홍미삼 알콜 추출박을 이용한 산성다당체 다량 함유 홍삼 엑기스 제조 (Preparation of Red Ginseng Extract Rich in Acidic Polysaccharide from Red Tail Ginseng Marc Produced After Extraction with 70% Ethyl Alcohol)

  • In this study, we investigated the appropriate conditions to extract acidic polysaccharide and to prepare red ginseng extract being rich in acidic polysaccharide from red tail ginseng marc produced after manufacturing alcoholic extract from red tail ginseng. Amount of acidic polysaccharide in red tail ginseng marc was about 11%. The best condition for the extraction of acidic polysaccharide from the marc was using of 3~5 mg of $\alpha$-amylase/g residue/15 ml of distilled water, and the amount of acidic polysaccharide in water extract of the residue treated with $\alpha$-amylase was about 27%. So, it is possible to manufacture red ginseng extract being rich in acidic polysaccharide using water extract of red tail ginseng alcoholic residue as extraction solvent. From the above results, we suggest that red tail ginseng residue produced by manufacturing alcoholic extract of red tail ginseng has high potencies in the utilization of waste material.

백삼 알코올 추출박을 이용한 산성다당체 다량 함유 백삼 농축액 제조 (Preparation of Ginseng Concentrate with High Content of Acidic Polysaccharide from White Tail Ginseng Marc)

  • 50% 에탄올 추출박으로부터 기존의 백삼농축액보다 산성다당체가 다량 함유된 백삼 농축액 제조 방법에 대해서 연구하였다. 50% 에탄올을 사용하여 제조한 백삼농축액의 추출 수율과 산성다당체 함량은 에탄올의 농도가 증가할수록 감소하였다. 반면에 백삼을 0∼90% 에탄올로 추출하고 남은 백삼박 내의 산성다당체 함량은 에탄올 농도가 증가할수록 증가하였다. 백삼박으로부터 산성다당체를 얻을 수 있는 최적의 조건은 $\alpha$-amylase를 390∼650 unit/g residue/15 mL 증류수의 농도로 4$0^{\circ}C$에서 5분 동안 처리하는 것으로 확인되었다. 50% 에탄을 추출 후 남은 백삼박에 $\alpha$-amylase를 처리함으로써 백삼박의 물추출액 중의 산성다당체 함량은 무처리구의 8.3%에서 10.5%로 증가하였다. 기존의 50% 에탄올 백삼추출농축액과 본 실험을 통하여 얻어진 백삼박 추출액을 8:2(w/w)로 혼합한 새로운 흔합백삼농축액을 제조한 결과 기존의 인삼농축액에 비해 조사포닌 함량은 비슷하며 약2배 정도의 산성다당체를 함유하고 색도가 짙은 농축액을 제조할 수 있었다.

인삼박으로부터 수용성 산성다당체의 추출 조건 분석 (Analysis of the Extraction Condition of Soluble Acidic Polysaccharides from Ginseng Marc)

  • This study was carried out to investigate the optimum conditions for extraction of soluble acidic polysaccharides from ginseng marc. Method of carbazole-sulfuric acid was applied to determine the amount of acidic polysaccharides in ginseng marc. The amounts of soluble acidic polysaccharides in water extract of ginseng marc were increased with increasing extraction temperature. The contents of acidic polysaccharides were not significantly different despite the extraction time increasing from 0.5 hours to 6 hours. To estimate the rehydration rate of the freeze dried polysaccharide, the extracted acidic polysaccharide fraction powder was determined the amount of soluble acidic polysaccharides by carbazole-sulfuric acid method again. The rehydration rate of acidic polysaccharides from water-extract of red ginseng marc at room temperature was 100%. On the other hand, the rehydration rate of acidic polysaccharide of red ginseng marc at boiling temperature was about 50%. The rehydration rate of acidic polysaccharides from water-extract of white ginseng marc at room temperature was 50%. The rehydration rate of acidic polysaccharide of red ginseng marc at boiling temperature was about 40%. The rate of soluble acidic polysaccharide of Red Ginseng is higher than that of White Ginseng. We can find out the maximum extraction method of soluble acidic polysaccharide from ginseng marc.

