. Sulfones and cyclohexyl tosylate are slowly reduced. Octyl bromide is reduced rapidly but octyl chloride and cyclohexyl bromide are reduced slowly.

Relationship between the values of blood parameters and physical status in Korean native calves with diarrhea

  • Dehydration, electrolyte disturbance, and acid-base imbalance are the most significant consequences of diarrhea in calves. We aimed to determine blood gas, hematological, electrolyte, and biochemical values and investigate the relationship between the physical status and blood parameters in Korean native calves (KNCs) with diarrhea. One hundred eighty KNCs with diarrhea (age < 75 days) were investigated. Blood samples were collected from the external jugular vein and analyzed using a portable clinical blood gas analyzer. The measured parameters were statistically compared according to the status of physical activity, dehydration, or prognosis. The mean values of parameters in the Calves with diarrhea showed metabolic acidosis, hyponatremia, and azotemia. The mean values of potassium, chloride, hematocrit, and hemoglobin were in the upper limit of their reference ranges. More than 75% of the calves had metabolic acidosis caused by bicarbonate loss, and 63.6% had high blood urea nitrogen (BUN) values. Moreover, BUN showed the highest correlation with the physical activity status and dehydration. pH, base excess of the extracellular fluid (BE), anion gap, potassium, hematocrit, bicarbonate, and hemoglobin were closely correlated with physical deterioration and dehydration (p < 0.001). BUN, pH, BE, and anion gap were closely correlated with physical deterioration and dehydration. These correlations between clinical symptoms and blood gas parameters can be clinically relevant in predicting the status of parameters according to clinical symptoms.

대체염 첨가가 유화형 소시지의 품질특성에 미치는 영향 (Effects of Replacement Sodium Chloride on the Quality Characteristices of Emulsion-type Sausages)

  • 본 연구는 유화형 소시지 제조 시 첨가되는 NaCl을 sodium citrate, potassium lactate 및 calcium ascorbate로 부분(40%) 대체하여 소시지를 제조한 다음 소시지의 품질특성에 미치는 영향을 비교하고자 실시하였다. 소시지의 pH는 sodium citrate 대체 소시지가 대조구와 다른 대체 소시지에 비해 높았다(P<0.05). 소시지의 명도(L*) 값은 sodium citrate 대체 소시지가 대조구와 다른 대체 소시지에 비해 높았다(P<0.05). 적색도(a*) 값은 potassium lactate 대체 소시지가 대조구와 다른 대체 소시지에 비해 높았지만, 황색도(b*) 값은 낮았다(P<0.05). 조직특성에서 단단함 정도를 나타내는 경도와 점착성은 sodium citrate 대체 소시지가 대조구와 다른 대체 소시지에 비해 높았다. NaCl 대체 소시지의 나트륨 함량은 대조구에 비해 낮았고, calcium ascorbate 대체 소시지가 가장 낮은 함량을 나타내었다(P<0.05). 소시지의 칼륨과 철 함량은 potassium lactate 대체 소시지가 대조구와 다른 대체 소시지에 비해 높았다(P<0.05). NaCl 대체 소시지의 칼슘 함량은 대조구에 비해 높았고, calcium ascorbate 대체 소시지가 가장 높은 함량을 나타내었다(P<0.05). 관능평가에서 짠맛은 sodium citrate 대체 소시지가 대조구와 다른 대체 소시지에 비해 높았다(P<0.05). 풍미는 calcium ascorbate 대체 소시지가 대조구에 비해 높았다(P<0.05). 이상의 결과에서, NaCl 대체 소시지는 pH, 색도 및 관능평가에서 대조구와 차이를 보였으며, 나트륨 함량을 낮추고 칼슘 함량이 증가한 건강지향적인 소시지를 생산할 수 있을 것으로 사료된다.

$K^+$ 통로개방제 Pinacidil이 종양이식 생쥐에서 Tl-201의 체내분포에 미치는 영향 (Effects of Pinacidil, a Potassium-Channel Opener, on Biodistribution of Thallium-201 in Tumor-Bearing Mice)

