Effect of Thesium Chinese Turczaninow Aqua-acupuncture Solution and Astragail Radix Aqua-acupuncture Solution on Promotion/ progression of Carcinogenesis (하고초와 황기 약침액이 발암 진행과정에 미치는 영향)

  • Cancer chemoprevention refers to the use of natural or synthetic substances to prevent initiational and promotional events that occur during the process of carcinogenesis. Thesium Chinese Turczaninow aqua-acupuncture solution (TCTAS) and Astragli Radix aqua-acupuncture solution (ARAS) were tested as the cancer chemopreventive agents using biochemical markers of carcinogenesis. The effects on the inhibition of phorbol 12- myristate 13-acetate(TPA)-induced free radical formation in HL-60 cells and the inhibition of polyamine metabolism were measured. There is significant inhibition of TAP-induced free radical formation in human leukemic cells with cells with ARAS. Proliferation of Acanthamoeba castellanii was inhibited by TCTAS and ARAS. TCTAS and ARAS positive in these assays may inhibit the carcinogenesis process and is considered promising cancer-preventing agents.

  • Pentaethylenehexaamine(penten) was reacted with a chloromethylated polystyrene resin on the purpose of the synthesis of polyamine chelating resin, $(P)_c$-penten. The stepwise dissociation constants of the synthesized polyamine chelating resin and the stability constants with metal ions were determined by Bjerrum's method. These stability constants were compared with those of the free penten. The adsorptivities and eluting tendencies of several metal ions on the chelating resin were studied.

Effects of $Ca^{2+}$ and Polyamine on Callose Contents in Carrot Suspension Cultured Cells (당근 현탁배양세포에서 $Ca^{2+}$과 Polyamine이 Callose 함량에 미치는 영향)

  • The effects of Ca2+ on polyamines on callose contents of carrot suspension cultured cells were studied. The regeneration process of the cell wall of carrot protoplast observed through the electron microscope. Treatment of the carrot suspension cultured cells with Ca2+ and polyamines resulted in considerable increase on callose contents at 0.1 mM of Ca2+ and polyamines, particulary spermidine. Poly-L-lysine and poly-L-ornithine increased about 30% and 100% of callose contents than that of the control respectively, whereas verapamil and flunarizine markedly decreased the callose contents. These effects of Ca2+ of free ion rather than as Ca2+-calmodulin complex. During the cultivation of the protoplast, the regeneration of the cell wall was somewhat observed on the 4th day, however, it was inhibited by verapamil. These results suggested that the promotive action of Ca2+ and polyamines were manifested in the callose contents and the regeneraton of the cell wall.

Effect of Polyamines on purified DNA Methyltransferase from Chlamydomonas reinhardtii (Chlamydomonas reinhardtii로부터 분리, 정제된 DNA Methyltransferase 활성에 대한 Polyamine의 영향)

  • DNA methyltransferase was purified 282.6-fold from Chlamydomonas reinhardtii 21gr (mt+) gametic cell to examine the effect of polyamine on the enzyme acctivity. Polyacrylamide gel electrophoresis(PAGE) revealed at least three bands(1 major band, 2 minor bands). Among these, the major band represents DNA methyltransferase. Polyacrylamide gel electrophoresis in the presence of 0.1% sodium dodecylsulfate(SDS-PAGE) revealed a major band with M.W. 60,000. DNA methyltransferase activity was inhibited more effectively by spermine than by spermidine, and the inhibition by putrescine was smaller than spermine and spermidine. DNA methyltransferase activity was inhibited by 40% and 53% at 5mM and 20mM spermine, respectively. In the case of spermidine, the inhibition was 35% at 10mM and 44% at 20mM. However, the inhibition by putrescine appeared only above 5mM and reached about 25% at 20mM.

Purification and Properties of $\beta-1$, 4-endoglucanase from Tobacco Suspension Cultured Cells (담배 현탁배양 세포로부터 $\beta-1$,4-endoglucanase의 정제 및 성질)

  • $\beta$-1, 4-endoglucanase (EC 3, 2, 1, 4) was isolated and purified from Nicotiana tabaccum L. Var. Virginia 115 suspensin cultured cells. The molecular weight as estimated by Sephadex G-100 was about 14, 000. The optimum pH for activity was 5.4. The Km value for carboxymethyl cellulose was 0.18 mg/unit and the enzyme was quite resistant to heating. Polyamine did not affect the activity of $\beta$-1, 4-endoglucanase in vitro but the activity increased drastically when polyamines were added in the suspension culture medium. It is suggested that increase in $\beta$-1, 4-endoglucenase activity due to polyamine might be related to growth of the plant.

  • The activity of Daucus carota L. root $\beta$-glucan synthetase II was observed to increase in the presence of polyamines such as putrescine, spermidine and spermine in vitro, whereas the activity of Daucus carota L. root $\beta$-glucan synthetase I was not affected by the polyamine. The activity $\beta$-glucan synthetase II from Daucus carota L. root protoplasts cultured on medium containing 10-6 M polyamines such as putrescine, spermidine and spermine was observed to be higher than that of the control. Daucus carota L. root protoplasts were observed to have the activities of arginine and ornithine decarboxylases and it was noted that they could produce polyamines, which might have an effect on $\beta$-glucan synthetase II activity.
