Pluronic F-68이 동결보존된 형질전환 담배세포의 해동 후 세포생장에 미치는 영향 (Effect of Pluronic F-68 on the Post-thaw Growth of Cryopreserved Transgenic Nicotiana tabacum Cells)

  • 본 연구에서는 Pluronic F-68을 해동 후 회복배지에 적용하여, 동결보존된 형질전환 식물세포의 생장 증진을 도모하였다. Pluronic F-68은 세포표면의 소수성을 감소시켰을 뿐만 아니라, 세포의 투과성을 증진시켜 세포와 직접 상호 작용할 수 있음을 관찰하였다. 또한 Pluronic F-68의 첨가에 의해 재조합단백질의 발현에 부정적인 영향을 보이지 않음을 확인하였다. 따라서 동결보존시 해동 후 회복배지에 Pluronic F-68의 첨가는 식물세포의 신속하고 효율적인 대량 현탁배양을 가능하게 할 수 있다.

북방대합, Spisula sachalinesisr 유생의 발생단계별 냉동-해동후 생존율 (Survival Rates of Frozen-thawed Surf Clam, Spisula sachalinensis Larvae in Five Developmental Stages)

  • 동해안산 북방대합의 유생을 냉동보존하기 위한 적절한 유생단계를 선택하기 위해, 담륜자, 초기 D상 유생, 후기 B상 유생, 초기 각정기, 후기 각정기 유생에 대한 냉동실험을 실시하였다. 동해방지제로는 dimethyl sulfoxide와 ethylene g1yco1를 사용하였다. 동해방지제에 각 단계의 유생을 10분간 두어 평형상태에 달하게 한 다음, 냉동하여 액체질소에 보존하였다. 해동결과, 가장 높은 생존율을 나타낸 유생은 담륜자로서 동해방지제 2.0 M BMSO와 2.0 M ethylene glycol에서 생존율은 각각 97.4%와 85.0%였다. 발생이 진행된 유생일수록 생존율이 낮아지는 경향을 보였다.

Unconfined compressive strength and freeze-thaw resistance of sand modified with sludge ash and polypropylene fiber

  • In recent years, the amount of sludge ash (SA) has considerably increased due to rapid urbanization and population growth. In addition, its storage in landfills induces environmental pollution and health problems. Therefore, its disposal in an environmentally friendly way has become more important. The main goal of this study is to investigate the reusability of sludge ash as an additive with polypropylene fiber (PF) to stabilize marginal sand based on the compressive strength performances from UCS tests. For this purpose, a series of UCS tests was conducted. Throughout the experimental study, the used inclusion rates were 10, 15, 20 and 30% for sludge ash and 0, 0.5 and 1% for polypropylene fiber by total dry weight of the sand+sludge ash mixture and the prepared samples were cured for 7 and 14 days prior to the testing. Freezing and thawing resistance of the mixture including 10% sludge ash and 0, 0.5 and 1% polypropylene fiber was also examined. On the basis of UCS testing results, it is said that sludge ash inclusion remarkably enhances UCS performance of sand. Moreover, the addition of polypropylene fiber to the admixtures including sand and sludge ash significantly improves their stress-strain characteristics and post-peak strength loss as well as UCS. As a result of this paper, it is suggested that sludge ash be successfully reused with polypropylene fiber for stabilizing sand in soil stabilization applications. It is also believed that the findings of this study will contribute to some environmental concerns such as the disposal problem of sludge ash, recycling, sustainability, environmental pollution, etc. as well as the cost of an engineering project.

Establishment and long-term culture of the cell lines derived from gonad tissues of Siberian sturgeon (Acipenser baerii)

  • To culture germline stem cells in vitro, establishment of the cell lines that can be used as the feeder cells is a prerequisite. In this study, we tried to establish gonad-derived cell lines in Siberian sturgeon (Acipenser baerii). Five 1-year-old A. baerii were used as a donor of gonad tissues, and gonad-dissociated cells were cultured in vitro. Subsequently, determination of growth conditions, long-term culture, characterization, and cryopreservation of the cell lines were also conducted. Five gonad-derived cell lines were stably established and cultured continuously over at least the 73th passage and 402 culture days under the media containing 20 % fetal bovine serum at $28^{\circ}C$. All cell lines consisted of two main cell types based on morphology even if the ratio of the two cell types was different depending on cell lines. Despite long-term culture, all cell lines maintained diploid DNA contents and expression of several genes that are known to express in the A. baerii gonad. After freezing and thawing of the cell lines, post-thaw cell viabilities between 57.6 and 92.9 % depending on cell lines were indentified, suggesting that stable cryopreservation is possible. The results and the cell lines established in this study will contribute to the development of an in vitro system for A. baerii germline stem cell culture.