  • 목적: 생체 내에서 potassium과 유사한 역학을 보이는 thallium은 종양의 영상에 널리 사용된다. $K^+$ 통로개방제는 세포 내의 potassium을 외부로 배출되게 하는 기능이 있어 Tl-201을 이용한 종양영상에도 영향을 미칠 수 있을 것이라 생각된다. 본 연구는 강력한 $K^+$ 통로개방제의 하나인 pinacidil이 Tl-201을 이용한 종양의 국소화에 어떠한 영향을 미치는 가를 알아보기 위하여, 종양을 가진 생쥐에서 pinacidil에 의한 Tl-201의 체내분포 변화를 알아보았다. 대상 및 방법: 생쥐 유방암세포주를 이식받은 Balb/c 생쥐를 3주간 사육한 후 실험에 이용하였다. Tl-201 185 KBq를 꼬리정맥을 통해 주입한 후 일정시간에 실험동물을 희생시켜 Tl-201의 체내 분포를 알아 보았으며, pinacidil $100{\mu}g$ 투여에 따른 분포 변화를 알아보았다. 또한 Tl-201 3.7 MBq를 꼬리정맥을 통해 주입하여 Tl-201의 시간에 따른 전신 잔류율을 측정하였고, pinacidil 투여에 의한 전신 잔류율 변화를 구하였다. 결과: pinacidil 투여시 대조군에 비해 혈액 내 Tl-201의 방사능치를 약간 감소시키나 신장에서는 현저한 감소를 일으켰다. 또한 간, 근육, 및 장관의 방사능은 pinacidil 투여에 의해 변하지 않았다. 종양 내 Tl-201 섭취율 및 종양조직/혈액 무게당 섭취비는 대조군에 비해 pinacidil 투여군에서 낮았으며, Tl-201의 24시간 전신 잔류율도 pinacidil 투여군에서 낮았다. 결론: $K^+$ 통로개방제는 Tl-201의 체외 배설을 촉진시키고, 신장 섭취를 감소시켜나, 종양 섭취량도 감소시켰다. 그러므로 Tl-201 종양영상 판독시 $K^+$ 통로개방제를 사용하는 경우에는 오히려 Tl-201 종양영상의 질이 향상되기 보다는 저하될 수 있다는 사실을 고려하여야 할 것으로 생각된다.

Expression, Characterization and Regulation of a Saccharomyces cerevisiae Monothiol Glutaredoxin (Grx6) Gene in Schizosaccharomyces pombe

  • Glutaredoxins (Grxs), also known as thioltransferases (TTases), are thiol oxidoreductases that regulate cellular redox state in a variety of organisms. In the budding yeast Saccharomyces cerevisiae, Grx1 and 2 are cytosolic dithiol Grxs, while Grx3, 4 and 5 are monothiol Grxs. A gene encoding a new monothiol Grx, Grx6, was cloned from the genomic DNA of S. cerevisiae by PCR. Its DNA sequence contains 1,080 bp, and encodes a putative protein of 203 amino acid residues containing Cys-Phe-Tyr-Ser at the active site. Grx6 is similar to other monothiol Grxs in the same organism and to Grx3 in the fission yeast Schizosaccharomyces pombe. and its predicted three-dimensional structure resembles that of S. pombe Grx3. S. pombe cells harboring plasmid pFGRX6 containing the Grx6 gene had about 1.3-fold elevated Grx activity in the exponential phase, and grew better than the control cells under some stressful conditions. Synthesis of ${\beta}$-galactosidase from a Grx6-lacZ fusion gene in S. pombe was enhanced by potassium chloride, aluminum chloride and heat ($37^{\circ}C$) treatment. S. pombe cells harboring plasmid pFGRX6 had elevated ROS levels whereas S. pombe cells harboring extra copies of Grx3 had reduced ROS levels.

전기화학증착 방법으로 성장시간에 따라 형성한 SnO2 나노세선의 구조적 성질

  • SnO2 나노세선은 n-형 전기적 성질과 화학적인 안정성 때문에 가스센서, 투명 전극 및 태양전지와 같은 전자소자와 광소자에 널리 사용되고 있다. 화학 기상 증착, 전자빔 증착과 전기화학증착법을 사용하여 SnO2 나노세선을 제작하고 있다. 여러 가지 증착 방법중에서 전기화학증착방법은 낮은 온도와 진공 공정이 필요하지 않으며 대면적 공정이 가능하고 빠른 성장 속도로 나노구조를 효과적으로 성장할 수 있는 장점을 가지고 있다. 본 연구에서는 전기화학증착법을 이용하여 Indium Tin Oxide (ITO) 기판 위에 SnO2 나노세선 성장하고 성장시간에 따라 형성한 SnO2 나노세선의 구조적 성질을 조사하였다. SnO2 나노세선을 성장하기 위하여 D.I. water와 Entanol을 7:3의 비율로 섞은 용액을 $65^{\circ}C$로 유지하였고, 0.015 M의 Tin chloride pentahydrate ($Cl4{\cdot}Sn{\cdot}5H2O$)를 타겟 물질로 이용하였고, 0.1 M의 Potassium chloride (KCl)를 완충 물질로 사용하였다. 전기화학증착 방법을 사용하여 제작한 ITO 기판위에 성장한 SnO2 나노세선 위에 전극을 제작하고 전류-전압 특성을 측정하였다. SnO2 나노세선이 성장되는 전기화학증착 전압을 1.2 V로 고정하고, 성장시간을 15분, 30분 및 1시간으로 변화하여 SnO2 나노세선의 구조적 특성을 분석하였다. X-선회절 (X-ray diffraction; XRD) 실험 결과는 $31^{\circ}$에서 (101) 성장방향을 갖는 SnO2 나노세선이 성장함을 확인하였고, 성장 시간이 길어짐에 따라(101) 성장방향의 XRD 피크의 intensity가 증가하였다. 전기화학증착 성장 시간이 길어짐에 따라 SnO2 나노세선의 지름이 60 nm에서 150 nm로 변화하는 것을 주사전자현미경으로 관측하였다. 이 실험 결과는 전기화학증착방법을 사용하여 제작한 SnO2 나노세선의 성장 시간에 따른 구조적 특성들을 최적화하여 소자제작에 응용하는데 도움이 된다.

