Expression of Human Thrombopoietin in Insect Cells by Polyhedrin-gp64 Dual Promoter-Based Baculovirus Vector System

  • A new baculovirus transfer vector (pPGP404) was constructed to increase the expression level of human thrombopoietin (hTPO) in insect cells. In pPGP404, hTPO was cloned next to the AcNPV polyhedrin-gp64 dual promoter and the leader sequence of hTPO was substituted with that of gp64. A recombinant baculovirus, AcPGP404, was constructed by using pPGP404 as a transfer vector. hTPO was expressed in AcPGP404-infected TN5 cells and it was observed that the expression levels of hTPO in TN5 cells increased three-fold ($6.0 {\mu}g/ml^{-1}$) compared to the level expressed under the control of the polyhedrin single promoter. These results indicate that the polyhedrin-gp64 dual promoter system would be useful for expression in large quantities of recombinant proteins in insect cells.

  • H. cunea nuclear polyhedrosis virus (HcNPV) 의 polyhedrin 아미노산 및 polyhedrin gene 의 염기서열을 분석하였다. Polyhedrin 은 SDS-PAGE 상에서 3개의 polypeptide band 가 나타났고 주요 polypeptide 는 약 25 Kd 의 분자량을 갖고 있었다. 또한 polyhedrin 은 17 개의 다른 아미노산으로 구성되어 있었다. HcNPV DNA를 EcoRI 효소로 절단하여 $\alpha^{32}P$로 labelling 된 Autographa californica (AcNPV) polyhedrin gene cDNA 의 probe DNA를 이용하여 hybridization 한 결과 polyhedrin gene은 EcoRI 절편들중 H 절편에 양성반응을 나타냈다. 또한 polyhedrin gene 을 포함하고 있는 EcoRI-H 절편을 pUC8 벡터에 cloning한 다음 이를 hPE-H라고 이름하였다. HcNPV genome DNA 의 promoter 부위를 sequence한 결과 TATA box의 염기배열은 polyhedrin gene 전사 개시위치로부터 위쪽으로 -79 bp 의 5' flanking 부위에서 발견되었다. polyhedrin gene 내 CAAT box는 TATA box 측면 염기 배열에서 나타나지 않았고, 4개의 tandem repeat 5'-CTAATAT-3' 와 5'-TAAATAA-3'의 염기는 polyhedrin gene내 전이 개시 위치로 부터 위쪽으로 -141 과 -108 bp 또는 -83 bp 부위에 존재하였으며, 다른 하나는 전이 개시위치로 부터 아래쪽으로 -52 bp 부위에서 발견되었다. 그리고 polyhedrin gene 내 전이 개시위치로 부터 위쪽으로 -141 bp 부위는 다량의 AT (78%) 염기가 존재하였다. 또한 polyhedrin 의 개시 coding region 은 ATG 였고 종결 coding region은 TAA 였다.

Analysis of Promoter Strength of Autographa californica Nuclear Polyhedrosis Virus IE1 Gene by Using Rreconmbinant Baculovirus

  • To analysis a promoter strength of Atographa californica nucler polyhedrosis virus (AcNPV) IE1 gene, an immediate viral gene, ${\beta}$-glactosidase gene as a reporter gene was introduced under the control of the IE1 promoter. The restriction fragment containing IE1 promoter and ${\beta}$-galctosidase gene from pAcIE1-gal were inserter into pBacPAK9 to yield transfer vector pAcNPV-IE1-gal. The pAcNPV-IE1-gal was cotransfected with AcNPV genomic DNA BacPAK6 into Sf9 cells to produce recombinant baculovirus AcNPV-IE1-gal. In addition, recombinant bacvulovirus AcNPV-gal, which express ${\beta}$-galac-tosidase under the control of the polyhedrin promoter, was constrer, was constructed to compared with AcNPV-IE1-gal. The recombinant viruses were respectively infected into Sf9 cells and characterized by the virus titer and expression of ${\beta}$-galactoxidase in Sf9 cells. The promoter strength of IE1 and polyhedrin promoters was determined by the amount of ${\beta}$-galactosidase secreted into medium by viral infection. The titer of AcNPV-IE1-Gal determined by plaque assays in Sf9 cells was similar to that of AcNPV-gal. However, expression level of ${\beta}$-galactosidase by AcNPV-IE1-gal was significantly lower than that by AcNPV-gal. In conclusion, promoter strength of IE1 was approximately 25-fold lower than that of polyhedrin.

