Pullulan과 Polysaccharides를 이용한 천연화장품용 필 오프 팩의 제조 및 특성 (Manufacture and Characteristics of Peel-off Pack for Natural Cosmetics Using Pullulan and Polysaccharides)

  • 본 연구에서는 천연 및 자연주의 화장품 시장에 대응하여 폴리비닐알코올(polyvinyl alcohol, PVA)을 이용한 필 오프 팩을 대체할 수 있는 성분을 탐색하고자 하였다. 수용성 다당류인 풀루란(pullulan)과 기타 다당류(sodium hyaluronate, cellulose gum, hydroxyethyl cellulose, sodium alginate, corn starch)와의 조합을 통한 필 오프 팩을 제조하였으며, 각 필 오프 팩의 pH와 점도, 온도별 안정도를 확인하였다. 제조한 필름의 두께와 인장 강도를 측정하여 기존의 PVA 필 오프 팩과 비교하였으며, 피부에서의 도포성과 건조속도, 제거성을 확인하였다. 그 중 pullulan-sodium hyaluronate 필오프 팩은 5.12% 얇은 두께와 4.23% 높은 필름 인장강도로 PVA 필 오프 팩을 대체할 수 있는 우수한 필름성을 보였다. 실제 피부에 적용 시 팩의 퍼짐 정도와 균일하게 도포 가능한 사용성, 건조 후 제거 시 필름의 형성 및 제거 강도 평가 또한 PVA 필 오프 팩과 흡사한 수준을 보였다. 이에 pullulan-sodium hyaluronate 필름은 천연 필 오프 팩으로 PVA 필 오프 팩을 대체할 수 있을 정도의 물성을 보임을 확인하였다.

Gluconobacter oxydans 생물전환을 통한 쌀 가수분해물 유래 dextran 합성 (The synthesis of dextran from rice hydrolysates using Gluconobacter oxydans KACC 19357 bioconversion)

  • 식물에서 유래하는 전분과 달리 dextran은 미생물 기원으로 바이오폴리머로 glucose 단위체가 α-1,6 결합 형태로 연결된 다당류이며, 주로 젖산균에 의해 설탕으로부터 생산되는 것으로 알려져 있으나 dextran-dextrinase 활성을 갖는 초산균(G. oxydans)을 통해서도 얻을 수 있다. 쌀로부터 dextrin 기반의 dextran을 얻기 위해 쌀 가수분해물을 이용하여 초산균 발효를 통해 dextran을 생산하고 생성물의 특성을 연구하였다. 발효조를 이용한 dextrin 및 쌀 가수분해물 첨가 배지 배양 시, 초산균 배양 20시간 후 모두 OD600값 5 수준을 유지하였으며, 배양 72시간 후 상등액의 분자량구성을 GPC 분석한 결과 배지 내 기질의 구성과 달리 DP 480 및 400 수준의 고분자 물질이 생성된 것을 확인하였다. 다당류의 glucose 결합패턴을 1H-NMR로 확인한 결과 α-1,4:1,6 결합비율이 각각 1:2.37 및 1:4.4로 증가하여 쌀 가수분해물을 이루는 주요 결합인 α-1,4 결합이 α-1,6 결합 물질로 전환된 것으로 확인되었고, rat 유래 alpha-glucosidase 소화효소 처리결과 glucose가 천천히 방출되는 것으로 나타났다. 이를 통해 쌀 가수분해물은 당전이 활성이 있는 초산균 발효를 통해서 미생물 유래의 dextran으로 전환시켜 수용성 식이섬유 소재와 같이 소화를 지연시키는 고부가 생물소재로 제조할 수 있음을 시사하였다.

