Preparation of Porous PLGA Microfibers Using Gelatin Porogen Based on a Glass Capillary Device (젤라틴 기공유도물질과 유리모세관 장치를 이용한 다공성 PLGA 미세섬유의 제조)

  • We present a method of fabricating poly (lactic-co-glycolic acid) (PLGA) porous microfibers using a pore template. PLGA microfibers were synthesized using a glass capillary tube in a poly-(dimethylsiloxane) (PDMS) microfluidic chip. Gelatin solution was used as a porous template to prepare pores in microfibers. Two phases of PLGA solutions in different solvents-DMSO (dimethyl sulfoxide) and DCM (dichloromethane)-were used to control the porosity and strength of the porous microfibers. The porosity of the PLGA microfibers differed depending on the ratio of flow rates in the two phases. The porous structure was formed in a spiral shape on the microfiber. The porous structure of the microfiber is expected to improve transfer of oxygen and nutrients, which is important for cell viability in tissue engineering.

  • Cellulose is an abundant biodegradable material in nature with excellent properties, but due to its poor processability, it has been widely studied for processing through modification. Cellulose acetate propionate (CAP) is a cellulose derivative in which the hydroxyl group of cellulose is replaced by acetyl and propionyl groups. CAP has several advantages, such as excellent solubility, structural stability, light and weather resistance, and good transparency. Porous nanofibers with excellent specific surface area, which can be applied in various fields, can be easily formed by the phase separation method using highly volatile solvents. High speed centrifugal spinning is a nano/micro fiber preparation method with advantages such as fast spinning and easy alignment control. In this study, a CAP/polybutylene succinate (PBS) spinning solution with chloroform as solvent was prepared to prepare porous microfibers and the fiber morphology was examined as a function of the disk rotation speed in an high speed centrifugal spinning device.

Highly-sensitive Detection of Salvianolic Acid B using Alumina Microfibers-modified Electrode

  • Alumina microfibers with porous structures were prepared through hydrothermal reaction, and then used to modify the surface of carbon paste electrode (CPE). After modification with alumina microfibers, the electrochemical activity of CPE was found to be greatly improved. On the surface of alumina microfibers-modified CPE, the oxidation peak current of salvianolic acid B, a main bioactive compound in Danshen with anti-oxidative and anti-inflammatory effects, was remarkably increased compared with that on the bare CPE surface. The influences of pH value, amount of alumina microfibers and accumulation time were studied. Based on the strong signal amplification effects of alumina microfibers, a novel electrochemical method was developed for the detection of salvianolic acid B. The linear range was from 5 ${\mu}gL^{-1}$ to 0.3 mg $L^{-1}$, and the detection limit was 2 ${\mu}gL^{-1}$ (2.78 nM) after 1-min accumulation. The new method was successfully used to detect salvianolic acid B in ShuangDan oral liquid samples, and the recovery was over the range from 97.4% to 102.9%.
