키가 큰 절화용 백합인 'Rialto'와 'Elodie' 및 'Raizan'을 분화 및 화단용으로 사용하기 위하여 ancymidol과 uniconazole 및 paclobutrazol을 농도별로 관주처리 하였다. 오리엔탈 백합인 'Rialto'의 경우, paclobutrazol을 처리한 구에서는 초장뿐만 아니라 잎과 꽃에 있어서도 변화를 관찰할 수 없었다. 그러나 ancymidol 처리구에서 생장은 농도에 관계없이 30% 이상 억제되었다. 한편 엽수나 개화수 등은 무처리구와 비슷한 경향이었다. 수국의 분화 생산을 위하여 온도, 입실시기 및 $GA_3$ 처리가 수국의 생장 및 개화에 미치는 영향을 구명하기 위하여 실험한 결과는 다음과 같다. 온도처리별 초장은 $20^{\circ}C$의 6.7 cm에 비해 $5^{\circ}C$에서 38cm로 현저히 길어졌으며 경장도 같은 경향이었다. 엽장 및 엽폭은 온도처리가 낮을수록 넓어졌으며, 줄기직경은 굵어지는 경향이었다. 개화시는 $5^{\circ}C$에 비해 $15^{\circ}C$ 및 $20^{\circ}C$에서 현저히 단축시킬 수 있었으며, 개화소요일수는 $5^{\circ}C$의 161일에 비해 $15^{\circ}C$에서 88일 소요되었다. 개화율은 온도 처리별 75.8~90.7% 정도이었다. 입실시기별 초장은 입실시기가 늦어질수록 길어졌으며, 엽장 및 엽폭도 같은 경향이었다. $GA_3$ 처리시 초장은 대조구에 비해 $GA_3$ 처리 농도가 높을수록 증가하였고, 엽장 및 엽폭도 같은 경향이었다. 개화시는 12월 30일 입실시 대조구 3월 17일에 비해 $GA_3$$50mgL^{-1}$ 처리시 7일 조기개화되었고, 개화소요일수는 입실시기가 늦을수록 크게 단축되었다. 화방폭은 12월 30일 입실시 대조구에 비해 $GA_3$ 처리시 증가되었고, 개화율은 1월 15일 $GA_3$$50mgL^{-1}$ 처리 97.9%에 비해 11월 15일 대조구에서 62.3%로 현저하게 감소하였다.
작물의 저온 및 고온 피해를 경감시킬 수 있는 방안을 강구하기 위하여 토마토 품종, Fireball과 Patio를 공시재료로 하여 0, 0.001, 0.01, $0.1mg{\cdot}pot^{-l}$의 uniconazole을 처리하였다. 주간/야간의 길이를 12시간으로 하고, 온도를 $25/25^{\circ}C$, $2.5/25^{\circ}C$, $25/2.5^{\circ}C$, $40/40^{\circ}C$로 조절하여 4일간 처리한 결과는 다음과 같다. 1. 가시피해율과 간장의 억제율은 $2.5/25^{\circ}C$ 처리에서 가장 높았고 엽장의 감소는 $40/40^{\circ}C$에서 가장 높았다. 주야간 고온($40/40^{\circ}C$)을 처리한 경우 개화를 지연시켰고 화뢰의 발달을 저해하고 낙뢰를 초래하였다. 온도처리에 따른 피해율은 Fireball에서 Patio에 비하여 높은 것으로 나타났다. 2. 처리농도에 관계없이 uniconazole은 식물체의 생장을 크게 억제하고 chlorophyll의 함량을 증가시켰으며 개화를 지연시키고 저온 및 고온에 의한 가시피해율을 크게 감소시켰다. 3. 일반적으로 uniconazole처리는 고온 및 저온에 의한 엽록소함량의 감소와 개화지연 및 화뢰의 낙뢰를 막아주는 효과는 크게 인정되지 않았다. 고농도 처리는 오히려 Fireball 품종에서 개화지연과 화뢰의 낙뢰를 조장하는 결과를 보였다. 4. Uniconazole의 저온 피해 경감은 free radical scavenger의 역할을 하는 물질의 생성 및 활성증대가 중요한 역할을 한 것으로 판단되나 고온피해에 대한 경감효과의 해석에는 더 진전된 연구가 요구된다.