  • Melanogenesis is a physiological process resulting in the synthesis of melanin pigment. We investigated the inhibitory effect of crude polysaccharide of water extract of Artemisia iwayomgi Kitamura (AI) on melanin biosystheies in B16/F10 melanoma cells. Crude polysaccharide of water extract of Artemisia iwayomgi Kitamura significantly inhibited tyrosinase activity and melanin contents with or without $\alpha$-MSH in vivo. Melanin contents and tyrosinase activity were decreased in a dose-dependent manner,These results show that crude polysaccharide of water extract of Artemisia iwayomgi Kitamura could be developed as skin whitening components of cosmetics.

미생물 발효 영지버섯 추출액의 다당체에 관한 연구 (Studies on the Polysaccharide of Ganoderma lucidium Extract by Microorganism Fermentation)

  • In order to investigate the totale yield, the content of soluble polysaccharide and the others of FEW extract from the yeast fermentated Ganoderma lucidium by supersonic method, the yeast strain was inoculated after pretreatment and subsequently followed fermentation and supersonic extract. The main construction of the extract method is composed of the main glucose and together with the xylose, fucose, galactose and mannose. Results show that because of the generated lactic acid and ethanol, pH value of extract decreases and the safety as well as the preservation is improved. The extract yield, the total soluble polysaccharide, SOD-like activity, cytotoxic effect and growth inhibitory effect against cancer cell line are much higher in FE method than RE method, especially FEW3 extracts fermented during 24hrs. It is concluded that yeast fermentation makes the extract yield increase because of the cell disintegration, the useful ingredients of the germ body, the metabolic products, the insoluble ingredients due to the generation of ethanol, and the cell fragmentation caused by the supersonic waves vibration. Content of generated ethanol, total soluble polysaccharide and extract yield all increase during the fermentation time from 24 to 72 hours and the optimum fermentation condition is at $27^{\circ}C$ for 72 hours. The bitter taste and smell of the Ganoderma lucidium extract is diminished, fragrant-bitter taste and smell is generated so that the whole functional quality is improved.

홍삼박으로부터 산성다당체의 최적 추출 조건 분석 (Conditioning of the Extraction of Acidic Polysaccharide from Red Ginseng Marc)

The contents of starch in water-extract of red ginseng marc were increased with increasing extraction temperature. The yields of polysaccharide precipitated from water-extract of red ginseng marc were increased with increasing extraction temperature. The hydration rate of acidic polysaccharides and starch from water-extract of red ginseng marc were decreased with increasing extraction temperature. The contents of starch were not significantly different despite of the extraction time increasing from 6 hours to 48 hours at $8^{\circ}C$. However, the rehydration rate of acidic polysaccharide for 48 hours were decreased at $8^{\circ}C$. The rehydration rate of acidic polysaccharide and starch extracted from 6 hours to 24 hours at $25^{\circ}C$ were not significantly different, but those extracted for 48 hours were increased. From the above results, we suggest that by altering the extraction conditions in red ginseng marc it is possible to develop optimum conditions for extraction that modulate the proportions of acidic polysaccharide and starch.

인삼 및 영지추출물이 유산균 생육에 미치는 영향 (Effects of Panax ginseng and Ganoderma lucidum Extract on the Growth of Lactic Acid Bacteria)

  • This study was carried out to Investigate the effects of Panax ginseng, Ganoderma lucidum extract and crude polysaccharide of G. lucidum on the growth of lactic acid bacteria. p. ginseng extract contained 60.7% carbohydrate and 27.5% protein, whereas G. lucidum contained 35.9% carbohydrate and 46.3% protein. The total sugar and protein content of crude polysaccharide of G. lucidum were 47.2% and 15.2%, respectively. Two amino acids(hg, Trp) were detected in p. ginseng extract and 11 amino acids (hg, Trp, Ua, Lys, Ser, etc.) in C. lucidum extract. By the addition of p. ginseng, 5. lucidum extract and crude polysaccharide, the cia. p. ginseng was more effective on the growth of 1. casei an: G. lucidum was more effective on that of S. thermophilus. The effect of free amino acids on the growth of tactic acid bacteria was also examined. Arginine and lysine stimulated the growth of L. casei, whereas Lysine, serine, arginine, and glutamic acid stimulated the growth of 5. thermophilus.

