Toward Complete Bacterial Genome Sequencing Through the Combined Use of Multiple Next-Generation Sequencing Platforms

  • PacBio's long-read sequencing technologies can be successfully used for a complete bacterial genome assembly using recently developed non-hybrid assemblers in the absence of second-generation, high-quality short reads. However, standardized procedures that take into account multiple pre-existing second-generation sequencing platforms are scarce. In addition to Illumina HiSeq and Ion Torrent PGM-based genome sequencing results derived from previous studies, we generated further sequencing data, including from the PacBio RS II platform, and applied various bioinformatics tools to obtain complete genome assemblies for five bacterial strains. Our approach revealed that the hierarchical genome assembly process (HGAP) non-hybrid assembler resulted in nearly complete assemblies at a moderate coverage of ~75x, but that different versions produced non-compatible results requiring post processing. The other two platforms further improved the PacBio assembly through scaffolding and a final error correction.

Deep Learning based Scrapbox Accumulated Status Measuring

  • In this paper, we propose an algorithm to measure the accumulated status of scrap boxes where metal scraps are accumulated. The accumulated status measuring is defined as a multi-class classification problem, and the method with deep learning classify the accumulated status using only the scrap box image. The learning was conducted by the Transfer Learning method, and the deep learning model was NASNet-A. In order to improve the accuracy of the model, we combined the Random Forest classifier with the trained NASNet-A and improved the model through post-processing. Testing with 4,195 data collected in the field showed 55% accuracy when only NASNet-A was applied, and the proposed method, NASNet with Random Forest, improved the accuracy by 88%.

  • A typical statistical parametric speech synthesis (text-to-speech, TTS) system consists of separate modules, such as a text analysis module, an acoustic modeling module, and a speech synthesis module. This causes two problems: 1) expert knowledge of each module is required, and 2) errors generated in each module accumulate passing through each module. An end-to-end TTS system could avoid such problems by synthesizing voice signals directly from an input string. In this study, we implemented an end-to-end Korean TTS system using Google's Tacotron, which is an end-to-end TTS system based on a sequence-to-sequence model with attention mechanism. We used 4392 utterances spoken by a Korean female speaker, an amount that corresponds to 37% of the dataset Google used for training Tacotron. Our system obtained mean opinion score (MOS) 2.98 and degradation mean opinion score (DMOS) 3.25. We will discuss the factors which affected training of the system. Experiments demonstrate that the post-processing network needs to be designed considering output language and input characters and that according to the amount of training data, the maximum value of n for n-grams modeled by the encoder should be small enough.

Seismic fragility curves of single storey RC precast structures by comparing different Italian codes

  • The seismic events in Northern Italy, May 2012, have revealed the seismic vulnerability of typical Italian precast industrial buildings. The aim of this paper is to present a seismic fragility model for Italian RC precast buildings, to be used in earthquake loss estimation and seismic risk assessment by comparing two building typologies and three different codes: D.M. 3-03-1975, D.M. 16-01-1996 and current Italian building code that has been released in 2008. Based on geometric characteristics and design procedure applied, ten different building classes were identified. A Monte Carlo simulation was performed for each building class in order to generate the building stock used for the development of fragility curves trough analytical method. The probabilistic distributions of geometry were mainly obtained from data collected from 650 field surveys, while the material properties were deduced from the code in place at the time of construction or from expert opinion. The structures were modelled in 2D frameworks; since the past seismic events have identified the beam-column connection as the weakest element of precast buildings, two different modelling solutions were adopted to develop fragility curves: a simple model with post processing required to detect connection collapse and an innovative modelling solution able to reproduce the real behaviour of the connection during the analysis. Fragility curves were derived using both nonlinear static and dynamic analysis.

  • In this paper, we describe a case of utilizing SaaS technology to build NC Post-processor(WBS) based cloud computing. Developed WPS system was implemented to provide stable and continuous system service by utilizing SCoD methodology. WPS is designed user interface module and control engine module. The interface module is downloaded in a client PC and the control engine is installed in cloud parm area. These modules are connected with computer network. WPS was completed a function test for sheet cutting field and mold manufacturing field, and it is processing a commercial service using with improve the user's convenience and adding a bill charge module.

