The production of extracellular polysaccharide, EPS WN9, from Paenibacillus sp. and its suitability as a viscosity modifying admixture for cement mortar mixing were investigated. After 48h culture in an optimized medium, cell growth and EPS production were 1.2g/L and 4.0g/L, respectively. By adding EPS WN9 to mortar, it was possible to prepare a homogeneous mortar without material segregation and excess air entrapment. The optimal amount of EPS addition to mortar found to be 0.02 to 0.05%(w/w) of the cement used. Increasing the dosage of EPS WN9 from 0 to 0.05%(w/w) resulted in a setting retardation of 0.14h to 0.8h and an increase in the compressive strength of mortar of 10 to 20%.

The immunomodulating activity of a polysaccharide isolated from Morus alba (PMA) root bark was examined in murine splenic lymphocytes. PMA enhanced proliferation of splenic lymphocytes in a synergistic manner in the presence of mitogens. However, PMA suppressed pri-may IgM antibody production from B cells, which was activated with lipopolysaccharide, a polyclonal activator, or immunized with a T-cell dependent antigen sheep red blood cells. Our observations showed that the immunomodulating activity of PMA increased lymphocyte proliferation and that PMA decreased antibody production from B cells, which was distinct from those of other plant-originated polysaccharides.
This study was concerned with the optimization of liquid culture conditions for mycelial growth and polysaccharide production and its physicochemical properties in Cordyceps militaris. The one factor at a time method was adopted to investigate the effects of medium composition, environmental factors and C/N ratio. Among the these varialbles, glucose 80g/L, yeast extract 10g/L, $MgSO_4{\cdot}7H_{2}O\;0.5g/L$, $KH_{2}PO_4\;0.5g/L$ were proved to be the most suitable carbon, nitrogen, and mineral sources, respectively. The optimal temperature, initial pH, working volume were identified to be $24^{\circ}C$, 7.0 and 100ml, respectively. Under the optimal conditions, the strategies in shake flask culture and 5L jar fermentor led to mycelial growth of 29.43 g/L, 28.88g/L and polysaccharide production of 2.53g/L, 6.38 g/L, respectively. Among the phisicochemical properties, relative concentrations(w/v) of total sugar, uronic acid, protein and hexoseamine were identified to be 74.07%, 1.13%, 0.91%, and 0.46%, respectively. The fraction of neutral and acidic polysaccharide were identified to be 81.9% and 18.1%, respectively.

To examine effects of agitation and aeration as well as adding of glucose and yeast extract on cell growth and polysaccharide production by Paecilomyces japonica, batch culture was carried out at 5L jar fermenter at $27^{\circ}C$ with the initial pH 7 for 7 days cultivation(innoculum size 2%, working volume 3L). Media compositions(g/L) were 30 glucose, 20 yeast extract, 0.5 $KH_2PO_4$, $0.1\;CuCl_2\;{\cdot}\;2H_2O$. Optimum culture conditions of agitation and aeration in batch culture were 400 rpm and 1.0 vvm, resulting in 23.1 g/L biomass and 2.5 g/L polysaccharide. Additional feeding of glucose and yeast extract with a pulse mode conferred an advantage on cell growth and polysaccharide production with showing the results of 29.2 g/L and 3.3 g/L, respectively.
The antitumor activities of the cell bound polysaccharide(CBP), water soluble polysaccharide(WSP) and sulfated polysaccharide(SP) of Zoogloea sp. were observe. The results obtained were as follows : 1) The CBP, WSP, and SP showed cytotoxic effect on the Meth A cells in vitro, however, the effect of CBP and WSP was more ten-fold greater than that pf SP. 2) When CBP, WSP, and SP was inoculated into the peritoneal cavity of the Meth A cells transplanted mice, the average survival days tended to prolonged slightly as compared with the control. 3) When Meth A cells were transplanted subcutaneously into the back side of mice, and then CBP, WSP, and Sp was inoculated into the peritoneal cavity of mice, the tumor growth inhibition ratio was 46.9% for WSP, 40.4% for CBP, and 16.2% for SP. 4) The phagocytic activity of peritoneal macrophages elicited with CBP, WSP, and SP was significantly increased than that of control. 5) The production of nitric oxide in the peritoneal macrophages stimulated with CBP, WSP, SP, and LPS aloneo was not increased than that of control. The production of nitric oxide in the peritoneal macrophages stimulated with IFN-r and CBP, IFN-r and WSP and IFN-r and SP was significantly increased than that of control, but in the case of stimulated with IFN-r and WSP was increased 50% for CBP and SP. These results suggest that the CBP, WSP and SP of Zoogloea sp. showed direct cytotoxic effect and tumor growth inhibition on Meth A cells in vitro and in vivo, and induced nitric oxide production of activated macrophages.