  • Bone marrow cell proliferating arabinogalactan-like polysaccharide (ALR-3IIa-1-1) has been purified from rhizomes of Atractylodes lancea DC. In order to characterize the essential structure of ALR-3IIa-1-1 for expression of the activity, sequential enzymatic digestion using ego-$\alpha$-L-arabinofurasidase (AFase) and ego-$\beta$-D-(1longrightarrow3)-galactanase (GNase) was employed. After ALR-3IIa-1-1 was digested with the AFase, the GNase digestion cleaved only 10% and 23% of 3-linked and 3,6-branched galactose, respectively, from arabinose-trimmed ALR-3IIa-1-1 (AT-ALR-3IIa-1-1), and gave small amounts of intermediate size (AT-G-2) and shorter oligosaccharides (AT-G-3) fractions in addition to a large amount of the GNase resistant fraction (AT-G-1). When AT-G-1 was redigested gradually with the AFase and GNase, it released trace amounts of oligosaccharides in addition to a large amount of the resistant fraction. When the final enzyme-resistant fraction from AT-G-1 was digested simultaneously with both AFase and GNase, the resistant fraction was significantly degraded into two long fragments (3AT-3G-1 and 2). The mixture of digestion products from the first GNase digestion of AT-ALR-3IIa-1-1 showed a significantly decreased bone marrow cell proliferation activity to about 30% of the activity of ALR-3IIa-1-1, but the GNase resistant fraction (AT-7-1) still had significant activity. Although the second gradual enzymatic digestion of AT-G-1 showed a marginal decrease in activity, the resulting fragments (3AT-3G-1 and 2) by the final simultaneous enzymatic digestion lost most of the activity. Component sugar, methylation and FAB-MS analyses indicated that the digestion products (AT-G-21 AT-G-31 2AT-2G-2 and 2AT-2G-3) released from AT-ALR-3IIa-1-1 by the sequential enzymatic digestion contained galactose-containing oligosaccharides mainly comprising 6-linked galactose, that some of which were partially arabinosylated, and these oligosaccharides were attached to $\beta$-D-(1longrightarrow3)-galactan backbone in its non-reducing terminal side as side chains.

Ganoderma lucidum IY009 균사체로부터 추출된 ganoderan의 항암 및 항보체 활성에 미치는 탄소원의 영향 (The effects of carbon sources on antitumor and anticomplementary activities of Ganoderan extracted from the mycelium of Ganoderma lucidum IY009)

  • 영지 균사체로부터 종양 면역 활성이 우수한 다당류를 얻기 위하여 여러 탄소원을 이용하여 균사체를 배양하였다. 배앙된 균사체로부터 ganoderan을 추출하고 이들의 조성과 항암 및 항보체 활성능을 알아보았다. 여러 탄소원은 영지 균사체에 의해 기질로 이용될 수 있으나 ${\beta}-glucan$성 다당류인 ganoderan의 생성에는 상당한 차이를 보였다. 균사체 생육이 우수한 탄소원에서 배양된 균사체의 경우, 비수용성(WI) 다당류의 생성이 증가하였고, 생육이 저하된 탄소원에서는 수용성(WS) ganoderan이 상대적으로 증가하였으며, 포도당을 탄소원으로 사용할 경우 약 $5.2{\sim}16%$가 알칼리 가용성 ganoderan으로 도입되었다. 탄소원에 따라 수획된 ganoderan은 포도당을 주 구성당으로 이루어진 ${\beta}-glucan$성 다당류이며 만노오스, 과당 등이 포함되어 있었으나, 비수용성 ganoderan의 경우 수용성보다 포도당의 함량이 약 $20{\sim}40%$ 더 많이 포함되었으며 약 16개의 아미노산으로 구성된 소량의 단백질이 결합되어 있었다. WI은 glucose가 주 구성당이었으나 WS는 여러 단당들이 상대적으로 많이 포함되어 있는 heteroglucan의 성격을 보였다. 탄소원에 따라 배양된 균사체의 알칼리 추출된 ganoderan 가운데 항암 및 항 보체 활성이 가장 우수한 것은 갈락토오스를 탄소원으로 이용한 ganoderan이었으며 항보체 활성(ITCH 50,%)치가 35.7%였고, 특히 균사체의 생장이 낮은 탄소원에서 수획한 ganoderan이 항보체 활성이 높게 나타났다. Sarcoma 180이 피하이식된 마우스에 20 mg/kg씩 복강 투여하여 항암 활성을 검토한 결과, 항보체 활성실험에서와 마찬가지로 갈락토오스를 탄소원으로 이용한 ganoderan-GAL이 83.6%의 암 저지율을 보여 항암 활성이 가장 높았으며, 이는 약 5%의 단백질이 결합된 ${\beta}-glucan$성 다당류로 55%의 포도당, 13%의 만노오스, 12%의 과당 및 소량의 galactose, xylose, ribose등으로 구성된 분자량 30 kD의 다당류이다.