돼지 동결 정액을 이용한 체외 수정란 생산 효율 (Effect of Production In Vitro Embryo using Boar Frozen Semen)

  • This study was carried out to investigate the effective genetic resources preservation system using the frozen boar semen. The porcine oocytes were matured for 44 hours in NCSU-23 medium with or without 10% Porcine Follicle Fluid (PFF), 0.5 ${\mu}g/ml$ porcine FSH, 0.5 ${\mu}g/ml$ equine LH, 1.0 ${\mu}g/ml$ 17 $\beta$-estradiol ($E_2$) and 10 ng/ml Epidermal Growth Factor (EGF) under mineral oil at $38.5^{\circ}C$ in humidified atmosphere of 5% $CO_2$ in air. After 44 h of culture, the oocytes were inseminated with frozen-thawed semen and fresh semen prepared with mTBM medium for 6 h. Later, set of 50 presumptive zygotes were transferred into 4-well dish (500 ${\mu}l$) of IVC medium. for embryos freezing, slow-freezing and vitrification methods were used as a cryopreservation. Differences among treatments were analyzed using General Linear Model Procedure by SAS Package (version 6.12) differences were considered significant when p<0.05. Following IVF and IVC, the rates of cleavage and blastocysts formation were significantly higher (p<0.05) in hormone supplemented group than that of hormone-free group (25.7 vs, 12.1). The development rates to cleavage and blastocysts were significantly higher in PZM-5 group than NCSU-23 group (60.3%, 46.6% vs 27.4%, 11.1%). Further improvement was achieved when PZM-5 was supplemented with FBS. Cleavage rates was significantly higher in fresh semen source group than frozen semen (66.7% vs 43.7%). However in blastocysts rates was similar two groups. Post-thaw survival rates of embryos were 1.2% and 2.2% in slow-frezing and vitrification groups, respectively. The results of our study suggest that it is still possible to improve the culture conditions and boar semen cryopreservation for enhance reproductive technology and animal genetic resources conservation.

Serum-Free Medium에서 배양한 한우 배의 내동성과 이식 (Transfer, Cryopreservation and Production of Bovine Embryos Cultured in Serum-Free System)

  • 난자의 체외성숙 및 체외배양에는 일반적으로 동물의 혈청을 사용하고 있다. 그러나, 채취한 소의 상태에 따라서 혈청의 질에 차이가 있어 실험데이터가 일정하지 않을 수 있고, 그것으로부터 바이러스, 세균, 마이코플라즈마 등에 오염될 가능성이 있다. 따라서, 본 실험에서는 완전 무 혈청 배양액에서 난자의 성숙, 배 발생율, 세포 수, 동결성을 검토하였다. 다음으로, 근래 혈청배지로 생산한 체외 배양 수정란은 과체중의 산자 생산, 초기 산자 사망률, caesarean section, dystocia 같은 발병으로 문제가 지적되고 있다. 그러나 무 혈청 배지로 생산한 체외 배양 수정란은 이러한 현상을 개선할 수 있다는 보고가 있어 본 실험에서는 완전 무혈청 배양액에서 생산된 동결란과 신선란을 젖소에 이식하여 그 결과를 검토하였다. 무혈청 배양액에는 에너지원과 세포성장인자가 첨가된 IVD101과 IVMD101, 대조군인 혈청 배양액에는 TCM199 + 10% FBS가 사용되었다. IVMD101에 의해 체외성숙이 이루어 졌고, 혈청이 첨가된 TCM199 + 10% FBS에서는 12.4% 배 발생율을 보인데 반해, 무혈청 배양액 IVD101과 IVMD101이 각 32.4%, 34.5%로 훨씬 높은 배 발생율을 보였다. 더욱이, 세포 수에 있어서도 무혈청 배양액에서 발생된 배반포의 세포수가 혈청이 첨가된 배양액에서 발생된 배반포의 세포수보다 월등하였으며, 이는 체내란과 비슷한 세포 수를 보이고 있다. 수정란의 내동성을 보면, 융해하고 24시간 배양기 정치 후에 IVD101과 IVMD101은 94.5%, 95.8%의 생존율을 보인 반면 TCM199 + 10% FBS는 52.5%에 불과하다. 융해하고 72시간후 탈출 배반포율이 IVD101과 IVMD101는 78.4%, 83.7%였는데 반해 TCM199 + 10% FBS는 32.0%였다. 마지막으로, 무혈청 배양액에서 발생한 동결란과 신선란을 젖소에 이식한 후에 임신율에 있어서는 유의적 차이는 없었지만, 무혈청 배지에서 생산된 수정란의 내동성의 탁월함이 증명되었다. 이러한 결과들로 종합해 보면, 무혈청 배양액 (IVD101, IVMD101)에서 발생된 수정란이 혈청 배양액에서 발생된 수정란보다 배발생율, 세포수, 동결성, 임신율에 있어서 우수함을 알 수 있었다. 이러한 무혈청 배양액은 배발생 과정의 연구뿐만 아니라 수정란 이식을 위한 고품질의 체외 수정란의 대량 생산, 복제, 형질전환 동물 생산 등에도 유익할 것으로 사료된다.