  • NC simulation has been used to replace the test cutting of NC machining. Although it can reduce the NC part programming effort, it still has a problem. If any error is found during the simulation, then the part has to be reprogrammed and it is time consumming. This paper presents a method for verifying and editing the NC code after the post-processing without going back to the part programming stage. A data structure and an algorithm to verify and edit the NC code interactively with the aid of graphics is introduced. Z-map method is used for the shaded image display and cross-sectional view display of the macined parts. The method was implemented in a IBM/PC-386 with MS-Windows software, and the multi-window function of the of the MS-Windows is used for the simultaneous editing and verification.

  • A continuum analysis has been performed for the application to the thermo-elasto-plastic behavior in a discontinuous metal matrix composite. an FEM (Finite Element Method) analysis was implemented to obtain the internal field quantities of composite as well as overall composite behavior and an experiment was demonstrated to compare with the numerical simulation . As the procedure, a reasonably optimized FE mesh generation, the appropriate imposition of boundary condition , and the relevant post processing such as elastoplastic thermomchanical analysis were taken into account. For the numerical illustration, an aligned axisymmetric single fiber model with temperature dependent material properties and precipitation hardening effect has been employed to assess field quantities. It was found that the residual stresses are induced substantially by the temperature drop during the thermal treatment and that the FEM results of the vertically and horizontally constrained model give a good agreement with experimental data.with non-woven carbon mat is about 24% higher than that of composite materials without non-woven carbon mat. Transverse tensile strength and torughness also increase by inserting non-woven carbon mat between layers.

  • We presented a method for Vad(Voice Activity Detection) using Bi-level HMM. Conventional methods need to do an additional post processing or set rule-based delayed frames. To cope with the problem, we applied to VAD a Bi-level HMM that has an inserted state layer into a typical HMM. And we used posterior ratio of voice states to detect voice period. Considering MFCCs(: Mel-Frequency Cepstral Coefficients) as observation vectors, we performed some experiments with voice data of different SNRs and achieved satisfactory results compared with well-known methods.

  • The information users of the middle of 1990s. who know the Internet as well as its useful information services, are now expecting the virtual library services. Especially the increasing demands on hypertext and hypermedia information in the internet settings have been centered on the W3 with the man-page information. In this manner, the paper describes the access methods with brief concepts of the W3 and explains URLs and HTML. It also gives the retrieval layouts of unformatted data including images and sounds and then provides the information sources and software of W3 Clients and Servers in order to catch up the most recently post version of W3.

  • 최근들어 제품을 개발하는 과정 중, 디자이너와 개발자, 의사 결정권자들이 FEM, CFD 시뮬레이션 결과를 리뷰할 때에 가상현실기술을 도입하는 사례가 늘고 있다. 몰입감을 높여주는 가상현실환경은 모델에 대한 해석 결과물을 정확하고 효과적으로 분석할 수 있도록 돕는다. 데이터의 실제 크기와 같게 혹은 그보다 더 크고 자세한 이미지를 제공하는 가상현실 몰입환경은 사용자가 데스크탑 환경만을 사용할 때 경험할 수 없는 높은 사실감을 제공함으로써 사용자에게 시각적인 만족감을 줄 수 있다. 하지만 데스크탑 환경에 비해 해상도가 낮고, 어두운 곳에서 스테레오 안경이나 HMD(Head Mounted Display), Data glove등을 착용해야 하는 불편함과 멀미, 시각적인 피로, 방향감각 상실로 대표되는 가상멀미 등으로 인해 장시간 사용에 어려움이 있다. 데스트탑 환경에서의 데이터 리뷰는 고해상도 이미지 분석은 가능하지만, 입체감이 떨어지기 때문에 리뷰 데이터의 실제감이 떨어진다. 이와 같은 문제점들을 보완하기 위해서 본 논문에서는 데스크탑 환경과 가상현실 환경 간의 협업이 가능한 FEM/CFD 가시화 시스템을 제시한다. 본 시스템은 가상현실 몰입환경에서 해석 데이터를 단순히 가시화하는 것뿐만이 아니라, 데스크탑 시스템과 동일한 3D 인터페이스 구조를 제공한다. 따라서, 해석 결과 분석을 위한 동일한 post-processing 작업이 네트워크로 연결된 원격 공간의 사용자들이 사용하는 시스템들 사이에서 실시간으로 진행될 수 있다.