Characteristic study on the chemical components of Korean curved ginseng products

  • Dried ginseng (DG) is in fact the representing ginseng product in the worldwide market. Although it is made in various packages depending on the processing method, size and age of DG, basic scientific data reporting the chemical components are limited. In this study, 4-year-old curved ginseng (CG), one of the domestic DG products, was selected for further investigation. Eighty-six samples of 30 and 50 piece-grade CG, which are the most widely distributed in the market, were collected for 5 yr. Their major components, such as moisture, total sugar, acidic polysaccharides, total phenolic compounds, and saponins, were analyzed to figure out the standard quality characteristics. The moisture content of all CG samples was less than 15%. The total water-soluble sugar contents were 22.9% to 47.8% and 23.2% to 49.5% in the 30 and 50 piece-grade CG, respectively. The acidic polysaccharide contents were 3.6% to 6.7% and 2.9% to 6.9% in the 30 and 50 piece-grade CG, respectively. The total phenolic compound content was 0.4% to 0.5% in CG, regardless of the piece-grade. The crude saponin content, which represents the active component of ginseng, was over 2% in all samples. In 30 piece-grade CG samples, the contents of major ginsenosides, Rb1, Rf, and Rg1, were 2.2 to 4.7 mg/g, 0.4 to 1.3 mg/g, and 1.6 to 4.0 mg/g, respectively. The ginsenoside contents in 50 piece-grade CG samples were 2.1 to 3.9 mg/g (Rb1), 0.5 to 1.2 mg/g (Rf), and 1.3 to 3.4 mg/g (Rg1). Overall, since there were relatively high standard deviation and coefficient of variation in all the chemical component contents that were assessed, we found some difficulties in showing the CG standard chemical component characteristics by average, standard deviation, and other statistical analysis factors.

감잎 유래 다당체가 면역억제 마우스의 면역조절에 미치는 영향 (Immunomodulatory Effects of Polysaccharides Derived from Persimmon Leaves on Cyclophosphamide-induced Immunosuppressed Mice)

  • 감잎으로부터 새로운 생리활성 다당 소재를 개발하기 위하여 감잎의 열수추출물로부터 다당획분 PLW-0, pectinase 처리 추출물로부터 다당획분 PLE-0를 분리하여 정상 및 면역 억제 동물모델에서 면역자극활성을 측정하였다. 감잎 유래 다당체들은 정상모델에서 마우스의 면역기관인 비장과 흉선의 무게를 증가시켰으며, 비장세포 유래 림프구의 증식에 농도 의존적으로 증식활성을 보였는데, pectinase 처리 다당 획분 PLE-0의 활성이 열수추출 다당 획분 PLW-0에 비해 전체적으로 높은 활성을 보이는 것을 확인할 수 있었다. 또한 CY에 의해 면역이 억제된 동물모델에서도 CY처리에 의해 저하된 임파구 증식, NK 세포 활성 및 백혈구 수를 현저하게 증가시켰다. 이러한 면역억제 극복 활성 역시 유의적인 차이는 보이지 않았으나 PLE-0가 PLW-0에 비해 높음을 확인할 수 있었다. CY는 악성 종양의 치료 및 장기이식 후 나타나는 거부반응의 억제제로 사용되고 있는 대표적인 약물로 알려져 있다. 그러나 CY의 비선택적인 독성으로 인해 암세포뿐만 아니라 정상세포에도 독성을 나타내어 백혈구 수 감소, 혈소판 감소, 빈혈 등과 같은 부작용을 나타내고 있다. 따라서 최근에는 이와 같은 부작용이나 면역독성을 최소화 할 수 있는 물질을 발굴하고자 하는 연구가 이루어지고 있는데, 본 실험의 결과로부터 감잎 유래 다당체는 정상모델에서의 면역자극 및 CY에 의해 초래된 면역억제를 감소시킴으로써 면역조절제로서의 개발가능성을 제시해 줄 수 있다고 생각된다. 또한 pectinase 처리 다당 PLE-0가 열수추출 다당 PLW-0에 비해 비교적 높은 활성을 보인 것을 감안해 보았을 때 효소처리가 감잎 유래 다당의 활성증가에 도움을 준다는 것을 확인하였다.