자생식물 중 실내분화용, 절화 및 화단용으로서 가치가 높고, 한약으로서 이용 가능한 용담의 기내개화 및 대량증식 체계를 확립하기 위하여 식물체의 부위 및 생장조절제의 효과를 시험한 결과는 다음과 같다. 화뢰의 출현은 0.5mg/l kinetin과 GA_${3}$를 첨가하였을 때 가장 많았으나 그 효과는 kinetine이 더 좋았다. 최적의 GA_${3}$나 kinetin농도일 때 두가지 생장조절제를 혼용하면 오히려 화뢰형성수가 감소되었고, 저농도에서는 상호작용의 효과가 있었다 화뢰가 50% 형성되는데 걸리는 주수(T_50는 0.1mg/l GA_3가 첨가되었을 때 Kinetin의 농도에 관계없이 11-14주이었다. 측지 발생은 GA_${3}$의 효과가 kinetin보다 좋았고, 농도는 모두 1.0mg/l까지 높을 수록 좋았다. 중간정도의 농도(0.5mg/l)에서 두 생장조절제간의 상호작용이 인정되었다. 뿌리의 형성은 GA$_{3}$의 농도가 높을 수록 감소하였다. 꼬투리의 크기가 8mm까지 클수록 발아력이 높았으며, 꼬투리의 크기가 작을 수록 비정상적인 식물체가 많았다. 배지에 GA_${3}$/l 첨가는 발아력을 현저히 증가시켰다.
Three horticultural herbaceous plants and a natural herbaceous plant were tested to determine the growth responses, biomass and uptake of cadmium(cd), lead(pb) by application of cd and pb soil treatment in pot culture. The ecological effects on the growth of the plants were investigated to determine the tolerance for the heavy metal pollutants cd and pb. the marginal concentrain of cd treatment on the growth of the each plant was below the 1, 000 ppm treatment of cd. The marginal concentration of pb treatment was below the 1, 000ppm treatment of pb in cultivation of salvia splendens ker., celosia cristata l. and below the 3, 000ppm treatment of pb in cultivation of portulaca grandiflora hook., sedum saramentosum bunge. the resistance for cd of sedum saramentosum bunge, celosia criastata l., portulaca grandiflora hook. and salvia splendens ker. was in the listed order. The resistence for pb was in order of sedum saramentosum bunge, portulaca grandiflora hook. Salvia splendens ker.and celosia criastata l.stems. The flowering of portulaca grandiflora hook. was sustained in the pb 1, 000ppm treated group only. The higher the concentration of pb in the soil cultivated the plants was, the less the content of leaf chlorophy11 in each plant was. The number of stomata per unit leaf area was the highest in salvia splendens ker. and in order of celosia l., sedum saramentosum bunge., portulaca grandiflora hook., the higher the cd and pb concentration of cd and pb treatment was, the more the concent of cd and pb in the part of each plant increased. the content of cd and pb in the stems of salvia spiendens ker. was the highest in the 1, 000ppm-treated ground and in order of the roots, the leaves and the flowers.
The most commonly grown economical and flue-cured tobacco cultivar Yellow Special A was used in pot-culture tests in order to study Ethrel (2-chloroethyl phosponic acid) effects on accelerating maturity of tobacco leaves in relation to the most adequate level of the chemical useful for field growing, nitrogen level in soil for the most pronounced response, and the most suitable spray period during the growth stages of pre-, post- and topping periods. The following conclusions, thus, were obtained from the present studies; 1. 500ppm Ethrel spray was reconfirmed to be adequate in the practical applications, although the extent of yellow-ripening of tobacco leaves was increased as the Ethrel level increased. The highest leevel treated resulted in causing chemically damaged lesions on leaves and early defoliation. 2. Ethrel-treated leaves showed deeper yellowish tinge to them than those without treatment, while different levels of the chemcial had less influence on the tinge. 3. An adequate level of nitrogen supply to plants favored the Ethrel response, whereas either very low or high level of nitrogen in the soil lowered the chemical effect on accelerating the yellow-ripening. When carbohydrates versus total nitrogen ratio became relatively high, the condition brought out some outstanding Ethrel effects. 4. Chlorophyll level of leaves increased as soil applications of nitrogen level increased, and that also increased carotenoid level of the tobacco leaves. Ethrel-treated leaves showed deeper orange tinge than those without treatment, while the highest level of nitrogen application showed the deepest orange tinge to tobacco leaves. 5. Pre-topping treatment (12 days before topping and flowering) resulted in almost no Ethrel response, and that treatment right on the day of topping, showed response of yellow-leaf ripening at nearly bottom-half leaves of a tobacco plant. The post-topping treatment (12 days after topping) made plants showing full response of Ethrel from bottom to the top leaves of tobacco plant in accelerating the leaf maturity. 6. The extent of Ethrel responses on accelerating yellow-ripening of tobacco leaves was discussed for the modifying influences brought about by certain environmental factors. Discussions were also made about the possible practical applications (particularly for pre-rice planting) and quality difference that may be caused by such growth environments.