Lentinus edodes 액체배양을 통한 세포외 다당체 생산 (Exo-Polysaccharide Production from Liquid Culture of Lentinus edodes)

  • Lentinus edodes의 액체배양을 통하여 균사체 및 polysaccharide의 최적 생산조건을 조사하였다. 플라스크 배양을 통하여 검토된 균체량 및 polysaccharede 생성을 위한 최적 배지의 조성은 glucose 60 g/L, yeast extract 10 g/L, $KH_2PO_4$ 2.0 g/L, $MgSO_4{\cdot}7H_2O$ 1.0 g/L이었다. 최대 균체량은 배양 10일째 11.01 g/L이었으며 최대 polysaccharide 생성량은 배양 6일째 1.64 g/L를 나타내었다. 생물반응기를 이용한 회분배양에서 균사체와 polysaccharide 생산량을 증가시키기 위하여 통기량을 조절하였다. 그 결과 통기량이 증가할수록 균체량과 polysaccharide의 생성량이 증가하였다. 결과적으로 통기량을 1.5 vvm으로 하여 배양하였을 때 배양 8일째 55.9 g/L의 균체량과 배양 7일째 7.34 g/L의 polysaccharide로 최대생성량을 얻을 수 있었다.

  • Polysaccharide fractions were prepared from Ginseng root, Mori Radicis Cortex (M. R. C. ), Phellodendri Cortex (Ph. C. ), Sappan Wood (S. W. ) and Tigli Semen (T. S.). Water extract was also prepared from the mixture of ph. C., S. W. and T. S. Ginseng polysaccharide and water extract of the mixture showed marked antitumor activity against sarcoma 180. Ginseng polysaccharide showed a mild increasing effect on the number of circulating leucocytes and a marked increasing effect on the number leucocytes and a marked increasing effect on the number of plaque forming cells (PEC). Polysaccharides from ginsing root, S. W., Ph. C. + T. S. and water extract of the mixture showed dramatic inducing activities of periotoneal exudate cells (PEC), polymorphonuclear leucocytes (PMN) and macrophages. These results suggest the possibility that water extract of the mixture may have the lentinan like effect and ginseng polysaccharide may have stimulating effects on the general immune system.

Exo-Polysaccharide Production in Liquid Culture of Pleurotus ferulae

  • Batch cultures were carried out to optimize the exo-polysaccharide production by liquid cultures of Pleurotus ferulae. Among the various carbon sources, when $5\%$ of glucose was used, the maximum mycelial growth and exo-polysaccharide concentration reached were 8.78 g/l and 3.59 g/l, respectively. Yeast extract and polypeptone were identified as the most suitable nitrogen sources. In particular, when a mixture of $1\%$ of polypeptone and $0.8\%$ of yeast extract was used, 9.52 g/l of mycelial growth and 4.09 g/l of exo-polysaccharide were obtained. In the case of mineral sources, K$_2$HPO$_4$ and MgSO$_4$$\codt$7H$_2$0 were found to be the best mineral sources for mycelial growth and exo-polysaccharide production. Under the optimized culture conditions, the agitation speed and aeration were investigated for mycelial growth and exo­polysaccharide production in a jar fermentor. The maximum mycelial growth and exo-polysaccharide concentration at 1.5 vvm and 200 rpm obtained were 13.2 g/l and 4.95 g/l, respectively, after 10 days of culture, which were $76\%$ and $79\%$ higher than those of the basal medium. The specific growth rate was decreased with the increase of mycelial growth. However, the specific production rate of the exo-polysaccharide was proportionally increased with the specific growth rate. The proposed model profiles showed good agreement with the experimental results for the mycelial growth and exo-polysaccharide production. The specific production rate using the optimized medium was higher than that of basal medium.