한약재 추출물의 Streptococcus mutans에 대한 항균효과 (Antimicrobial Activity of Streptococcus mutans by Herbal Medicine Extracts)

  • 본 연구에서는 국내에서 유통되는 63종의 한약재 열수 추출물의 S. mutans에 대한 항균효과를 paper disc method로 검색하여, 항균효과가 우수한 지부자, 황금의 S. mutans 생장에 관한 영향을 평가하였다. 1. 63종의 한국산 한약재 열수 추출물의 향균효과를 탐색한 결과 S. mutans에 대한 향균력이 우수한 순서는 지부자, 황금순이었다. Paper disc method에 의한 항균활성 측정(20mg/ml 투여)결과는 지부자가 6.3mm, 황금이 3.5mm이었다. 2. 한약재 추출물의 S. mutans 생장에 대한 효과는 대조군이 8시간에서 1.05로 최대 생장률을 보였으나, 지부자, 황금 추출물 2mg/ml을 투여한 배지에서는 16시간 만에 0.91, 0.87로 최대 생장을 보였다. 3. S. mutans를 8시간 동안 배양한 후 배지내 pH를 측정한 결과 대조군은 5.08 이었으며, 지부자, 황금 추출물이 투여된 배지는 5.80, 5.74이었다. 4. S. mutans를 8시간 동안 배양한 후 배지내 탄수화물 변화량을 측정한 결과 대조군은 0.81mg/ml이었으며, 지부자, 황금 추출물이 투여된 배지는 1.70mg/ml, 1.84mg/ml 이었다. 5. S. mutans를 8시간 동안 배양한 후 배지내 단백질 변화량을 측정한 결과 대조군은 8.39mg/ml 이었으며, 지부자, 황금 추출물이 투여된 배지는 9.52mg/ml, 9.28mg/ml 이었다. 6. S. mutans의 다당류 생성에 미치는 효과는 대조군의 경우 배양 8시간에 300mg/100ml을 생성하였으나, 지부자, 황금 추출물이 투여된 배지에서는 200mg/100ml, 220mg/100ml을 생성하였다.

Effects of Nutrient Specifications and Xylanase Plus Phytase Supplementation of Wheat-based Diets on Growth Performance and Carcass Traits of Broiler Chicks

  • The simultaneous addition of xylanase (5,600 EXU/kg) and phytase (500 FTU/kg) feed enzymes to wheat-based broiler diets was investigated. Starter, grower and finisher diets, with three tiers of nutrient specifications, were fed to 1,440 broiler chicks kept on deep litter from 1-42 days post-hatch, without and with xylanase plus phytase, to determine the effects of diet type and enzyme supplementation on growth performance. The nutrient specifications of type A diets were standard; energy density and protein/amino acid levels were reduced on a least-cost basis to formulate type B diets and further reduced to type C diets. Phosphorus (P) and calcium (Ca) levels were adjusted in supplemented diets. From 1-42 days post-hatch, diet type significantly influenced growth performance. Birds on type C diets had lower growth rates (2,429 vs. 2,631 g/bird; p<0.001), higher feed intakes (4,753 vs. 4,534 g/bird; p<0.005) and less efficient feed conversion (1.96 vs. 1.72; p<0.001) than birds offered type A diets. Enzyme supplementation increased growth rates by 3.2% (2,580 vs. 2,501 g/bird; p<0.005) and improved feed efficiency by 2.7% (1.80 vs. 1.85; p<0.05) over the entire feeding period. There were no interactions between diet type and enzyme supplementation. At 21 days, 5 out of 30 birds per pen were transferred to cages to ascertain treatment effect on apparent metabolisable energy (AME) and nitrogen (N) retention. Xylanase plus phytase enhanced AME (13.48 to 13.91 MJ/kg DM; p<0.001) and N retention (56.3 to 59.7%; p<0.005). Carcass and breast weights of the caged birds were determined following commercial processing. Diet type significantly influenced breast weight, carcass weight and yield. Birds offered Type A diets, in comparison to Type C diets, supported heavier breast (467 vs. 424 g; p<0.001) and carcass weights (1,868 vs. 1,699 g; p<0.001) with superior carcass yields (71.8 vs. 70.6%; p<0.005). Enzyme addition increased carcass weight by 3.9% (1,752 vs. 1,821 g; p<0.005) and breast weight by 5.8% (431 vs. 456 g; p<0.01) without influencing yields. Feed ingredient costs per kg live weight gain and per kg carcass weight indicated that enzyme addition was economically feasible, where supplementation of Type A diets generated the most effective results. Importantly, soluble and total non-starch polysaccharide and phytate contents of the wheat used were typical by local standards. This study confirms the potential of supplementing wheat-based broiler diets with xylanase plus phytase but further investigations are required to define the most appropriate inclusion rates and dietary nutrient specifications in this context.