A simple and efficient protocol was developed for culturing Cu-tolerant and Cu-accumulating plants via pre-adaptation to Cu during plant tissue culture. We induced multiple shoots from begonia (Begonia evansiana Andr.) leaf explants on MS medium supplemented with naphtaieneacetic acid and benzyladenine. After 3 months, small plantlets were transferred to MS medium supplemented with $100{\mu}M\;CuCl_2$ for pre-adaptation to Cu and cultured for 5 months. Then, these plantlets were individually planted in pots containing artificial soil. An additional 500 mg of Cu dissolved in 1/4 strength MS solution was applied to each pot during irrigation over the course of 2 months. We planted pre-adapted and control begonias in soil from the II-Kwang Mine, an abandoned Cu mine in Pusan, Korea, to examine their ability to tolerate and accumulate Cu for phytore-mediation. Pre-adapted begonias accumulated $1,200{\mu}g$ Cu/g dry root tissue over the course of 45 days. On the other hand, non-Cu-adapted controls accumulated only $85{\mu}g$ Cu/g dry root tissue. To enhance Cu extraction, chelating agents, ethylenediamine tetraacetic acid (EDTA)-dipotassiun and pyridine-2,6-dicarboxylic acid (PDA), were applied. While the chelating agents did not enhance accumulation of Cu in the roots of control begonias, EDTA application increased the level of Cu in the roots of pre-adapted begonias twofold (to $2,500{\mu}g$ Cu/g dry root tissue). Because pre-adapted begonias accumulated a large amount of Cu, mainly in their roots, they could be used for phytostabilization of Cu-contaminated soils. In addition, as a flowering plant, begonias can be used to create aesthetically pleasing remediation sites.
This study was conducted to determine the efficacy of using uniconazole,[(E)-1-(4-chlorophenyl)-4,4-dimethyl-2-(1,2,4-triazole-1-yl)-1-penten-3-ol)] as a phytoprotectant against $SO_2$ injury in snap been (Phaseolus vulgaris L. 'Strike'). Thirteen days prior to $SO_2$ fumigation, plants were given a 100 ml soil drench of uniconazole solution at concentrations of 0.02, 0.10, 0.25 and 0.50 mg/pot. All four uniconazole concentrations were significantly effective in providing protection against $SO_2$ exposure(3 h at 1.5 ppm), but uniconazole treatment above 0.02 mg/pot severely reduced stem elongation, leaf enlargement, flowering date and pod number and weight. Uniconazole treatment had little or no effect on stomatal conductance but reduced transpiration rate on a whole plant basis by nearly 40%. This may reflect an alteration in canopy structure by reducing stem elongation and leaf enlargement. Although uniconazole did not increase the activities of superoxide dismutase(SOD) and peroxidase(POD) in non-$SO_2$-fumigated plants, it significantly increased those enzyme activities in $SO_2$-fumigated plants. Chlorophyll concentration on the basis of unit area was increased 50-60% by uniconazole. However, the difference was not detected on the basis of dry weight. $SO_2$ increased variable chlorophyll fluorescence (Fv) 48% after 1.5 h of exposure in non-uniconazole treated plants but decreased Fv in the plants after 3 h of exposure. By appliing uniconazole, it was possible to maintain high Fv values in the latter group of plants. These results suggest that the phytoprotective effects of uniconazole are related to its growth-retarding properties as an anti-gibberellin as well as the increase of activites of free radical scavengers such as SOD and POD.
