Reduction of Cell Membrane Toxicity of Amphotericin B Using Micelle,Liposome and Polyethyene Glycol (Micelle, Liposome, Polythylene Glycol을 이용한 Amphotericin B의 세포막 독성저하)

  • Micelle, liposome and polyethylene glycol(PEG) were employed to reduce the cell mem- brane toxicity of Amphotericin B(Amp. B). Cholesterol-sulfate which can form a mixed micelle with Amp. B molecules was found very effective for the reduction of Amp. B toxicity. 0.01% of cholesterol-sulfate could reduce the toxicity of 5X 10$^{-6}$ M Amp. B by 90%. The required concent- ration of cholesterol-sulfate for the toxicity reduction was proportionally increased with increasing Amp. B concentration. PEG was also effective on the reduction of Amp. B toxicity. 2% PEG was required for the reduction of toxicity by 50%, regardless of Amp. B concentration. The liposome system showed an effective reduction of Amp. B toxicity on RBC, maintaining the antibiotic effect on Candida albicans as free drugs. This seems to be due to the fact that liposome bilayer plays a role of buffer system between ergosterol of fungi cell membrane and cholesterol of red blood cell membrane, which leads the redistribution of Amp. B between them, as the result, the reduction of drug toxicity on cell membrane.

Biosorption of Copper by the Immobilized biomass of Barine Brown Algae(Phaeophyta) Hizikia fusiformis (해양 갈조류인 톳의 고정화된 생물질에 의한 구리의 생흡착)

  • It was investigated the biosorption performances of copper by the immobilized biomass of nonliving marine brown alge h. fusiformis by each of the Ca-alginate method(Ca-ALG), Ba-alginate method(Ba-ALG), polyethylene glycol method(PEG), and carrageenan method (CARR). The copper removal performance increased but the copper uptake decreased as the biomass amount was increased. However, the copper uptake by the immobilized biomass increased with increasing initial copper concentration. The copper uptake by the immobilized biomass of the immobilization method decreased in the following sequence; Ca-ALG>Ba-ALG>PEG>CARR among the immoblization emthods. The copper uptake by the immobilized biomass followed the Langmuir isotherm better than the Freundlich isotherm.

Effects of Polyethylene Glycol Treatment for Improvement of Preservative Penetration and Prevention of Drying Check of Preservative Treated Round Post (방부처리 원주가공재의 방부제 침윤도 향상 및 건조 할렬 방지를 위한 폴리에틸렌 글리콜 처리 효과)

  • We investigated the effect of polyethylene glycol(PEG) treatment of preservative treated green japanese cedar(Cryptomeria japonica) round posts on their preservative penetration and check formation during the air drying. The round rods were dip-treated in 10% preservative solution of Basilit CFK for 1 day, followed by application(3, 5, and 10 times) with 50% PEG-400 solution, and then dried for 30 days under natural conditions. The rate of preservative penetration into sapwood was a low by 23.2% without PEG treatment, whereas that was a high by about 51.2~64.5% with PEG treatment. From these results, it was assumed that PEG played an affirmative role in the penetration of preservative components loaded onto the surface of round rods by dipping. During the air drying, the formation of drying checks decreased significantly with increasing application times of PEG. Even though some drying checks in PEG treated rods were developed, the number and size of checks was reduced remarkably by PEG treatment. After 2 months of outdoor exposure, PEG treatment failed to reduce checking in preservative-treated rods whereas finishing of polyurethane resin lacquer after PEG treatment was significantly effective in preventing check development.

  • The release of angiotensin and ${\alpha}-amylase$ from monolithic devices of different molecular weight of polyethylene glycol (PEC) grafted polyurethane copolymer was investigated. Water-soluble PEG grafted polymer provided a controlled release of angiotensin and ${\alpha}-amylase$. The release rate of angiotensin and ${\alpha}-amylase$ could be controlled by varying the molecular weight of PEC grafted. The release mechanism may be associated with the creation of pore or domain through the devices following the gel swelling and self-aggregation by PEC grafted polymer. Hydrophobic polyurethane grafted with PEG can provide a biomaterial for prolonged release of angiotensin and ${\alpha}-amylase$ from angiotensin and ${\alpha}-amylase$ blended system.