폐섬유자원의 발효공학적 이용에 관한 연구 (제3보) 볏짚, 왕겨및 전분박 당화액을 이용한 효모배양 (Studies on the Fermentative Utilization of Cellulosic Wastes (part III) Production of Yeast from the Hydrolyzate of Rice straw, Rice hull and Corn Starch Pulp.)

  • 전분박을 첨가한 왕겨및 볏짚의 산당화액을 이용하여 효모생육조건을 검토하여 다음과 같은 결론을 얻었다. 1. 왕겨와 볏짚에 전분박을 첨가하여 당화시킴으로써 당화액의 당농도가 각각 9.12%, 7.98%까지 증가되었다. 2. 산당화액을 중화할 때 사용한 중화제로서는 CaCO$_3$, Ca(OH)$_2$, NH$_4$OH순으로 효모생육이 좋았다. 3. 당화액을 기질로 하여 무기영양요구성을 검토한 결과 (NH$_4$)$_2$SO$_4$0.3%, $K_2$HPO$_4$0.4%, MgSO$_4$ㆍ7$H_2O$0.02%, NaCl 0.02%, CaCl$_2$0.02%를 가하였을 때 효모의 생육도가 가장 좋았다. 4. 전분박을 첨가한 왕겨와 볏짚의 당화액을 기질로 하여 효모를 배양하였을 때 48시간 이내에 각각 91.2% 90.8%의 당을 소화하였고 당에 대한 수율은 각각 46.5 %, 45.4%이었다.

구강점막 병소 환자의 치과재료에 대한 과민반응 평가 (Hypersensitivity Reactions to Dental Materials in Patients with Oral Mucosal Lesions)

  • This study was to find dental materials causing hypersensitivity reactions by carrying out patch tests in the patients with oral mucosal lesions to investigate the possibility of hypersensitivity reactions in etiology of oral mucosal lesions. 31 patients (female 26, male 5, age range 24-72 years) with oral mucosal lesions were classified as patient group, and 41 volunteers (female 24, male 17, age range 23-40 years) without oral mucosal lesion, systemic disease and history of allergy as control group. The obtained results were as follows: 1. There were various dental restorations in most of patient group and control group, 29(94%) in 31 patient group, 35(85%) in 41 control group. 2. Among sites of oral mucosal lesions, buccal mucosa was the most common site with 60%, followed by gingiva with 24%, tongue with 16%. Lesions in contact with restorations were highly 90% in tongue and 89% in buccal mucosa, but comparatively lower 53% in gingiva. 3. The ratio of positive reactions to the patch test in patient group was significantly higher than the control group (p<0.05). 4. Dental materials causing positive reactions to the patch test were mainly mercury(19%), potassium dichromate(16%), cobalt chloride(16%) in patient group, cobalt chloride(17%) in control group. 5. In 20 patients with lichen planus, 8 patients(40%) showed positive reactions to the patch test.

Impact of partial substitution of NaCl by KCl, and MgCl2 on physicochemical and sensory properties of cooked sausages during storage

  • Objective: To determine the effect of three salting treatments (control, 100% NaCl; T1, 60% NaCl and 40% KCl; T2, 50% NaCl, 40% KCl, and 10% MgCl2) on meat quality of sausages during storage. Methods: Various types of curing salts were used for processing of salamis. Physico-chemical and sensory evaluation of the sausages were analyzed during 4 weeks of ripening. Results: The pH values and salinity contents were the highest in control, while they were the lowest in T2 at 4 weeks (p<0.05). Control and T1 had less thiobarbituric acid reactive substances than T2 at 2 weeks (p<0.05). The volatile basic nitrogen levels in T1 were similar to control and lower than T2 at 2 and 4 weeks (p<0.05). Moreover, T1 samples were redder and had a higher saturation index (C) value when compared to the others. T2 showed greater hardness, gumminess and chewiness when compared with the control (p<0.05). Control was comparable to T1 for sensory attributes. Conclusion: Sodium chloride may be partially substituted with potassium chloride without negatively influencing meat quality of sausages up to 4 weeks of storage. These results could help to develop low-sodium sausages.
