생체적합성 및 생분해성의 장점을 지닌 PLGA는 우수한 기계적 성질과 분해속도를 조절할 수 있는 장점을 가지고 있지만 소수성으로 인해 세포의 초기부착률이 낮고, 지지체 내부에 영양액의 공급이 원활하지 않다. 본 연구에서는 이러한 단점을 보완하고자 탈미네랄화된 골분(DBP, demineralized bone particle) 젤을 PLGA 지지체에 함침시켰다. 다양한 형태의 지지체 압축강도를 측정한 결과 PLGA/DBP 지지체가 연골과 비슷한 강도를 가지고 있었으며, 이를 바탕으로 DBP젤을 함침시킨 PLGA/DBP젤 지지체를 제작하였다. 연골세포를 파종하여 in vivo 실험을 진행하였고 조직화 정도를 확인하기 위해 H&E, Safranin-O, Alcian blue, Collagen type I 및 Collagen type II 염색을 실시하였다. 그 결과 DBP를 함침시킨 PLGA 지지체에서 GAG, Collagen type I 및 Collagen type II의 높은 발현과 조직화를 보여 연골조직으로 대체할 수 있는 가능성을 확인하였다.
The achievement of tissue engineering can be highly depending on the capability to generate complicated, cell seeded three dimensional (3D) micro/nano-structures. So, various fabrication techniques that can be used to precisely design the architecture and topography of scaffolding materials will signify a key aspect of multi-functional tissue engineering. Previous methods for obtaining scaffolds based on top-down are often not satisfactory to produce complex micro/nano-structures due to the lack of control on scaffold architecture, porosity, and cellular interactions. However, a bioprinting method can be used to design sophisticated 3D tissue scaffolds that can be engineered to mimic the tissue architecture using computer aided approach. Also, in recent, the method has been modified and optimized to fabricate scaffolds using various natural biopolymers (collagen, alginate, and chitosan etc.). Variation of the topological structure and polymer concentration allowed tailoring the physical and biological properties of the scaffolds. In this presentation, the 3D bioprinting supplemented with a newly designed plasma treatment for attaining highly bioactive and functional scaffolds for tissue engineering applications will be introduced. Moreover, various in vivo and in vitro results will show that the fabricated scaffolds can carry out their structural and biological functionality.
젤란검은 천연재료로써 내열성, 내산성, 내효소성 등이 우수하여 용도가 광범위하나 기계적 강도가 약하다는 단점이 있다. 따라서 본 연구에서는 기계적 성질을 개선하고자 젤란검 스폰지에 PLGA 미립구를 혼합하였다. PLGA 미립구의 다양한 함량의 젤란검 스폰지는 기계적 강도를 알아보고자 압축강도를 측정하였고, MTT 분석, SEM, 생체활성조직학적 평가 및 RT-PCR을 통해 세포의 증식 및 ECM 분비 효과를 확인하였다. 그 결과, PLGA 미립구가 50% 함유된 젤란검 지지체에서 섬유륜세포의 꾸준한 증식과 세포외기질 분비가 우수한 것을 확인할 수 있었다. 이 연구를 통하여 PLGA 미립구가 함유된 젤란검이 디스크조직 재건을 위한 지지체로서 적합함을 알 수 있었으며, 젤란검의 다양한 응용가능성을 제시하였다.
단백질 및 펩타이드의 서방형 약물전달체로서 소장점막하조직(SIS)으로 개질된 PLGA 담체를 제조하고자 하였으며, SIS/PLGA 담체는 용매 캐스팅/염 추출법에 의해 준비된 PLGA 담체에 SIS 용액을 첨가하여 단순 함침방법으로 제조하였다. 본 실험에서 사용된 돼지의 소장 점막층에서 유래된 SIS는 면역거부반응이 적어 생체재료로 널리 사용되고 있다. 제조된 PLGA 및 SIS/PLGA 담체를 SEM을 통한 표면 및 내부 관찰결과 두 담체 모두 열린 다공구조를 이루며, 특히 SIS/PLGA 담체는 PLGA 담체의 다공 내부에 SIS가 침투되어 작은 네트워크를 형성하고 있음을 확인하였다. 또한 단백질의 방출경향을 확인하기 위하여 형광이 결합된 소 혈청 알부민(FITC-BSA)을 PLGA 및 SIS/PLGA 담체에 담지시킨 후, 형광광도계를 통해 이들의 방출거동을 확인하였다. PLGA 담체와 비교할 때 SIS/PLGA 담체에서의 BSA의 방출은 초기방출량이 적고 지속적으로 일정량이 방출되는 거동을 확인할 수 있었으며 함량별 BSA 농도에 따른 SIS/PLGA 담체에서의 방출은 BSA의 양이 증가할수록 빠르고 많은 양이 방출되는 경향성 있는 방출패턴을 보임을 확인하였다. 결론적으로 PLGA 담체에 침투한 SIS 젤이 BSA의 급격한 초기방출을 억제하며, SIS로 개질된 PLGA 담체는 방출조절이 가능한 약물전달체로서 매우 유용할 것으로 사료된다.
Conventional methods for fabricating three-dimensional (3-D) scaffolds have substantial limitations. In this paper, we present 3-D scaffolds that can be made repeatedly with the same dimensions using a microstereolithography system. This system allows the fabrication of a pre-designed internal structure, such as pore size and porosity, by stacking photopolymerized materials. The scaffolds must be manufactured in a material that is biocompatible and biodegradable. In this regard, we synthesized liquid photocurable biodegradable TMC/TMP, followed by acrylation at terminal ends. And also, solidification properties of TMC/TMP polymer are to be obtained through experiments. Cell adhesion to scaffolds significantly affects tissue regeneration. As a typical example, we seeded chondrocytes on two types of 3-D scaffold and compared the adhesion results. Based on these results, the scaffold geometry is one of the most important factors in chondrocyte adhesion. These 3-D scaffolds could be key factors for studying cell behavior in complex environments and eventually lead to the optimum design of scaffolds for the regeneration of various tissues, such as cartilage and bone.