  • The superoxide dismutase (SOD) mimetic activity of copper complex of 3,5-disopropylsalicylic acid (Cu(II)-DIPS) was tested and compared to those of human recombinant SOD (hrSOD) and its conjugate form with polyethyleneglycol (PEG) using fer- ricytochrome c reduction assay. Stability constant of Cu(II)-DIPS was measured po- tentiometrically using SCOGS2 program. In the presence of 10 g/L albumin, Cu(II)-DIPS lost most of its SOD mimetic activity. HrSOD was modified with polyethylene glycol (PEG) of M.W. 5000. These conjugates have markedly prolonged plasma half-lives of enzymatic activity (15.5 hr) compared to native hrSOD (5 min). In summary, efficient SOD mimetics should be stable enough not to dissociate in blood by serum protein. HrSOD could have longer half-life by conjugation with inert PEG for sustained SOD effect.

Formation of Magnetic Structures for Trapping of Breast Cancer Cell

  • This work focuses on the fabrication of excellent magnetic structures for trapping breast cancer cells. Micromagnetic structures were patterned for trapping cancer cells by depositing 30 nm of permalloy on a silicon substrate. These structures were designed and fabricated using two fabrication techniques: electron beam lithography and laser direct writing. Two types of magnetic structures, rectangular wire and zig-zagged wire, were created on a silicon substrate. The length of each rectangular wire and each straight line of zig-zagged wire was 150 ㎛ with a range of widths from 1 to 15 ㎛ for rectangular and 1, 5, 10 and 15 ㎛ for zigzag, respectively. The magnetic structures showed good responses to the applied magnetic field despite adding layers of silicon nitride and polyethylene glycol. The results showed that Si + Si3N4 + PEG exhibited the best adhesion of cells to the surface, followed by Si + Py + Si3N4 + PEG. concentration of 5-6 with permalloy indicates that this layer affected silicon nitride in the presence of Polyethylene glycolPEG.

Anti-tumor Activity of Paclitaxel Prodrug Conjugated with Polyethylene Glycol

  • The purpose of this study was to develop a method for increasing the solubility of paclitaxel in water to reduce its toxicity and make the drug more feasible for chemotherapy. A series of highly water soluble paclitaxel polyethylene glycol (PEG) esters were synthesized and evaluated for their anti-tumor activity and toxicity. The solubility of 7-polyethylene glycol paclitaxel carbonate 5 was 840 mg/mL and the acute toxicity ($LD_{50}$) was 286 mg/kg. Because of its reduced toxicity, compound 5 showed a dramatic reduction of tumor volume without any loss of animals in long-term treatment (daily consecutive injections for 15 days).

Dehydration of Carrot Slice Using Polyethylene Glycol and Maltodextrin and Comparison with Other Drying Methods (폴리에틸렌 글리콜과 말토덱스트린 첨가에 의한 당근 절편의 탈수, 건조와 기존 건조 방법과의 비교)

  • Carrots were dried using 30, 50, and 80% polyethylene glycol (PEG) or maltodextrin as a dehydrating agent, and the dried carrots were compared with the freeze dried and hot-air dried in terms of rehydration ratio, color, carotene content, and sensory evaluation. The amount of moisture loss during drying of carrots using PEG or maltodextrin increased with increasing concentration of dehydrating agent. Rehydration ratio as well as carotene content of the PEG-treated carrots were greater than those of freeze-dried or hot-air dried carrots. Regarding color and sensory evaluation of the dried carrots, the PEG-treated and maltodextrin-treated carrots were better than freeze-dried or hot-air dried carrots. These results suggest that drying of carrots using PEG or maltodextrin is a very efficient method because of its good rehydration capacity and minimal destruction of nutrients and cell structure.