누에 핵다각체병 바이러스를 이용한 새로운 전이 벡터의 제작 (Construction of New Transfer Vector of Nuclear Polyhedrosis Virus of the Silkworm, Bombyx mori)

  • 국내 분리주의 BmNPV를 이용하여 유용 단백질을 생산할 수 있는 새로운 전이벡터의 제작과 외내 유전자의 발현에 대한 연구결과는 다음과 같다. 1. PCR 기법에 의하여 다각체 단백질 유전자의 +1 - -194에 해당하는 promoter 부위를 증폭하고 클로닝 하였다. 2. 다각체 단백질 유전子의 5' 및 3'의 leader 부위를 promoter 부위와 함께 단계적으로 클로닝하여 전이벡터를 완성하였다. 3. 완성된 전이벡터는 pBmKSK1으로 명명하였으며, 염기서열 결정을 통하여 다각체 단백질 유전자의 +2 부터 +597까지 제거되고 외내 유전자의 클로닝 sites로 EcoRI, KpnI과 SacI을 가짐을 확인하였다. 4. 외래 유전자로서 E. coli의 $\beta$-galactosidase 유전자를 pBmKSKl에 클로닝하고 누에 세포주에 전이시킨 후, X-gal 염색 및 SDS-PAGE에 의하여 pB- mKSKl이 제 기능을 수행함을 확인하였다.

Expression of Bombyx mori Nucleopolyhedrovirus ORF4 under the Control of BaculoviruS Ie1 Promoter by a Novel Bac-to-Bac/BmNPV Baculovirus Expression System

  • Open reading frame 4 of Bombyx mori nucleopolyhedrovirus (BmNPV), designated as Bm4, is a gene whose function is completely unknown. With the recently developed BmNPV bacmid and a modified pFastBac1 whose polyhedrin promoter was replaced with ie1 promoter, a recombinant bacmid expressing Bm4-EGFP fusion protein under the control of ie1 promoter in BmN cells was successfully constructed. The result not only showed that the polyhedrin promoter can be replaced efficiently with other promoters to direct the expression of foreign gene in BmN cells by using Bac-to-Bac/BmNPV baculovirus expression system but also laid the foundation for rescue experiment of Bm4 deletion mutant due to the ability of ie1 promoter to direct gene expression throughout the infection cycle.

Comparison Promoter Activity of the p10 Gene between Bombyx mori Nucleopolyhedrovirus Variants

  • To compare the p10 promoter activity of Bombyx mori nucleopolyhedrovirus (BmNPV)K1 and K4, recombinant viruses Bm101-LacZ and Bm104-LacZ with a lacZ gene under the control of each p10 promoter were constructed. The $\beta$-galactosidase activity due to Bm101-LacZ was about 5.5- and 1.1-fold higher than that due to Bm104-LacZ and BmK1-LacZ, respectively. expressing ${\beta}$-galactosidase under the control of a polyhedrin promoter. The recombinant virus BmK1-104LacZ with the same genome structure as Bm101-LacZ, except for a p10 promoter region, produced a similar ${\beta}$-galactosidase activity to that due to Bm104-LacZ and 5.5-fold lower than that due to Bm101-LacZ. The virus yield, expression level of polyhedrin, and polyhedra productivity for each recombinant virus was almost similar. These results suggested that the difference in the expression level of ${\beta}$-galactosidase resulted from a difference in the p10 promoter regions, and that an expression vector using the p10 promoter of BmNPV-K1 could be usefully exploited in the mass production of foreign proteins with silkworm larvae by means of oral ingestion.

Characterization of an Improved Recombinant Baculovirus Producing Polyhedra that Contain Bacillus thuringiensis Cry1Ac Crystal Protein

  • A novel recombinant baculovirus, Bactrus, was constructed by the insertion of the Bacillus thuringiensis cry1Ac gene between two polyhedrin genes of Autographa californica nucleopolyhedrovirus (AcNPV) under the control of the polyhedrin gene promoter. Polyhedra produced by Bactrus in insect cells were incorporated with 130 kDa of polyhedrin-Cry1Ac-polyhedrin fusion protein, and 30 kDa of intact polyhedrin, resulting from a homologous recombination between two polyhedrin genes, was also expressed. The insecticidal activity of Bactrus against Spodoptera exigua larvae was similar to that of AcNPV, but it showed significantly higher toxicity towards Plutella xylostella larvae in comparison with that of AcNPV. The expression level of fusion protein and the insecticidal activity of recombinant polyhedra produced by the Bactrus against P. xylostella larvae were decreased after serial passages. In conclusion, the Bactrus had improved insecticidal activity and returned to wild-type AcNPV after several passages.