The cultural condition of Pseudomonas mendocina for polysaccharide production was examined. The optimal medium contains following composition per liter of distilled water: Sucrose 23.75g, $(NH_4)_2SO_4$ 1.57g, Yeast extract 0.5g, $KH_2PO_4\;2.9g,\;MgSO_4.\;7H_2O\;1.0g,\;CaCO_3$ 2.5g. The optimum temperature and pH were $30^{\circ}C$ and 6.5. At the condition. Ps mendocina produced 5.98g/l of polysaccharide. The culture viscosity after 3 days was 191mPa.s at $70sec^{-1}$. The product yield $(Y_{p/s})$ and specific productivity $(Q_p)$ were 25.18% and 32.83mg/g-cell/h.
The effect of cultural conditions on mycelial growth, polysaccharide production in Phellinus linteus and its monosaccharide composition was studied. P. linteus showed the highest growth (0.9 g/100 ml) on glucose but the polysaccharide production was the highest (13.7%) on mannose. The fungus grew very well at neutral pH (0.9 g/100 ml) but the growth was reduced to 0.47 g per 100 ml at alkaline pH. For the different pH, the yield of polysaccharide was in the range of $5{\sim}8%$. The highest yield of 7.94% was obtained at pH 5. Also a variation in monosaccharide composition was observed for different carbon sources and pH. The composition ranges of glucose, mannose, and galactose of polysaccharide were $80{\sim}95%,\;3{\sim}12%,\;and\;2{\sim}10%$ depending on carbon sources, respectively. In contrast, the variation of composition range of three monosaccharides was narrower for different pH than that for carbon sources. These results suggested the possibility of the improvement of production and the physiological modification of the polysaccharide.
As a link in the studies on the extracellular polysaccharide by lactic acid bacteria, the experiment was conducted to investigate the viscosity variations of 10% reconstituted skim milk and optimum cultural conditions for the production of extracellular polysaccharide. 1. The viscosity of 10% reconstituted skim milk cultured by 8tr. thermophilu8 510 reached 5,000 CP at $41^{\circ}C$ and on 5 days. Because it was the highest value among the microorganisms tested, Str. thermophilu8 510 was chosen as a main test microoganism and studied on factors affecting maximum viscosity depending upon production of exopolysaccharide. 2. Absorbance of synthetic culture medium was highest at $41^{\circ}C$. Sucrose, in a member of carbohydrates, was the most effective carbon source for the production of exopolysaccharide within 5% concentration. 3. The production of exopplysaccharide was stimulated by $Mg^{++}$.
A strain GP32 which produces a highly viscous extracellular polysaccharide was conducted with soil samples and identified as Pseudomonas species. The culture flask conditions for the production of extracellular polysaccharide by Pseudomonas sp. GP32 were investigated. The most suitable carbon and nitrogen source for extracellular polysaccharide production were galactose and (NH4)2SO4. The optimum carbon/nitrogen ratio for the production of extracellular polysaccharide was around 50. The optimum pH and temperature for extracellular polysaccharide production was 7.5 and 32℃, respectively. In batch fermentation using a jar fermentor, the highest extracellular polysaccharide content (15.7 g/l) was obtained after 70 hr of cultivation. The extracellular polysaccharide produced by Pseudomonas sp. GP32 (designated Biopol32) was purified by ethanol precipitation, cetylpyridinium chloride (CPC) precipitation, and gel permeation chromatography. Biopol32, which has an estimated molecular weight of over 3×107 datons, is a novel polysaccharide derived from sugar components consisting of galactose, glucose, gulcouronic acid and galactouronic acid in an approximate molar ratio of 1.85 : 3.24 : 1.00 : 1.42. The solution of Biopol32 showed non-Newtonian characteristics. The viscosity of Biopol32 exhibited appeared to be higher at all concentration compared to that of zooglan from Zoogloea ramigera. An analysis of the flocculating efficiency of Biopol32 in industry wastewater (food, textile, and paper wastewater) revealed chemical oxygen demand (COD) reduction rates 58.4-67.3% and suspended solid (SS) removal rates 82.6-91.3%. Based on these results, Biopol32 is a possible candidate for industrial applications such as wastewater treatment.
The polysaccharide (BB-pol) extracted from Bifidobacterium bifidum BGN4 showed growth inhibitory effects on several colon cancer cell lines such as HT-29 and HCT-116. To increase the yield of polysaccharide, B. bifidum BGN4 was cultured in various culture media with different compositions. When B. bifidum BGN4 was cultured in modified MRS broth containing phytic acid, the cells showed increased branch formation and enlarged morphology. The content of total carbohydrate and the ability of adhesion to intestinal epithelial cells were also increased by phytic acid. The polysaccharide obtained from the cells grown in the presence of phytic acid inhibited the proliferation of cancer cell lines such as HT-29 and MCF-7 cells but not normal colon cell line, FHC. Taken together, Bifidobacterium grown in the presence of phytic acid may confer enhanced beneficial function for the host.