Effects of Enzyme Addition to Broiler Diets Containing Varying Levels of Double Zero Rapeseed Meal

  • Maize-soybean meal diets with 0, 100, 200 and 300 g/kg double zero rapeseed meal ('00' RSM) with and without an enzyme mixture (xylanase, pectinase, cellulase) at a level of 1.6 g/kg were evaluated with 624 day-old broiler chicks for 5 weeks. The birds were randomly allocated to eight dietary treatments with three replicates of 26 birds each. Average daily gain (ADG) and feed intake (FI) were recorded weekly and ileal viscosity, organ weights, serum enzyme activity, hormonal profile and hematological parameters were measured at the end of week 5. Average daily gain during the weekly periods was significantly influenced by the dietary level of '00'RSM (p<0.01). Inclusion of '00' RSM improved the ADG up to day 28 with the increased level; beyond that time no improvement was recorded when compared to control groups. However, ADG from 1-35 days was significantly different between 300 g/kg inclusion level of '00' RSM and the control diet. Inconsistent decline in feed intake and feed conversion ratio was observed up to day 21 and the trend was reversed thereafter. The proportion of '00' RSM in the diet had a significant ($p{\leq}0.05$) influence on thyroid weight but had no effect on the relative weights of liver and heart, serum enzyme activities (${\gamma}$-glutamyl transferase, alanine amino transferase and aspartate amino transferase), thyroid hormones ($T_3$ and $T_4$), hemoglobin level and hematocrit. Significant improvement in ADG was recorded during the 2nd week of age with the addition of enzyme, whereas for all other periods, including the whole period of the trial, higher but non-significant ADG was observed. FI and FCR were not affected by the addition of enzyme but there was a numerical reduction in FCR during the whole period. The addition of enzyme reduced the ileal viscosity at all levels of '00' RSM inclusion. The results suggest that '00' RSM can be included up to 300 g/kg in broiler diets without any adverse effects on health and performance. The addition of commercial enzyme mixture containing xylanase, pectinase, cellulase to broiler diets containing '00'RSM has some effect on growth rate and feed conversion efficiency.

Effects of Post-harvest Storage Duration and Variety on Nutrient Digestibility and Energy Content Wheat in Finishing Pigs

  • This study was conducted to investigate the effects of post-harvest storage duration and wheat variety on the digestibility and energy content of new season wheat fed to finishing pigs. Two wheat varieties (Shi and Zhong) were harvested in 2013 and stored in the warehouse of the Fengning Pig Experimental Base at China Agricultural University for 3, 6, 9, or 12 mo. For each storage period, 12 barrows were placed in metabolism crates and allotted to diets containing 1 of the 2 wheat varieties in a randomized complete block design. The experimental diets contained 97.34% wheat and 2.66% of a vitamin and trace mineral premix. With an extension of storage duration from 3 mo to 12 mo, the gross energy (GE) and crude protein (CP) of the wheat decreased by 2.0% and 12.01%, respectively, while the concentration of neutral detergent fiber (NDF), acid detergent fiber (ADF) and starch content increased by 30.26%, 19.08%, and 2.46%, respectively. Total non-starch polysaccharide, total arabinose, total xylose and total mannose contents decreased by 46.27%, 45.80%, 41.71%, and 75.66%, respectively. However, there were no significant differences in the chemical composition between the two wheat varieties with the exception of ADF which was approximately 13.37% lower in Shi. With an extension of storage duration from 3 mo to 12 mo, the digestible energy (DE), metabolizable energy (ME) content and the apparent total tract digestibility of GE, CP, dry matter, organic matter, ether extract, ADF and metabolizability of energy in wheat decreased linearly (p<0.01) by 5.74%, 7.60%, 3.75%, 3.88%, 3.50%, 2.47%, 26.22%, 27.62%, and 3.94%, respectively. But the digestibility of NDF changed quadratically (p<0.01). There was an interaction between wheat variety and storage time for CP digestibility (p<0.05), such that the CP digestibility of variety Zhong was stable during 9 mo of storage, while the CP digestibility of variety Shi decreased (p<0.05). In conclusion, the GE, DE, and ME of wheat was stable during the first 3 to 6 mo of post-harvest storage, and decreased during the following 6 to 12 mo of storage under the conditions of this study.