Clinically-favored materials for bone regeneration are mainly based on bioceramics due to their chemical similarity to the mineral phase of bone. A successful scaffold in bone regeneration should have a 3D interconnected pore structure with the proper biodegradability, biocompatibility, bioactivity, and mechanical property. The pore architecture and mechanical properties mainly dependent on the fabrication process. Bioceramics scaffolds are fabricated by polymer sponge method, freeze drying, and melt molding process in general. However, these typical processes have some shortcomings in both the structure and interconnectivity of pores and in controlling the mechanical stability. To overcome this limitation, the rapid prototyping (RP) technique have newly proposed. Researchers have suggested RP system in fabricating bioceramics scaffolds for bone tissue regeneration using selective laser sintering, powder printing with an organic binder to form green bodies prior to sintering. Meanwhile, sintering process in high temperature leads to bad cost performance, unexpected crystallization, unstable mechanical property, and low bio-functional performance. The development of RP process without high thermal treatment is especially important to enhance biofunctional performance of scaffold. The purpose of this study is development of new process to fabricate ceramic scaffold at room temperature. The structural properties of the scaffolds were analyzed by XRD, FE-SEM and TEM studies. The biological performance of the scaffolds was also evaluated by monitoring the cellular activity.
조직 형성을 위한 생리학적 환경을 제공하는 새로운 생활성 지지체의 디자인은 생체재료 연구에서 중요한 분야이다. 본 연구에서는 3차원적 실크/PLGA 지지체의 물성평가를 위해 압축강도, 수분친화도, SEM 분석을 하였으며, 세포친화성 평가를 위해 RAW 264.7과 NIH/3T3의 부착, 증식 및 표현형유지와 염증반응에 미치는 영향을 조사하였다. 지지체는 용매 캐스팅/염 추출법으로 제조하였고, 압축강도, 수분친화도 면에서 실크 함량이 20 wt%에서 우수함 확인했으며, 표면의 거침도활 높여 세포부착에 긍정적인 구조임을 확인하였다. 세포친화성 분석 결과 실크함량이 20 wt%인 실크/PLGA 지지체에서 높은 초기부작도 및 증식률을 보였으며, 실크함량이 20 wt%에서 염증반응이 눈에 띄게 감소함을 확인하였다. 조직공학적 응용에 실크/PLGA 지지체가 유용할 것이라 판단하였다.
본 연구에서는 세포 배양시 감염방지용으로 널리 쓰이는 겐타마이신 설페이트(GS)가 서방화된 다공성 PLLA 지지체를 유화동결건조방법으로 제조하였다 이들의 물성을 전자현미경 및 수은다공도계로 특성결정하였고, 적심성은 푸른염색 수용액으로 관찰하였으며, 방출거동은 high performance liquid chromatography (HPLC)를 이용하여 측정하였다. GS가 5, 10 및 20%가 포접된 PLLA 지지체의 다공도는 80~90%이었으며 평균다공크기 범위는 30~57 ${\mu}{\textrm}{m}$ 그리고 가장 큰 것으로는 150${\mu}{\textrm}{m}$이상의 것도 관찰되었다. 전체적인 다공도의 모양은 다공과 다공사이의 연결이 양호하고 대부분이 열린 셀 구조를 하고 있는 것으로 나타났다. 대조군에 비해 GS의 농도가 증가함에 따라 PLLA 지지체의 다공도가 감소하는 것으로 보아 GS내의 설페이트 부분은 친수성 역할 그리고 겐타마이신 부분은 소수성 역할을 수행하는 계면활성제의 역할에 기인한 것으로 사료된다. PLLA 지지체의 방출거동은 GS의 농도가 증가함에 따라 방출되는 양이 증가하였고, 적심성 또한 향상되어 세포배양시 긍정적 효과를 끼칠 것으로 예상되었다.
늑연골세포는 초기 증식능력이 관절연골세포보다 우수하며, 또한 세포 채취에 용이하다는 장점이 있어, 관절연골의 대안으로 사용된다. FDA의 승인을 받은 합성고분자인 폴리락산-글리콜산 공중합체(PLGA)는 조직공학적 생체재료로 사용되고 있으나, 세포 부착률이 낮고, 염증반응을 야기시킨다고 보고되고 있다. 본 연구에서는 세포의 증식에 영향을 주며 염증반응이 감소된다고 보고된 탈미네랄골분(DBP)을 이용하여, 함량별 DBP/PLGA를 제조한 뒤, 세포를 파종하여 연구를 실시하였다. 세포의 부착 및 증식률을 측정하기 위하여 세포 파종한 후 MTT와 SEM 분석을 수행하였으며, DBP가 세포외 기질 형성에 미치는 영향을 확인하고자 글리코스아미노글라이칸(sGAG)과 콜라겐 함량을 측정하였다. 생체 내 외 환경에서 세포의 부착과 증식에 미치는 영향을 관찰하기 위해 연구를 진행하여, PLGA 지지체보다 DBP/PLGA 지지체가 세포의 성장과 증식에 영향을 주는 것을 확인하였으며, DBP의 함량이 증가할수록 효과가 좋은 것으로 나타났다.