Construction of a Novel Recombinant Bombyx mori Nuclear Polyhedrosis Virus Producing the Fluorescent Polyhedra

  • We have constructed a novel recombinant Bombyx mori nuclear polyhedrosis virus (BmNPV) producing the green fluorescent polyhedra. For the production of the fluorescent polyhedra, partial polyhedrin gene containing KRKK as nuclear localization site from the BmNPV polyhedrin gene and the green fluorescent protein (gfp) gene were introduced under the control of p10 promoter of BmNPV. The recombinant BmNPV was stably produced fluorescent polyhedra in the infected Bm5 cells and the morphology of the fluorescent polyhedra was similar to that of wild-type BmNPV. The fluorescent polyhedra had 32 kDa native polyhedrin and 41 kDa fusion protein. From these data, we have further developed a novel BmNPV p10-based transfer vector producing recombinant polyhedra with foreign gene Product. The novel BmNPV P10-based transfer vector is composed of partial polyhedrin gene, factor Xa, and multiple cloning sites.

Construction of a Novel Baculovirus Autographa californica Nuclear Polyhedrosis Virus Producing the Fluorescent Polyhedra

  • A novel recombinant baculovirus Autographa californica nuclear polyhedrosis virus (ACNPV) producing the green fluorescent polyhedra was constructed and characterized. The recombinant virus was stably produced fluorescent polyhedra in the infected cells and the morphology of the polyhedra was nearly similar to that of wild-type AcNPV. For the production of the fluorescent polyhedral the green fluorescent protein (GFP) gene was introduced under the control of polyhedrin gene promoter of AcNPV by translational fusion in the front and back of intact polyhedrin gene. The recombinant baculovirus was named as CXEP, As expected, the 93 kDa fusion protein was expressed in the CXEP-infected cells. Interestingly, however, the cells infected with CXEP also showed a 33 kDa protein band as cells infected with wild-type AcNPV. The results of Southern blot analysis and plaque assay suggested that two types of baculoviruses expressing the GFP fusion protein or only native polyhedrin were formed through homologous recombination between two polyhedrin genes in the same orientation. Thus, this system can be applied for the production of recombinant polyhedra with foreign gene product of diverse interest.

곤충세포주에서 Autographa californica 핵다각체병 바이러스의 다각체 단백질과 Bacillus thuringiensis cryIA(c) 내독소 단백질의 융합 단백질 발현 (Expression of Fusion Protein with Autographa californica Nuclear Polyhedrosis Virus Polyhedrin and Bacillus thuringiensis cryIA(c) Crystal Protein in Insect Cells)

  • Autographa californica 핵다각체병 바이러스(AcNPV)의 다각체 단백질과 Bacillus thuringiensis(Bt) cryIA(c) 내독소 단백질의 융합단백질을 생산하는 새로운 재조합 바이러스를 제작하고, 곤충세포주(Spodoptera frugiperda 9)에서 발현된 융합단백질의 특성을 분석하였다. Bt kurstaki HD-73의 cryIA(c) 내독소 단백질 유전자의 N-발단 AcNPV의 완전한 다각체 단백질 유전자의 앞쪽에 융합함에 의하여 또는 다닥체 단백질 유전자내의 제한효소 HindII부위에 삽입함에 의하여 다각체 단백질 유전자의 프로모터 조절하에 도입하였다. 이렇게 작성된 재조합 바이러스를 각각 Btrusl 또는 BtrusII라고 명명하였다. BtrusI은 분명히 단일 전사체를 보임에도 92kDa의 융합 단백질과 다각체 단백질의 두 단백질을 생산하였다. 또한 Btrusl에 의해 만들어진 융합 단백질은 다각체를 형성하지 않았다. 한편, BtrusII에 의해 감염된 곤충세포주에서는 33kDa의 다각체 단백질은 보이지 않았고 단지 융합 단백질만 생산하였으나 다각체는 형성하지 않았다. 따라서 Btrusl에 의해 생산된 융합 단백질의 독성을 조사하기 위하여, Btrusl으로 감염된 곤충세포주를 2령 누에(Bombyx mori)에 접종한 결과 융합 단백질에 의한 독성이 관찰되었다. 결론적으로 다각체 단백질과 Bt cryIA(c) 내독소 단백질에 의한 융합 단백질이 독성을 가지고 있음을 확인하였다.
